Most cases involving soil in criminal investigations in Brazil have focused on the chemical and mineralogical analyses of soil fractions without including the organic matter. The organic fraction contains plant-wax markers which may be useful to “fingerprint” forensic soils due to their chemical diversity, relative longevity and resistant nature. The aim of this study was to test the long- (kilometre), medium- (metre) and short- (centimetre) scale variability of plant-wax biomarkers in a forensic context in anthropised urban soils and soils developed under subtropical conditions. Two areas from the Curitiba municipality and two areas from the Colombo municipality, Paraná State, South Brazil, were selected. Soil colour analysis was carried out to obtain reflectance data over the 360–740 nm wavelength range. Furthermore, plant-wax marker compounds (n-alkanes and fatty-alcohols) were assessed by extraction and separation into different classes and an analysis of the compounds by gas chromatography (GC/MS). The compositions of the wax-marker profiles were different in samples collected side-by-side, showing sensitivity to local variations under subtropical conditions and in areas under intense human urban disturbance. Under these conditions, biomarkers may be used in real crime scenes, even on a micrometric scale of variation.

Colocasia esculenta L. (Schoot) is a widely used plant in developing countries in Asia, Africa, and Central America. The parts of the Colocasia plant that can be used are the tubers, stalks, and leaves. There has not been much research on the content of this plant. The study aims to determine the flavonoids and terpenoids in the ethanol extract of Colocasia stalk and leaves. The maceration method with 70% ethanol solvent and drying with vacuum evaporator. Determination of flavonoid levels based on quercetin marker compounds using AlCl3 reagent by UV-Vis spectrophotometry. The terpenoid levels were carried out gravimetrically with petroleum ether as a solvent. The results showed that the ethanol extract of Colocasia stalk and leaf contained flavonoids, respectively 3.18 +0.0581% and 4.33 +0.0285%, while the results of the terpenoid levels for stalks were 7.10 +0.0676% and leaves were 8.39 +0.0023%.

Lobelia chinensis Lour. (L. chinensis) has traditionally been used as a treatment for snake bites, high fever, jaundice, edema, and diarrhea, and modern studies have reported its anti-inflammatory, antioxidant, and antiviral activities. L. chinensis contains various compounds, such as flavonoids and coumarins, and its flavonoid components have been identified in many studies. In this study, a high-performance liquid chromatograph equipped with a photodiode array (PDA) detector and an Aegispak C18-L reverse-phase column (4.6 mm × 250 mm i.d., 5 μm) was used to simultaneously analyze four marker components in L. chinensis for standardization purposes. HPLC-PDA (detection at 340 nm), performed using a 0.1% formic acid-water/0.1% formic acid-acetonitrile gradient, separated the four marker compounds: luteolin-7-O-β-d-glucuronopyranosyl (1→2)-O-β-d-glucuronopyranoside, clerodendrin, chrysoeriol-7-O-diglucuronide, and diosmin. The developed analytical method showed excellent linearity values (r2 > 0.9991), limits of detection (LODs: 0.376–2.152 μg/mL), limits of quantification (LOQs: 1.147–6.521 μg/mL), intra- and inter-day precisions (RSD < 1.96%), and analyte recoveries (96.83–127.07%; RSD < 1.73%); thus, it was found to be suitable for the simultaneous analysis of these four marker compounds in L. chinensis.

Methods for a dissolution study by ultra-high performance liquid chromatography/triple quadrupole mass spectrometry (UHPLC-QqQ/MS) analysis of grape polyphenol dietary supplements, namely, grape seed extract (GSE) and resveratrol (RSV) capsules, were developed following the guidance of United States Pharmacopeia (USP) &lt;2040&gt;. Two dissolution media, 0.1 N hydrochloric acid (pH 1.2) and 0.05 M acetate buffer (pH 4.6), were evaluated with dissolution apparatus (USP 1), 100 rpm rotation speed, and 900 ml dissolution medium volume. Dissolution profiling was performed over 120 min. Major phenolic compounds of gallic acid, catechin, epicatechin, and procyanidin B2 were quantitated to obtain the dissolution profile of GSE capsules, and trans-RSV was used for RSV capsules. Results indicated that the released trans-RSV for RSV capsules in both of the dissolution media meets the USP standards, and that for the GSE capsules, all the four marker compounds passed the dissolution test in the HCl medium but did not reach a 75% release within 60 min in the acetate buffer. These promising results suggest that the general USP dissolution protocols are adequate for the successful release of RSV capsules in HCl medium and acetate buffer and GSE capsules (in HCl medium), but may be inadequate for GSE capsules in acetate buffer. These results showed that under a low pH of 1.2 (simulated stomach environment), bioactive compounds were released on time from the GSE capsules and met the USP guidelines; however, under a higher pH of 4.6 (simulated duodenum environment), the same biomarkers failed, suggesting the need to further improve the dissolution of GSE over a wider range of pH environments to enhance bioavailability and efficacy.

