Pollution is the main cause that effects the environment in a degradable condition. For the presence of heavy metals in the atmosphere birds are considered as tremendous bio indicator. Blue pigeon and domestic pigeon (Coloumba livia) of Faisalabad are selected to examine the level of heavy metals including Manganese (Mn), zinc (Zn), Cobalt (Co), lead (Pb) cadmium (Cd) and Copper (Cu) in several tissues such as (heart, lungs, bones, muscles and feathers. Six adult birds were collected rural, urban and residential and traffic areas of Faisalabad. After collection of sample fresh weight will be measured of each sample, then digestion of sample was done by conventional acid digestion method and digested samples were analyzed in the lab by Atomic Absorption Spectrophotometer for the determination of heavy metals.  It was concluded that accumulation of heavy metals in the birds depends upon the environmental conditions. Our result showed that concentration of heavy metals more in rock pigeon as compared to domestic pigeon which may be due to more exposure of industrial waste to rock pigeon. It has inferred from our study that high concentration of heavy metals in the excreta of rock pigeon was due to its high trophic level in food chain.

Recent advances in single-cell proteomics highlight the promise of sensitive analyses in limited cell populations. However, technical challenges remain for sample recovery, throughput, and versatility. Here, we first report a water droplet-in-oil digestion (WinO) method based on carboxyl-coated beads and phase transfer surfactants for proteomic analysis using limited sample amounts. This method was developed to minimize the contact area between the sample solution and the container to reduce the loss of proteins and peptides by adsorption. This method increased protein and peptide recovery 10-fold as well as the number of quantified transmembrane proteins compared to an in-solution digestion (ISD) method. The proteome profiles obtained from 100 cells using the WinO method highly correlated with those from 10000 cells using the ISD method. We successfully applied the WinO method to single-cell proteomics and quantified 462 proteins. Using the WinO method, samples can be easily prepared in a multi-well plate, making it a widely applicable and suitable method for single-cell proteomics.

Abstract Perai river is one of the important rivers to Penang and Kedah states. Many industries are located along the river that flows to the Strait of Malacca. In order to gather the status of pollution along the river, a total of 47 stations were established to measure the heavy metals content and the sediment characteristics. Findings show that finer sediments had dominated all sampling sites as samples are mostly muddy sediment. The heavy metal concentration in the surficial sediment was analyzed using the Inductively Coupled Plasma Mass Spectrometer after the Teflon Bomb digestion method. The average concentration for Zn, Cr, Cu, Cd, Pb were 74.7±33.3, 66.0±28.1, 21.8±9.05, 0.42±0.32, and 28.6±6.84 μg/g dry weights, respectively. Generally, the concentration of metals studied seemed to be controlled by natural processes. This is proved by the enrichment factor, categorized as a deficiency to minimal enrichment, and can be concluded to be the main source from the lithogenous in origin. The sources of the heavy metals in some of the stations in the Perai river are speculated from the nearby urban runoff, industry activities along with the river, fishing and shipping activities, and land transportation emission may also accumulate for the pollution in the river.

Prickly rose (Rosa acicularis Lindl.) is the most distributed rose species in the Northern Hemisphere, used by indigenous people for various food purposes. The lack of detailed information about the chemical composition of R. acicularis has led us to study the phytochemical composition and metabolic profile of prickly rose extracts using chromatographic techniques. Many groups of phenolic and non-phenolic compounds were quantified in the leaves, flowers, roots and fruits of R. acicularis. Phenolic compounds were the dominant phytochemicals in the aerial parts and roots of R. acicularis. A precise study by high-performance liquid chromatography with photodiode array detection and electrospray ionization triple quadrupole mass spectrometric detection showed the presence of 123 compounds, among which ellagic acid derivatives, ellagitannins, gallotannins, catechins, catechin oligomers, hydroxycinnamates and flavonoid glycosides of kaempferol, quercetin and dihydroquercetin were all identified for the first time. The most abundant phenolic compounds were ellagitannins and flavonoid glycosides, with a maximal content of 70.04 mg/g in leaves and 66.72 mg/g in flowers, respectively, indicating the great ability of R. acicularis organs to accumulate phenolic compounds. By applying a standardized static, simulated gastrointestinal digestion method, we found the inhibitory potential of the leaf extract against digestive α-amylases. A pancreatic α-amylase activity-inhibiting assay coupled with HPLC microfractionation demonstrated high inhibition of enzyme activity by ellagitannin rugosin D, which was later confirmed by a microplate reaction with mammalian α-amylases and the simulated digestion method. This study clearly demonstrates that R. acicularis leaf extract and its main component, ellagitannin rugosin D, strongly inhibit digestive α-amylase, and may be a prospective antidiabetic agent.

