Background: Alzheimer’s disease (AD) is a progressive neurodegenerative disorder, and one of the most predominant causes of dementia in older people. Preventing acetylcholine from breakdown by an excess amount of acetylcholinesterase is a proven therapy for AD. Traditionally Curculigo orchioides is known for its antioxidant activity, enzyme inhibition activity, and other medicinal uses. In this study, in vitro acetylcholinesterase, inhibitory activity was investigated, and methanolic extract of the plant showed significant activity. This study aims to investigate cholinesterase inhibitory activity, learning, and memory-enhancing activity of Curculigo orchioides. Materials and Methods: The crude methanol extract of the dried powder was prepared by the cold extraction method. In-vitro acetylcholinesterase (AChE) inhibitory activities were determined by the modified Ellman’s method. To confirm learning and memory-enhancing effects, the scopolamine-induced memory impairment Swiss-albino mice were used, and to find the anti-amnesic effect of the extract, a passive shock avoidance task was applied. Results: Results proved that scopolamine-induced cognitive dysfunction decreased significantly by administration of the plant extract solution in the passive avoidance task, and inhibited brain acetylcholinesterase activity. While running an in-vitro AChE inhibitory test the IC50 was 0.782±0.067μg/ml. Conclusion: The results suggested that methanolic extract of Curculigo orchioides can inhibit AChE in-vitro. Besides this, it provides data that proves its ability to improve whole brain AChE activity and enhance memory.

Polyphenolic enriched extracts from two species of Cyperus, Cyperus glomeratus and Cyperus thunbergii, possess mammalian arginase inhibitory capacities, with the percentage inhibition ranging from 80% to 95% at 100 µg/mL and 40% to 64% at 10 µg/mL. Phytochemical investigation of these species led to the isolation and identification of two new natural stilbene oligomers named thunbergin A-B (1–2), together with three other stilbenes, trans-resveratrol (3), trans-scirpusin A (4), trans-cyperusphenol A (6), and two flavonoids, aureusidin (5) and luteolin (7), which were isolated for the first time from C.thunbergii and C. glomeratus. Structures were established on the basis of the spectroscopic data from MS and NMR experiments. The arginase inhibitory activity of compounds 1–7 was evaluated through an in vitro arginase inhibitory assay using purified liver bovine arginase. As a result, five compounds (1, 4–7) showed significant inhibition of arginase, with IC50 values between 17.6 and 60.6 µM, in the range of those of the natural arginase inhibitor piceatannol (12.6 µM). In addition, methanolic extract from Cyperus thunbergii exhibited an endothelium and NO-dependent vasorelaxant effect on thoracic aortic rings from rats and improved endothelial dysfunction in an adjuvant-induced arthritis rat model.

 Medicinal plants are safe and the oldest natural products used for many years to conserve food, to treat health disorders and to prevent diseases. The active chemical compounds formed during secondary vegetal metabolism is usually responsible for the biological properties of some plant species used throughout the world for various purposes including treatment of diabetes, cancer, infectious diseases etc. The present study was undertaken to analyze the phytochemicals by colour differentiation method, to evaluate the toxic effect by phytotoxic assay, antidiabetic activity by α amylase enzyme inhibition and antioxidant potential by DPPH (2,2-diphenyl-1-picrylhydrazyl) scavenging activity of methanolic extract of Ageratum houstonianum. Treatment of problem in carbohydrate uptake needed the inhibition of α-amylase plays a role in the digestion of polysaccharide and glycogen, is made a strategy for controlling diabetes. For this study whole plant was collected, dried and the powder was made. The extraction was carried out by cold percolation in which methanol was used as a solvent. The methanolic extract was subjected to In-vitro phytotoxic activity by adopting the standard protocol. The α-amylase enzyme inhibition activity of plant extract was carried out by using starch as substrate, pancreatic α amylase as the enzyme, and acarbose as standard. The result of in-vitro phytotoxic bioassay revealed that the plant extract showed moderate activity with percentage growth regulation 80 and 25 percent in a concentration-dependent manner. The α-amylase enzyme inhibition was 74.13 to 99.39 percent in a dose-dependent manner. The antioxidant potential of Ageratum houstonianum extract showed mild activity with IC50 123.67 μg/ml as compared to the standard ascorbic acid IC50 5.38 μg/ ml. It is concluded from the present study that Ageratum houstonianum could be used as a natural source to isolate antioxidant, anti-hyperglycemic agent, herbicide and weedicide as it shows a good α amylase inhibition, radical scavenging and phytotoxic activity respectively.

We demonstrate that NMR-based metabolomics studies can be used to identify xanthine oxidase-inhibitory compounds in the diethyl ether soluble fraction prepared from a methanolic extract of Sophora flavescens. Loading plot analysis, accompanied by direct comparison of 1H NMR spectra exhibiting characteristic signals, identified compounds exhibiting inhibitory activity. NMR analysis indicated that these characteristic signals were attributed to flavanones such as sophoraflavanone G and kurarinone. Sophoraflavanone G showed inhibitory activity towards xanthine oxidase in an in vitro assay.

Pistacia integerrimais one of twenty species among the genusPistacia. Long horn-shaped galls that develop on this plant are harvested and used in Ayurveda and Indian traditional medicine to make “karkatshringi”, a herbal medicine used for the treatment of asthma and different disorders of respiratory tract. However, until now, the molecular mechanisms of action of “karkatshringi” and its chemical characterization are partially known. This study deals with the isolation and characterization of the active constituents from the methanolic extract ofP. integerrimagalls and it was also oriented to evaluatein vitroandin silicotheir potential enzymatic inhibitory activity against phosphodiesterase-1 (PDE1), a well-known enzyme involved in airway smooth muscle activity and airway inflammation. Our results showed that the methanolic extract ofP. integerrimagalls and some of its active constituents [naringenin (1) and 3,5,7,4′-tetrahydroxy-flavanone (2)] are ablein vitroto inhibit PDE1 activity (59.20 ± 4.95%, 75.90 ± 5.90%, and 65.25 ± 5.25%, resp.) and demonstratein silicoan interesting interaction with this enzymatic site. Taken together, our results add new knowledge of chemical constituents responsible for the biological activity ofP. integerrimaand contextually legitimate the use of this plant in folk medicine.

