Aerial tissues of the woody lianas- Combretum sp. was selected and assessed for study of endophytic fungal diversity from three forest areas of West Medinipur and Jhargram districts of West Bengal. In monsoon study it was observed that out of 225 various tissue segments ,165 segments had been colonized by endophytic fungi Combretum roxburghii and 209 endophytic isolates were isolated from them. Average colonization frequency (CF)was 73.32 percent. Highest CF was in plant of Belpahari (76%) and in petiole (82.66%). Out of isolated fungal genera Pestalotiopsis sp. was highest in number. Diplodia sp., Beltrania sp., Chaetomium sp., Fusarium sp., Arthrinium sp. were also greater in number. Dominance index was highest in Belpahari (0.1547). Simpson’s diversity was maximum in Godapiasal (0.8693) and Shannon-Wiener index was also highest (2.152) in plant of Godapiasal. Highest diversity of endophytic fungi was in plants of Chilkigarh. Isolated endophytic fungi were Lasiodiplodia sp., Diplodia sp., Fusarium sp., Chaetomium sp., Arthrinium sp., Aspergillus sp., Pestalotiopsis sp. etc. Simpson’s diversity and Shannon-Wiener index were maximum in plants of Godapiasal. The study of antimicrobial activity of isolated fungal endophytes indicated that few plugs showed antimicrobial activity against few pathogenic bacteria. Among all isolates tested only seven showed antimicrobial activity. Aspergillus sp. and one unidentified fungi showed the antimicrobial activity against three bacteria i. e., Bacillus cereus, Escherechia coli and Vibrio cholera and maximum inhibition zone was observed against E. coli, diameter of inhibition zone is 1.4 cm. Other fungi showed less antimicrobial activity. Diameter of zone of inhibition varied (0.7 – 1.4) and this variation might be due to the difference of bioactive compound produced or the varied concentrations or amount degree of same compound.

Objective:To study the antimicrobial activity of crud ethylacetate  extract from endophytic fungus in Calotropis procera root.Methods:Endophytic fungus was screened for production of antimicrobial metablites.fermentation was carried out in 500 ml Erlenmeyer flask. Disc diffusion method was used to test antimicrobial activity of crude extract using chloramphenicol sulphate and Flucnazole as positive controleRessults: A total of fourteen Endophytic fungi were isolated (CPR1- CPR14). Among these fourteen isolates, CPR5 was found to show maximum antimicrobial activity, in compare to other isolates, against gram positive, gram negative bacteria and fungi. The antimicrobial activity was tested against Escherichia coli. Pseudomonas aeroginosa, Bacillus subtilis, Staphylococcus aureus, Ralstonia solanacearum, Xanthomonas oryzae, Penicillium chrysogenum, Candida albicans, Phoma exigua, Sclerotium rolfsii and Sclerotinia scleratiourum. Minimum inhibitory concentration (MIC) of crude extract against test microorganisms was determined. Fungus was identified as aspergillus sp. Prduction parameters (Temprature, pH, Carbon source, Nitrgen surce, Sdium Chlride concentration) were optimized.Conclusion:  Crud  extract produced by the isolated endophytic fungus could be an important source of broad spectrum antimicrobial metabolites. Keywords: Endophytic fungi, Antimicrobial metabolites, Process optimization, Calotropis procera, Inhibition zone.

Twenty fungal endophytes were isolated from different medicinal plants of Biligirirangana Hill, Chamarajanagar dist. Phytochemical analysis of ethyl acetate extract showed positive results for tanins, flavonoids, steroids, terpenoids, glycosides, saponins and alkaloids except saponin and flavonoid tests. The crude extracts of the fungal endophytes were tested against two gram positive and two gramnegetive bacteria for its antibacterial activity. The highest zone of inhibition was produced by Fusarium sps (AB9)35mm. All the crude extracts were found to be effective against Escherichia coli, Bacillus subtilis and Staphylococcus aureus rather in Pseudomonas aerogenosa. Among them the highest inhibition zone was produced in Penicillium sp (AB11) 24mm, Cladosporium sp(AB3) 21mm, and Aspergillus sp(AB12) 30mm. Therefore, endophytic fungi can be a good source to inhibit the growth of harmful pathogens.

