Aim. The aim was to estimate the efficiency of cell-containing and non-cellular dermal coverages in animal model in vivo. Methods. We carried out the application of gel wound coverages with three different cell lines and cultural medium conditioned by these cells on the third degree burns of ICR line mice. In the control group animals were treated with fresh medium-containing gel. Photo fixation of burn wound status was carried out once a day. The results were estimated by ANOVA approach. Results. There was a statistically significant difference of burn wounds development and healing within three experimental groups treated with cell suspensions of different lines. Cell-free gels with media conditioned by these cells were shown to be effective as well. Conclusions. It has been shown the dependence of wound healing properties of coatings containing cells from the origin of these cells. The effectiveness of these coverages is supposed to be intermediated by biologically active substances secreted by the cells.Keywords: burn wound, dermal equivalent, stem cells, skin equivalent, skin substitute, tissue engineering.

Aim. The aim was to estimate the influence of cell suspension quality on the therapeutic efficiency of cell-containing dermal coverages in animal model in vivo. Methods. We carried out the application of gel wound coverages with different quality of cellular compound on the third degree burns of ICR line mice. In the negative control group animals were treated with fresh medium-containing gel, and in positive control – by gel containing high quality cell suspension. Photo fixation of burn wound status was carried out once a day. The results were estimated by ANOVA approach. Results. There was a statistically significant difference of burn wounds development and healing between positive control and both experimental groups. The decreasing of alive cell fraction in prepared gel within 5 hours has been shown as well. Conclusions. It has been shown the dependence of wound healing properties of coatings containing cells on the cell compound quality, in particular, from the viability and integrity of cells. These results are important for developing of clinical protocols using such cell‑containing dermal equivalents. AbstractThe purpose of this study was to evaluate whether cryopreserved (frozen) adipose-derived regenerative cells (ADRCs) have a therapeutic effect on burn wound healing as well as freshly isolated (fresh) ADRCs.Full thickness burns were created on dorsum of nude mice and burn wound was excised. The wound was covered by artificial dermis with; (i) fresh ADRCs, (ii) frozen ADRCs, and (iii) PBS (control). The assessment for wound healing was performed by morphological, histopathological and immunohistochemical analyses.In vivo analyses exhibited the significant therapeutic effect of frozen ADRCs on burn wound healing up to the similar or higher level of fresh ADRCs. There were significant differences of wound closure, epithelized tissue thickness, and neovascularization between the treatment groups and control group. Although there was no significant difference of therapeutic efficacy between fresh ADRC group and frozen ADRC group, frozen ADRCs improved burn wound healing process in dermal regeneration with increased great type I collagen synthesis compared with fresh ADRCs.These findings indicate that frozen ADRCs allow us to apply not only quickly but also for multiple times, and the cryopreserved ADRCs could therefore be useful for the treatment of burn wounds in clinical settings.

Background. For centuries, medicinal plants have been extensively used in wound healing of burn injuries. The aim of this study is to analyze comparatively the effects of curcumin and Hypericum perforatum (HP) on second-degree burn wounds in rats. Materials and Methods. This experimental study was conducted on 24 male Sprague-Dawley rats with second-degree burns. The animals were randomly divided into three groups. The burns were treated with curcumin (Group B) and Hypericum perforatum (Group C) with daily application. Group A was considered as the control group and received no medication. Histological parameters, reepithelization, granulation tissue formation, inflammation, and angiogenesis were assessed after the scar biopsy at the end of research. Results. All histological parameters of the control group showed statistically significant difference than the other groups (p<0.05). There was no statistically significant difference between Groups B and C in terms of reepithelization and inflammation (p=0.351, p=0.067). In the curcumin group compared with the HP group, a significant difference was observed for two parameters of fibrosis and angiogenesis (p<0.05). Conclusion. Both curcumin and Hypericum perforatum oil are effective in burn wound healing. Our findings showed a better quality of healing in curcumin-treated rats.

