Nilaparvata lugens proclotting enzymes (NlPCEs) belong to the clip domain serine protease (clip-SP) family, which is a characteristic protease family in arthropods. NlPCE3 was previously reported to regulate egg production and development in female N. lugens, but its role in male N. lugens is unclear. In the present study, qPCR analysis showed that NlPCE3 was expressed in three different tissues (gut, testis and fat body). RNAi revealed that dsNlPCE3 injection made the male vas deferens thinner and reduced the oviposition level of the females that mated with dsNlPCE3-treated males, causing eggs not to hatch. Furthermore, immunofluorescence staining showed that NlPCE3 was widely expressed in the male internal genitalia. However, after dsNlPCE3 injection, expression of NlPCE3 was diffuse in the male internal genitalia, whose peripheral cells seemed degraded. Overall, these results indicate that NlPCE3 is important for reproduction in male N. lugens.

Clip domain serine proteases play vital roles in various innate immune functions and in embryonic development. Nilaparvata lugens proclotting enzymes (NlPCEs) belong to this protease family. NlPCE1 was reported to be involved in innate immunity, whereas the role of other NlPCEs is unclear. In the present study, N. lugens proclotting enzyme-3 (NlPCE3) was cloned and characterized. NlPCE3 contains a signal peptide, a clip domain, and a trypsin-like serine protease domain. NlPCE3 was expressed in all tissues examined (gut, fat body, and ovary), and at all developmental stages. Immunofluorescence staining showed that NlPCE3 was mainly expressed in the cytoplasm and cytomembrane of follicular cells. Double stranded NlPCE3 RNA interference clearly inhibited the expression of NlPCE3, resulting in abnormal egg formation and obstruction of ovulation. These results indicate that NlPCE3 plays an important role in egg production in N. lugens.

AbstractIn the present paper, four cDNAs encoding the alpha and gamma subunits of elongation factor 1 (EF-1) were cloned and sequenced from Nilaparvata lugens, named NlEF-1α, NlEF-1γ, and its yeast-like symbiont (YLS), named YsEF-1α and YsEF-1γ, respectively. Comparisons with sequences from other species indicated a greater conservation for EF-1α than for EF-1γ. NlEF-1α has two identical copies. The deduced amino acid sequence homology of NlEF-1α and NlEF-1γ is 96 and 64%, respectively, compared with Homalodisca vitripennis and Locusta migratoria. The deduced amino acid sequence homology of YsEF-1α and YsEF-1γ is 96 and 74%, respectively, compared with Metarhizium anisopliae and Ophiocordyceps sinensis. Reverse transcription-quantitative polymerase chain reaction (RT–qPCR) analysis revealed that the expression level of NlEF-1α and NlEF-1γ mRNA in hemolymph, ovary, fat body and salivary glands were higher than the midgut and leg tissue. YsEF-1α and YsEF-1γ was highly expressed in fat body. The expression level of NlEF-1α was higher than that of NlEF-1γ. Through RNA interference (RNAi) of the two genes, the mortality of nymph reached 92.2% at the 11th day after treatment and the ovarian development was severely hindered. The RT–qPCR analysis verified the correlation between mortality, sterility and the down-regulation of the target genes. The expression and synthesis of vitellogenin (Vg) protein in insects injected with NlEF-1α and NlEF-1γ double-stranded RNA (dsRNA) was significantly lower than control groups. Attempts to knockdown the YsEF-1 genes in the YLS was unsuccessful. However, the phenotype of N. lugens injected with YsEF-1α dsRNA was the same as that injected with NlEF-1α dsRNA, possibly due to the high similarity (up to 71.9%) in the nucleotide sequences between NlEF-1α and YsEF-1α. We demonstrated that partial silencing of NlEF-1α and NlEF-1γ genes caused lethal and sterility effect on N. lugens. NlEF-1γ shares low identity with that of other insects and therefore it could be a potential target for RNAi-based pest management.

1. There are two adult-specific proteins in the haemolymph of Rhodnius. They appear to be formed in the fat body. 2. The two proteins are absorbed by the oocytes and form the bulk of the soluble egg proteins. 3. The changes in the concentration of total protein in the haemolymph and of four protein fractions, as separated on cellulose acetate, do not reflect egg production. This may be a consequence of the hormonal control of reproduction.

