Kombucha adalah hasil fermentasi teh dan gula oleh SCOBY (symbiotic culture of bacteria and yeast). Kombucha dapat dibuat dari daun yang tinggi fenol salah satunya adalah daun salam. Penelitian ini bertujuan menentukan aktivitas antioksidan, total fenol, dan kadar vitamin C dari kombucha daun salam. Pada penelitian ini menggunakan kombucha daun salam dibuat dengan konsentrasi 12,5; 25; 37,5; dan 50 g/l. Kombucha daun salam yang diperoleh di uji karakteristik kombucha daun salam meliputi uji fitokimia, pH serta pengujian aktivitas antioksidan, total fenol dan kadar vitamin C secara in vitro. Aktivitas antioksidan kombucha daun salam pada penelitian ini diperoleh 38,58%-75,05% dengan nilai IC50 sebesar 27 g/l, total fenol kombucha daun salam berkisar 4,74-51,972 ppm, dan kadar vitamin C antara 16,426-30,506 mg/100g. Berdasarkan uji total fenol, kadar vitamin C dan uji antioksidan maka kombucha daun salam memiliki aktivitas antioksidan.

Gastric cancer is a multifactorial disease with complex interplay of environmental and genetic factors. Helicobacter pylori (H. pylori) infestation has been identified as the most important etiological agent in the pathogenesis of gastric cancer. Also, the role of dietary factors that is low consumption of fruits and vegetables have been found to be associated with gastric cancer. Among the dietary factors, antioxidants especially vitamin C has been found to confer the strongest protection against gastric cancer. Its anti-proliferative and pro-apoptotic action has been suggested in vitro. Because of its antioxidant activity, it protects cells against oxidative DNA damage caused by toxic effects of reactive oxygen species. It also inhibits production of carcinogenic N-nitroso compound in the stomach. The person with H. pylori infection has low levels of vitamin C in their gastric juice and levels of vitamin C normalizes on eradication of H. pylori. Vitamin C levels are high in gastric mucosa and gastric juice, sometimes more than that of in plasma. But gastric pathological conditions cause lowered secretion of vitamin C into gastric juice. Effect of H. pylori on vitamin C in gastric juice is reversible and on eradication of H. pylori, it returns to normal level. Hence, eradication of H. pylori and chemoprevention with antioxidant supplementation will be an effective preventive strategy to reduce the incidence of gastric cancer and related mortality. Resistant maltodextrin (RMD) is a water-soluble and fermentable functional fiber. RMD is a satiating prebiotic, reducer of glucose and triglycerides in the blood, and promoter of good gut health, and its addition to food is increasingly frequent. Therefore, it is necessary to study its potential effects on intrinsic bioactive compounds of food and their bioaccessibility. The aim of this study was to evaluate the effect of adding RMD on the bioactive compounds of pasteurized orange juice with and without pulp, and the bioaccessibility of such compounds. RMD was added at different concentrations: 0 (control sample), 2.5%, 5%, and 7.5%. Ascorbic acid (AA) and vitamin C were analyzed using HPLC, whereas total phenols, total carotenoids (TC), and antioxidant capacity were measured using spectrophotometry. After that, sample in vitro digestibility was assessed using the standardized static in vitro digestion method. The control orange juice with pulp presented significantly higher values of bioactive compounds and antioxidant capacity than the control orange juice without pulp (p < 0.05). RMD addition before the juice pasteurization process significantly protected all bioactive compounds, namely total phenols, TC, AA, and vitamin C, as well as the antioxidant capacity (AC) (p < 0.05). Moreover, this bioactive compound protective effect was higher when higher RMD concentrations were added. However, RMD addition improved phenols and vitamin C bioaccessibility but decreased TC and AA bioaccessibility. Therefore, the AC value of samples after gastrointestinal digestion was slightly decreased by RMD addition. Moreover, orange pulp presence decreased total phenols and TC bioaccessibility but increased AA and vitamin C bioaccessibility.

