Major Nutritional Metabolic Alterations Influencing the Reproductive System of Postpartum Dairy Cows

Early successful conception of postpartum dairy cows is crucial in determining the optimum reproductive efficiency and profitability in modern dairy farming. Due to the inherent high production potential of modern dairy cows, the extra stress burden of peri-parturient events, and associated endocrine and metabolic changes causes negative energy balance (NEBAL) in postpartum cows. The occurrence of NEBAL is associated with excessive fat mobilization in the form of non-esterified fatty acids (NEFAs). The phenomenon of NEFA mobilization furthers with occurrence of ketosis and fatty liver in postpartum dairy cows. High NEFAs and ketones are negatively associated with health and reproductive processes. An additional burden of hypocalcemia, ruminal acidosis, and high protein metabolism in postpartum cows presents further consequences for health and reproductive performance of postpartum dairy cows. This review intends to comprehend these major nutritional metabolic alterations, their mechanisms of influence on the reproduction process, and relevant mitigation strategies.

Effects of Isochronal Induced Feed Restriction With Transition Period On mRNA Abundance of The Hepatic Genes Related to Lipid Metabolism in Fat-Tailed Ewes

The process of fat mobilization during the transition period requires deep re-orchestration of the energy indices, and understanding its mechanism has generated considerable interest among the TP-related studies. The present study aims to validate the effect of feed restriction and TP on the mRNA abundance of hepatic genes related to fat metabolism in fat-tailed sheep. Twenty pregnant ewes with the age of 40.8 ± 6.2 (mean ± standard error) month were randomly assigned to Control (n=10) or Restriction (n=10), and investigated from week -5 to 5 relative to parturition. Control animals received 100% DM during the trial. Restriction animals received 100% DM through weeks -5, -1, 1 and 5 and were fed with 50, 65, and 80% DM in the weeks -4, -3, -2 and 2,3, and 4, respectively. On the third week of experiment (65%) during both pre and post-partum, the hepatic tissue was biopsied, and the mRNA load of the fatty acid synthase, acetyl-CoA carboxylase, carnitine palmitoyltransferase (CPT) 1, CPT2, and acyl-CoA synthase long-chain family member-1 genes was quantified by the TaqMan qPCR technique. Data were analyzed using the Mixed Model procedure of SAS. The mRNA abundance of the target genes was not influenced by feed restriction, during the pre and postpartum periods. Parturition suppressed the mRNA abundance of target genes in both groups. It can be concluded that the liver of the fat-tailed sheep would have a higher capacity for the metabolism of free fatty acids mobilization during the feed insufficiency and the challenging period of transition.

Responses of Outdoor Housed Dairy Cows to Shade Access during the Prepartum Period under Temperate Summer Conditions

Cows are affected by environmental factors associated with warm weather conditions; however, little is known about the effect of shade access especially during the prepartum period of dairy cows in temperate regions. This study assessed the effect of shade on the behavior (lying, rumination, feeding, and drinking), body fat mobilization, and health status of outdoor-housed dairy cows during the prepartum period under temperate summer conditions. During the 3 weeks prior to calving, 24 multiparous Holstein cows were grouped (4 cows/group) and assigned to either an open corral without shade or with access to shade until calving. We daily measured shade use, lying, rumination, feeding, and drinking behavior. Weekly, prepartum non-esterified fatty acids (NEFA) and postpartum b-hydroxybutyrate (BHB) concentrations were measured. Clinical examination was periodically performed individually until 21 postpartum days. Shade use averaged 45.6, 46.0, and 19.8% during the hottest hours of the day (11–18 h) in weeks 3, 2, and 1 prior to calving, respectively. Shaded cows had higher values for rumination time and feeding time during the morning but spent less time drinking during the warmest hours than unshaded cows. NEFA and BHB concentrations and clinical diseases were similar between both treatments. These findings suggest that under temperate summer conditions the access to an artificial shade is an important resource, observing beneficial effects mainly on behavioral variables.

Endocannabinoids Produced by White Adipose Tissue Modulate Lipolysis in Lean but Not in Obese Rodent and Human

White adipose tissue (WAT) possesses the endocannabinoid system (ECS) machinery and produces the two major endocannabinoids (ECs), arachidonoylethanolamide (AEA) and 2-arachidonoylglycerol (2-AG). Accumulating evidence indicates that WAT cannabinoid 1 receptors (CB1R) are involved in the regulation of fat storage, tissue remodeling and secretory functions but their role in controlling lipid mobilization is unclear. In the present study, we used different strategies to acutely increase ECS activity in WAT and tested the consequences on glycerol production as a marker of lipolysis. Treating lean mice or rat WAT explants with JLZ195, which inhibits ECs degrading enzymes, induced an increase in 2-AG tissue contents that was associated with a CB1R-dependent decrease in lipolysis. Direct treatment of rat WAT explants with AEA also inhibited glycerol production while mechanistic studies revealed it could result from the stimulation of Akt-signaling pathway. Interestingly, AEA treatment decreased lipolysis both in visceral and subcutaneous WAT collected on lean subjects suggesting that ECS also reduces fat store mobilization in Human. In obese mice, WAT content and secretion rate of ECs were higher than in control while glycerol production was reduced suggesting that over-produced ECs may inhibit lipolysis activating local CB1R. Strikingly, our data also reveal that acute CB1R blockade with Rimonabant did not modify lipolysis in vitro in obese mice and human explants nor in vivo in obese mice. Taken together, these data provide physiological evidence that activation of ECS in WAT, by limiting fat mobilization, may participate in the progressive tissue remodeling that could finally lead to organ dysfunction. The present findings also indicate that acute CB1R blockade is inefficient in regulating lipolysis in obese WAT and raise the possibility of an alteration of CB1R signaling in conditions of obesity.

