A Erythrina velutina Willd. é conhecida popularmente como “mulungu”, suas cascas são utilizadas principalmente pela população do nordeste brasileiro devido a suas propriedades calmante, sudorífica, emoliente e anestésica local. A pesquisa teve como propósitos o isolamento de substâncias químicas, a partir de técnicas cromatográficas, presentes no extrato bruto e frações deste (hexano, diclorometano, acetato de etila e hidroalcoólica remanescente). A identificação dos compostos isolados foi realizada por meio da análise dos dados das espectroscopias de ultravioleta, infravermelho e ressonância magnética nuclear de carbono e hidrogênio; a verificação da atividade antibacteriana do extrato bruto foi realizada através dos testes de difusão em disco e concentração inibitória mínima e, da fração hexano por teste de difusão em disco, além disso também foi procedida a verificação da atividade alelopática na germinação e alteração do crescimento da radícula e hipocótilo das sementes de Lactuca sativa para diferentes concentrações do extrato bruto e frações, os dados foram analisados estatisticamente pela análise de variância e teste de comparação múltipla de médias de Scott & Knott. Na fração hexano foram identificados o ácido fênico, ácido cinâmico, a-amirina, estigmasterol, b-amirina, b-sitosterol e lupeol por cromatografia gasosa e isolados o estigmasterol em mistura com o b-sitosterol. Da fração diclorometano foram isolados dois alcalóides e as estruturas propostas são eritralina, que foi identificada por comparação de seus dados espectrais com os dados publicados e já foi isolada desta espécie, 8-oxo-11b-metoxieritralina e eritrinina (não relatadas para a espécie); da fração hidroalcoólica remanescente isolou-se um composto que exibe algumas características de açúcar monocíclico, ainda não completamente identificado. O extrato bruto e a fração hexano demonstraram atividade antibacteriana moderada, em relação aos antibióticos controle, contra o Staphylococcus aureus e Streptococcus pyogenes. A germinação das sementes de Lactuca sativa não foi inibida e o crescimento da radícula e hipocótilo destas plântulas para o extrato bruto e frações foi estatisticamente significativo e diferente dos controles com água e etanol, a fração hidroalcoólica remanescente teve a maior atividade e alterou a morfologia das plântulas causando enovelamento, ausência de pêlos radiculares e ausência de folíolos. Cabe salientar que a atividade antibacteriana, a atividade alelopática e a presença de estigmasterol e o composto isolado da fração hidroalcoólica remanescente ainda não haviam sido citadas na literatura conforme levantamento bibliográfico para Erythrina velutina. PHITOCHEMICAL AND BIOLOGICAL STUDY FROM ERYTHRINA VELUTINA WILLD., FABACEAE (LEGUMINASAE- PAPILIONOIDEAE) BARK. Abstract Erythrina velutina Willd. is popularly known as mulungu, and its bark is used especially by the population of the Brazilian northeast owning to its sedative, sudorific, emollient and topic anaesthetic properties. This research is aimed at isolating the chemical substances in the crude extract and its partitions (hexane, dichloromethane, ethyl acetate and hydro alcoholic remaining fractions), through chromatographic techniques. The analysis of the spectrum copies of ultraviolet, infrared and nuclear magnetic resonance of carbon and hydrogen were used in the identification of the isolated compounds. The antibacterial activity of the crude extract was verified by using disk diffusion method and minimal inhibition concentration (MIC), and hexane fraction activity was verified through the disk diffusion method, and besides that the allelopathic activity in the germination and growth of the radicle and plantule of the Lactuca sativa seeds was checked at different concentrations of crude extract and fractions. Data were statistically examined by analysis of variance (ANOVA) and Scott Knott multiple comparison test. The hexane fraction GC showed fenic acid, cinnamic acid, a-amirin, stigmasterol, b-amirin, b-sitosterol and lupeol by gas chromatography; stigmasterol was isolated and identified as the major compound in the mixture with b-sitosterol. Two alkaloids were isolated from the dichloromethane fraction and the structures found were: erythraline that has been identified by comparing its spectral characteristics to its published ones and has been isolated from this gender, and 8–oxo–11(-methoxyerythraline and erythrinin which had not been related to this gender before; from the remaining hidroalcoholic fraction it was possible to segregate a compound that seems to have monocyclic sugar characteristics, but has not been thoroughly identified so far. The crude extract and the hexane fraction gave evidence of moderate antibacterial activity in relation to the antibiotics control, against Staphylococcus aureus and Streptococcus pyogenes. The germination of Lactuca sativa seeds was not inhibited and the growth of the radicle and hypocotyls to the amount of crude extract and fractions were statistically significant and different from the water and ethanol control. The remaining hidroalcoholic fraction was the most active and that caused the plantules morphology to show alterations such as curling, radicle hair absence and follicle absence. It seems important to emphasize that antibacterial activity, allelopathic activity, stigmasterol presence and the isolated compound from the remaining hidroalcoholic fraction had never been mentioned before in literature according to a bibliographic survey of Erythrina velutina.

