Forty-three metabolites including several methoxylated flavonoids, tremetones, and ent-clerodane diterpenes were accurately identified for the first time in the ethanolic extract of P. quadrangularis by means of hyphenated UHPLC-quadrupole Orbitrap mass spectrometry, and seven isolated compounds were tested regarding gastroprotective activity using the HCl/EtOH-induced lesion model in mice. A new tremetone (compound 6) is reported based on spectroscopic evidence. The isolated clerodanes and tremetones showed gastroprotective activity in a mouse model, evidenced by compound 7 (p-coumaroyloxytremetone), which showed the highest gastroprotective activity (76%), which was higher than the control drug lansoprazole (72%). Our findings revealed that several constituents of this plant have gastroprotective activity, and particularly, p-coumaroyloxytremetone could be considered as a lead molecule to explore new gastroprotective agents. This plant is a rich source of biologically active tremetones and terpenoids which can support the ethnobotanical use of the plant.

Phytochemical investigation of the ethanolic extract of leaves of Polyalthia longifolia var. pendula has led to the isolation of seven clerodane diterpenoids and five alkaloids. (-)-14, 15-bisnor-3, 11 E-kolavadien-13-one (1), (-)-16-oxocleroda-3,13(14) E-dien-15-oic acid (2), (-)-16α-hydroxycleroda-3,13 (14) Z-dien-15,16-olide (3), (+)-(4→2)-abeo-16(R/S)-2, 13 Z-kolavadien-15, 16-olide-3-al (4), (-)-3β, 16α-dihydroxycleroda-4(18), 13(14) Z-dien45,16-olide (5), (-)-3, 12 E-kolavadien-15-oic acid-16-al (6), (-)-labd-13 E-en-8-ol-15-oic acid (7), liriodenine (8), (-)-anonaine (9), (+)-isoboldine (10), (-)-asimilobine (11) and hordenine (12) have been isolated. This is the first report of 1, 6 and 10 from this plant species while 12 is reported for first time from this genus. Clerodane derivatives 1-7 were evaluated for their antimicrobial activity. Diterpene 3 was found to be most potent agent with MIC value of 6.25 μg/mL against Staphylococcus aureus and Sporothrix schenckii.

Bioassay guided fractionation of the ethanolic extract of Asphodelus microcarpus Salzm.et Vivi (Asphodelaceae) resulted in the isolation of one new metabolite, 1,6-dimethoxy-3-methyl-2-naphthoic acid (1) as well as nine known compounds: asphodelin (2), chrysophanol (3), 8-methoxychrysophanol (4), emodin (5), 2-acetyl-1,8-dimethoxy-3-methylnaphthalene (6), 10-(chrysophanol-7’-yl)-10-hydroxychrysophanol-9-anthrone (7), aloesaponol-III-8-methyl ether (8), ramosin (9) and aestivin (10). The compounds were identified by 1D and 2D NMR and HRESIMS. Compounds 3, 6 and 10 were isolated for the first time from this species. Compounds 3 and 4 showed moderate to weak antileishmanial activity with IC50 values of 14.3 and 35.1 μg/mL, respectively. Compound 4 exhibited moderate antifungal activity against Cryptococcus neoformans with an IC50 value of 15.0 μg/mL, while compounds 5, 7 and 10 showed good to potent activity against methicillin resistant Staphylococcus aureus (MRSA) with IC50 values of 6.6, 9.4 μg/mL and 1.4 μg/mL respectively. Compounds 5, 8 and 9 displayed good activity against S. aureus with IC50 values of 3.2, 7.3 and 8.5 μg/mL, respectively. Compounds 7 and 9 exhibited a potent cytotoxic activity against leukemia LH60 and K562 cell lines. Compound 10 showed potent antimalarial activities against both chloroquine-sensitive and chloroquine-resistant strains of Plasmodium falciparum with IC50 values in the range of 0.8-0.7 μg/mL without showing any cytotoxicity to mammalian cells.

Objective: To investigate the phytochemicals and GC-MS analysis of ethanol extracts of Silybum marianum. Methods: The air-dried leaves were powdered and subjected to selective sequential extraction using solvents of increasing polarity through percolation, ethanol to obtain an ethanolic extract. Then, each of the extracts was further subjected to gas chromatography-mass spectrometry. Results: Qualitative determination of the different biologically active compounds from crude extracts of Silybum marianum using gas chromatography-mass spectrometry revealed different types of high and low molecular weight chemical entities with varying amounts present in each of the extracts. These chemical compounds are considered biologically and pharmacologically important. Conclusions: The study established the chemical composition and anticancer activity of the plant. Objective: Chemical investigation of the extract of sponge-derived fungus, Aspergillus niger, was performed by liquid chromatography coupled with QExactive mass spectrometry for the first time. Method: A total number of 444 constituents were detected, 288 of which were identified positively or tentatively by the comprehensive utilization of accurate molecular weight and fragmentation information acquired from quadrupole orbitrap mass spectrometry. The identified compounds were divided into several types, namely, organic acid, alkaloid, saccharide, amino acid and cyclopeptide, terpenoid, polyketone, phenylpropanoid and other types of compounds. Systematic diagnostic ions and featured fragment patterns were summarized for each type, based on which 8 novel compounds belonging to the same type were characterized. Results: This work provided a rapid approach for the research of microconstituents in a complex analyte. Furthermore, the anti-tumor activity of the extract was evaluated on two different cell lines—Bel-7402 and Hela-S3 in vitro. The tumor-inhibitory effect of the Aspergillus niger extract was confirmed, and may be mainly derived from its pro-apoptotic action. Moreover, the extract exerted more significant cytotoxicity in Bel-7402 cells than Hela-S3 cells, indicative of its selectivity on specific tumor cells. Conclusion: The evidence suggested that the Aspergillus niger extract may potentially serve as a remedy for prevention and therapy of hepatic and breast carcinoma.