Для интенсификации производства животноводческой продукции и повышения ее качества необходимо обогащать рацион сельскохозяйственных животных и птицы кормовыми добавками, содержащими комплекс биологически активных веществ. Исследованы образцы комплексной кормовой добавки, содержащей масляную композицию БАВ в-каротина, витаминов Е и С, жидкого рапсового лецитина и селена, и пробиотическую добавку (ПД) Целлобактерин в соотношениях соответственно от 1 : 1 до 1 : 5. Установлено, что при соотношении масляная композиция БАВ : ПД Целлобактерин 1 : 3 количество пробиотической культуры Enterococcus faecium 1-35, содержащейся в ПД Целлобактерин, в опытном образце достигает максимального значения 6,4 Ч 105 КОЕ/г. При этом обеспечивается равномерное распределение масляной композиции БАВ на поверхности ПД. В опытах in vivo определена эффективность применения комплексной добавки с установленным соотношением компонентов для нормализации обменных процессов в организме животных. Доказано, что введение в рацион лабораторных крыс опытной группы многокомпонентной кормовой добавки в количестве 3 к основному рациону (ОР), контаминированному микотоксинами, позволяет достичь опережения прироста массы тела на 15-е и 45-е сутки эксперимента на 6,45 и 33,05 соответственно снижения содержания мочевины в сыворотке крови на 58,43 по сравнению с аналогичными показателями у животных контрольной группы, получавших только ОР. Степень снижения аланинаминотрасферазы и аспартатаминотрансферазы по отношению к аналогичным показателям контрольной группы составила 18,53 и 29,89 соответственно. Следовательно, комплексная кормовая добавка при соотношении масляная композиция БАВ : ПД Целлобактерин 1 : 3 обладает ярко выраженными гепатопротекторными и антитоксическими свойствами. Enriching the diet of farm animals and poultry by feed additives containing a complex of biologically active substances is necessary to intensify the production of livestock products and improve their quality. Samples of a complex feed additive containing an oil composition of BAS в-carotene, vitamins E and C, liquid rapeseed lecithin and selenium, and a probiotic additive (PA) Cellobacterin in ratios from 1 : 1 to 1 : 5 respectively were studied. It was found that the ratio of the oil composition of BAS : PA Cellobacterin 1 : 3 respectively the amount of probiotic culture Enterococcus faecium 1-35 contained in PA Cellobacterin in the experimental sample reaches a maximum value 6,4 Ч 105 CFU/g. In this case, the distribution of BAS oil composition on the PA surface is uniform. The effectiveness of using a complex additive with a set ratio of components to normalize metabolic processes in animals was determined in in vivo experiments. It is established that introduction in a diet of laboratory rats of the experimental group multicomponent feed additive in an amount of 3 to the basic ration (BR), contaminated with mycotoxins, can achieve compared to the same indicators in animals of the control group who received only the BR, the timing of weight gain on 15-th and 45-th day of the experiment by 6,45 and 33,05 respectively reduction of urea content in blood serum at 58,43. The degree of reduction of alanine aminotransferase and aspartate aminotransferase in relation to similar indicators of the control group was 18,53 and 29,89 respectively. Therefore, a complex feed additive with the ratio of oil composition of BAS : PA Cellobacterin 1 : 3 has pronounced hepatoprotective and antitoxic properties.

Background: Uncemented implants are still associated with several major challenges, especially with regard to their manufacturing and their osseointegration. In this study, a novel manufacturing technique—an optimized form of precision casting—and a novel surface modification to promote osseointegration—calcium and phosphorus ion implantation into the implant surface—were tested in vivo. Methods: Cylindrical Ti6Al4V implants were inserted bilaterally into the tibia of 110 rats. We compared two generations of cast Ti6Al4V implants (CAST 1st GEN, n = 22, and CAST 2nd GEN, n = 22) as well as cast 2nd GEN Ti6Al4V implants with calcium (CAST + CA, n = 22) and phosphorus (CAST + P, n = 22) ion implantation to standard machined Ti6Al4V implants (control, n = 22). After 4 and 12 weeks, maximal pull-out force and bone-to-implant contact rate (BIC) were measured and compared between all five groups. Results: There was no significant difference between all five groups after 4 weeks or 12 weeks with regard to pull-out force (p > 0.05, Kruskal Wallis test). Histomorphometric analysis showed no significant difference of BIC after 4 weeks (p > 0.05, Kruskal–Wallis test), whereas there was a trend towards a higher BIC in the CAST + P group (54.8% ± 15.2%), especially compared to the control group (38.6% ± 12.8%) after 12 weeks (p = 0.053, Kruskal–Wallis test). Conclusion: In this study, we found no indication of inferiority of Ti6Al4V implants cast with the optimized centrifugal precision casting technique of the second generation compared to standard Ti6Al4V implants. As the employed manufacturing process holds considerable economic potential, mainly due to a significantly decreased material demand per implant by casting near net-shape instead of milling away most of the starting ingot, its application in manufacturing uncemented implants seems promising. However, no significant advantages of calcium or phosphorus ion implantation could be observed in this study. Due to the promising results of ion implantation in previous in vitro and in vivo studies, further in vivo studies with different ion implantation conditions should be considered.