The plant Chloranthus japonicus Sieb is known for its anticancer properties and mainly distributed in China, Japan, and Korea. In this study, we firstly investigated the diversity and antimicrobial activity of the culturable endophytic fungi from C. japonicus. A total of 332 fungal colonies were successfully isolated from 555 tissue segments of the medicinal plant C. japonicus collected from Qinling Mountains, China. One hundred and thirty representative morphotype strains were identified according to ITS rDNA sequence analyses and were grouped into three phyla (Ascomycota, Basidiomycota, Mucoromycota), five classes (Dothideomycetes, Sordariomycetes, Eurotiomycetes, Agaricomycetes, Mucoromycetes), and at least 30 genera. Colletotrichum (RA, 60.54%) was the most abundant genus, followed by Aspergillus (RA, 11.75%) and Diaporthe (RA, 9.34%). The Species Richness Index (S, 56) and the Shannon-Wiener Index (H′, 2.7076) indicated that C. japonicus harbored abundant fungal resources. Thirteen out of 130 endophytic fungal ethyl acetate extracts exhibited inhibitory activities against at least one pathogenic bacterium or fungus. Among of these, F8158, which was identified as Trichoderma cf. harzianum, exhibited good antagonistic capacities (the percent inhibition of mycelial growth ranged from 47.72~88.18) for different pathogens and has a potential application in biological control. In addition, it is noteworthy that the strain F8157 (Thanatephorus cucumeris, an opportunistic pathogen) showed antibacterial and antifungal activity, which is reported firstly in this study, and should be investigated further. Taken together, these results indicated that the endophytic fungi from C. japonicus may be of potential interest in screening bio-control agents and discovering of new bioactive compounds.

Endophytic fungi, which colonize within a host plant without causing any apparent diseases, have been considered as an important source of bioactive secondary metabolites containing antimicrobial and antioxidant activities. The aim of this research was to isolate the endophytic fungi ofCinnamomum loureiroiand then to screen their antimicrobial and antioxidant activities. A total of 11 fungal endophytes were isolated from healthy leaves ofCinnamomum loureiroibelonging to six genera:Botryosphaeria,Colletotrichum,Diaporthe,Fusarium,Neopestalotiopsis, andPestalotiopsis. All isolated strains were cultured and further extracted with ethyl acetate solvent. Antimicrobial activity of all crude endophytic fungal extracts was analyzed using disc diffusion assay against six bacterial and two fungal pathogens. Crude extracts of strains MFLUCC15-1130 and MFLUCC15-1131 showed broad-spectrum antimicrobial activity against all tested pathogens. Activity againstBacillus cereusandStaphylococcus epidermidiswas notable, showing the lowest minimum inhibitory concentration at 3.91 μg/mL. Antioxidant activity of all crude endophytic fungal extracts was also evaluated based on 2,2-diphenyl-1-picrylhydrazyl assay. Significant antioxidant activity was detected in the crude extracts of fungus MFLUCC15-1130 and MFLUCC15-1131 with IC50of 22.92 ± 0.67 and 37.61 ± 0.49 μg/mL, respectively. Using molecular identification, MFLUCC15-1130 and MFLUCC15-1131 were identified asNeopestalotiopsissp. andDiaporthesp., respectively. The major chemical constituents produced by both crude extracts were identified by gas chromatography-mass spectrometry. Eugenol, myristaldehyde, lauric acid, and caprylic acid were the primary antimicrobial and antioxidant compounds in both crude extracts. This is the first report of eugenol being a biologically active compound ofNeopestalotiopsissp. andDiaporthesp. fungal endophytes. Eugenol has been reported as antimicrobial and antioxidant agents with agronomic applications. Thus the two newly-isolated endophytes may be used for eugenol production, which in turn can be used in a variety of applications.

