Nigella sativa (kalonji) has been used since ancient times as a nutritional supplement and for treating various infections and chronic ailments. As pathogens become resistant to most drugs, kalonji can be used as an alternative compound in modern medicines. The use of herbal extracts as endodontic irrigants might be beneficial as a part of a growing trend to seek natural remedies for dental treatment. Aim:  To compare the antibacterial potency of Aqueous Nigella sativa extract, Aqueous Neem leaf extract, and 3% Sodium Hypochlorite. Materials and Methods: Test solutions were tested against E. faecalis (ATCC 29212) to check for their Minimum Inhibitory Concentration (MIC) by double dilution method and Kill time to measure their antimicrobial potency to be used as an intracanal irrigant. Results: The study's limitations show that Aqueous Nigella sativa extract has a better antimicrobial effect than Aqueous Neem leaf extract and 3% Sodium Hypochlorite solution against E. faecalis.

ABSTRACT Introduction Debridement and disinfection of the root canal is a crucial step in the success of endodontic treatment. Several antimicrobial agents alone or in combination are used to achieve this. The objective of this in vitro study was to assess the antimicrobial efficacy of neem (Azadirachta indica) extract against Enterococcus faecalis. Materials and methods Neem leaf extract, 2% chlorhexidine, 3% sodium hypochlorite were used to assess the antimicrobial efficiency. Agar well diffusion test was used to study the antimicrobial efficacy with saline as control. The zone of inhibition was recorded, tabulated, and analyzed statistically with the help of IBM Statistical Package for the Social Sciences statistics version 20 using analysis of variance test. Results All the three medicaments showed well-defined and comparable zones of inhibition around their respective wells. All values were significantly higher than the control group. Analysis of variance showed significant difference between zone diameters of chlorhexidine, neem leaf extract, and 3% sodium hypochlorite against E. faecalis (p < 0.05). Conclusion From the present study, it can be concluded that neem leaf extract shows comparable zones of inhibition with that of chlorhexidine and sodium hypochlorite. Clinical significance Neem leaf extract has significant antimicrobial activity against E. faecalis and thus opens the perspectives for the use of neem extract as an intracanal medication. Research has been conducted on the treatment of phlegm cough with miana leaf extract in vitro (effective dose of miana leaf extract as an expectorant and antibacterial agent causing cough with phlegm). The study aims to compare the antioxidant activity of miana leaves from 3 locations where it grows, determine the types of contaminant bacteria in the sputum of cough sufferers, determine the minimum value of inhibitor concentration (MIC) and MKC of miana leaves against the test bacteria causing cough with phlegm, determine the effective dose of miana leaves that can used as a reference for coughing up phlegm and proving the potential of miana leaves as a sputum thinner. Miana leaf extraction is done by the juicer method. Antioxidant activity testing uses the DPPH method. Determination of test bacteria is done by isolating and identifying contaminant bacteria in the sputum sample of cough with phlegm. Testing the effectiveness of miana leaves against test bacteria is determined by the liquid dilution method. Expectorant activity testing was determined by measuring the viscosity of mucus viscosity of cow intestine treated with miana leaf extract. The results found that antioxidant activity was influenced by the location where miana leaves grew with an antioxidant potential of IC72 0.072 mg/ml - 0.76 mg/ml. Contaminant bacteria from sputum samples of cough patients are Streptococcus pneumonia, Klebsiella pneumonia, Staphylococcus aureus, Staphylococcus epidermidis and Enterobacter agglomerans. MIC values for contaminant bacteria are 0.1% - 0.75% and MKC values are found between 0.25% - 1.75%. miana leaf extract has the potential as a sputum thinner at a concentration of 0.01% - 0.1%. The recommended dose of miana leaf extract as a cough with phlegm is 1.75% w/v.

Acrylic resin dentures are a type of denture that is often used. At present the use of disinfectants to clean dentures from microorganisms, such as sodium hypochlorite and green betle leaves. Sodium hypochlorite contains chlorine, and green betle leaf extract contains tannins that containable to provide color changes in artificial teeth. The purpose of this study was to study the change in color on the hot polymerization acrylic plate after soaking with sodium hypochlorite solution and green betle leaf extract (Piper betle Linn). This study was a pre-post test control group design, the sample was heat-cured resin acrylic base plate and divided into 3 treatment groups: a group immersed in sodium hypochlorite, extract of green betel leaf and control group with aquades. The discoloration measurement was using Spectrophotometer. The sample was immersed for 15 days. Data were analyzed by paired T-test, followed by One way Anova and LSD. The results showed that there were significant color changes in the 2 treatment groups, while the control group was not significant. The mean of discoloration after immersion with Sodium Hypochlorite was 0.668 + 0.018, green betle leaf extract 30% was 0.625 + 0.024, and aquades was 0.534 + 0.0224. Color changes on heat-cured resin acrylic base plate occur by the presence of chlorine in sodium hypochlorite and tannin contained in the green betel leaf extract. This research concluded that immersion of heat-cured resin acrylic plate in sodium hypochlorite solution and 30% betle leaf extract may cause discoloration.

Abstract: Noni (Morinda cittrifolia L.) leaves contain antraquinon, atsiri oil, saponin, tannin, alkaloid, flavonoid, polifenol, and sterol that have been proved can inhibit the growth of Candida albicans. This study was aimed to establish the minimum inhibitory concentration (MIC) of noni leaf extract against Candida abicans. This was a true experimental study with a randomized pretest-posttest control group design. We used serial dilution method with turbidimetry and spectrophotometry tests. Noni leaves were extracted by using maceration with 96% ethanol. Candida albicans fungi were obtained from Microbiology Laboratory of Faculty of Mathematics and Natural Sciences, Sam Ratulangi University. The turbidimetry test using three repetitions showed that the MIC of noni leaf extract against Candida albicans was 6.25% meanwhile the spectrophotometry test established 12.5% as the MIC of noni leaf extract. Conclusion: Minimum inhibitory concentration of noni (Morinda cittrifolia L.) leaf extract against the growth of Candida albicans was 12.5%.Keywords: noni (Morinda citrifolia L.), Candida albicans, minimum inhibitory concentration (MIC) Abstrak: Daun mengkudu (Morinda citrifolia L.) mengandung antraquinon, minyak atsiri, saponin, tannin, alkaloid, flavonoid, polifenol dan sterol yang terbukti dapat menghambat pertumbuhan Candida albicans. Penelitian ini bertujuan untuk mendapatkan konsentrasi hambat minimum (KHM) dari ekstrak daun mengkudu terhadap Candida albicans. Jenis penelitian ialah eksperimental murni dengan randomized pretest-posttest control group design. Metode yang digunakan dalam penelitian yaitu serial dilusi dengan pengujian turbidimetri dan spektrofotometri. Daun mengkudu diekstraksi dengan metode maserasi menggunakan pelarut etanol 96%. Jamur Candida albicans diambil dari stok jamur Laboratorium Mikrobiologi Program Studi Farmasi Fakultas MIPA Universitas Sam Ratulangi. Hasil penelitian menunjukkan bahwa pengujian turbidimetri dengan tiga kali perlakuan mendapatkan KHM pada konsentrasi 6,25% sedangkan pengujian spektrofotometri mendapatkan KHM pada konsentrasi 12,5%. Simpulan: Konsentrasi hambat minimum (KHM) ekstrak daun mengkudu (Morinda citrifolia L.) terhadap pertumbuhan Candida albicans terdapat pada konsentrasi 12,5%.Kata kunci: mengkudu (Morinda citrifolia L.), Candia albicans, konsentrasi hambat minimum (KHM)
