Determination of total flavonoids content of Jinxianlian from different sources and its in vitro antioxidant and hypoglycemic activities

Background Salvia eigii., Salvia hierosolymitana and Salvia viridis are native to the Mediterranean region, and are used in traditional medicine for the treatment of many ailments. In the current investigation, the methanolic extracts obtained from the air dried aerial parts of S. eigii, S. hierosolymitana and S. viridis from Jordan were screened for their total phenolics content (TPC), total flavonoids content (TFC) and their in vitro antioxidant activity. Additionally, the presence of four bioactive phenolic acids including gallic acid, caffeic acid, rosmarinic acid and salvianolic acid B and other seven flavonoids including luteolin-7-O-glucoside, apigenin, apigenin-7-O-glucoside, rutin, nariginin, hesperidin and quercetin was determined using Liquid chromatography-Electron Spray Ionization-Tandom Mass Spectrometry (LC-ESI-MS/MS). Methods Antioxidant activity of the obtained three extracts were examined via the DPPH•, ABTS• + radical scavenging methods in addition to Ferrous Ion Chelating (FIC) effect. TFC and TPC of the extracts were measured using the aluminum chloride colorimetric method and the Folin-Ciocalteau method, respectively. The presence and concentration of the selected 11 compounds was further determined through LC-ESI-MS/MS. Results The results indicated that three Salvia species had high total flavonoids content expressed in mg quercetin/g dry extract (S. heirosolymitana: 770.85 ±  5.26; S. eigii: 520.60 ±  6.24, S. viridis: 311.36 ±  4.41). S. heirosolymitana had the highest DPPH• activity (0.184 ±  1.22 × 10−2 mg/ml) and FIC effect (0.354 ±  0.018 mg/ml). S. heirosolymitana had slightly higher ABTS• + scavenging activity than S. eigii (0.176 ±  1.16 × 10−2 mg/ml; 0.183 ±  0.031 mg/ml, respectively). All 11 compounds were detected in the extracts of the three Salvia species. Luteolin-7-O-glucoside was detected in high concentration levels in the three species (1756.73, 21651.36, and 26125.14 mg/kg dry plant; S. eigii, S. hierosolyimitana and S. viridis, respectively), yet rosmarinic acid had the highest contribution to both S. hierosolymitana (27124.93 mg/kg) and S. eigii (15783.33 mg/kg). Notably, S. hierosolymitana and S. viridis contained salvianolic acid B (896.11; 890.9 mg/kg). Conclusions The three Salvia species exhibited good antioxidant activity, especially S. heirosolymitana due to its high TPC, TFC, and the presence of high concentration levels of romarinic acid and other phenolic acids and flavonoids. This is the first phytochemical and antioxidant evaluation of S. eigii, S. hierosolymitana and S. viridis from Jordan. Prior to this investigation, no phytochemical investigation on S. eigii was reported.

In the last decades, it has been intensively studied natural alternatives to synthetic products. Chassalia kolly is one of the medicinal plants used traditionally for treatment of skin infections. This study aimed at identifying the diversity secondary metabolites, to investigate the antioxidant and anti-inflammatory activities of ethanolic extract of Chassalia kolly leaves. Total phenols were determined by Folin Ciocalteu method. The aluminum trichloride method has been used to quantify total flavonoids, while the determination of condensed tannins was carried out by the hydrochloric vanillin method. The antioxidant capacity was evaluated by 2, 2-diphenyl-l-picrylhydrazyl (DPPH), 2, 2'-Azino-bis- (3-ethylbenzothiazolin-6-sulfonic acid) (ABTS) and phosphomolybdenum methods. In vitro anti-inflammatory activity of ethanolic extract of Chassalia kolly leaves was evaluated by method membrane stabilization. In this results, Chassalia kolly leaves contain flavonoid, mucilages, anthocyans, sterols and terpenes, saponosides, alkaloids and reducing compounds. The contents of total phenols, total flavonoids and total tannins are respectively (20.41±2.11) µgGAE/mgDM; (30.29±2.18) µgQE/mgDM and (1.211±0.122)µgCE/mgDM. The results obtained during this work allowed us to assert that all extracts of the studied plant have very good antioxidant properties (IC50=0.05µg/µL). The ethanolic extract of the leaves of Chassalia kolly showed an anti-inflammatory activity more interesting than aspirin which is the reference compound used in this study.

