Aim. Testing the haploproduction ability of 30 hybrids of winter soft wheat. Methods. In vitro culture of isolated anthers of wheat. The percentage of callus and regeneration of green plants for each genotype calculated as a percentage of the planted anthers. Results. The differences in the frequency of callus induction and the ability to regenerate plants in the process of androgenesiss in vitro of winter soft wheat were detected. The microspores of 17 of 30 hybrids formed callus by in vitro anther culture were shown. The intensity of one process was different: more than half of the genotypes (18 pcs.) were characterized by a low percentage of callus (from 0.10 to 1.0%), 6 genotypes - medium (from 1.0 to 3.0%), and three - high (4.36%; 15.11% and 15.81%, respectively). Conclusions. Genotype-specific of microspores morphogenetic reactions of soft winter wheat in the process of androgenesis in vitro were revealed Samples P26 and P27 showed the highest level of callus formation. The percentage of callus and regeneration of green plants for each genotype calculated as a percentage of the planted anthers. Results. The differences in the frequency of сallus induction and the ability to regenerate plants in the process of androgenesiss in vitro of winter soft wheat were detected. The range of variation haploproduction activiti was broad. The sign of "the formation of сallus" for varieties was in limited from 0 to 21.2 % and for hybrids – from 0 to 38.8 %; on the sign of "regeneration of green plants" for varieties – from 0 to 9.4 %, and for hybrids from 0 to 9.1 %. The positive effect of 1BL/1RS translocation on signs of haploproduction in anther culture Triticum aestivum L. Conclusions. Genotype-specific of microspores morphogenetic reactions of soft winter wheat in the process of androgenesis in vitro were revealed. It has been found that F1 hybrids have a higher potential production potential in anther culture than varieties. During the period from 2011 to 2017, the 580 lines of winter wheat were transferred to the breeding departments. Keywords: soft winter wheat, anther culture in vitro, callus, regeneration.

Aim. Study the the efficiency of androgenesis in vitro in anther culture of different genotyps winter wheat. Methods. Obtaining of soft winter wheat double haploid lines by anther culture in vitro. The statistical methods. Results The ability to androgenesis in an in vitro anthers culture of 30 soft winter wheat genotypes was tested. The differences in the frequency of callus induction and the ability to regenerate plants in the process of androgenesiss in vitro of winter soft wheat were detected. The range of variation haploproduction activiti (from % of implanted anthers) was broad. The sign of "the formation of callus" was in limited from 0 to 13.2 % and on the sign of "regeneration of green plants" – from 0 to 1.8 %. The 126 green plants-regenerants were received. Conclusions. Genotype-specific of microspores morphogenetic reactions of soft winter wheat in the process of androgenesis in vitro were revealed. It is advisable to use the anthers from the F2 hybrid population plant as a donor material to create an effective technology for producing double haploids of soft winter wheat was shown. To estimate the level of in vitro haploproduction androgenic ability of different generations populations (F1, F2) soft winter wheat hybrids of the same combination to maximize the use of biotechnology cereals. Methods. Obtaining of bread wheat double haploid lines by anther culture in vitro. The statistical methods. Results. The influence different generation of wheat hybrids on the processes of induction and regeneration in anther culture of winter bread wheat were studied. The 21 green plants-regenerants were received. Conclusions. Aims. To study the effect of chemically modified starch D–5aM in the culture medium on the efficiency of androgenesis in vitro in anther culture of rice. Methods. Obtaining of rice double haploid lines by anther culture in vitro. The statistical methods. Results The influence different variants of gellatyne source in culture medium on the processes of induction and regeneration in anther culture of rice were studied. The 119 green plants-regenerants were received. Conclusions. The negative effect on the formation of green regenerants using a gel-forming components of the chemically modified starch D–5aM was shown. The callus-promoting activity of most commonly known as well as some rarely tested auxins was compared with that of 2,4-dichlorophenoxyacetic acid (2,4-D) for in vitro culture of the excised embryo of spring and winter wheat (Triticum aestivum L.), cv. Chinese Spring and cv. Fredrick. Different auxins in a concentration range from 1 to 50 μM showed widely different activities. Also the two wheat cultivars responded differently to the auxins. When rapid callus formation with limited root growth was used as the basis for comparison, 2-(2-methyl-4-chlorophenoxy)propionic acid (2-MCPP), α-naphthaleneacetic acid, 3,6-dichloro-2-methoxybenzoic acid (dicamba), 4-amino-3,5,6,trichloropicolinic acid (picloram), γ-(2,4-dichlorophenoxy)butyric acid, 2,4,5-trichlorophenoxyacetic acid, and 2,4,5-trichlorophenoxypropionic acid, in the order of effectiveness, were superior to 2,4,-D for callus induction from the embryo of 'Chinese Spring,' although the concentration required was higher than that of 2,4-D. For the winter wheat 'Fredrick,' however, only picloram, dicamba, and 2-MCPP performed as well as 2,4-D. All auxins tested promoted shoot growth; 2-methyl-4-chlorophenoxypropionic acid was most effective for 'Chinese Spring,' whereas picloram was most effective for 'Fredrick.'

