Exogenous carbohydrases are commonly added to monogastric animal feed to degrade non-starch polysaccharides (NSP) to improve zootechnical performance and nutrient digestion. In the current study, the effects of dietary supplementation of a new, intrinsically thermostable, monocomponent xylanase on intestinal parameters, zootechnical performance and carcass traits of broilers were evaluated. A total of 720 1-day-old broilers were randomly allotted to four wheat-based diets with different levels of xylanase (T1: control; T2: 30,000 U/g; T3: 45,000 U/g; T4: 90,000 U/g) and fed from 1 to 35 days of age. The data showed that xylanase supplementation reduced the intestinal viscosity significantly, especially in the ileum. Duodenum and cecum pH was not influenced by xylanase supplementation, while T3 treatment showed the lowest pH value in jejunum and ileum. Xylanase supplementation reduced significantly feed conversion ratio (FCR) without affecting feed intake (FI). T2 treatment exhibited a higher body weight gain (BWG) compared the other treatments. Live weight, carcass weight, legs, and liver weight were significantly higher in T3 and T4 treatments compared to T1. No significant differences on foot pad lesions were observed among the four treatments. In conclusion, dietary supplementation of this new xylanase significantly improves intestinal tract viscosity and affects beneficially broilers’ performance and carcass traits.

Soluble adhesion molecules (mainly VCAM-1 and E-selectin) have a vital role in the pathogenesis of the rheumatoid arthritis (RA) and consider as angiogenic mediators for this disease. The main goal for this research is to evaluate the efficacy of orally administer niclosamide (NCS) in prevention the angiogenic mediators (VCAM-1 and E-selectin) using collagen induced arthritis model in rats (CIA). Fifty male Spraque-Dawley rats underwent collagen induced arthritis (CIA) model. When arthritis was fully developed, the rats were either treated orally with low-dose (50mg/kg) NCS or high-dose (100mg/kg) NCS or treated intrapertonially (IP) with 30mg/kg NCS or leave without treatment for 4 weeks. Body weight measurement and arthritis index were monitor before and after treatment in all groups. At the end of the treatment period serum level of vascular cell adhesion molecule-1 (VCAM-1), E-selectin and TNFα were measured together with collection of articular synovial tissue to evaluate the pathological changes. The experiment showed that NCS significantly reduce the arthritis index, foot pad thickness and ankle swelling (p value < 0.05) when given orally in a high dose and IP to the experimental animal. Comparing to the CIA model group, significant reduction in the serum level of VCAM1 and E-selectin has been observed in those rats treated with high dose of oral NCS or IP injection of NCS (p value < 0.05). Niclosamide can effectively decrease, in dose dependent manner, the clinical scores, joint swelling, VCAM1, E-selectin and pathological changes in arthritic rats induced by collagen type II.

Poultry producers accustomed to use light colours in broiler production as atrial to increase their productivity, especially in the developing countries to cover the animal protein gap. This experiment was planned to study the effect of different blue light colour intensities (high, medium and dim) on a recently imported Indian River (IR) broilers to Egypt. In this study, 270 one-day old Indian River broiler chicks were used. The birds were exposed to high blue light intensity (HBLI), medium blue light intensity (MBLI) and dim blue light intensity (DBLI), through a monochromatic lighting system that was installed in different rooms for 24 hours daily. The birds were randomly divided and housed into three well controlled pens of 5.46 m2 with three replicates of 30 each using a density of 17 birds/m2 in the room. The results showed that the broilers reared under DBLI had a significantly (p< 0.05) higher body weight, body weight gain, Newcastle disease virus antibody titer and foot pad dermatitis with obviously, economic Feed conversion ratio (FCR) and low activities and heterophyil/lymphocyte ratio in comparing with (MBLI) and (HBLI). In conclusion, poultry producer can use dim blue light in their farms to reduce the activities and increase the productivity of the birds.

