Effect of Armillaria mellea on Blood Lipid Levels and Antioxidant Enzymes Activity in High Fat Mice

The effect of Armillaria mellea on blood lipid levels and oxidative stress in mice fed on high-fat diet was investigated. Animals were allocated to the Armillaria mellea polysaccharides-treatment groups (I, II) and Armillaria mellea oligosaccharides-treatment groups (I, II). All mice were fed with high-fat diet for 40 days but control mice with basic diet. TC, TG, HDL-c, LDL-c were measured by enzymatic and colorimetric methods. The same, MDA,SOD, GSH-PX were measured. Results showed that administration of Armillaria mellea polysaccharides and oligosaccharides significantly increased antioxidant enzymes GSH-Px activities and decreased TC, TG, LDL-c, MDA level in mice (P < 0.05) compared with model group. In conclusion Armillaria mellea polysaccharides and oligosaccharides were able to protect mices antioxidative and improve abnormal blood lipid levels.

Abstract Background This study aims to compare whether two different routes of Renal denervation (RDN) from the intima and adventitia of the renal artery can reduce renal fibrosis in a pig model of hypertension induced by a high-fat diet and to explore possible molecular mechanisms. Methods Twenty-four Bama miniature pigs were randomly divided into a control group (normal diet, n=6) or a hypertension model group (high-fat diet, n=18). The model group was randomly divided into the intima-RDN group (n=6), the adventitia-RDN group (n=6), or the renal arteriography only group (sham group, n=6). All animals were fed separately. The model group was fed a high-fat diet after the operation, and the control group was fed conventionally for 6 months. After 6 months, renal artery angiography was performed again to observe the condition of the renal arteries, after which all animals were euthanized. The blood pressure (BP) and blood biochemical results of each group were evaluated 6 months after the operation; kidney tissue morphology and collagen fiber content were examined by hematoxylin-eosin (HE) staining and Masson staining; Superoxide dismutase (SOD) activity and the malondialdehyde (MDA) content of kidney tissue were assessed by a biochemical enzyme method; the protein expression level of transforming growth factor-β 1 (TGF-β1), α smooth muscle actin (αSMA) and Smad3 were assessed by Western blot; and electron microscopy was used to examine changes in kidney microstructure. Results After 6 months of a high-fat diet, the blood lipid levels of the model group were significantly higher compared to baseline and to that of the control group during the same period (all showed P<0.05); the blood lipid levels of the control group did not change significantly from baseline (P>0.05). The degree of glomerular damage caused by hyperlipidemia in the intima-RDN group and the adventitia-RDN group was significantly lower than that of the sham and control groups, and the renal fibrosis area percentage was also significantly lower (P<0.05). Electron microscopy showed that both the intima-RDN group and the adventitia-RDN group had a more even distribution of chromosomes and less mitochondrial swelling compared with the sham group. Conclusion RDN from the adventitia of the renal artery and RDN from the intima of the renal artery have the similar advantages of delaying high fat-induced renal fibrosis. The anti-fibrotic effect of RDN may be related to inhibition of the TGF-β1/smad3 pathway.

This research was carried out to investigate the effects of flavonoids ingredient from Polygonum capitatum (FPC) on blood lipid levels, vascular inflammation, and oxidative stress in high-fat diet (HFD) rats, as well as their mechanism of action. Rats fed with HFD for 6 weeks obviously displayed hyperlipidemia ( P < 0.01). Treatment with FPC at 90 and 180 mg/kg body weight significantly increased serum apolipoprotein A (ApoA) and high-density lipoprotein-cholesterol (HDL-C) levels and decreased serum apolipoprotein B (ApoB), triglyceride (TG), total cholesterol (TC), and low-density lipoprotein-cholesterol (LDL-C) levels of hyperlipidemia rats. FPC also improved the serum superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activities and decreased serum malondialdehyde (MDA), tumor necrosis factor-alpha (TNF-α), and interleukin-6 (IL-6) levels. Meanwhile, the result of reverse transcription polymerase chain reaction (RT-PCR) manifested that FPC upregulated the messenger RNA (mRNA) expression of low-density lipoprotein receptor (LDLR) and peroxisome proliferator–activated receptor α (PPARα) and downregulated the mRNA expression of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), acetyl CoA carboxylase (ACC), and sterol regulatory element-binding protein 1c (SREBP-1C) in the hepatic. These results demonstrated that FPC exerted anti-atherosclerosis effect in hyperlipidemia rats by regulating blood lipid metabolism, improving antioxidant ability, and modulating a proinflammatory profile and the expression levels of genes referred to lipogenesis and lipid oxidation, which might be attributed to flavonoid ingredients such as luteolin-7-O-glucoside, rutin, and quercitrin.