AbstractThis study was performed to explore the efficacy of combining more than one postharvest treatment in maintaining some quality attributes and reducing fungal pathogenicity in cold-stored guava fruits. The investigated postharvest treatments included the control, CaCl2(4%), lemongrass oil (2 dm3kg−1), gamma (γ) irradiation (0.2, 0.4 and 0.6 kGy), 0.4 kGy γ irradiation + CaCl2(4%), and 0.4 kGy γ irradiation + lemongrass oil (2 dm3kg−1). The studied physiochemical attributes included weight loss, decay percentage, fruit firmness, total soluble solids (TSS), titratable acidity (TA), and vitamin C content. Different fungal species were also isolated from decayed fruits and were identified asAlternaria alternata,Alternaria solani,Aspergillus niger,Botrytis cinerea,Fusarium solaniandRhizopus stolonifer. The severity of infection for the different fungi was determined, and anin vitroantifungal assay was conducted for lemongrass oil. All the investigated treatments generally reduced decay and water loss percentages, and controlled TSS, TA and vitamin C decrements that occurred during cold storage. On the other hand, higher irradiation doses generally increased fruit softness, and the 0.4 kGy γ dose did not contribute to the overall fruit quality when coupled with CaCl2and lemongrass oil, compared to CaCl2and lemongrass oil treatments alone.

To determine whether Vitamin C might relate to the increased platelet sensitivity in the diabetic, we have measured levels of platelet Vitamin C and studied the effects of Vitamin C on platelet aggregation. Ascorbic acid levels in washed platelets from diabetics were significantly lower than from normals (4s.2±3 μg/1010 platelets vs. 2s.s±2 μg/1010 platelets, p<.001). The effects of ascorbic acid on platelet aggregation in vitro were studied by adding ascorbic acid in buffered solution (pH 7.35) prior to-aggregating agents. Ascorbic acid in platelet-rich plasma consistently inhibited platelet aggregation with threshold concentrations of ADP, epinephrine, and collagen. With washed platelets, ascorbic acid inhibited arachidonic, acid-induced aggregation. When platelets were incubated at 37°C for 10 minutes with varying concentrations of ascorbic acid, rewashed, and aggregation with arachidonic acid tested, aggregation was inhibited in a linear dose-dependent fashion. Oral ingestion of ascorbic acid (2 gm/day) for seven days by normal non-smoking males produced a marked inhibition of aggregation. In a similar study, platelets from an insulin-dependent diabetic showed no change in aggregation. These results suggest that platelet levels of ascorbic acid may relate to the hyperaggregat ion of platelets from diabetics.

Diarrhea is a common health complaint occurring with mild, temporary conditions to a potentially life-threatening condition. <i>Cucurbita maxima</i> (Cucurbitaceae) seed is reportedly used traditionally for the treatment of diarrheal and thus this study aimed to evaluate the in vitro antioxidants, total tannins, phenolics contents, and antidiarrheal potentials of <i>n</i>-hexane and aqueous <i>C. maxima</i> seed extracts in castor oil induced diarrheal rats. The <i>n</i>-hexane extract mainly contains oils while the aqueous extract was thick, brown solid. The aqueous and <i>n</i>-hexane seed extract of <i>C. maxima</i> expressed significant 2,2, diphenyl-1-picrythdrazyl (DPPH) scavenging activities at 6.25 – 1000 mg/mL with IC<sub>50</sub> values of 104.01 mg/mL, 29.27 mg/mL and 26.78 mg/mL for <i>n</i>-hexane, aqueous and vitamin C respectively. The hydroxyl radical scavenging activities of the <i>n</i>-hexane and aqueous seed extract of <i>C. maxima</i> were significantly lower at higher concentrations compared with that of vitamin C. Furthermore, the total antioxidant capacities of <i>n</i>-hexane (22.08mg/mL) and aqueous seed extract (11.03 mg/mL) of <i>C. maxima</i> were found to be higher than that of vitamin C (134.46 mg/mL). The aqueous extracts (658.33±380.08 mg QE/g) total tannins were not significantly different from the <i>n</i>-hexane extract (468.33±102.55 mg QE/g) while the <i>n</i>-hexane extract contains significantly higher total phenolics (2.93±1.25 mg GAE/g) compared with the aqueous extract (0.19±0.04 mg GAE/g). Percentage inhibition of stooling was found to be 57±22%, 41±16%, and 46±11% for loperamide, <i>n</i>-hexane, and aqueous respectively. The study concludes that aqueous and n-hexane seed extracts of <i>C. maxima</i> (pumpkin) possess in vitro antioxidant activities and antidiarrheal properties.