Winter soft wheat is one of the most important crops. New varieties of wheat should be characterized by a set of valuable economic characteristics, but the most relevant is the creation of varieties with high yields and grain quality, resistant to a complex of diseases and other negative factors. The aim of the study was to evaluate new promising varieties of winter soft wheat breeding of the NSC "Institute of Agriculture NAAS" for grain yield, winter hardiness, disease resistance and grain quality. Experimental studies were conducted at the NSC "Institute of Agriculture NAAS" in 2016-2020, research fields are located in the Fastiv district of Kyiv region. Different weather conditions over the years of research have contributed to a better and more comprehensive assessment of varieties on a set of valuable economic characteristics. During field experiments, the generally accepted technology of growing winter wheat was used. The area of the competitive variety testing site was 20.0 m2, repeated four times. Field, measuring and weighing, laboratory and mathematical and statistical methods were used during the research. In 2018, new varieties of soft winter wheat Krasunia Poliska, Mokosha, Pyriatynka, Fortetsia Poliska, Efektna and in 2020 the variety Zemlerob were transferred to the Ukrainian Institute of Plant Variety Examination for qualification examination. Varieties created in NSC "Institute of Agriculture NAAS" by the method of hybridization using as parent components varieties and lines of local and foreign breeding. The variety Krasunia Poliska, belongs to the Lutescens type, is characterized by high drought resistance, resistance to lodging, germination on stumps, grain shedding. The Mokosha variety, belongs to the Lutescens type, has high drought resistance, is resistant to lodging, germination on stumps and grain shedding. Variety Pyriatynka, belongs to the Lutescens type, has high drought resistance, resistance to lodging, germination on the stump, shedding. Fortetsia Poliska, belongs to the Alborbrum type, is characterized by high drought resistance, resistance to lodging, germination on the stump and shedding of grain. Variety Efektna, belongs to the Erythrospermum type, is short (plant height 64-80 cm), characterized by high drought resistance, resistance to lodging, germination on the stump, shedding. The variety of winter soft wheat Zemlerob, belongs to the Lutescens type, has high drought resistance, resistant to lodging, germination on the stump, shedding of grain. The highest grain yield on average over the years of testing was obtained in the variety Fortetsia Poliska - 7.31 t / ha, varieties Pyriyatynka and Zemlerob with indicators of 7.10 and 7.06 t / ha, respectively, were distinguished by high yields. The yields of Mokosha, Effektna and Krasunia Poliska ranged from 6.27 to 6.95 t / ha. According to the increased winter hardiness, the best varieties are Pyriatynka, Krasunia Poliska, Mokosha and Efektna with a score of 8.0-8.2 points. In the varieties Fortetsia Poliska and Zemlerob the score for winter hardiness was 7.8-7.9 points. Estimation of powdery mildew showed that the maximum percentage of lesions on average over the years of research was found in the variety Mokosha (25.5%). As the most stable selected varieties Efektna, Lisova pisnia, Pyriatynka and Fortetsia Poliska (1.7-11.6%). In terms of resistance to brown rust, all varieties showed high resistance, the damage did not exceed 8.0%. The most stable variety was Efektna (0.3%). The greatest damage to winter wheat varieties was found in leaf septoria, which ranged from 19.0 to 39.3%. The greatest resistance to this disease is determined in the cultivar Zemlerob. New wheat varieties should also be characterized by high levels of protein, gluten and other valuable traits that ensure grain quality. The highest protein content was determined in the grains of the varieties Efektna (11.67%), Mokosha (11.62%) and Krasuni Poliska (11.54%). According to the increased content of gluten, the varieties Krasunia Poliska, Lisova pisnia, Mokosha (19.22-19.66%) were distinguished. According to the highest indicators of sedimentation, the varieties Krasunia Poliska, Pyriatynka and Mokosha (34.50-34.84 %) should be noted, these varieties are also the best in terms of a comprehensive assessment of grain quality. New varieties of soft winter wheat Krasunia Poliska, Mokosha, Pyryatynka, Fortetsia Poliska, Efektna and Zemlerob breedibg of NSC "Institute of Agriculture NAAS" are characterized by high grain yield (up to 7.31 t / ha), increased winter hardiness and disease resistance, as well as good indicators of grain quality: protein content - up to 11.67%; gluten - up to 19.66%. The introduction of these varieties into production will allow to obtain high yields of quality grain.

