Flavobacterium columnare immersion challenges are affected by water-related environmental parameters and thus are difficult to reproduce. Whereas these challenges are typically conducted using flow-through systems, use of a recirculating challenge system to control environmental parameters may improve reproducibility. We compared mortality, bacterial concentration, and environmental parameters between flow-through and recirculating immersion challenge systems under laboratory conditions using 20 rainbow trout families. Despite identical dose concentration (1:75 dilution), duration of challenge, lot of fish, and temperature, average mortality in the recirculating system (42%) was lower (p < 0.01) compared to the flow-through system (77%), and there was low correlation (r = 0.24) of family mortality. Mean days to death (3.25 vs. 2.99 d) and aquaria-to-aquaria variation (9.6 vs. 10.4%) in the recirculating and flow-through systems, respectively, did not differ (p ≥ 0.30). Despite 10-fold lower water replacement rate in the recirculating (0.4 exchanges h-1) compared to flow-through system (4 exchanges h-1), differences in bacterial concentration between the 2 systems were modest (≤0.6 orders of magnitude) and inconsistent throughout the 21 d challenge. Compared to the flow-through system, dissolved oxygen during the 1 h exposure and pH were greater (p ≤ 0.02), and calcium and hardness were lower (p ≤ 0.03), in the recirculating system. Although this study was not designed to test effects of specific environmental parameters on mortality, it demonstrates that the cumulative effects of these parameters result in poor reproducibility. A recirculating immersion challenge model may be warranted to empirically identify and control environmental parameters affecting mortality and thus may serve as a more repeatable laboratory challenge model.

Many countries around the world are resorting to mandatory funded components in their multi-pillar pension systems with the purpose of catering for the financial pressure from ageing. This paper aims at analysing the possible replacement rates for such a scheme, by choosing different assumptions and setting the best combined area for the expected result. Then, an approach for analysing the potential for the implementation of such a scheme in Greece is presented along with the actuarially projected expected benefit expenditure and respective accrued capital. A result of the introduction of such a component is expected to be the elevated replacement rate at retirement with a concurrent alleviation of the fiscal burden for the state. The projected scale of savings will also provide domestic financing for investments generating growth.

Fishmeal (FM) is still the most important protein source in aquafeeds. However, due to the reduction of wild fish stocks used for FM production, its manufacturing it is now unsustainable. Insect meal represents a valid alternative to FM, due to the low carbon footprint of its production and its high nutritional value. The aim of this study was to investigate the potentials of replacing FM with black soldier fly (Hermetia illucens) meal (HIM) in aquafeeds, using zebrafish as the animal model. Four diets were formulated with increasing HIM/FM replacement rate (the control diet contained 20% FM). Mortality rates, growth performances, and feed consumptions were measured, and fish intestine samples were collected for histological and enzymatic analysis. After 49 days, all groups almost tripled their initial body weight (cumulative body weight gain ranged between 143.9 and 155.2 mg), and no statistically significant differences among treatments were observed in relation to growth performances and histological traits. Contrarily, trypsin, alkaline phosphatases, and alpha amylase–glucoamylase activities were significantly reduced when the FM replacement rate was increased. In conclusion, HIM may represent an alternative to FM, since no adverse effects were observed when it was included up to 20% in the zebrafish diet, even when replacing 100% of the FM.

Abstract We propose models for describing replacement rate variation in genes and proteins, in which the profile of relative replacement rates along the length of a given sequence is defined as a function of the site number. We consider here two types of functions, one derived from the cosine Fourier series, and the other from discrete wavelet transforms. The number of parameters used for characterizing the substitution rates along the sequences can be flexibly changed and in their most parameter-rich versions, both Fourier and wavelet models become equivalent to the unrestricted-rates model, in which each site of a sequence alignment evolves at a unique rate. When applied to a few real data sets, the new models appeared to fit data better than the discrete gamma model when compared with the Akaike information criterion and the likelihood-ratio test, although the parametric bootstrap version of the Cox test performed for one of the data sets indicated that the difference in likelihoods between the two models is not significant. The new models are applicable to testing biological hypotheses such as the statistical identity of rate variation profiles among homologous protein families. These models are also useful for determining regions in genes and proteins that evolve significantly faster or slower than the sequence average. We illustrate the application of the new method by analyzing human immunoglobulin and Drosophilid alcohol dehydrogenase sequences.

The objectiveof this study was to elucidatereproductive characteristics, potential output and dynamics population of Ongole Gradecattle in Kebumen Regency, Province of Central Java. The studywasconductedfromSeptemberto November 2015.The materials used in the study were165 farmers as respondentshavingOngole Gradecattle. The research method used was survey. Respondent samples were taken from five districts in which the location was determined by using purposive sampling. Data were collected through direct interviews with livestock farmers as respondents. The analysis used wasdescriptive analysis. The results showed that the age at first mating was 26.87 month, S/C was 1.64, post-partum mating was4.52 month, weaning age was 3.97 month, calving interval was 14.32 month, and the birth rate topopulation was45.5%. Reproductive efficiency value was 91.8%, natural increase was 44.68%, and net replacement rate of bull and heifer were 1,209% and 253%,respectively, andtotal output 44.11%. Population dynamics of Ongole Gradecattle from 2009 to 2014 fluctuated with average growthwas -4.84%. In 2019, cow population of Ongole Gradewasestimated about 68.381 heads. In conclusion Kebumen Regencyhasa relatively high potential output of Ongole Gradecattle thatis44.11% and population dynamics from 2010 to 2014 fluctuated with average of decrease growth is 4.84%. Estimation of cow’s population of Ongole Grade cattle willincrease 16.7% per year.

The study analyzed the possibility of using the acoustic emission method to analyse the reaction of alkali with aggregate in the presence of lithium nitrate. Lithium nitrate is a chemical admixture used to reduce adverse effects of corrosion. The tests were carried out using mortars with reactive opal aggregate, stored under the conditions defined by ASTM C227. The acoustic activity of mortars with a corrosion inhibitor was referred to linear changes and microstructure of specimens in the initial reaction stages. The study found a low acoustic activity of mortars with lithium nitrate. Analysis of characteristic parameters of acoustic emission signals, combined with the observation of changes in the microstructure, made it possible to describe the corrosion processes. As the reaction progressed, signals with different characteristics were recorded, indicating aggregate cracking at the initial stage of the reaction, followed by cracking of the cement paste. The results, which were referred to the acoustic activity of reference mortars, confirmed that the reaction of opal aggregate with alkali was mitigated in mortars with lithium nitrate, and the applied acoustic emission method enabled the detection and monitoring of ASR progress.

Abstract. Recycled aggregate—rural building material wastes pretreated by cement mortar—are applied into concrete with different replacement rates: 0, 25%, 50%, 75%, and 100%. Results from measurements of compressive strength, cleavage tensile strength, mass loss after fast freeze-thaw cycles, and compressive strength loss indicate that a different recycled aggregate replacement rate certainly influences concrete mechanical properties and frost resistance. Recycled aggregate replacement rates less than 75% performs better than common concrete. Data from the 100% replacement rate is worse than that of rates less than 75% but still satisfy the general demands of GB standard on C30 concrete.
