Kersen leaves (Muntingia calabura L) contain metabolites such as alkaloids, anthrax, polyphenols, tannins, saponins and are rich in flavonoid components such as flavones, flavonones, flavan and biflavan which have antidiabetic activity. According to a study conducted by Apriyanti (2016), the ethanol extract of kersen (Muntingia calabura L) at a dose of 250 mg / kg BB significantly reduced blood glucose levels in male wistar rats. In this study aimed at determining IC50 ethanol extract of Muntingia calabura L as an inhibitor of α-glucosidase enzymes using ELISA reader. The method was divided into 3 category is 1 (sample extract), 2 (blank) and 3 (akarbose). Each group added 25 µL α-glucosidase solution (0.25 units / mL then measured using ELISA reader 405 nm. The results showed that the ethanol extract of kersen (Muntingia calabura L) had activity as an inhibitor of α-glucosidase enzyme with IC50 34,197 µg / mL and can be categorized as active.

Excessive energi consumption play a role in increasing blood glucose levels (hyperglycemia), due to the inability of the hormone insulin to compensate the high blood glucose levels. Blood glucose receptor found in the hypothalamus of VMH (Ventro Medial Hypothalamus) and LH (the Lateral Hypothalamus) is able to detect and change the settings for the feeding activity. The aim of this study was to obtain the relationship between high sucrose feeding on male Wistar rats of behavior and activity. This method uses Wistar rats (Rattus norvegicus) as an animal model. Sixteen rats were grouped into 4 treatment groups. The first group was given additional feed 20% sucrose, the second group was given additional feed 40% sucrose, a third group was given additional feed 60% sucrose, and the last as a control group. This feed given continuously for 70 days.Observation of activities conducted using Opto-varimex® auto-track system ver.4.31. The results showed an corellation between activity based doses of sucrose.

Abstract Objectives Diethylnitrosamine (DEN) is found in workplaces, processed meats, tobacco smoke, whiskey, etc. It is capable of forming DNA-adducts. Fluted pumpkin (Telfairia occidentalis [To]) is a medicinal plant, and its herbal preparations have been employed variously in ethnomedicine. Furthermore, it has been reported to possess anti-oxidant, anti-cancer, anti-inflammatory properties. We investigated the possible mitigating effect of the leaf paste of To on DEN-induced deleterious effects in male Wistar rats. Methods Forty-five rats weighing between 100 and 150 g were equally divided into nine groups and treated thus: Group 1 (negative control), Group 2 (0.05 mg/kg carboxymethyl cellulose [CMC] daily), Group 3 (positive control, 25 mg/kg bw DEN administered intraperitoneally thrice per week), Group 4 (25 mg/kg bw quercetin [QUE] daily alone), Groups 5 and 6 (100 and 200 mg/kg bw To daily, respectively), Group 7 (25 mg/kg bw DEN and QUE), Groups 8 and 9 (25 mg/kg bw DEN with 100 and 200 mg/kg bw To, respectively). Blood glucose levels, liver damage biomarkers (aspartate aminotransferase [AST], alanine aminotransferase [ALT] and gamma-glutamyltransferase [γ-GT]), frequency of micronucleated polychromatic erythrocyte (mPCEs), and liver histology were assessed. Results DEN significantly (p<0.05) increased blood glucose levels, activities of ALT, AST and γ-GT, and frequency of mPCEs. Histologically, DEN caused a severe architectural anarchy. However, the intervention groups demonstrated the remarkable protective properties of To by ameliorating the adverse effects caused by DEN. Conclusions Taken together, the leaf paste of To is capable of mitigating DEN-induced hepatotoxicity and clastogenicity in male Wistar rats.