Excessive food consumption and insufficient exercise lead to the prevalence of metabolic syndrome in modern life, which consequently increases the risk of many chronic diseases. Magnesium lithospermate B (MLB) from Danshen has been demonstrated to improve metabolic changes in high-fat diet–fed rats with metabolic syndrome. In this study, Mg2+ in MLB was successfully replaced with Zn2+ to form zinc lithospermate B (ZLB) complex. MLB (10 mg/kg /day) and ZLB of various concentrations (1, 2.5, 5, and 10 mg/kg/day) were prepared and examined for their therapeutic effects on metabolic syndrome induced in rats fed with a high-fat diet. The results showed that both MLB and ZLB were able to recover or alleviate the abnormal physiological states of high-fat diet–fed rats including weight gain, epididymal fat accumulation, fatty liver, retarded blood lipid and glucose metabolism putatively caused by insulin resistance, and elevated levels of proinflammatory cytokine, leptin, and oxidative stress. In an overall view of the animal study, the effectiveness of ZLB supplementation seemed to be better than that of MLB supplementation for the recovery of high-fat-fed rats from metabolic syndrome.

In this study, we investigated the effects of a Chinese herbal medicine formula Xiao-Gao-Jiang-Zhuo (XGJZ) in obese rats induced by a high-fat diet. Ten male rats in the normal group were fed with a standard diet. Another 50 male obese rats were induced by a 12-week high-fat diet feeding, and were randomly divided into five groups (n = 10 per group): the model group, the high-dose XGJZ group, the middle-dose XGJZ group, the low-dose XGJZ group, and the sibutramine group. After 14 weeks of treatment, body weight, abdominal fat, blood lipid and serum insulin level were measured, and the protein and gene expression of PTP1B in liver tissue was tested. Our data showed that the body weight of the high-dose and middle-dose groups and the sibutramine group had significant differences in comparison with the model group (p < 0.05), and all three dose groups had significantly reduced abdominal fat (p < 0.05). The triglyceride level of the three dose groups and the sibutramine group, and the total cholesterol level of the middle-dose group were all significantly reduced (p < 0.05). The serum insulin of the high-dose and middle-dose groups also decreased significantly (p < 0.05). The expression of hepatic PTP1B mRNA of the three dose groups decreased significantly in comparison with the model group (p < 0.05 or 0.01). The expression of hepatic PTP1B protein of the high-dose and middle-dose groups decreased significantly (p < 0.05). Our data suggested that XGJZ can modulate the body weight, abdominal fat and blood lipid in the obese rats, and this modulation might improve insulin resistance by inhibiting the PTP1B signal pathway.

In the present study, we aimed to examine the effects of soya oligosaccharides (SOS) and soya oligopeptides (SOP) on blood lipid levels, release of vasoactive substances, antioxidant activity and faecal bile acid (FBA) excretion in rats fed a high-fat diet (HFD). Male Sprague–Dawley rats were evenly divided into five groups according to diets as follows: regular diet (control), HFD, HFD enriched with 2 % of SOS (SOS), HFD enriched with 3 % of SOP (SOP) and HFD enriched with 2 % SOS and 3 % SOP (SOSP). The results showed that SOS and SOP significantly reduced plasma total cholesterol, LDL-cholesterol and TAG, whereas HDL-cholesterol concentration was significantly increased. Furthermore, SOS and SOP reduced plasma apoB, apoE and the apoB:apoAI ratio, whereas apoAI was significantly increased. Moreover, SOS and SOP also reduced plasma thromboxane A2(TXA2) and the TXA2:prostacyclin (PGI2) ratio, whereas plasma PGI2and nitric oxide were significantly increased. In addition, SOS and SOP significantly reduced serum and liver malondialdehyde concentrations and increased FBA excretion. However, we did not observe obvious influences of SOS and SOP on superoxide dismutase activities in the liver of HFD-fed rats. The combination of 2 % SOS and 3 % SOP showed a more marked effect than SOS or SOP alone in improving the lipid profile, release of vasoactive substances and increasing FBA excretion (P < 0·05). In summary, SOS and SOP might help prevent atherosclerosis through improving abnormal blood lipid levels, regulating vasoactive substances and protecting against oxidative stress.

TheSiraitia grosvenoriipolysaccharide (SGP) from theSiraitia grosvenorii(Swingle) was isolated and purified. The therapeutic effects of SGP on diabetic rabbits induced by feeding high fat/high sucrose chow were studied. After administration of SGP for 4 weeks, the fasting blood glucose (FBG), plasma insulin levels (INS), plasma total cholesterol (TC), triglyceride (TG), and HDL-C were assayed. The results showed that administration of SGP can significantly decrease plasma total cholesterol, triglyceride, and glucose levels; and increase HDL-C levels after 4 weeks of treatment. The antihyperglycaemic effect of SGP at dose of 100 mg⋅kg−1bw was the most significant in three dosage groups. Furthermore, SGP could restore the blood lipid levels of diabetic rabbits (P<.05). These data indicate that SGP not only ameliorates the lipid disorder, but also lowers plasma glucose levels. So SGP have obvious glucose-lowering effect on hyperglycaemic rabbits induced by feeding high fat/high sucrose chow, its mechanism may be related to amelioration of lipid metabolism and restoring the blood lipid levels of hyperglycaemic rabbits.