ABSTRACT Environmental conditions affect bacterial behavior and can greatly influence the course of an infection. However, the environmental cues that elicit bacterial responses in specific infection sites are relatively unknown. Pseudomonas aeruginosa is ubiquitous in nature and typically innocuous. However, it is also one of the most prevalent causes of fatal sepsis in burn wound patients. The aim of this study was to determine the impact of environmental factors, specifically the availability of arginine, on the pathogenesis of P. aeruginosa in burn wound infections. Comparison of burned versus noninjured tissue revealed that l-arginine (l-Arg) was significantly depleted in burn wounds as a consequence of elevated arginase produced by myeloid-derived suppressor cells. We also observed that l-Arg was a potent chemoattractant for P. aeruginosa, and while low concentrations of l-Arg increased P. aeruginosa’s swimming motility, high concentrations resulted in diminished swimming. Based on these observations, we tested whether the administration of exogenous l-Arg into the burn wound could attenuate the virulence of P. aeruginosa in thermally injured mice. Administration of l-Arg resulted in decreased P. aeruginosa spread and sepsis and increased animal survival. Taken together, these data demonstrate that the availability of environmental arginine greatly influences the virulence of P. aeruginosa in vivo and may represent a promising phenotype-modulating tool for future therapeutic avenues. IMPORTANCE Despite our growing understanding of the pathophysiology of burn wounds and the evolution of techniques and practices to manage infections, sepsis remains a significant medical concern for burn patients. P. aeruginosa continues to be a leader among all causes of bacteremic infections due to its tendency to cause complications in immunocompromised patients and its ubiquitous presence in the hospital setting. With the unforgiving emergence of multidrug-resistant strains, it is critical that alternative strategies to control or prevent septic infections in burn patients be developed in parallel with novel antimicrobial agents. In this study, we observed that administration of l-Arg significantly reduced bacterial spread and sepsis in burned mice infected with P. aeruginosa. Given the safety of l-Arg in high doses and its potential wound-healing benefits, this conditionally essential amino acid may represent a useful tool to modulate bacterial behavior in vivo and prevent sepsis in burn patients. Objectives: Biopure MTAD™, a new root canal irrigant has shown promising results against the most common resistant microorganism, E. faecalis, in permanent teeth. However, there is lack of studies comparing its antimicrobial effectiveness with NaOCl in primary teeth. The purpose of this study was to compare the in vivo antimicrobial efficacy of NaOCl 2.5% and Biopure MTAD™ against E. faecalis in primary teeth. Study design: Forty non vital single rooted primary maxillary anterior teeth of children aged 4-8 years, were irrigated either with NaOCl 2.5% (n=15), Biopure MTAD™ (n=15) and 0.9% Saline (n=10, control group). Paper point samples were collected at baseline (S1) and after chemomechanical preparation (S2) during the pulpectomy procedure. The presence of E. faecalis in S1 & S2 was evaluated using Real time Polymerase Chain Reaction. Results: Statistical significant difference was found in the antimicrobial efficacy of NaOCl 2.5 % and BioPure MTAD™ when compared to saline (p&gt;0.05). However, no statistical significant difference was found between the efficacies of both the irrigants. Conclusions: NaOCl 2.5% and BioPure MTAD™, both irrigants are equally efficient against E. faecalis in necrotic primary anterior teeth. MTAD is a promising irrigant, however clinical studies are required to establish it as ideal root canal irrigant in clinical practice.

