Abstract Background Enteritis is a common cause of morbidity and mortality in lorikeets that can be challenging to diagnose and treat. In this study, we examine gut microbiota in two lorikeet flocks with enteritis (Columbus Zoo and Aquarium—CZA; Denver Zoo—DZ). Since 2012, the CZA flock has experienced repeated outbreaks of enteritis despite extensive diet, husbandry, and clinical modifications. In 2018, both CZA and DZ observed a spike in enteritis. Recent research has revealed that the gut microbiota can influence susceptibility to enteropathogens. We hypothesized that a dysbiosis, or alteration in the gut microbial community, was making some lorikeets more susceptible to enteritis, and our goal was to characterize this dysbiosis and determine the features that predicted susceptibility. Results We employed 16S rRNA sequencing to characterize the cloacal microbiota in lorikeets (CZA n = 67, DZ n = 24) over time. We compared the microbiota of healthy lorikeets, to lorikeets with enteritis, and lorikeets susceptible to enteritis, with “susceptible” being defined as healthy birds that subsequently developed enteritis. Based on sequencing data, culture, and toxin gene detection in intestinal contents, we identified Clostridium perfringens type A (CZA and DZ) and C. colinum (CZA only) at increased relative abundances in birds with enteritis. Histopathology and immunohistochemistry further identified the presence of gram-positive bacilli and C. perfringens, respectively, in the necrotizing intestinal lesions. Finally, using Random Forests and LASSO models, we identified several features (young age and the presence of Rhodococcus fascians and Pseudomonas umsongensis) associated with susceptibility to clostridial enteritis. Conclusions We identified C. perfringens type A and C. colinum associated with lorikeet necrohemorrhagic enteritis at CZA and DZ. Susceptibility testing of isolates lead to an updated clinical treatment plan which ultimately resolved the outbreaks at both institutions. This work provides a foundation for understanding gut microbiota features that are permissive to clostridial colonization and host factors (e.g. age, prior infection) that shape responses to infection.

I wanted to tell a story from the point of view of a data entry worker, someone not typically considered when the advantages and disadvantages of eldercare technologies are being assessed. The worker, remotely located and unknown to the people she watches, offers a perspective that points out some of the strangeness of watching from a distance a living person being injured, struggling, or just needing human care and attention. As we move into more widespread use of eldercare technologies, it’s worth considering how surveillant and robotic technologies to support independent elder living could change our relationships with and sensitivity to older populations.

AbstractDespite the growing need to use conservation breeding and translocations in species’ recovery, many attempts to reintroduce animals to the wild fail due to predation post-release. Released animals often lack appropriate behaviours for survival, including anti-predator responses. Anti-predator training—a method for encouraging animals to exhibit wariness and defensive responses to predators—has been used to help address this challenge with varying degrees of success. The efficacy of anti-predator training hinges on animals learning to recognize and respond to predators, but learning is rarely assessed, or interventions miss key experimental controls to document learning. An accurate measure of learning serves as a diagnostic tool for improving training if it otherwise fails to reduce predation. Here we present an experimental framework for designing anti-predator training that incorporates suitable controls to infer predator-specific learning and illustrate their use with the critically endangered Hawaiian crow, ‘alalā (Corvus hawaiiensis). We conducted anti-predator training within a conservation breeding facility to increase anti-predator behaviour towards a natural predator, the Hawaiian hawk, ‘io (Buteo solitaries). In addition to running live-predator training trials, we included two control groups, aimed at determining if responses could otherwise be due to accumulated stress and agitation, or to generalized increases in fear of movement. We found that without these control groups we may have wrongly concluded that predator-specific learning occurred. Additionally, despite generations in human care that can erode anti-predator responses, ‘alalā showed unexpectedly high levels of predatory wariness during baseline assessments. We discuss the implications of a learning-focused approach to training for managing endangered species that require improved behavioural competence for dealing with predatory threats, and the importance of understanding learning mechanisms in diagnosing behavioural problems.

The last few decades have seen an outpouring of gastrointestinal (GI) microbiome studies across diverse host species. Studies have ranged from assessments of GI microbial richness and diversity to classification of novel microbial lineages. Assessments of the “normal” state of the GI microbiome composition across multiple host species has gained increasing importance for distinguishing healthy versus diseased states. This study aimed to determine baselines and trends over time to establish “typical” patterns of GI microbial richness and diversity, as well as inter-individual variation, in three populations of western lowland gorillas (Gorilla gorilla gorilla) under human care at three zoological institutions in North America. Fecal samples were collected from 19 western lowland gorillas every two weeks for seven months (n = 248). Host identity and host institution significantly affected GI microbiome community composition (p < 0.05), although host identity had the most consistent and significant effect on richness (p = 0.03) and Shannon diversity (p = 0.004) across institutions. Significant changes in microbial abundance over time were observed only at Denver Zoo (p < 0.05). Our results suggest that individuality contributes to most of the observed GI microbiome variation in the study populations. Our results also showed no significant changes in any individual’s microbial richness or Shannon diversity during the 7-month study period. While some microbial taxa (Prevotella, Prevotellaceae and Ruminococcaceae) were detected in all gorillas at varying levels, determining individual baselines for microbial composition comparisons may be the most useful diagnostic tool for optimizing non-human primate health under human care.