This study aimed to investigate the possibility of rearing newly hatched chicks with immediate access to feed and water in the same hatching unit one week prior to transferring them to the conventional broiler house with special regards to foot pad health and growth performance. Two trials were performed with a total of 6900/6850 (trials 1/2) broiler chickens (ROSS 308). A total of 3318/3391 chicks (trials 1/2) were transported from the hatchery (duration of about 3 h) and reared in a conventional broiler house (control group: delayed nutrition on-hatchery hatched). The control group did not receive any form of nutrition until they were taken to conventional broiler housing. Additionally, a total of 3582/3459 (trials 1/2) embryonated eggs (d 18) were obtained from the same parent flock of the same commercial hatchery and taken to the farm facility. After on-farm hatch, the chicks had immediate access to water and feed (experimental group: early nutrition on-farm hatched). After d 6/7 of life, the on-farm hatched chicks (trials 1/2) were transferred to the broiler house on the same facility. The delayed nutrition on-hatchery hatched groups displayed a significantly lower dry matter content in the litter compared to the early nutrition on-farm hatched groups (two-factorial analysis) at d 6/7 and d 14 of life. However, thereafter, no significant differences were noted. Based upon two-factorial analysis, the early nutrition on-farm hatched groups revealed lower foot pad lesions from d 14 of life onwards and showed a higher body weight (BW) throughout the rearing period compared to the delayed nutrition on-hatchery hatched groups (p < 0.05). Overall, early nutrition on-farm hatched chickens is of critical importance together with using new litter at d 7 to maintain healthy foot pads as well as to enhance nutrient utilization and optimize the growth performance.

The poultry sector is moving towards antibiotic-free production, both to challenge the increasing spread of the antibiotic resistance phenomenon and to meet market demands. This could negatively impact the health and welfare of the animals. In this study, the welfare of 14 batches of 41–47-day-old broilers raised by the same integrated company with and without antibiotics was assessed using the Welfare Quality® protocol. The total welfare score did not significantly differ between the two systems: the good-feeding principle was, on average, higher in the conventional batches, with statistical significance (t = −2.45; p = 0.024), while the other welfare principles (good housing, good health and appropriate behaviour) were slightly better in the antibiotic-free batches. Despite stocking densities averagely higher in the antibiotic-free batches, the absence of antibiotics did not seem to impact the good-health principle; in particular, hock burns, foot pad dermatitis and lameness were significantly less severe in the antibiotic-free batches (p < 0.0001, p = 0.018, p < 0.0001, respectively), which showed also a lower death rate (2.34% vs. 2.50%). Better management of antibiotic-free batches was reported, particularly concerning litter conditions. Further studies would be required to identify and standardise a set of managerial methodologies in order to improve the health of broilers raised without antibiotics.

We compared closed- and open-sided industrial houses with respect to the welfare of broiler chickens in southern Brazil. Ten flocks from each design were evaluated and measures divided into the following categories: i) bird health: contact dermatitis on the breast and abdominal areas, bird soiling, foot-pad dermatitis, hock burn, lameness, fractures, bruising, scratches, dead on arrival, diseases; ii) environmental measurements: relative humidity, temperature, air velocity, ammonia (NH3), carbon dioxide (CO2), light intensity, litter moisture; iii) behaviour: bird behaviour, touch test; and iv) affective states: qualitative behaviour assessment. Closed-sided houses showed worse contact dermatitis on the breast and abdominal areas, lower exploratory behaviour prevalence, higher NH3 (11.2 [± 6.8] vs 7.5 [± 3.9] ppm) and CO2 (1,124.9 [± 561.5] vs 841.0 [± 158.0] ppm), lower light intensity (6.9 [± 6.3] vs 274.2 [± 241.9] lux), while open-sided houses had a higher prevalence for scratches and panting behaviour, and lower air velocity (2.1 [± 0.7] vs 1.1 [± 1.0] m s–1). Stocking densities of 13.9 (± 0.4) and 12.0 (± 0.3) per m2 for closed- and open-sided houses, respectively, likely influenced some results. All values shown are means (± SD). Even though open-sided houses presented fewer animal welfare restrictions (according to five indicators as opposed to three for closed-sided houses), both revealed important welfare problems, evidenced by poor environmental indicators, behavioural restrictions and injuries.