ANTIHIPERGLIKEMIK EKSTRAK TUMBUHAN SURUHAN (Peperomia pellucida [L.] Kunth) TERHADAP TIKUS WISTAR (Rattus norvegicus L.) YANG DIINDUKSI SUKROSAABSTRAK Telah dilakukan penelitian untuk menentukan dosis optimum dari ekstrak tumbuhan suruhan (Peperomia pellucida [L.] Kunth) sebagai antihiperglikemik tikus wistar yang diinduksi sukrosa. Penelitian ini menggunakan metode uji toleransi glukosa. Digunakan tikus putih jantan wistar berjumlah 20 ekor tikus yang dibagi dalam 5 kelompok perlakuan yaitu kelompok perlakuan kontrol negatif (K-) diberi CMC 0,5%, kelompok perlakuan kontrol positif (K+) diberi Glibenklamid dosis 0,45 mg/kgBB, kelompok perlakuan ekstrak tumbuhan suruhan  dosis 20; 40; 80 mg/kgBB. Data diperoleh dari pemeriksaan kadar gula darah puasa (basal), 30 menit setelah diindukasi sukrosa dan pada menit ke 30, 60, dan 120 setelah diinduksi sediaan per oral. Data diolah dengan uji ANOVA dan Ducan untuk membandingkan antara kelompok kontrol negatif, ekstrak dan kontrol positif. Data hasil pengujian kadar glukosa darah dengan dosis 20; 40; 80 mg/kgBB memberikan penurunan kadar glukosa darah. Pemberian ekstrak tumbuhan suruhan dengan dosis 40 mg/kgBB pada menit ke-120 memberikan hasil  yang signifikan dan bebeda nyata dibandingkan kelompok kontrol negatif (CMC 0,5%) dan tidak berbeda nyata dengan glibenklamid dosis 0,45 mg/kgBB. Kesimpulannya pemberian ekstrak tumbuhan suruhan dosis 40 mg/kgBB pada menit ke 120  memberikan penurunan kadar glukosa darah yang efektif dan berbeda nyata (signifikan) dibandingkan pemberian ekstrak tumbuhan suruhan dosis 20 dan 80 mg/kgBB. Used 20 male Wistar rats were divided into 5 groups, they are the negative control treatment group (K-) were given 0.5% CMC, the positive control groups (K+) given 0,45 mg/kgBB  Glibenklamid, and Suruhan extract treatment group of 20; 40; 80 mg/kgBB dose. Data obtained from the examination offasting blood sugar (basal) content, 30 minutes after induced bysucroseandat minute of 30, 60, and120 after-induced oraldosage.The data were processed by ANAVA and LSD testtocompare betweenthe negativecontrol group, the extractandpositive control. Dataresults of testing blood sugar levels with doses of 20, 40, 80 mg/kgBB gavea decreasein blood glucose levels were significant and significantly different than the negative control group (CMC 0.5%) and did not give a clear difference with 0.45 mg/kgBB dose of Glibenclamide. Agents of plant extract at a dose of 40 mg/kgBB body weight gives better results to the decline in blood glucose levels compared with doses of 20 and 80 mg/kgBB. In conclusion, Suruhan plant extract has an effect on blood glucose levels decreased against white male Wistar rats were induced sucrose. Abstract: Herbal and natural products of folk medicine have been used for centuries in every culture throughout the world. One of folks medicine that has been used in Indonesia is the noni fruit. According to research conducted by I Ketut Adnyana, et al, there is a decrease in blood glucose levels from noni fruit extract with a dose of 500 mg/kg BW and 1000 mg/kg BW. This research aims to know the minimal dose of noni fruit (Morinda citrifolia L.) extract against a decrease in blood glucose levels in Wistar rats (Rattus norvegicus) induced alloxan. This study used 15 Wistar rats with diabetic induced by 130 mg/kg BW alloxan intraperitoneal. The rats were randomly divided into 5 groups; each group were given Aquades, Insulin Novomix 0,4 IU/100 gr BW, Noni fruit extract 125 mg/kg BW, 250 mg/kg BW, and 500 mg/kg BW. The result data obtained by the results of blood glucose levels evaluation on each rats on the first, second, and second day before the treatment after 6 hours, 12 hours, 18 hours, and 24 hours. Menurut penelitian yang dilakukan oleh I Ketut Adnyana, dkk terdapat penurunan kadar glukosa darah dari ekstrak buah mengkudu dengan dosis 500 mg/kg BB dan 1000 mg/kg BB. Penelitian ini bertujuan untuk mengetahui dosis minimal dari ekstrak buah mengkudu (Morinda citrifolia L.) terhadap penurunan kadar glukosa darah pada tikus Wistar (Rattus norvegicus) yang diinduksi aloksan. Tikus Wistar sejumlah 15 ekor diinduksi dengan aloksan intraperitoneal 130 mg/kg BB dan dibagi secara acak dalam 5 kelompok penelitian yaitu kelompok perlakuan dengan Aquades, Insulin Novomix 0,4 IU/100 gr BB, ekstrak buah mengkudu 125 mg/kg BB, 250 mg/kg BB dan 500 mg/kg BB. Data yang diperoleh dari hasil pemeriksaan kadar glukosa darah pada semua kelompok tikus Wistar pada hari pertama, hari kedua, dan hari kedua jam ke-0, 6, 12,18 dan 24. Hasil penelitian menunjukkan bahwa ekstrak buah mengkudu dengan dosis 125 mg/kg BB sudah memiliki efek untuk menurunkan kadar glukosa darah pada tikus Wistar yang diinduksi aloksan. Insulin secretion is impaired in type 2 diabetes (T2D). The insulin and glucose responses to central autonomic activation induced by excitation of brain medullary TRH receptors were studied in T2D Goto-Kakizaki (GK) rats. Blood glucose levels in normally fed, pentobarbital-anesthetized GK and nondiabetic Wistar rats were 193 and 119 mg/100 ml in males and 214 and 131 mg/100 ml in females. Intracisternal injection (ic) of the stable TRH analog RX 77368 (10 ng) induced significantly higher insulin response in both genders of overnight-fasted GK rats compared with Wistar rats and slightly increased blood glucose in female Wistar rats but significantly decreased it from 193 to 145 mg/100 ml in female GK rats. RX 77368 (50 ng) ic induced markedly greater glucose and relatively weaker insulin responses in male GK rats than Wistar rats. Bilateral vagotomy blocked ic RX 77368-induced insulin secretion, whereas adrenalectomy abolished its hyperglycemic effect. In adrenalectomized male GK but not Wistar rats, ic RX 77368 (50 ng) dramatically increased serum insulin levels by 6.5-fold and decreased blood glucose levels from 154 to 98 mg/100 ml; these changes were prevented by vagotomy. GK rats had higher basal pancreatic insulin II mRNA levels but a lower response to ic RX 77368 (50 ng) compared with Wistar rats. These results indicate that central-vagal activation-induced insulin secretion is susceptible in T2D GK rats. However, the dominant sympathetic-adrenal response to medullary TRH plays a suppressing role on vagal-mediated insulin secretion. This unbalanced vago-sympathetic activation by medullary TRH may contribute to the impaired insulin secretion in T2D.