Trichinellosis is a zoonotic meat-borne disease caused by the nematodes of the genus Trichinella. Meat containing live Trichinella larvae is a source of infection. The examination of meat for Trichinella was introduced in 1869, but the digestion method for this did not appear in Poland until the late 1970s. Nowadays, the meat of all food animals susceptible to Trichinella spp. is examined in the frame of official post mortem control with the digestion method. The majority of laboratories in Poland meet the requirements of the ISO/IEC 17025 Standard (352 field laboratories). Laboratory personnel participate in quality control programs. This paper presents the results of proficiency tests (PTs) organized within 2015–2019 in Poland. Over this period, the laboratories examined 7580 samples (contamination levels: zero, one, three, and five larvae). Each laboratory was provided with a set of samples (one negative and three positive). Over 95% of the samples were considered correct in qualitative assessments, though the results of the quantitative evaluations were slightly lower, with 89% of samples being considered correct. Based on a sample evaluation, 88% of laboratories passed the PT comparison. A slight decrease was observed in the examination of samples spiked with five larvae, and great progress was achieved in samples containing three larvae. Low levels of sample contamination are sought after in laboratories but may make evaluations difficult. For this reason, we must consider increasing the number of larvae added to the samples in the next PTs.

Trichinella nematodes continue to circulate in various hosts both in the domestic and sylvatic cycles. In the majority of countries in Europe, wild boars have been noticed as a primary source of Trichinella spp. infections in humans. However, in some regions, the meat of pigs containing Trichinella spp. larvae can still be a cause of trichinellosis. Therefore, in the present study, we aimed to determine and present actual data on the occurrence of Trichinella spp. on pig farms (Sus scrofa f. domestica) in Poland. In this study, over 194 million pigs, slaughtered for commercial and personal purposes between 2012 and 2020, were tested with a digestion method according to the official rules for Trichinella control. Positive results were noticed in 172 pigs which gives an overall prevalence of 0.000088%. On seven farms, rats (Rattus norvegicus) infected with Trichinella spp. were also discovered. The species identification showed pigs were infected with Trichinella spiralis on 26 farms, and on four farms pigs with Trichinella britovi infections were found. Therefore, it is important to constantly monitor pigs for the presence of these parasites, especially in view of the growing interest in organic meat originated from ecological farms.

Abstract Sewage sludge after urban wastewater treatment needs to be treated intelligently to get maximum benefits. This study proposes a method of anaerobic co-degradation of agricultural sludge and residues to both solve environmental problems and recover a large amount of biogas and organic fertilizers. The results show the potential for energy recovery from biogas and impact assessment of the application directly on the rice fields. It is estimated that biogas recovery from sludge treatment from wastewater treatment plants gives a calorific value of about 76 × 106 MJ/year. Results from the trial crop showed that fertilizer shows good supportability to the plant. The results show that the fertilizer from sewage sludge can be reduced and replaced by 50% to 100% of the number of chemical fertilizers but still give the same yield. The data show that the anaerobic co-digestion method is a suitable method for sludge treatment. The dual objective is to provide valuable benefits from recovered biogas and anaerobic digestion products.

Abstract: Diatoms are unique algae. They are scientifically known as Bacillariophyceae. Diatoms have been used in forensic science in a variety of ways, the most frequent being the diagnosis of death by drowning. When a person drowns, water will enter the lungs and then enter the bloodstream through ruptures in the peripheral alveoli before being carried to the other organs such as the liver and heart. Naturally, the microscopic contents of the water, which will include diatoms, will pass into the blood as well. The detection of diatoms in the organs can contribute to a diagnosis of death by drowning, a process referred to as the ‘diatom test’. A study was conducted in the department of Forensic Science, SHUATS, Prayagraj which included the extraction and identification of diatoms from the collected water samples from three different sites of Kaalesar Ghat of Rapti River in Gorakhpur. The acid digestion method was used for diatom extraction. And, after analysis total 13 diatom species were found. The identified diatoms were of great ecological assessment that plays an important role in criminal investigations related to premortem or ante-mortem drowning.