Abstract BackgroundOrofacial granulomatosis (OFG) is a granulomatous inflammation (GI) disease in maxillofacial region, the underlying cause of it remains unknown. Our previous study demonstrated that tooth apical periodontitis (AP) plays a significant role in the pathogenesis of OFG, we aimed here to characterize the AP bacterial signatures of OFG patients, and identify bacteria that may be important pathogens capable of inducing OFG.ResultsThe composition of AP microbiota in OFG cases and common AP controls was compared using 16S rRNA gene sequencing, the results showed a specific AP microbiota signature in OFG patients, characterized by domination of phyla Firmicutes and Proteobacteria , notably members of Streptococcus, Lactobacillus and Neisseria. To assess the pathogenicity of the potential pathogens in OFG, we isolated and successfully in vitro cultured Streptococcus, Lactobacillus casei, Neisseria subflava, Veillonella parvula and Actinomyces from OFG patients, and injected the clinical isolates into mice respectively. Ultimately, foot pad injection with N. subflava elicited granulomatous inflammation, and the virulence of N. subflava was verified based on Koch’s postulates.ConclusionsOur findings confirmed the role of bacteria in OFG, and first suggested that the component of the host normal microbiota, N. subflava is likely a pathogen for GI.

The RNA-binding protein tristetraprolin (TTP) is an anti-inflammatory factor that prompts the mRNA decay of target mRNAs and is involved in inflammatory diseases such as rheumatoid arthritis (RA). TTP is regulated by phosphorylation, and protein phosphatase 2A (PP2A) can dephosphorylate TTP to activate its mRNA-degrading function. Some small molecules can enhance PP2A activation. Short interfering RNA (siRNA) targeting TTP expression or PP2A agonist (Arctigenin) was administered to monosodium urate (MSU) crystal-induced J774A.1 cells, and the expression of inflammatory related genes was detected by RT-PCR and Western blot assays. The effects of Arctigenin in mouse models of acute inflammation induced by MSU crystals, including peritonitis and arthritis, were evaluated. The data indicated that TTP expression levels and endogenous PP2A activity were increased in MSU-crystal treated J774A.1 cells. TTP knockdown exacerbated inflammation-related genes expression and NLRP3 inflammasome activation. However, PP2A agonist treatment (Arctigenin) suppressed MSU crystal-induced inflammation in J774A.1 cells. Arctigenin also relieved mitochondrial reactive oxygen species (mtROS) production and improved lysosomal membrane permeability in MSU crystal-treated J774A.1 cells. Moreover, TTP knockdown reversed the anti-inflammatory and antioxidant effects of Arctigenin. Oral administration of Arctigenin significantly alleviated foot pad swelling, the number of inflammatory cells in peritoneal lavage fluids and the production of IL-1β in the mouse model of inflammation induced by MSU crystals. Collectively, these data imply that targeting TTP expression or functional activity may provide a potential therapeutic strategy for inflammation caused by MSU crystals.

AbstractThe use of modern chicken genotypes with high egg or meat performance results in the ethically unacceptable practice of culling day-old male layer chicks because of their inefficient fattening performance. Dual-purpose genotypes with a balanced performance profile for both eggs and meat are one option to avoid this practice. In this study, four chicken crosses of a layer breed (White Rock or New Hampshire) and the meat breed Bresse Gauloise were compared under the conditions of organic agriculture. Purebred Bresse Gauloise and the layer hybrid Lohmann Sandy served as controls. Part 1 of this study focused on the fattening performance of the cockerels, which were reared together with the pullets. The birds were housed in a floor system (9.9–20.7 kg live weight per m2 at the end of week 15) with access to a green outdoor run. Live weight of the crosses before slaughter at the age of 15 weeks ranged between 2355 and 2447 g and did not differ significantly between the genotypes. With average daily gains of 22.1–22.8 g, the crosses grew slower than Bresse Gauloise males (26.1 g) but faster than Lohmann Sandy males (15.9 g). Welfare assessment indicated a generally high level of welfare with no foot pad lesions or hock burns on any of the cockerels. Fattening dual-purpose cockerels can therefore be an ethically desirable option with a high level of animal welfare and a better fattening performance than in male layer chicks, but still requires more resources than the fattening of specialised broilers.
