Abstract Background This research paper contributes to presenting a proposed framework for the formal and structural specifications of parametric digital architecture in order to increase knowledge and know-how in this field closely to specialists and those interested in it through the research methodology, which is divided first into the theoretical background to include a presentation of the definition of digital architecture and the most important pioneers of digital architecture, digital building materials and construction mechanisms parametric digital and what are the most important computer software used in parametric digital architecture in terms of design and construction implementation and identifying the most important digital specifications and characteristics that were mentioned in previous studies in this field and what are the unconventional digital esthetic values, then comes the role of the applied study in formulating a comprehensive matrix of parametric design modern and its impact on the development of traditional architectural models in digital architecture. Results The research study found that both (unrealistic shape) and (typical interaction) ranked first with the highest percentage reaching 100% in all study cases. The ratio of the volume of each to the total volume of all elements of morphological characteristics and non-traditional esthetic values was 8%, the most important characteristic of buildings in parametric digital architecture, in most of them, is the lack of realism to a very significant degree, and we always find a clear increase in the interaction, vitality and dynamism of buildings with the surrounding environment. Conclusions The most important characteristic of buildings in parametric digital architecture, in most of them, is the lack of realism to a very significant degree, and we always find a clear increase in the interaction, vitality and dynamism of buildings with the surrounding environment. While we conclude that the least characteristic of the parametric digital buildings is the simulation of nature or the tendency to everything that is traditional, as well it is often inclined to everything strange and unfamiliar.

In this paper, a versatile drum setup for measuring rolling resistance of small wheels is presented. The purpose is to provide a flexible setup for testing of models for rolling resistance under controlled circumstances. To demonstrate this, measurements of rolling resistance with a series of sandpapers of different grit sizes representing surface textures were carried out. The measurements show a clear increase in the rolling-resistance coefficient with increasing surface roughness, rolling speed and load. Numerical calculations in the time domain for a visco-elastic contact model run on equivalent surfaces agree with the trends found experimentally. We conclude that this approach to simplifying the experiment in order to obtain a high degree of control, accuracy and repeatability is useful for validating and testing models for calculating the rolling resistance for a given surface texture.

AbstractThe SorLA protein, encoded by the SORL1 gene, is a major player in Alzheimer’s disease (AD) pathophysiology. Functional and genetic studies demonstrated that SorLA deficiency results in increased production of Aβ peptides, and thus a higher risk of AD. A large number of SORL1 missense variants have been identified in AD patients, but their functional consequences remain largely undefined. Here, we identified a new pathophysiological mechanism, by which rare SORL1 missense variants identified in AD patients result in altered maturation and trafficking of the SorLA protein. An initial screening, based on the overexpression of 70 SorLA variants in HEK293 cells, revealed that 15 of them (S114R, R332W, G543E, S564G, S577P, R654W, R729W, D806N, Y934C, D1535N, D1545E, P1654L, Y1816C, W1862C, P1914S) induced a maturation and trafficking-deficient phenotype. Three of these variants (R332W, S577P, and R654W) and two maturation-competent variants (S124R and N371T) were further studied in details in CRISPR/Cas9-modified hiPSCs. When expressed at endogenous levels, the R332W, S577P, and R654W SorLA variants also showed a maturation defective profile. We further demonstrated that these variants were largely retained in the endoplasmic reticulum, resulting in a reduction in the delivery of SorLA mature protein to the plasma membrane and to the endosomal system. Importantly, expression of the R332W and R654W variants in hiPSCs was associated with a clear increase of Aβ secretion, demonstrating a loss-of-function effect of these SorLA variants regarding this ultimate readout, and a direct link with AD pathophysiology. Furthermore, structural analysis of the impact of missense variants on SorLA protein suggested that impaired cellular trafficking of SorLA protein could be due to subtle variations of the protein 3D structure resulting from changes in the interatomic interactions.

The noise caused by rail corrugation seriously affects the operation quality of metro vehicles. In this work, the rail corrugation, interior noise and wheel–rail noise of a metro line were tested, and the test results were compared with those after two kinds of treatments. The results show that rail corrugation is the main cause of the abnormal interior noise. The interior noise in many sections exceeds the limit, where clear rail corrugations from 31.5~63 mm are found. When the train passes through the rail corrugation section, the interior noise shows a clear increase, and the maximum increase is higher than 25 dB(A). After increasing the lateral stiffness of the track and rail grinding, the interior noise is reduced by 11.4 dB(A). After a long renovation time, the interior noise is effectively equal to that when the renovation was completed. The research results of this work can provide a reference for rail corrugation treatment and noise control.

The purpose of this study was to determine the evolution of the age of badminton players in the top 100 of the World Ranking for men and women from 1994 to 2020. Data were collected from badminton players participating in the top 100 World Rankings (4800 entries: 1233 players; 595 men and 638 women) from 1994 to 2020. The mean age of the top 100 and the average highest ranking of the players were analysed for both genders. The mean age of the male players in the World Ranking increased from 23.7 ± 3.2 years in 1994 to 26.3 ± 4.4 years in 2020 (p < 0.001) and in female players, from 22.8 ± 3.8 years in 1994 to 24.7 ± 3.3 years in 2020 (p < 0.001). In addition, women recorded a younger age at entry into the top 100 and when reaching their best ranking. Additionally, there has been a clear increase in Asian players in the top 100 of the World Ranking in recent years, reaching over 60%. These data could be used to develop and organise training plans in this sport, optimising and maximising players’ performance.