Introduction: Staphylococcus aureus causes a wide range of infections and as a multivalent pathogen is one of the causative agents of nosocomial and community infections. Therefore, the aim of this study was to identify and determine the pattern of antibiotic resistance of methicillin-resistant Staphylococcus aureus (MRSA) isolates from patients in hospitals and medical centers in Marand city and also to evaluate the presence of mecA gene. Materials and Methods: In this cross-sectional descriptive study, 385 samples of S. aureus were collected from different clinical samples of patients in hospitals and medical centers of Marand city. S. aureus was identified using standard biochemical methods.  Methicillin resistance was determined by disk diffusion method in the presence of oxacillin and cefoxitin. The pattern of antibiotic resistance of the strains was determined by disk diffusion method and according to CLSI recommendation and also PCR method was used to evaluate the frequency of MecA gene. Results: In the present study, out of 385 samples of S. aureus, 215 (55.84%) samples were methicillin resistant. PCR results for mecA gene showed that 110 samples had mecA gene.  The highest antibiotic resistance was observed against penicillin (100%) and erythromycin (83.63%). Most MRSA were isolated from urine and wound samples. Conclusion: The results of this study indicate the prevalence of methicillin-resistant species and also the increase in antibiotic resistance of MRSA to various antibiotics.  Therefore, in order to prevent increased resistance to other antibiotics, it is recommended to avoid inappropriate use of antibiotics.

The essential oil of the leaves of Espeletia nana Cuatrec, obtained by hydrodistillation, was analyzed by GC-MS, which allowed the identification of 24 components, which made up 99.9% of the oil. The most abundant compounds were α-pinene (38.1%), β-pinene (17.2%), myrcene (15.0%), spathulenol (4.2%), bicyclogermacrene (4.0%), α-zingiberene (4.0%), and γhimachalene (3.7%). Antibacterial activity was tested against Gram-positive and Gram-negative bacteria using the agar disk diffusion method. Activity was observed only against Gram-positive bacteria. MIC values were determined for Staphylococcus aureus ATCC 25923(200 μg/mL) and Enterococcus faecalis ATCC 29212 (600 μg/mL).

In this study we aimed to characterize antimicrobial resistance in methicillin-resistant Staphylococcus aureus (MRSA) isolated from bloodstream infections as well as the associated genetic lineages of the isolates. Sixteen MRSA isolates were recovered from bacteremia samples from inpatients between 2016 and 2019. The antimicrobial susceptibility of these isolates was tested by the Kirby–Bauer disk diffusion method against 14 antimicrobial agents. To determine the macrolide–lincosamide–streptogramin B (MLSB) resistance phenotype of the isolates, erythromycin-resistant isolates were assessed by double-disk diffusion (D-test). The resistance and virulence genes were screened by polymerase chain reaction (PCR). All isolates were characterized by multilocus sequence typing (MLST), spa typing, staphylococcal chromosomal cassette mec (SCCmec) typing, and accessory gene regulator (agr) typing. Isolates showed resistance to cefoxitin, penicillin, ciprofloxacin, erythromycin, fusidic acid, clindamycin, and aminoglycosides, confirmed by the presence of the blaZ, ermA, ermC, mphC, msrA/B, aac(6’)-Ie-aph(2’’)-Ia, and ant(4’)-Ia genes. Three isolates were Panton–Valentine-leukocidin-positive. Most strains (n = 12) presented an inducible MLSB phenotype. The isolates were ascribed to eight spa-types (t747, t002, t020, t1084, t008, t10682, t18526, and t1370) and four MLSTs (ST22, ST5, ST105, and ST8). Overall, most (n = 12) MRSA isolates had a multidrug-resistance profile with inducible MLSB phenotypes and belonged to epidemic MRSA clones.