Tomatoes, members of the Solanaceae plant family, produce biologically active secondary metabolites, including glycoalkaloids, which may have both adverse and beneficial biological effects. Using the linear ion trap (LIT) mass spectrometry, multi-stage collision induced dissociation (CID) experiments (MSn) were performed to elucidate characteristic fragmentation pathways of the glycoalkaloid, tomatidine and of β1-hydroxytomatine. High resolution with high accuracy mass analysis using an Orbitrap fourier transform MS with higher-energy collisional induced dissociation (HCD) was used to produce mass spectra data across a wide spectral range for confirmation of proposed ion structures and formulae.

Relevance. A number of scientific studies confirm that consumption of fruits and vegetables can reduce the risk of certain chronic diseases, such as cancer and cardiovascular diseases, for example, consumption of fresh tomatoes and tomato products is inversely proportional to the development of certain types of cancer. Tomato Solanum lycopersicum L. contains a large number of polyphenolic complexes, which are biologically active compounds. In this article, the authors have attempted for the first time to present the complete metabolomic composition of Solanum lycopersicum extracts.Materials and methods: As an object of research, authors used the extracts of Solanum lycopersicum L., from the collection of the Federal Research Center N.I. Vavilov All-Russian Institute of Plant Genetic Resources, grown and collected at the Far Eastern Experiment Station Branch of the Federal State Budgetary Scientific Institution in September 2020 (varieties: k-5351 Ont77 13, Canada; k-3149 Rehovoth, Israel; 2698 Ukraine). High performance liquid chromatography (HPLC) in combination with a BRUKER DALTONIKS ion trap (tandem mass spectrometry) was used to identify target analytes in extracts obtained by the maceration method.Discussion: The results of initial studies revealed the presence of 36 biologically active compounds, of which 22 were identified for the first time in Solanum lycopersicum L. These are Apigenin, Luteolin, Kaempferol, Taxifolin, Myricetin, Coutaric acid, Caffeoylmalic acid, Caftaric acid, Dicaffeoylquinic acid, coumarins Fraxetin, and Fraxetin-7-O-beta-glucuronide, Pelargonidin, Salvianolic acid D, Rosmanol, Colnelenic acid, Ethyl rosemary, lignan Medioresinol-O-hexoside, Squalene, etc. The findings will help to intensify future research into the development and production of various functional food products containing targeted extracts of Solanum lycopersicum L.

Haloarchaea are extreme halophilic microorganisms belonging to the domain Archaea, phylum Euryarchaeota, and are producers of interesting antioxidant carotenoid compounds. In this study, four new strains of Haloarcula sp., isolated from saline lakes of the Atacama Desert, are reported and studied by high-resolution mass spectrometry (UHPLC-Q-Orbitrap-MS/MS) for the first time. In addition, determination of the carotenoid pigment profile from the new strains of Haloarcula sp., plus two strains of Halorubrum tebenquichense, and their antioxidant activity by means of several methods is reported. The effect of biomass on cellular viability in skin cell lines was also evaluated by MTT assay. The cholinesterase inhibition capacity of six haloarchaea (Haloarcula sp. ALT-23; Haloarcula sp. TeSe-41; Haloarcula sp. TeSe-51; Haloarcula sp. Te Se-89 and Halorubrum tebenquichense strains TeSe-85 and Te Se-86) is also reported for the first time. AChE inhibition IC50 was 2.96 ± 0.08 mg/mL and BuChE inhibition IC50 was 2.39 ± 0.09 mg/mL for the most active strain, Halorubrum tebenquichense Te Se-85, respectively, which is more active in BuCHe than that of the standard galantamine. Docking calculation showed that carotenoids can exert their inhibitory activity fitting into the enzyme pocket by their halves, in the presence of cholinesterase dimers.

Through a combination of thermogravimetry, mass spectrometry and differential thermal analysis, we demonstrate for the first time that all four zeolites show experimental differences in their host-guest interactions with 18C6. In addition, we have estimated the kinetics of 18C6 decomposition, which is a technique that has not been applied to zeolites previously. Using these findings as a toolkit, a more rational use of OSDAs can be utilised to prepare designer zeolites. Furthermore, the new methodologies presented herein can be applied to current zeolites, such as MFI-type zeolites used in the petrochemical industry.