A rose hip is the fruit of a rose plant and mostly belongs to Rosa canina and Rosa dumalis. These species are genuine found as wild in natural conditions, while in some countries are cultivated for their hips that are picked and used in a wide variety of preparations. Because they contain a variety of antioxidants, carotenoids, flavonoids, polyphenols, leucoanthocyanins and catechins, rose hips are considered to be a good cancer preventative. The Rosa genus has been the focus of many recent studies due to its potential benefic effects on treatment and prevention of several diseases. However, there are few in vitro studies concerning its composition and antioxidant capacity. The aim of the present study was to determine bioactive content, including ascorbic acid, total phenolics, total flavonoids, total carotenoids and antioxidant activity of Rosa dumalis genotypes growing wild in Erzurum province located eastern Anatolia. The results revealed that fruits of Rosa dumalis different genotypes were rich rich in terms of vitamin C, which ranged from 402 to 511 mg/100 g fresh weight base. The total phenolic content varied from 297 to 403 mg/100 g fresh weight. The genotype 'E-09' had the highest total flavonoids content (229 mg quercetin equivalent/100 g fresh weight) and the lowest value was attributed to genotype 'E-04' with 143 mg/ quercetin equivalent/100 g fresh weight. The antioxidant activity of the genotypes was between 12.9-28.6 μg Trolox/ml samples. The results revealed that there was enough diversity among Rosa dumalis genotypes for bioactive content and promising genotypes, with high bioactive content, were determined, which can become study material for future breeding activities.

Artemia or brine shrimp is a crustacean animal that lives in conditions of high salinity in the tropical and sub-tropical regions of the world. There is great potential for the culture and production of artemia in Iran. Artemia biomass is a good source of protein that can be used in poultry diets (Zarei et al., 2006). The objective of this study was to determine the protein digestibility of different sources of artemia in vitro and in vivo in comparison with fish meal.

Infectious diseases are the second leading cause for worldwide death. Treatment of infections continues to be difficult in modern time because of the severe side effects of some drugs and the growing resistance to antibacterial agents. Over the past few decades the use of antibiotics is under threat as many commonly used antibiotics have become less effective against certain illnesses due to emergence of multi drug-resistant bacteria. In the present study the effects of 3 types of solvents, chloroform, ethyl acetate and methanol were investigated to determine the presence of various phytochemical constituent, total phenolic content, total flavonoids content and in vitro antimicrobial activity from rhizomes of Iris kashmiriana (Kashmir Iris) and Iris ensata (Japanese Iris), belong to family Iridaceae. The reason for selecting in vitro method was to minimize the usage of experimental animals. The antimicrobial activity of chloroform, ethyl acetate and methanol extract of rhizomes of Iris kashmiriana and Iris ensata were evaluated on bacterial strains of Bacillus cereus, Pseudomonas auregenosa, Proteus vulgaris and Eschirichia coli and fungal strains of Candida albicans and Aspergillus niger by agar well diffusion method. The preliminary phytochemical studies and quantitative analysis of alkaloids, phenol and flavonoids were performed by well reported method. These extracts were further subjected to TLC (Thin layer chromatography analysis).  The chemical contents of the Iris kashmiriana and Iris ensata were presented as total phenolic content and total flavonoids content. Phytochemical screening of the extract showed the presence of some common compounds like phenols, terpenoids, flavonoids, carbohydrate etc. The antimicrobial potential of the plant extract was evaluated against different bacterial species which shows significant inhibitory action against all the tested bacterial and fungal strain. Methanolic extract was found to be more active than chloroform and ethyl acetate extracts. It reveals that the methanol soluble components of the plant are highly active against the above mentioned microorganism. Establishment of in vitro cultures from Astragalus glycyphyllos, determination of biomass and analysis of total flavonoids, rutin and camelliaside A were performed. To increase flavonoid production various combinations of plant hormones and light/dark regimen were investigated. Suspension cultures with exogenous quercetin were evaluated for possible increase in flavonoid production. Shoots, calli and suspensions were successfully established. Rutin and camelliaside A were proved in highest amount in shoots. Calli, cultivated on modified G48 medium, with double amount of Ca2+ and Mg2+, achieved higher total flavonoid content (2.37 and 2.03 mg/g DW). Suspensions cultures, cultivated on modified G48 medium with 10, 20 and 30 mg/mL quercetin achieved higher total flavonoid content (0.09, 0.10 and 0.13 mg/mg DW). Biotransformation of quercetin to isoquercitrin was achieved. The highest concentration of isoquercitrin (56.73 ng/mg DW) was observed on suspensions cultures cultivated on modified G48 medium, with 20 mg/mL quercetin.