The anatomy and histology of the male and female internal genitalia of Lytta nuttalli Say and the functions of the various organs during copulation and oviposition are described. In addition to the ovaries, lateral and common oviducts, and vagina, the female system includes a spermatophoral receptacle, accessory gland, and spermatheca. The most distinctive feature is the voluminous spermatophoral receptacle, which seems to be homologous with the bursa copulatrix of other Coleoptera, but serves to store and digest old spermatophores. The accessory gland is not a colleterial gland, but instead produces materials that probably are involved in the transfer of the spermatozoa into the spermatheca. The epithelia of the calyces and oviducts secrete the frothy, mucilaginous material that coats the eggs at oviposition. In the absence of a separate spermathecal gland, the epithelium of the spermatheca apparently has taken over its functions. The ovaries contain several hundred ovarioles of the telotrophic type. The chief structures of the male system are three pairs of accessory glands plus the testes, vasa deferentia, and ejaculatory duct. Each vas deferens consists of an enlarged portion that serves as an additional accessory gland and a narrow part, which is the seminal vesicle. Materials produced in the three pairs of accessory glands and the glandular portions of the vasa deferentia are used in spermatophore formation. The testes contain several hundred short sperm tubes similar to those of other insects. The arrangement, form, and functions of the internal reproductive organs of L. nuttalli are compared with those of other insects. Observations made on the reproductive systems of four species of Epicauta are also discussed in this context.

SUMMARYThe tickRhipicephalus (Boophilus) microplusis an important parasite of cattle in many areas of the tropics. Characterization of molecules involved in mechanisms such as vitellogenesis and embryo development may contribute to a better understanding of this parasite's physiology. The vitellin-degrading cysteine endopeptidase (VTDCE) is the most active enzyme involved in vitellin hydrolysis inR. micropluseggs. Here we show an association between VTDCE and vitellin in an additional site, apart from the active site. Our data also demonstrate cysteine endopeptidase activity in different tissues such as ovary, gut, fat body, salivary gland and female haemolymph, where it is controlled by a physiological inhibitor. InR. microplusfemale gut, VTDCE is localized in areas of protein synthesis and trafficking with the underlying haemolymph. VTDCE is also localized in the ovary basal region, in vesicle membranes of ovary pedicel cells and in oocyte cytosol. These results suggest that VTDCE plays a role in vitellin digestion during tick development.

SUMMARYThe life-cycles of two closely related cephalobaenid pentastomids, Raillietiella gehyrae and Raillietiella frenatus, which utilize geckos as definitive hosts and cockroaches as intermediate hosts, have been investigated in detail. Early development in the fat-body of cockroaches involves 2 moults to an infective, 3rd-stage larva which appears from 42–44 days post-infection. Complete development in geckos involves a further 5 moults in the case of males and 6 for females. Males mature precociously and copulation is a once-in-a-lifetime event which occurs around day 80 post-infection when both sexes are the same size but the uterus of the female is undeveloped. Sperm, stored in the spermathecae, is used to fertilize oocytes which slowly accumulate in the developing saccate uterus. Patency commences when the uterus carries approximately 4000–5500 eggs but only 25–36 % of these contain fully developed primary larvae. Since only mature eggs are deposited, we postulate that the vagina (?) of the female must be equipped with a selective filter that allows through large eggs but retains smaller, immature eggs. Thus the only limit on fecundity is the total number of sperms in the spermathecae and this is precisely the same factor that constrains egg production in the advanced order Porocephalida.

Growth and reproduction are the two most basic life processes of organisms and the distribution of energy in these processes is a core issue of the life history of organisms. Nilaparvata lugens (Stål), the brown planthopper (BPH), is a single-feeding rice pest. In the present study, this species was used as a model for testing the effects of nutritional conditions on various growth and reproduction indicators. First, the third-instar nymphs were fed with three different concentrations (100, 50, and 25%) of artificial diet until the second day of adulthood. The results showed that as the nutrient concentration decreased, the body development and oviposition of BPH were hindered. The total lipid content in the fat bodies was also significantly reduced. RT-PCR analysis showed compared to the 100% concentration group, the expression levels of vitellogenin (Vg) genes in the fifth-instar nymphs, adults, and in different tissues (ovary, fat body, and other tissues) were significantly decreased in the 50 and 25% treatment groups. Western blot analysis showed that Vg protein expression was highest in the 100% group, followed by the 50% group, with no expression in the 25% group. These results indicate that growth and reproduction in the BPH are regulated by, or correlated with, nutrient concentration. This study is of great significance as it reveals the adaptive strategies of the BPH to nutritional deficiencies and it also provides valuable information for the comprehensive control of this pest.

AbstractOvarian development and egg production in the larch sawfly, Pristiphora erichsonii (Hartig), were studied in the development of a method for measuring reproductive capacity for population studies. Most of the oöcytes are mature at the time of adult emergence but some mature in the following few days. Quantitative differences in foliage consumption during the larval stages have an appreciable effect on egg production but normal environmental temperature differences and exposure to flooding during the spring development period influence ovarian development only to a limited extent.Reproductive capacity can be determined by oöcyte counts of dissected adults of uniform age. The sample size necessary for estimates of various degrees of precision was calculated.Several size measurements have a significant correlation with the numbers of oöcytes produced. The regression of numbers of oöcytes on adult weight is of limited usefulness in the estimation of reproductive capacity. This is largely due to the individual variability in the extent to which fat body tissue is utilized in ovarian development particularly in the case of the larger adults.