Purpose The inclusion of natural antioxidants and nanoparticles into edible coatings can improve their structure, functionality and applicability in foods. This study aims to determine the antibacterial effect of new nano-composite based on bean pod shell gum (BPSG), silver nanoparticle (SNP) and herbal essential oil (EO). Design/methodology/approach For this purpose, BPSG (4 per cent w/v), SNP (0.5-1 per cent w/v) and Helichrysum arenarium EO (1-2 per cent v/v) were mixed. After preparation the edible film, the antibacterial activity was tested on five food-borne pathogenic bacteria in two categories including two-gram positives and three-gram negatives bacteria. The EO composition was determined by GC–mass spectrometry and the antibacterial activity was tested using disk diffusion test. Findings Results showed that gram-positive bacteria were more susceptible than gram-negative bacteria. Increasing Helichrysum arenarium EO and SNP content increased antimicrobial activity of the edible film based on BPSG, so that the treatment containing 2 per cent v/v Helichrysum arenarium EO and 1 per cent w/v SNP led to the highest inhibition zone (8.1-13.1 mm) compared to treatment containing 1 per cent v/v Helichrysum arenarium EO and 0.5 per cent w/v SNP with inhibition zone range of 5.4-9.9 mm and the difference was statistically significant (p = 0.41). Also, the chemical composition analysis of the EO identified a total of 38 compounds in which a-pinene (32 per cent), 1,8-cineole (16 per cent), α-humulene (15 per cent) and ß-caryophyllene (8 per cent) were the main fractions. Other separated components accounted less than 29 per cent of the oil. Originality/value In general, SNP and Helichrysum arenarium EO improved the functional properties including the antimicrobial activity of the edible film based on BPSG, which increases the potential to be used as active packaging for fresh products.

The study was undertaken to investigate the antimicrobial activity both individually and in combination of two Nigerian plants (Azadirachta indica and Dodonea viscosa) against a variety of pathogenic bacteria. Five bacterial species namely; Staphylococcus aureus, Escherichia coli, Salmonella typhi, Klebsiella pneumoniae and Pseudomonas aeruginosa were selected for the assays. The powdered plant materials of the two plants were extracted using sohxlet extraction technique with methanol water and petroleum ether as solvents. The crude extracts of the two plants were subjected to phytochemical screening for qualitative detection of plant secondary metabolites. The extracts were further tested for antibacterial activity against the selected pathogens singly and then combined using agar well diffusion method. Antibacterial activity of the two plants singly indicates that the methanolic extract possess the highest antibacterial activity at a concentration of 50mg/ml with an inhibition zone of 23.3±0.5mm compared to aqueous extract with 21.7±0.5mm inhibition zone at a concentration of 50mg/ml. The least activity was observed with the petroleum ether extract with an inhibition zone of 7.3±0.5mm at a concentration of 12.5mg/ml. Combination of the plants extracts exhibited lower antibacterial activity on the test isolates compared to single plants as evidenced by the production of lower inhibition zones. ABSTRACT Introduction The main goal of endodontics is the prevention of apical periodontitis. This is due to the presence of persistent pathogenic microorganisms, such as Enterococcus faecalis, and its ability to directly cause acute and chronic inflammation in the periapical tissues. Lactobacillus has been shown to promote health in the intestines as well as to inhibit the growth of certain problematic oral bacteria. This study explores shifting the established paradigm of endodontic treatment, which has focused on eliminating all bacteria from the canal system and on elimination of the problematic bacteria through introducing probiotics. A preliminary work was performed to evaluate the possible effectiveness of probiotics in preventing the growth of E. faecalis. Materials and methods Two probiotic cultures Lactobacillus plantarum ATCC 8041 and Lactobacillus rhamnosus ATCC 7408 were selected to check their antimicrobial activity against E. faecalis ATCC 29212 by two methods: agar cup/well diffusion method and deferred antagonism test. Agar cup method A total of 0.5 mL of requisite test pathogen culture was inoculated into 20 mL of molten sterile Mueller and Hinton agar and cooled to 45 ± 2°C. Circular wells of diameter 10 mm were punched in each of the poured plates. Appropriately diluted test samples were added to the above-punched wells. The plates were incubated upright position at 37°C for 24 hours in aerobic conditions. Postincubation, zone of inhibition was measured. The cell-free supernatant of Lactobacillus species was also evaluated for antimicrobial activity. Deferred antagonism test The test probiotic strain was standardized to 0.1 optical density (OD) at 600 nm and inoculated in a 1 cm wide diametric streak across the surface of trypticase soy agar + yeast extract + calcium carbonate (TSYCa) agar using a sterile cotton swab. Then, the plate agar containing the test strain was incubated at 37°C for 24 hours under aerophilic condition and then standardized to 0.1 OD at 600 nm for overnight (18 hours, 37°C). A purified culture of indicator strain (pathogen) was streaked at right angles to the line of original producer growth. Postincubation plates were observed for the inhibition zone width of the indicator strain. Results Under the conditions of this study, Lactobacilli had an inhibitory effect on the growth of E. faecalis by agar cup method but not by deferred antagonism test. Conclusion This pilot study demonstrated that probiotics show a potential in root canal therapy. Clinical Significance If probioitcs are effective against endodontic pathogens, they can be potentially used as intracanal medicaments. This will be a novel concept of introducing bacteriotherapy in endodontics and replacing pathogenic bacteria by healthy bacteria, normal flora. How to cite this article Bohora AA, Kokate SR. Abstract: The aim of this study is to investigate the antibacterial effect of endophytic fungi originated from the root of mangrove Rhizophora stylosa growing on Mangrove Plantation Area around DAS Simpang 5 Jl. Piere Tendean Manado. The method of this research was using combination of PDA and NA media. Two species of endophytic fungi were extracted from the root of R. stylosa, black mycelium fungi as isolate A and brown mycelium fungi as isolate B. The activities of both isolates were tested against pathogenic bacteria Staphylococcus aureus and Escherichia coli. The result showed the same inhibition zone of isolate A and B to S. aureus, while, isolate A showed bigger the inhibition zone in comparison to isolate B against E. coli. As a conclusion, the endophytic fungi taken from the root of R. stylosa have antibacterial activity toward S. aureus and E. coli. Piere Tendean Manado. Metode penelitian dilakukan dengan menggunakan kombinasi media Potato Dextrose Agar (PDA) dan Nutrient Agar (NA). Dari akar bakau R. stylosa diperoleh dua  jenis jamur endofit yaitu isolat A dengan karateristik miselium jamur berwarna hitam dan isolat B yang dengan karakteristik miselium berwarna coklat. Kedua isolat ini selanjutnya diujikan aktivitasnya terhadap bakteri Staphylococcus aureus dan Escherichia coli. Hasil penelitian menunjukkan isolat A dan B memberikan  zona hambat yang sama terhadap bakteri Staphylococcus aureus dan pada bakteri Escherichia coli isolat A menunjukkan diameter zona hambat yang lebih besar dibandingkan isolat B. Kesimpulan, Jamur endofit yang diisolasi dari akar bakau R. Stylosa memiliki efek antibakteri terhadap pertumbuhan bakteri S. aureus dan E. coli. Kata kunci: jamur endofit, Rhizophopra stylosa, antibakteri, Staphylococcus aureus, Escherichia coli.