Diabetes mellitus is a metabolic disease that always increasing in prevalence. Excessive glucose intake and oxidative stress that can lead to damage to the pancreatic cell or insulin resistance, are such factors that influence the occurrence of diabetes mellitus. Graptophyllum pictum (L.) Griff leaf contains active metabolites such as flavonoid which able to decrease free radical levels. This study aimed to prove the effectiveness of Graptophyllum pictum (L.) Griff leaves extract on blood glucose levels in alloxan-induced Wistar rats. In this study, twenty-five male Wistar rats were divided into five groups. On day 0, the negative control and each treatment group injected with 125 mg/kg dose of alloxan and would be checked on 4th day. On the 4th day after the rats had diabetes, normal group (K-1) was given only standard feed, Negative control (K-2) was given only alloxan intraperitoneally on day 0, first treatment group (K-3) was given 50 mg/kg dose of Graptophyllum pictum (L.) Griff, second treatment group (K-4) was given 100 mg/kg dose of the leaf extract, and the third treatment group (K-5) was given the leaf 200 mg/kg dose of the leaf extract. On the 10th day, fasting blood glucose was taken from the rats. Post-test only group design was used to be the experimental design. There were significant differences in the treatment groups (K-3, K-4, and K-5) compared to the negative control group (K-2) after treatment. The mean of blood glucose levels decreased with the increased doses of Graptophyllum pictum (L.) Griff leaf extract treatment. The treatment groups (K-3, K-4, and K-5) were not significant compared to the normal group (K-1).

The present study aims to study the hypoglycemic effect of methanol extract of Andrographisechioides (MEAE) in streptozotocin (STZ)-induced diabetic Wistar rats. Hyperglycemia was induced in rats by single intraperitoneal injection of STZ (55 mg/kg bodyweight). Three days after STZ induction, the hyperglycemic rats were treated with MEAE orally at the doses of 200, 500, and 800 mg/kg body weight daily for 21 days. Glibenclamide (1 mg/kg, orally) was used as reference drug. The fasting blood glucose levels were measured on each 7th day during the 21 days of treatment.