We aimed to investigate the apoptotic effects of the methanolic extract of Dorema glabrum seed on WEHI-164, cancerous cells in comparison with L929, normal cells and compared them with the cytotoxic effects of Taxol. So, MTT test and DNA fragmentation assay were performed on cultured and treated cells. Also electrophoresis which was followed by immunoblotting was done to survey the production of Caspase-3 and Bcl2 proteins, and to inquire into their relative genes expression, RT-PCR was used. According to our findings, the methanolic extract of Dorema glabrum seed can alter cells morphology as they shrink and take a spherical shape and lose their attachment too. So, the plant extract inhibits cell growth albeit in a time- and dose-dependent manner and results in degradation of chromosomal DNA. Induction of apoptosis by the plant extract was proved by the reduction of pro-Caspase-3 and Bcl2 proteins and increase in Caspase-3 gene expression and decrease in that of bcl2 too. Our data well established the antiproliferative effect of methanolic extract of Dorema glabrum seed and clearly showed that the plant extract can induce apoptosis and not necrosis in vitro. These results demonstrated that Dorema glabrum seed might be a novel and attractive therapeutic candidate for tumor treatment.

In this study, the methanolic extracts of leaves of Citrus maxima were evaluated for their carminative and antidiarrheal activities. By soxhlet method, the methanolic extract of leaves of Citrus maxima was prepared. Carminative activity of methanolic extract was studied in vitro by acid-base titration technique and antidiarrheal activity was studied in rats by castor oil induced diarrhoea method. The phytochemical screening was done using various chemical tests. Alkaloids, tannins, carbohydrates, flavonoids, terpenoids, amino acids, anthraquinones, glycosides are confirmed to be present in the leaf. Carminative activity of extract was found significantly less with 5mL, when compared with 10mL extract. The antidiarrheal activity observed in the animals treated with 200mg/kg showed significant activity compared with the animals treated with 100mg/kg. The study revealed that carminative activity and antidiarrheal activity of methanolic extract of C. maxima. May be the extract acts on circular and longitudinal intestinal muscle by inhibiting acetylcholine and prostaglandins, which reduces peristaltic movements and prolongs transmit time, reduces fecal volume, increases viscosity and viscous fluid and electrolyte loss. The compound in the extract causes mild irritation resulting in vasodilation of gastrointestinal muscles, thereby relieving cramping and expels gas

  Objective: This communication explores a lead fraction from methanolic extract of novel Stevia species raised under in vitro conditions for its various biological activities.Methods: The dried Stevia leaves were crushed in methanol to get the polar extract. This methanol extract was tested for pancreatic lipase and alpha-amylase inhibitory activity using quantitative plate assays. Antibacterial property of the extract was also evaluated against Staphylococcus epidermidis, Escherichia coli, Bacillus subtilis, Staphylococcus aureus, and Pseudomonas aeruginosa. Further, the antioxidant potential was evaluated using 1,1-diphenyl-2-picrylhydrazyl.Results: The methanolic extract inhibited pancreatic lipase with IC50 of 5.74 μg/ml in a similar manner to a well-known anti-obesity drug in the market orlistat. The methanolic extract also showed a better pancreatic α-amylase inhibitory activity (IC50=88 μg/ml) than acarbose. Further, the lead fraction exhibited 88.48% antioxidant activity. It also exhibited broad spectrum antimicrobial activity against the spectrum of Gram-positive and Gram-negative bacteria tested under laboratory conditions with a minimal inhibitory concentration ranging from 1.95 to 31.25 μg/ml.Conclusion: Thus, this study signifies the vast potential of the lead fraction from a novel Stevia species for further development into a herbal formulation for prevention of various infectious and non-infectious diseases.

Aim:The aim of this study was undertaken to assessin vitro H+-K+ ATPase inhibitory activity of different extracts of Cissus quadrangularis Linn. Materials and Methods: Phytochemical and Phyto-analytical studies like total phenolic compound and total flavonoid contents from extract were quantified and H+-K+ ATPase inhibition assay was performed in presence of different concentrations(10,20,50 and 100 μg/ml) of standard (omeprazole),chloroform and methanolic extract. Results:The extract has shown dose dependent significant (*P < 0.05) proton pump inhibitory activity in the goat gastric mucosal homogenate which was compared to standard Omeprazole. Conclusions: Hence, from this study we have concluded that methanolic extract of C.quadrangularis has more inhibitory effect than chloroform extract.

 Objectives: The purpose of the present study was to determine the anthelmintic properties of chloroform, methanol and aqueous extracts of Avicennia marina leaves on Indian earthworm- Pheretima posthuma. Methods: Three concentrations of each extract (25 mg/ml, 50 mg/ml, and 75 mg/ml) were studied in this activity to determine the time of paralysis (vermifuge) and the time of the death of the worms (vermicidal). Piperazine citrate in the same concentrations was taken a reference standard and normal water was taken as control. The time taken for paralysis and death of the worms was recorded.Results: All the extracts showed dose-dependent and a significant activity was shown at higher concentration (75 mg/ml). Methanolic extract showed peak activity at a concentration of 75 mg/ml.Conclusion: Based on the results it was found that the leaves of A. marina possess vermifuge and vermicidal activities and can be used as an effective anthelmintic.