Black mulberry leaves teas (BMLTs) were prepared using boiled water and different steeping time (5 10, 20, 35 and 45 minutes). In order to establish the connection between steeping time and tea quality, total phenolics content (TPC), total flavonoids content TFC contents were measured, as well as two antioxidant assays (DPPH and reducing power assays), alone with antimicrobial and cytotoxic tests. The obtained results showed that TPC, TFC, IC50 and EC50 values increased with the increase in steeping time, while antimicrobial and cytotoxic activity exhibited different tendency. Based on the obtained results, the 10-minute steeping time was the optimum for tea preparation and this tea was used for the determination of polyphenolic profile using HPLC-MS technique. The results showed that the main compounds in BMLT were chlorogenic and caffeic acids with the contents of 7226.00 and 537.52 ?g/g, respectively.

Antioxidants are substances that hamper or inhibit the oxidation of oxidizable substrates in the cells. The aerial parts of Blepharis maderaspatensis dry powder was extracted with various solvents (PE, EA and methanol) through Soxhlet extractor. The aerial parts of different concentrates (Pet.ether, ethyl acetate and methanol) of Blepharis maderaspatensis was evaluated for its in-vitro antioxidant potential by hydroxyl radical, total antioxidant activity taking ascorbate used as standard for the both methods and total flavonoids content was estimated as equivalent to rutin. The methanolic concentrates of Blepharis maderaspatensis & standard exhibited antioxidant potential possessing IC50 210µg/ml & 62µg/ml (hydroxyl radical) 200µg/ml & 57µg/ml (Total antioxidant activity) respectively. An IC50 value was originate that methanolic concentrates of Blepharis maderaspatensis more efficient in hydroxyl radical, total antioxidant activity compared EA & PE concentrates. The methanolic and EA concentrates of Blepharis maderaspatensis showed the total flavonoids content (11.230±0.22 and 2.858±0.56 mg of rutin equivalent/g of powder) respectively. The difference in scavenging potential of the extracts can be due to variation in the percentage of bioactive compound flavonoids present in methanolic extracts. Invitro antioxidant studies obviously show methanolic concentrates of Blepharis maderaspatensis have better antioxidant activity due to the presence of total flavonoids content. The findings from the study reveal that the antioxidative potentials of methanolic concentrates Blepharis maderaspatensis could serve as free radical inhibitors, possibly acting as natural antioxidants.

Cinnamon plants (Cinnamomum burmani Blume), including plants that have medicinal properties for diabetes mellitus because they contain flavonoid compounds. This study aimed to determine the total flavonoids content of the ethanol extract of cinnamon leaf ethanol extract using UV-Vis spectrophotometry method. The ethanol extract of Cinnamon leaf was obtained by maceration using ethanol as a solvent.The qualitative analysis used FeCl3by forming yellowish green complex. The determination of total flavonoids content is conducted basedon AlCl3method with total flavonoidse xpressed in QE (Quercetin equivalent) at the maximum wavelength of 435 nm. The result showed that the average content of flavonoid total is 19.544 mg QE/g extract.
