The purpose of the study was to assess the metabolic status of the body when correcting the diet with a concentrated natural product. Materials and methods. The diet of athletes was analyzed. The leading group (n = 15) received concentrated food products (CNP) for 20 days: No. 1 - 30.0 g and No. 2 - 20 g each. Blood indices including (total protein, uric acid, urea, creatinine, total cholesterol, high and low-density lipoprotein cholesterol, triglycerides, glucose, amylase) were assessed metabolic status: taken before, after, and in one month of taking CNP Results. The diet’s calorie content was 5953.2-6494.4 kcal/day in the main and the comparison group corresponding to this cohort’s physiological need. The proportions of protein and fat exceeded the recommended ones; deficiency of carbohydrates accounted for 8.7%. The inclusion of CNP increased the ratio of slow carbohydrates, vitamins, minerals, and minor food components. In the leading group, the increase in urea and uric acid was less significant in terms of average indices, and the proportion of such people increased. Creatinine was significantly higher than baseline within one month after CNP administration. The increase in the proportion of people with increased total cholesterol is 2.9 times less; the share with a borderline high level decreased by 15.5%, in the comparison group, it increased by 5.6%. The proportion with low-density lipoproteins exceeding the reference value decreased from 61.5% to 30.0% in the comparison group - without dynamics (55.6%) and signs of dyslipidemia increased. In the main group, within the normal range, 76.9% of individuals had an increase in glucose, amylase by 5.7% (p = 0.049). Conclusion. The diet is not macronutrient balanced. The use of CNP affected the metabolism of proteins, fats, and carbohydrates. It proved a more adequate energy supply for the activities of the persons of the main group. There has been shown the possibility of correcting the body’s metabolic processes with food products that reduces the risk of developing prenosological conditions during physical exertion, particularly fatigue and cardiovascular pathology.

AbstractCombinations of fructose- and fat-rich diets in experimental animals can model the human metabolic syndrome (MS). In rats, the increase in blood pressure (BP) after diet manipulation is sex related and highly dependent on testosterone secretion. However, the extent of the impact of diet on rodent hypophysial-testicular axis remains undefined. In the present study, rats drinking a 10% fructose solution or fed a high-fat (35%) diet for 10 weeks had higher plasma levels of luteinizing hormone (LH) and lower plasma levels of testosterone, without significant changes in circulating follicle-stimulating hormone or the weight of most reproductive organs. Diet manipulation brought about a significant increase in body weight, systolic BP, area under the curve (AUC) of glycemia after an intraperitoneal glucose tolerance test (IPGTT), and plasma low-density lipoprotein cholesterol, cholesterol, triglycerides, and uric acid levels. The concomitant administration of melatonin (25 μg/mL of drinking water) normalized the abnormally high LH levels but did not affect the inhibited testosterone secretion found in fructose- or high-fat-fed rats. Rather, melatonin per se inhibited testosterone secretion. Melatonin significantly blunted the body weight and systolic BP increase, the increase in the AUC of glycemia after an IPGTT, and the changes in circulating lipid profile and uric acid found in both MS models. The results are compatible with a primary inhibition of testicular function in diet-induced MS in rats and with the partial effectiveness of melatonin to counteract the metabolic but not the testicular sequelae of rodent MS.

Mouse peritoneal macrophages can be induced to accumulate cholesteryl esters by incubating them in the presence of acetylated low density lipoprotein. The cholesteryl esters are sequestered in neutral lipid droplets that remain in the cell even when the acetylated low density lipoprotein is removed from the culture media. Previous biochemical studies have determined that the cholesterol component of cholesteryl ester droplets constantly turns over with a half time of 24 h by a cyclic process of de-esterification and re-esterification. We have used morphologic techniques to determine the spatial relationship of cholesteryl ester, free cholesterol, and lipase activity during normal turnover and when turnover is disrupted. Lipid droplets were surrounded by numerous 7.5-10.0-nm filaments; moreover, at focal sites on the margin of each droplet there were whorles of concentrically arranged membrane that penetrated the matrix. Histochemically detectable lipase activity was associated with these stacks of membrane. Using filipin as a light and electron microscopic probe for free cholesterol, we determined that a pool of free cholesterol was associated with each lipid droplet. Following incubation in the presence of the exogenous cholesterol acceptor, high density lipoprotein, the cholesteryl ester droplets disappeared and were replaced with lipid droplets of a different lipid composition. Inhibition of cholesterol esterification caused cholesteryl ester droplets to disappear and free cholesterol to accumulate in numerous myelin-like structures in the body of the cell.

The purpose of this study was to analyze the association between APOE alleles and serum lipemia in adolescents with low and adequate aerobic fitness. The sample was comprised of 105 boys and 151 girls (49% and 46% from rural area) of European ancestry, aged 11 to 17 years, and classified according to: 1) APOE genotype: group ε2 (ε2/3+ε2/2), ε3 (ε3/3), and ε4 (ε3/4+ε4/4); 2) aerobic fitness: adequate or low; 3) serum lipemia: elevated total cholesterol (TC), low-density lipoprotein (LDL) and triglycerides, and low high-density lipoprotein (HDL). The results showed that aerobic fitness modulates the association between APOE alleles and serum lipemia in adolescents, suggesting that adequate aerobic fitness levels exert a greater effect of reducing TC and LDL in ε2 carriers, as well as of increasing HDL and reducing triglycerides in ε3 and ε4 carriers.