Clove (Syzygium aromaticum) is one of the most precious spices that has been used since ages. Cloves are well-known for their many medicinal properties. It is also widely used in food, flavours and cosmetics industry. One of the main usage of cloves is in the dentistry field to treat bad odour, toothache etc. Though it is native to Indonesia, it has been cultivated in several parts of the world. Clove oil is the richest source of phenolic compounds such as eugenol, eugenyl acetate. It is most powerful source of antioxidant as per oxygen radical absorption capacity. Many review articles had already been published on clove oil which elucidate its morphology, biological and pharmacological benefits. The characteristics of this review study is the exploration and elaborate discussion about extraction methods, discrepancies between the chemical composition due to geographic conditions and identification of marker compounds in leaf, stem and bud clove oil. The comprehensive review revealed that the clove oil mainly consists of phenolic compounds such as eugenol, eugenyl acetate followed by sesquiterpene namely beta-caryophyllene.

The leaves of African pumpkins (Momordica balsamina L.) are a commonly consumed traditional vegetable. They are a good source of polyphenolic antioxidants and carotenoids, which are, however, affected by cooking or digestion. We investigated the effect of household cooking methods (stir-frying or boiling) on the changes in bioactive metabolites, antioxidant capacity, release and accessibility of β-carotene and also inhibition of inhibitory activity against α-amylase and α-glucosidase enzymes during in vitro digestion of African pumpkin leaves compared to the raw leaves. Compared to boiled or raw leaves, stir-frying improved the availability of bioactive metabolites at the gastrointestinal phase. Quercetin 3-galactoside and rhamnetin 3-O-glucoside (marker compounds) discriminated the stir-fried leaves from raw leaves and boiled leaves after digestion. Stir-frying improved the release and accessibility of β-carotene and enhanced the antioxidant activities compared to boiling. Dialysable fractions of stir-fried leaves exhibited the greatest inhibitory activity against α-amylase and α-glucosidase enzymes compared to the raw and boiled leaves, as well as acarbose. Stir-frying, therefore, is recommended for use in household cooking to benefit consumers by increasing the intake of phenolics and β-carotene.

This work investigated the microbiological quality and chemical profiles of two different dairy creams obtained by centrifugation vs. natural creaming separation systems. To this aim, an untargeted metabolomics approach based on UHPLC-QTOF mass spectrometry was used in combination with multivariate statistical tools to find potential marker compounds of the two different types of two dairy creams. Thereafter, we evaluated the chemical, microbiological and sensorial changes of a ricotta cheese made with a 30% milk cream (i.e., made by combining dairy creams from centrifugation and natural creaming separation) during its shelf-life period (12 days). Overall, microbiological analysis revealed no significant differences between the two types of dairy creams. On the contrary, the trend observed in the growth of degradative bacteria in ricotta during shelf-life was significant. Metabolomics revealed that triacylglycerols and phospholipids showed significant strong down-accumulation trends when comparing samples from the centrifugation and natural creaming separation methods. Additionally, 2,3-Pentanedione was among the best discriminant compounds characterising the shelf-life period of ricotta cheese (VIP score = 1.02), mainly related to sensorial descriptors, such as buttery and cheesy. Multivariate statistics showed a clear impact of the shelf-life period on the ricotta cheese, revealing 139 potential marker compounds (mainly included in amino acids and lipids). Therefore, the approach used showed the potential of a combined metabolomic, microbiological and sensory approach to discriminate ricotta cheese during the shelf-life period.

This work aims to explore the contribution of prickly ash (Zanthoxylum bungeanum Maxim) on the taste perception of stewed sheep tail fat. Liquid chromatography-tandem quadrupole time of flight mass spectrometry (LC-QTOF-MS) was applied to analyze the taste-related compounds. A total of 99 compounds in different sheep tail fat samples were identified. The semi-quantitative results showed that there were differences between the samples. The partial least squares discriminant analysis (PLS-DA) model without overfitting was used to investigate the effect of prickly ash. Eleven marker compounds were predicted with a variable importance for projection > 1, fold change > 2 and p < 0.05. An additional experiment showed that guanosine 5′-monophosphate, malic acid, inosine and adenosine 5′-monophosphate could improve the umami and saltiness taste of stewed sheep tail fat.