Abstract Background: We investigated the in-vitro effects of vitamin C on delta-9-tetrahydrocannabinol (THC) -induced reduction in spermatozoa motility and kinematics. Methods: Six rats were used for the study. Semen from each of the 6 rats was randomly divided into 6 groups such that each rat’s semen was in all of the groups. Groups I-III received placebo, THC (1 mM), and vitamin C (5 mM) respectively. Group IV was pretreated with cannabinoid receptors’ blockers (CBs-) 1 and 2, followed by THC. Groups V and VI received THC and vitamin C, but group VI was additionally pre-treated with CBs-. Results: The spermatozoa progressive motility, average path velocity (VAP), curvilinear velocity (VCL), straight-line velocity (VSL), amplitude of lateral head (ALH) and beat cross frequency (BCF) were reduced by THC (6.08±1.16%; 5.64±0.82 µm/s; 6.96±0.74 µm/s; 2.75±0.23 µm/s; 0.31±0.02 µm; and 0.78±0.08 Hz respectively) but increased by vitamin C (51.20±1.32 %; 17.90±0.21 µm/s; 25.11±0.96 µm/s; 8.80±0.27 µm/s; 0.75±0.01 µm; and 3.15±0.03 Hz respectively) when compared to control (39.72±0.38 %; 13.70±0.29 µm/s; 18.04±0.58 µm/s; 7.54±0.34 µm/s; 0.65±0.02 µm; and 2.79±0.01 Hz respectively). Vitamin C inhibited the THC-induced reduction in these parameters (37.36±0.73 %; 10.98±0.45 µm/s; 13.58±0.30 µm/s; 7.11±0.22 µm/s; 0.58±0.01 µm; and 2.60±0.01 Hz respectively) in the absence of CBs- 1 and 2, and even caused additional increases in progressive motility (49.54±1.01 %), VAP (15.70±0.38 µm/s) and VCL (22.53±0.29 µm/s) above the control levels with CBs-.Conclusion: Vitamin C ameliorates the THC-induced reduction in spermatozoa motility in-vitro by modulation of their kinematics.

Dennettia tripetala fruit is a popular Nigerian fruit from the family of plant known as Annonaceae. The whole fruit (flesh and seed) is usually consumed as snacks and it is oftentimes consumed with local gin (ufofop in Ibibio or kaikai in Igbo) or added to dishes as spice due to its peculiar strong pepperish taste and sweet aroma. The present study is aimed at evaluating the antioxidant potentials of ethanol extract of ripe and unripe D. tripetala (DT) fruit in vitro. The antioxidant activity of the ethanol extract of DT was evaluated spectrophotometrically using various in vitro models like 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) and hydrogen peroxide scavenging activity; metal chelating activity and reducing power. Vitamin C was used as the standard antioxidant.Unripe and ripe DT fruits, as well as vitamin C showed a competitive inhibition of DPPH and H2O2 free radicals. As concentration of the extracts increased from 20 to100 µg/mL, the % scavenging activity for vitamin C increased from 87.86 ± 0.11 to 90.66 ± 0.07 and for ripe DT fruits from 15.15 ± 0.24 to 25.52 ± 0.23, while for unripe, fruits values increased from 12.09 ± 0.35 to 23.06 ± 0.12. The IC50 values was highest in unripe (549.23) followed by ripe (276.63) and lowest in vitamin C (12.92) indicating that vitamin C was the best scavenger of DPPH radical. Similar trend was obtained for H2O2 scavenging activity as well as reducing power. Unripe DT fruit extract was more potent at chelating metal ions (IC50 was 95.38), followed by the standard ascorbic acid with IC50 of 97.03 and was lowest in ripe DT fruit extract with IC50 value of 124.66. Unripe and ripe DT are potent antioxidants in nature and may be used to supplement our diets as rich sources of natural antioxidants for health protection.