Background: Diabetes mellitus (DM) is a syndrome consisting of metabolic, vascular and neuropathic components that are interrelated. Diabetes mellitus is associated with a considerably increased risk of premature atherosclerosis, particularly coronary heart disease (CHD) and peripheral arterial disease. Dyslipidemia is a common feature of diabetes. There is an association between atherosclerotic cardiovascular disease and serum cholesterol and triglyceride levels in both type 1 and type 2 diabetes. Methods: The study was done on 50 adult diabetes mellitus (T2) patients from IPD of General Medicine wards at SMS Hospital, Ahmedabad, Gujarat. 50 healthy age and sex matched healthy volunteers were taken as control. They were evaluated for lipid profile i.e., Total Cholesterol (TC),Triglyceride (TG), Low-density lipoprotein (LDL), High density lipoprotein (HDL), Very low density lipoprotein (VLDL) and glycemic status i.e., Fasting blood glucose (FBS), Postprandial 2 hours blood glucose (PP2BS) & Glycosylated haemoglobin(HbA1C).Results: Diabetic cases had statistically highly significant (p<0.001) elevated levels of total Cholesterol, Triglycerides and VLDL as compared to controls. Serum TG, serum TC, LDL-C and VLDL-C had positive correlation with the postprandial plasma glucose, fasting plasma glucose and HbA1c.Conclusions: Significant correlations between HbA1c levels and lipid levels point towards the usefulness of HbA1c for screening high-risk diabetic patients. High TC, TG, LDL-C and HbA1c with normal or low HDL-C is seen in almost all diabetic patients either alone or in combinations.

Statin side effects are not a rare occurrence, in particular dyspeptic disorders, insomnia, headache, skin erythema, rash are often noted. All of this determines scientists to find new effective and low-toxic hypolipidemic agents. Various natural and synthetic xanthine derivatives have been recognized as therapeutically potential compounds and reported to control various diseases. Therefore, the study of new xanthine derivatives and their hypolipidemic effects, which would have a significant therapeutic effect with minimal side effects, is relevant. The aim of the study was to examine the effect of 7-β-hydroxy-γ-aryloxypropylxanthinyl-8-thioalkanic acid derivatives on lipidogram parameters in experimental laboratory rats. Materials and methods. The objects of the study were 7-β-hydroxy-γ-aryloxypropylxanthinyl-8-thioalkanic acid derivatives. The experiments were performed in white laboratory Wistar rats weighing 180–220 g. Experimental modeling of hyperlipidemia – tween model: intraperitoneal administration of tween-80 at a dose of 200 mg/100 g body weight. The test compounds were administered orally, simultaneously with tween, at a dose of 1/10 of LD50 (previously calculated by Prozorovsky express method) for 6 days. The following indicators of lipidogram were determined: total cholesterol (TC), high-density lipoprotein cholesterol (HDL cholesterol), low-density lipoprotein cholesterol (LDL cholesterol), triglycerides (TG) and atherogenic index of plasma: TC – HDL cholesterol / HDL cholesterol. The experiments were carried out with respect to Bioethical rules and norms. Results. The studies have shown data on the hypolipidemic activity of 7-β-hydroxy-γ-aryloxypropylxanthinyl-8-thioalkane acid derivatives. According to the conditional efficiency index Ʃ, which included the overall percentage of the following indicators – total cholesterol, low-density lipoprotein cholesterol and triglycerides, the leading compounds were 2439 (87.47 %), 6047 (82.30 %). The reference drug atorvastatin had a value of 82.98 %. Conclusions. The major compound was 2439 identified among all compared to the control group. The prospect of further research is a more detailed study on the ability of xanthine derivatives to exhibit hypolipidemic effects and to influence oxidative stress in various hyperlipidemic models.

Abstract This study evaluated the effect of the temperament of 75 water buffaloes of three genetic groups (GG: Jafarabadi, Mediterranean, and Murrah; n = 25 for each GG) on serum lipid levels. The animals were classified for temperament through the temperament score (TSc) obtained by the mean of the sum of the scores of the entry time into the squeeze chute (1 to 5: 1=greater time spent for entry, and 5=less time spent for entry) and the exit velocity score (1 to 5: 1=lower speed, and 5=higher speed) calculated after the period of adaptation to the feedlot (d0). The animals were categorized into adequate (ADQ; TSc≤3) or excitable (EXC; TSc >3) temperaments. Blood samples were collected from the jugular vein on days -28, 0, and 84, and evaluated for serum levels of total cholesterol, triglycerides, high-density lipoprotein (HDL), low-density lipoprotein (LDL), and very low-density lipoprotein (VLDL) using colorimetric enzymatic analyses performed by commercial kits. Data were analyzed using MIXED procedure in SAS, considering the GG, temperament, day, and the resulting interactions as fixed effects. The term day was used for repeated measures in time. Correlations were estimated using the CORR procedure in SAS. There were no interactions for the variables evaluated (P ≥ 0.11). ADQ animals showed lower levels of total cholesterol (65.95 vs 70.96mg/dL for ADQ and EXC, respectively; SEM=2.33; P = 0.04) and LDL (26.74 vs 30.40mg/dL for ADQ and EXC, respectively; SEM=1.57; P = 0.02) than EXC animals. There was no effect of temperament for the triglycerides (P = 0.41), HDL (P = 0.47), and VLDL (P = 0.46). Positive correlations were verified between both the TSc and triglycerides (r=0.32; P < 0.01), and TSc and VLDL (r=0.33; P < 0.01). Tendency for a positive correlation was detected between TSc and LDL (r=0.20; P = 0.09). In conclusion, there are differences in serum lipid levels between water buffalo temperaments which deserves further investigation. Supported by FAPESP (#2018/25939-1; #2014/05473-7).