Aims: To determine the chemical composition and antibacterial activity of Kenyan Ganoderma lucidum. Study Design: Structural determination of the isolated compound was done using spectral evidences and in comparison with literature. The antibacterial properties of the compound was done using disc diffusion method. Place and Duration of Study: Department of Pure and Applied Chemistry, Masinde Muliro University of Science and Technology, between January and November, 2019. Methodology: Sequential extraction of dried samples of Kenyan G. lucidum were done using solvents hexane, ethyl acetate and methanol. Chromatographic separation of hexane extract of Ganoderma lucidum was done using spectroscopic data. The compound was assayed against Escherichia coli, Klebsiella pneumoniae, Methicillin–Resistant Staphylococcus aureus (MRSA), Pseudomonas aeruginosa and Streptococcus pyogenes. Standard antibiotic namely; ampicillin was used as the control. Disc diffusion method was used and zones of inhibition, after respective incubation periods, were used to quantify antibacterial activity. Results: From hexane extract of Ganoderma lucidum, Ergosta-5, 7, 22-triene-3β, 14α – diol (22Z) was isolated. Ethylacetate and methanol extracts produced a mixture of complex compounds. Ergosta-5,7,22-triene-3β,14α-diol (22Z) exhibited significant activity against Methicillin-Resistance Staphylococcus aureus (MRSA) (p=0.022) and Streptococcus pyogenes (p = 0.05). The most sensitive microbe was Streptococcus pyogenes. Conclusion: One major compound, Ergosta-5, 7, 22-triene-3β, 14α – diol (22Z) was isolated, characterized and antibacterial activity determined.

ABSTRACT Neem (Azadirachta indica A.Juss) is a plant that potentially developed for antibacterial agent for both the leaves and barks. The aims of this study were to compare the effectiveness of the antibacterial activity of neem leav es and stem barks extract and to identify the antibacterial compounds of the most active fractions. The extraction method was done using sonication method. Antibacterial activity was evaluated using wells solid diffusion method and TLC-Bioautography. Extract fractionation was conducted using liquid-liquid partitioning method. The chemical compounds of extracts and fractions were analyzed using TLC and GCMS. The result of sonication extraction obtained neem leaves oil (12,02%), leaves crude extract (4,3%) and stem barks crude extract (16,85%). The major chemical constituents of GCMS analysis are 2,3-Dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one (6,06%), L-proline,1-Acetyl-(CAS) Acetylproline (5,85%), 4-hydroxy-2-methyl-pyrrolidine-2-carboxylic acid (21,42%), 2,3-Dyhidrobenzofuran (2,69%), alpha-D-methylglucopyranoside (4,54%), palmitic acid (2,92%), Arabino-hex-1-enitol, 1,5-Anhydro-2-deoxy-(CAS) glucal (31,69%). Phytochemical screening of neem leaves oil, leaves and barks crude extract revealed the presence of alkaloids, flavonoids, phenols, saponins, triterpenoids, steroids and sterols. Antibacterial test results showed neem leaves oil was more effective than leaves and stem barks crude extract against Escherichia coli. The n-hexane fraction showed higher antibacterial activity than ethyl acetate fraction and ethanol fraction. Phytochemical screening of n-hexane fraction showed the presence of triterpenoids, steroids, sterols and phenols.   Mimba (Azadirachta indica A.Juss) merupakan tanaman yang berpotensi dikembangkan sebagai antibakteri baik bagian daun maupun kulit batang. Penelitian ini bertujuan untuk membandingkan efektivitas antibakteri ekstrak daun dan kulit batang mimba terhadap Escherichia coli dan untuk mengidentifikasi golongan senyawa potensial antibakteri pada fraksi teraktif. Pengujian aktivitas antibakteri dilakukan dengan metode difusi padat menggunakan sumuran dan KLT-Bioautografi. Fraksinasi ekstrak dilakukan dengan metode partisi. Komponen kimia ekstrak dan fraksi dianalisis menggunakan KLT dan GCMS. Hasil ekstraksi sonikasi diperoleh minyak daun (12,02%), ekstrak kasar daun (4,3%) dan ekstrak kasar kulit batang (16,85%). Skrining fitokimia menunjukkan minyak daun, ekstrak kasar daun dan ekstrak kulit batang mimba mengandung senyawa alkaloid, flavonoid, fenolik, saponin, triterpenoid, steroid dan sterol. Komponen kimia mayor hasil analisis GCMS minyak daun mimba adalah 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one (6,06%), L-proline,1-Acetyl-(CAS)Acetylproline (5,85%), 4-hydroxy-2-methyl-pyrrolidine-2-carboxylic acid (21,42%), 2,3-dyhidrobenzofuran (2,69%), Alpha-d-methylglucopyranoside (4,54%), Asam Palmitat (2,92%), Arabino-hex-1-enitol, 1,5-anhydro-2-deoxy-(CAS)glucal (31,69%). Hasil uji antibakteri menunjukkan minyak daun lebih efektif menghambat pertumbuhan Escherichia coli dibandingkan dengan ekstrak kasar daun dan kulit batang. Fraksi n-heksan menunjukkan aktivitas antibakteri paling besar dibandingkan dengan fraksi etil asetat dan etanol. Hasil skrining fitokimia fraksi n-heksan menunjukkan adanya senyawa triterpenoid, steroid, sterol dan fenolik.