There is an urgent need for anti-viral agents that treat and/or prevent Covid-19 caused by SARS-Coronavirus (CoV-2) infections. The replication of the SARS CoV-2 is dependent on the activity of two cysteine proteases, a papain-like protease, PL-pro, and the 3C-like protease known as main protease Mpro or 3CLpro. The shortest and the safest path to clinical use is the repurposing of drugs with binding affinity to PLpro or 3CLpro that have an established safety profile in humans. Several studies have reported crystal structures of SARS-CoV-2 main protease in complex with FDA approved drugs such as those used in treatment of hepatitis C. Here, we report the crystal structure of 3CLpro in complex Vitamin C (L-ascorbate) bound to the protein's active site at 2.5 Angstrom resolution. We also demonstrate that L-ascorbate inhibits the 3CLpro in vitro at mmol/L concentrations. The crystal structure of the Vitamin C 3CLpro complex may aid future studies on the effect of Vitamin C not only on the coronavirus main protease but on related proteases of other infectious viruses. Since ascorbate is readily available, as an over-the-counter vitamin supplement, our results have the potential for development of a global and inexpensive antiviral treatment.

A research  to study the sterilization   method  and application   of Kinetin  and IAA to induce the Durian  young  leaf (Durio zibethinus) in MS  medium   was conducted in Balai Benih Induk Hortikultura in Salaman  Magelang  district  of Central  Java  started  on September  until December 2003. The Laboratory experiment   was arranged  in two phases,  which were  the optimation  phase of sterilization   and  induction   phase.  At  the  first  phase,  the  sterilization method  used  was  the modification   of Mulya  (2001) method.  The modification   use of sterilant,  vitamin  C antioxidant, Alcohol  70 %, Benlate, Agrept,  Tween-20  and Betadine  were done to obtain  effectiveness   of the sterilization.  Explants  planted  then in MS medium  for two weeks. Contamination   time, percentage of contamination   and viabilitas  (percentage of living explants)  were observed  then.  At the second phase,  the treatments were arranged  in a 3 x 3 factorial  completely   randomized   design  (CRD)  to observed  the influence  of Kinetin  and IAA combination.   The concentration   of Kinetin  observed were 2, 4, and 6 mg/I, where  as the IAA concentration   were 0.5,  1.0, and  1.5 mg/I. All treatments were  repeated  three  times,  with three samples  on each  replication.   The percentage   of browning explants, percentage  of contaminated   explants,  site of  contamination   and percentage of explants live were observed  at the end of incubation. The results  showed that sterilization  of Durian young leaves explants  with 1  g/l deterjent  for 15 minutes  then by 2 g/l Benlate  and Agrept  for 10 minutes,  then by 1  g/200 mg Vitamin C, then by Alcohol  70 % for 1  minute, then by 20% Clorox,  then by 2 drip of Tween-20  for 10 minute and then by Betadine  decreased  the contamination down to 50 %, and this kind of sterilization  was relatively better than  the other  kinds.  Application   of growth  regulators   were  not  able  to induce  explants growth,  but stimulated  callus formation  at the cutting surface though,  in the application  of Kinetin 4 mg/1 + IAA 0,5 mg/I, Kinetin 4 mg/1 + IAA  1,5 mg/1, Kinetin  6 mg/I+  IAA 0,5  mg/1 and Kinetin 6 mg/l+IAA   1,0 mg/I.