The production of the double haploid plant in vitro through anther culture technique is a plant breeding technique used to obtain pure strain rapidly. A variety of pretreatment has been reported to induce callus and regenerate planlets efficiently. This study aims at describing the influence of cold anther pretreatment towards the callus formation of curly red chili pepper (Capsicum annuum L.). This research was conducted in the laboratory of Genetics and Breeding, Faculty of Agriculture and Animal Science, Universtas Islam Negeri Sultan Syarif Kasim Riau. The explants used are anther of local genotype of curly red chili pepper. The anthers are stored at low temperatures (4 °c) with different time intervals of 0, 24, 48 and 72 hours. The results showed that the percentage of highest callus formation was obtained at 24 and 72 hours length storage, ie 50%. Cold pretreatment of 72 hours anther storage results in a faster callus with a percentage of the highest yellowish white callus color of 17.65% and a compact structure. The cold pretreatment with 72 hours anther storage is the most optimal acceleration in the development stage of anther culture and induces te formation of curly red chili pepper (Capsicum annuum L.) local genotypes.

Calli cultures of stems and leaves explants excised from field -grown rubber, Ficus elastica Decora, plants were formed on agar-solidified Murashige and Skoog (MS) medium.The results proved that MS medium supplemented with 1.0 mg L-1 benzyl adenin (BA) and 0.8mg L-1 2,4-dichloro-phenoxy acetic acid (2,4-D) was suitable to stimulate stem’s callus at ratio 87.5% . Whereas supplementation of MS medium with 0.5 mg L-1 of both BA and Indole -3-butyric acid (IBA)encouraged callus formation to reach76.6%. Leaves showed responses for callus initiation up to 75% at the same media. Stem calli showed limited ability to regenerate shoots on both agar solidified MS medium containing 1.0 mg L-1 2,4-D and kinetin (kin) 0.5 mg L-1 and MS medium supplemented with 0.5 mg L-1 of both BA and IBA. Transferring of shoots to differentiation medium (MS+0.5 mg L-1 BA+ 0.1 mg L-1 IBA) stimulated the growth and elongation of these shoots. Shoots were transferred to agar-solidified MS medium free from growth regulators failed to form roots. Because of the less number of shoots, other rooting media were not tested. The data showed, that shoot tips succeeded to regenerate shoots when they were cultured in different MS. The results proved clear increase in chlorophyll and protein content of the rubber shoots as compared with content of field grown rubber plants. It was noticed that agar-solidified MS medium supplemented with 4.0 mg L-1 BA was considered the optimum medium for shoots regeneration. All plants regenerated from shoot tips were readily rooted in agar-solidified MS medium with increasing Potassium Nitrate KNO3 from 1900 mg L-1 to 2000 mg L-1, and at the same medium supplemented with 3.0 mg L-1 IBA and 1.0 mg L-1 BA. All these plants were successfully acclimated and transferred to peat moss.

We investigated the anther culture (AC) efficiency of thirteen F<sub>4</sub> combinations of winter wheat (Triticum aestivum L.). The genotype dependency was assessed during the induction of the androgenic entities, i.e. embryo-like structures (ELS), regenerated-, green-, albino-, and transplanted plantlets. The number of green plantlets per 100 anthers (GP/100A) varied from 0.36 to 24.74 GP/100A with a mean of 8.31 GP/100A. Albino plantlets (AP) occurred in each combination, ranging from 0.20 to 22.80 AP/100A with an average value of 5.59 AP/100A. Between 25–87.76 doubled haploid (DH) plants per 100 acclimatised plantlets (DH/100ADP), depending on the combination, with a mean of 59.74% were recovered. We have found the highest DH production in the combinations Béres/Midas, Kalász/Tacitus, Béres/Pamier, and Premio/5009. This improves remarkably the choice of basic genetic material in subsequent crossing programmes. These observations emphasise the usability and efficiency of in vitro AC in producing a large number of DH lines for breeding and the applied researches of winter wheat. Although albinism was found in each combination, it was mitigated by the in vitro AC application.

The improvement of genetic diversity and acceleration of breeding process are the most important tasks in wheat breeding. In vitro biotechnological methods are of practical interest for breeding process. The current paper has considered the effects of genotypes and nutritious medium on the formation of regenerants. The purpose of the research was to study the ability to androgenesis in winter soft wheat anthers and to identify promising winter soft wheat genotypes. The estimation of the ability to androgenesis was carried out among ten winter wheat samples. 8711 anthers were planted on medium of N6 and PII induction; the 190-2 medium was used for regeneration. As a result of the study, it was found that the process of neoplasms occurrence depends both on the variant of the nutrient medium and on the chosen genotype. It has been proven that the most favorable medium for the cultivation of winter soft wheat anthers is medium N6. It was found that the maximum percentage of neoplasms (5.21%) from winter soft wheat anthers was recorded in the sample “595/13”. It was found that the samples “595/13” (9 plants) and “Niva Dona” (6 plants) developed by the FSBSI “Agricultural Research Center “Donskoy” showed the highest responsiveness to the formation of calluses and plants-regenerants. There were obtained 6 green regenerants and 3 albino forms from the pollen callus of the sample “595/13” and 4 green regenerants and 2 albino regenerants from the sample “Niva Dona”.