ABSTRACT Lactic acid bacteria (LAB) were isolated from of cocoa beans fermentation Forestero variety from West Sumatera, that were eleven isolates. The isolates were tested to antimicrobial activity against pathogenic bacteria E.coli NBRC 14237, Staphylococcus aureus NBRC 13276, Bacillus subtilis BTCCB 612, listeria m. dan S. Typhii. Results the research showed that, isolates had inhibition zone to pathogenic bacteria, that were 7 mm till 12 mm at 48 hours observation. R2.4 isolate was most potential to inhibition zones growth pathogenic bacteria, that was 11mm till 12 mm to five pathogens. R2.4 isolates was the highest to against pathogenic bacteria (Bacillus subtilis BTCCB, Listeria monocytogenesis and Staphylococcus aureus NBRC) had inhibition zones, that was 12.00 mm till 48 hours. Listeria monocytogenesis had been known as pest bacterium of food born, so that R2.4 isolate can be used as food biopreservative. Crude of R2.4 isolate molecular weight was 10 kDa by SDS-PAGE.  Key words: Lactic acid bacteria, Antimicrobial activity, SDS-PAGE, Cocoa fermentation and food biopreservative

Endophytes or endophytic fungi have been investigated as a store house of bioactive compound. This study was aimed to evaluate the potential antibacterial activity of endophytic fungi isolated from Medinilla speciosa Blume. The antibacterial test was determined by measuring the inhibition zone with disk-diffusion method. Twenty endophytes were isolated from the leaves of Medinilla speciosa Blume and identified morphologically. The results demonstrated that ten isolates showed variation in their antibacterial activity against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Bacillus subtilis ATCC 6633, and Shigella dysenteriae ATCC 13313. Further investigation will be needed to explore and identify the bioactive molecules of the isolated endophytic fungi.