AbstractDespite the considerable amount of attention presently devoted to the high accessibility of online pornography, very little formal analyses have been carried out to show how the advent and proliferation of Internet technology has changed the prevalence of pornography use in populations. We conducted a preliminary analysis based on objective website traffic data, representing the changes in the number of (1) Internet users generally and (2) online pornography users specifically, between 2004 and 2016 in Poland. We observed a clear increase in the estimated number of people using online pornography in the analyzed period. The estimated number of general population members viewing pornography on the Internet increased over three times (310%) between October 2004 and October 2016–starting from an estimated 2.76 million in the first period to 8.54 million in the last. At the same time, we did not observe a clear increase in the percentage of Internet users who viewed online pornography in the same time period. Additionally, pornography viewership on the Internet was almost 2 times more prevalent among male (47%) than female Internet users (27%), and most popular in the 18–27 age group. Since our analysis is based on objective data, it does not share the limitations inherent in self-reports. However, our approach also has several important limitations (e.g., the analysis does not include online activity generated on mobile devices and under a private browsing mode); thus, the results should be interpreted with caution.

Size-segregated particulate matter (PM) including the PM0.1 fraction, particles ≤ 0.1 µm, was monitored during the rainy and dry seasons at three different cities in Sumatra island, Indonesia in 2018. In order to identify possible emission sources, carbonaceous components in the particles collected by a cascade air sampler that is capable of collecting PM0.1 particles were analyzed by applying a thermal/optical reflectance (IMPROVE-TOR) protocol. The PM0.1 levels in the Jambi and Pekanbaru areas were similar to those in large cities in East Asia, such as Bangkok and Hanoi. During the rainy season, local emissions in the form of vehicle combustion were the main sources of PM. The influence of peatland fires in the dry season was more significant in cities that are located on the east coast of Sumatra island because of the larger number of hotspots and air mass trajectories along the coast. A clear increase in the carbonaceous profiles as OC, TC, and OC/EC ratios in the dry season from the rainy season was observed, particularly in fine fractions such as PM0.5–1. In both seasons, EC vs. OC/EC correlations and soot-EC/TC ratios showed that the PM0.1 fraction in Sumatra island was heavily influenced by vehicle emissions, while the effect of biomass burning was more sensitive with respect to the PM0.5–1 fraction, particularly in Jambi and Pekanbaru sites during the dry season.

Peening techniques are nowadays attracting more research attention due to their association with the extending of the service life and improving surface texture of engineering components. Ultrasonic pulsating water jet peening represents a new way of mechanical surface treatment. Accelerated water droplets via hammer effect cause small elastic-plastic deformations on the surface. This work deals with peening of aluminum alloy using an ultrasonic pulsating water jet, where periodically acting water droplets were used as the peening medium. The aim of the work was the feasibility study of the peening process and to observe the effects of pressure (p = 10, 20 and 30 MPa) and pattern trajectory (linear hatch and cross hatch). The peened surfaces were analyzed by the surface roughness profile parameters Ra and Rz and the microhardness along the peening axis into the material. Graphically processed results show a clear increase of measured values with increasing pressure (p = 10, 20 and 30 MPa), where the roughness values ranged from 1.89 µm to 4.11 µm, and the microhardness values ranged from 43.3 HV0.005 to 47 HV0.005, as compared to 40.3 HV0.005 obtained for the untreated sample. The achieved results indicate potential using of an ultrasonic pulsating water jet as a new method of surface treatment of metals. By controlled distribution of water droplets, it is possible to achieve a local distribution of surface roughness, and at the same time, strengthening of the subsurface layers in the material without thermal influence on the material.

Microbreweries and beer tourism are a good example of the „neolocalism” turn in the globalized world. As a reaction to the unified beer offer of the large transnational brewery groups, there is a clear increase of the consumers’ demand for the bigger diversity of the beer offer. Seeking for the new beer styles in unique places – the beer tourism – is targeted not just to the microbreweries, but also to multitap restaurants with the large beer offer and to the beer festivals. The goal of this paper is to map the growing popularity of these beer tourism destinations in the Czech Republic and Poland and also among the big cities and smaller municipalities, and to compare the openness to the foreign beer brands and styles among the consumers in both countries.

Abstract Background In typical cases of Bartter syndrome (BS), assessing response to diuretics (furosemide and thiazide), hereinafter referred to as diuretic loading test, may be used to diagnose the type by detecting which part of the kidney tubule is not functioning correctly. However, the diuretic loading test may not always agree with the results of genetic analyses. Case presentation A 5-year-old boy was admitted due to lower extremity weakness and abnormal gait. He had a recurrent episode of muscle weakness and laboratory results showed severe hypokalemia. The direct genomic sequencing of the case revealed a new mutation in the SLC12A1 gene, which is associated with type I Bartter syndrome. Because there was the difference between the phenotype and genotype, we conducted a diuretic loading test to confirm the diagnosis. However, the results showed a clear increase in urine excretion of Na and Cl. These results were not consistent with typical type I BS, but consistent with the patient’s phenotype. Conclusion The diuretic loading test has limited utility for diagnosis especially in atypical cases. On the other hand, this test, which allows assessment of channel function, is useful for better understanding of the genotype-phenotype correlation.