Abstract Introduction Thermal burns account for 5–10% of casualties sustained in present-day conflicts and are expected to be one of the most common wounds to occur in future conflicts. In prolonged field care (PFC) situations, medical evacuation could be delayed for days. During this time, burn wounds can become infected, detrimentally impact neighboring tissue, and cause systemic immune responses. Therefore, it is essential to test and evaluate non-surgical debridement agents that could be implemented prior to reaching a Role 3 military treatment facility. This work details how the proprietary proteolytic gel SN514 impacts burn debridement when applied within a PFC-like timeline. SN514 contains an enzyme formulation that is thermostable, easy to apply, and selectively degrades non-viable tissue in vitro and in vivo. Methods Deep-partial thickness contact burns were created using an established porcine model and covered with gauze or an antimicrobial incise drape. Four days later, the burns were treated with one of five treatments: 0.2% SN514, 0.8% SN514, a vehicle control, gauze, or an antimicrobial silver dressing. Treatments were re-applied every 24 hours for 72 to 96 hours. The effects of the treatment regiments were compared histologically. Biopsies were also taken to monitor bacterial contamination levels. Results Burns treated with SN514 were partially debrided and visually distinct from those treated with gauze, the silver dressing, or the vehicle control. Preliminary analyses suggest that SN514-treated burns that had been covered with “dry” gauze had a much lower debridement efficiency than those treated with the incise drape. This suggests that SN514 debridement efficiency may depend on the presence of a moist eschar. Preliminary analyses also suggest that there was little difference in burn wound bacterial counts among the five treatment groups. Conclusions SN514 is able to debride burns that experienced delayed treatment, without any evidence of harm to the surrounding tissue or evidence of exacerbating the original burn injury. SN514-treated wounds displayed little to no blood loss and did not increase burn wound infection levels compared to wounds treated with gauze or an antimicrobial silver dressing.

Abstract Introduction Burn is one of the most severe traumas that causes coagulative destruction of the skin. The use of various products that accelerate wound healing in patients with burn may affect the patient’s survival and reduce the complications that may be seen. In the present study we aimed effects of subcutaneous ozone injection on second degree burn wound. Methods A total of 72 Sprague-Dawley male rats included in the study were divided randomly into three groups (control group (CG), silver sulfadiazine group (SG), ozone group (OG)) and each group was divided randomly two subgroups (as sacrificed on d7 and on d14).A deep second degree scald burns were created on the lower back. In CG subcutaneous 0.9% serum saline was injected daily into the burn area. In SG, burns were dressed with silver sulfadiazine daily and in OG subcutaneous ozone was injected daily into the burn area. Tissue hydroxyproline level measurement and histopathological evaluation were done. Results When the groups were compared in terms of weight change, no significant difference was found on the 7th and 14th days. In the evaluation made in terms of tissue hydroxyproline, tissue hydroxyproline level in OG was found to be significantly higher on both the 7th and 14th days (p &lt; 0.001). In histopathological evaluations, it was determined that wound healing in OG was significantly higher than in the other groups. Conclusions According to the results, subcutaneous ozone therapy is more effective than silver sulphadiazine in the healing process of second-degree burn wounds and it can be safely used in the treatment of burn wounds.

ABSTRACT This study demonstrated the capacity of bacteriocin-producing lactic acid bacteria (LAB) to reduce intestinal colonization by vancomycin-resistant enterococci (VRE) in a mouse model. Lactococcus lactis MM19 and Pediococcus acidilactici MM33 are bacteriocin producers isolated from human feces. The bacteriocin secreted by P. acidilactici is identical to pediocin PA-1/AcH, while PCR analysis demonstrated that L. lactis harbors the nisin Z gene. LAB were acid and bile tolerant when assayed under simulated gastrointestinal conditions. A well diffusion assay using supernatants from LAB demonstrated strong activity against a clinical isolate of VRE. A first in vivo study was done using C57BL/6 mice that received daily intragastric doses of L. lactis MM19, P. acidilactici MM33, P. acidilactici MM33A (a pediocin mutant that had lost its ability to produce pediocin), or phosphate-buffered saline (PBS) for 18 days. This study showed that L. lactis and P. acidilactici MM33A increased the concentrations of total LAB and anaerobes while P. acidilactici MM33 decreased the Enterobacteriaceae populations. A second in vivo study was done using VRE-colonized mice that received the same inocula as those in the previous study for 16 days. In L. lactis-fed mice, fecal VRE levels 1.73 and 2.50 log10 CFU/g lower than those in the PBS group were observed at 1 and 3 days postinfection. In the P. acidilactici MM33-fed mice, no reduction was observed at 1 day postinfection but a reduction of 1.85 log10 CFU/g was measured at 3 days postinfection. Levels of VRE in both groups of mice treated with bacteriocin-producing LAB were undetectable at 6 days postinfection. No significant difference in mice fed the pediocin-negative strain compared to the control group was observed. This is the first demonstration that human L. lactis and P. acidilactici nisin- and pediocin-producing strains can reduce VRE intestinal colonization.