Prepartum Fat Mobilization in Dairy Cows with Equal Body Condition and Its Impact on Health, Behavior, Milk Production and Fertility during Lactation

The objective of this study was to evaluate the effect of two levels of fat mobilization at the close-up period in dairy cows with an equal body condition score (BCS = 3.0) on the circulating concentrations of metabolic, inflammatory, and oxidative stress biomarkers, incidence of diseases, behavior, milk production, and fertility during the postpartum. Late-gestation multiparous Holstein cows (n = 59) with a body condition score of 3.0 (5-point scale) were enrolled at the beginning of the close-up period and then were followed during the entire lactation. Cows were retrospectively allocated into two groups: animals with prepartum non-esterified fatty acids concentration over 0.3 mmol/L were categorized as high fat mobilization (HFM) (n = 26), and below this threshold as low fat mobilization (LFM) (n = 33). Blood samples were collected 21 d before expected calving and once weekly for 3 wk postpartum in order to analyze β-hydroxybutirate, haptoglobin, fibrinogen, total proteins, and malondialdehyde. Health was observed daily for 21 d postpartum. Behavioral data was collected with an accelerometer and milk production and fertility were obtained from the farm records. An increased fat mobilization in dairy cows with equal BCS modified the inflammatory and oxidative stress responses during the early postpartum without impairing their health status and fertility. Moreover, milk production and behavior were markedly affected by excessive prepartum fat mobilization through lactation.

Metabolic Stress in the Transition Period of Dairy Cows: Focusing on the Prepartum Period

All modern, high-yielding dairy cows experience a certain degree of reduced insulin sensitivity, negative energy balance, and systemic inflammation during the transition period. Maladaptation to these changes may result in excessive fat mobilization, dysregulation of inflammation, immunosuppression, and, ultimately, metabolic or infectious disease in the postpartum period. Up to half of the clinical diseases in the lifespan of high-yielding dairy cows occur within 3 weeks of calving. Thus, the vast majority of prospective studies on transition dairy cows are focused on the postpartum period. However, predisposition to clinical disease and key (patho)physiological events such as a spontaneous reduction in feed intake, insulin resistance, fat mobilization, and systemic inflammation already occur in the prepartum period. This review focuses on metabolic, adaptive events occurring from drying off until calving in high-yielding cows and discusses determinants that may trigger (mal)adaptation to these events in the late prepartum period.

Circadian Rhythms in Hormone-sensitive Lipase in Human Adipose Tissue: Relationship to Meal Timing and Fasting Duration

Background Fat mobilization in adipose tissue (AT) has a specific timing. However, circadian rhythms in the activity of the major enzyme responsible for fat mobilization, hormone-sensitive lipase (HSL), have not been demonstrated in humans. Objective To analyze in a cross-sectional study whether there is an endogenous circadian rhythm in HSL activity in human AT ex vivo and whether rhythm characteristics are related to food timing or fasting duration. Methods Abdominal AT biopsies were obtained from 18 severely obese participants (age: 46 ± 11 years; body mass index 42 ± 6 kg/m2) who underwent laparoscopic gastric bypass. Twenty-four-hour rhythms of HSL activity and LIPE (HSL transcript in humans) expression in subcutaneous AT were analyzed together with habitual food timing and night fasting duration. Results HSL activity exhibited a circadian rhythm (P = .023) and reached the maximum value at circadian time 16 (CT) that corresponded to around midnight (relative local clock time. Similarly, LIPE displayed a circadian rhythm with acrophase also at night (P = .0002). Participants with longer night fasting duration >11.20 hours displayed almost double the amplitude (1.91 times) in HSL activity rhythm than those with short duration (P = .013); while habitual early diners (before 21:52 hours) had 1.60 times higher amplitude than late diners (P = .035). Conclusions Our results demonstrate circadian rhythms in HSL activity and may lead to a better understanding of the intricate relationships between food timing, fasting duration and body fat regulation.

Butaphosphan Effects on Glucose Metabolism Involve Insulin Signaling and Depends on Nutritional Plan

Butaphosphan is an organic phosphorus compound used in several species for the prevention of rapid catabolic states, however, the mechanism of action remains unclear. This study aimed at determining the effects of butaphosphan on energy metabolism of mice receiving a normal or hypercaloric diet (HCD) and submitted or not to food restriction. Two experiments were conducted: (1) during nine weeks, animals were fed with HCD (n = 28) ad libitum, and at the 10th week, were submitted to food restriction and received butaphosphan (n = 14) or saline injections (n = 14) (twice a day, for seven days) and; (2) during nine weeks, animals were fed with a control diet (n = 14) or HCD (n = 14) ad libitum, and at the 10th week, all animals were submitted to food restriction and received butaphosphan or saline injections (twice a day, for seven days). In food restriction, butaphosphan preserved epididymal white adipose tissue (WAT) mass, increased glucose, NEFA, and the HOMA index. In mice fed HCD and submitted to food restriction, the butaphosphan preserved epididymal WAT mass. Control diet influences on PI3K, GCK, and Irs1 mRNA expression. In conclusion, butaphosphan increased blood glucose and reduced fat mobilization in overweight mice submitted to caloric restriction, and these effects are influenced by diet.