ABSTRAK Kunyit (Curcuma longa L.) merupakan salah satu jenis tanaman obat yang termasuk dalam keluarga Zingiberaceae. Senyawa aktif yang terkandung dalam rimpang kunyit (Curcuma longa L.) mampu bekerja sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari ekstrak etanol rimpang kunyit (Curcuma longa L.) terhadap Staphylococcus aureus dan Pseudomonas aeruginosa. Ekstraksi dilakukan dengan cara refluks menggunakan pelarut etanol 96%. Pengujian aktivitas antibakteri dilakukan menggunakan metode difusi paper disk dengan masing-masing konsentrasi ekstrak 10%, 20%, 30%, 40% b/v. Kontrol positif yang digunakan adalah Ciprofloxacin sedangkan kontrol negatif yang digunakan adalah DMSO. Hasil skrining fitokimia ekstrak etanol rimpang kunyit mengandung alkaloid, flavonoid, fenol, tanin dan terpenoid. Berdasarkan hasil penelitian, ekstrak etanol rimpang kunyit dapat menghambat bakteri Staphylococcus aureus dan Pseudomonas aeruginosa pada konsentrasi 10%, 20%, 30%, 40% dan konsentrasi 40% merupakan konsentrasi yang memberikan diameter zona hambat terbesar terhadap kedua bakteri uji yaitu 8,63 mm dan 7,8 mm. The active compounds contained in the Curcuma longa L. rhizome can work as antibacterial. This study aims to determine the antibacterial activity of the ethanol extract of Curcuma longa L. Antibacterial activity testing was carried out using the paper disk diffusion method with each extract concentration of 10%, 20%, 30%, 40%. The positive control used was Ciprofloxacin while the negative control used was DMSO. The results of phytochemical screening of the ethanol extract of Curcuma longa L. rhizome contain alkaloids, flavonoids, phenols, tannins and terpenoids. Based on the results of the study, the ethanol extract of turmeric rhizome can inhibit Staphylococcus aureus and Pseudomonas aeruginosa bacteria at concentrations of 10%, 20%, 30%, 40% and 40% concentrations which give the largest inhibition zone diameter of the two test bacteria which is 8.63 mm and 7.8 mm. The aim of the present research work to investigate antimicrobial activity of some honey samples six winter honeys six summer honeys collected from different regions of Western Ghats. The microbes used in this study are Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa, and Proteus mirabilis. Antibacterial activity of the honeys was assayed using the Disc diffusion method. Noticeable variations in the antibacterial activity of the different honey samples were observed. Among the microbes Staphylococcus aureus is the most sensitive against all honey samples shows the maximum inhibitor zone compare to summer honeys.

 ABSTRACT The essential oil composition of the Toona sureni (Blume) Merr leaf was analyzed by GC-MS. More than 68 peaks, representing 99.99% of total oil, forty three components were identified, this represents 80.65% of the total oil component. The major components were α-terpinene (9.58%), α-copaene (8.39%), bicyclogermacrene (7.61%), δ-cadinene (6.65%), β-elemene (4.88%), germacrene-D (4.65%), δ-selinene (3.58%), caralene (3.10%), β-caryophyllene (2.88%), α-cubebene (2.82%), δ-gurjunene (2.20%), and (-)-isoledene (2.05%). The antibacterial activity of the essential oil of Toona sureni (Blume) Merr leaf was evaluated using disk diffusion method. The oil was effective on the inactivation of Escherichia coli, Staphylococcus aureus and Bacillus subtilis. Keywords: Toona sureni (Blume) Merr, antibacterial activity, GC-MS, essential oil