Abstract Background Elephant endotheliotropic herpesvirus haemorrhagic disease (EEHV HD) is the leading cause of death in captive Asian elephant calves in Asia, North America, and Europe with a mortality rate of ~ 65% in calves that are under human care. Although EEHV HD was first found in elephant camps, more recently it was identified in wild populations which poses a greater threat to the elephant population. Deaths due to EEHV HD have been seen in wild elephants, but the in-situ prevalence and mortality rate is unknown. We report the first EEHV HD cases in Malaysia from 3 wild born endangered Bornean elephant calves from Sabah with known typical clinical signs. Case presentation The first calf died within 24 h of the onset of clinical signs; the second calf died within 12 h of the onset of clinical signs. The third calf succumbed within 72 h. Necropsies revealed that all 3 calves had similar presentations of EEHV HD but in the third calf with less severity. We conducted conventional polymerase chain reaction (cPCR) assays and found EEHV DNA at all 7 loci in the 3 calves; it was identified as EEHV1A, the virus type that has been found in most other reported cases. Conclusion Typical EEHV HD clinical signs and the molecular confirmation of EEHV by cPCR and sequencing point to EEHV as the cause of death. Further genetic investigation of the strain is in progress.

Emotions are an essential part of how we experience our world. Humans can express emotions by telling others how we feel—but what about animals? How can we tell whether they experience emotions and, if they do, which ones? When we think about the animals under human care, it is not only scientifically interesting but also ethically important to understand how these animals experience their worlds. Over the last 20 years, researchers have made considerable progress by identifying ways to assess emotions in animals. For example, researchers can look at the facial expressions of animals, record their vocalisations, or measure body processes such as changes in the heartbeat or hormone concentrations in the blood. This information can tell us more about how animals feel, why and how emotions have evolved, and what we, as humans, share with animals in our emotional experience of the world around us.

Abstract Objective Here we tested the application of a full mouth speculum to sedated elephants in human care to gain access to the oral cavity, the trachea (bronchi) and esophagus (stomach) and therefore improve diagnostic and therapeutic options in elephant medicine. The construction of this oral speculum for elephants and the procedure are described. Material and methods The oral speculum is a steel construction consisting of 2 bite plates of 0.8 × 60.0 × 8.0 cm attached between 2 threaded guiding poles (40 cm). Through crank handles, the metal plates are dispersed once placed between the elephant‘s jaws in front of the molars. The oral speculum was applied in 26 elephants (6,16 Asian elephants, and 1,3 African elephants) during standing sedation. Results All sedated elephants tolerated the positioning of the mouth opener and subsequent manipulations well. The mouth opener was applied for the following procedures: inspection of the oral cavity (n = 2), placing a stomach tube (n = 16), and/or performing endoscopic examinations such as bronchoscopy (n = 20) and/or gastroscopy (n = 8). Conclusion This method provides a new possibility to open the jaws to gain access to the molars, larynx and pharynx in captive elephants without full immobilization. Valuable samples for diagnostics may be obtained or animals medicated via stomach tube with this application. Clinical relevance The mouth opener provides veterinarains with a new option to perform necessary diagnostic and therapeutic procedures around the oral cavity, airways and stomach in captive elephants during standing sedation with no need for a full anaesthesia.

To optimize the health evaluation of Giant and Neotropical otters in the wild and under human care and  to guarantee the quality of the information collected and maximize efforts in research projects, Brazilian veterinarians developed two protocols: 1) Protocol for collection, processing and storage of biological samples from live Giant and Neotropical otters, 2) Protocol for collection of biological material during necropsy.

Studies on how visitors affect penguins in human care report a mixture of negative, neutral, and positive impacts on behavior and physiology. Swimming is a highly motivated behavior that may promote positive welfare in penguins. We investigated how visitor crowd size, composition, and noise levels impact pool use in a mixed-species colony housing king (Aptenodytes patagonicus; n = 20), gentoo (Pygoscelis papua; n = 14), and southern rockhopper (Eudyptes chrysocome; n = 24) penguins. We used video and sound loggers to record if penguins were on land or in water, the number of human adults and children present, and noise levels using 5-minute scan samples from 09:00-15:00 over 36 continuous days. Data were analyzed using linear mixed models with proportion of penguins in the water as the dependent variable and crowd size, composition, and noise levels in A-weighted (dBA) and C-weighted (dBC) scales as independent variables. Crowd size was positively associated with pool use in gentoo penguins. Crowd composition did not predict pool use in any species. Noise levels in dBA, which is adjusted to the higher frequencies of human hearing, positively predicted pool use in southern rockhopper penguins. Noise levels in dBC, which captures lower frequencies, did not predict pool use in any species. No evidence of negative visitor effects was observed. Instead, these results suggest visitors are a neutral stimulus to king penguins and may be enriching to gentoo and southern rockhopper penguins.

Toxoplasma gondii infection has been reported in numerous species of marine mammals, some of them with fatal consequences. A serosurvey for T. gondii infection was conducted in pinnipeds from an oceanographic park in Portugal (n = 60); stranded pinnipeds on the Portuguese coast (n = 10); and pinnipeds captured in Lorenzensplate, Germany (n = 99). Sera from 169 pinnipeds were tested for the presence of antibodies to T. gondii by the modified agglutination test with a cut-off titre of 25. An overall seroprevalence of 8.9% (95% confidence interval: 5.1–14.2) was observed. Antibody titres of 25, 50, 100, 1600 and ≥3200 were found in five (33.3%), two (13.3%), five (33.3%), one (6.7%) and two (13.3%) animals, respectively. Pinnipeds under human care had a seroprevalence of 20.0% (12/60), in contrast to 2.8% (3/109) in wild pinnipeds (p < 0.001). General results suggest a low exposure of wild pinnipeds to T. gondii, while the seroprevalence found in pinnipeds under human care highlights the importance of carrying out further studies. This is the first serological survey of T. gondii in pinnipeds in Portugal and the first infection report in South African fur seal (Arctocephalus pusillus pusillus).
