The integration of untargeted metabolomics and transcriptomics from the same population of cells or tissue enhances the confidence in the identified metabolic pathways and understanding of the enzyme–metabolite relationship. Here, we optimised a simultaneous extraction method of metabolites/lipids and RNA from ependymoma cells (BXD-1425). Relative to established RNA (mirVana kit) or metabolite (sequential solvent addition and shaking) single extraction methods, four dual-extraction techniques were evaluated and compared (methanol:water:chloroform ratios): cryomill/mirVana (1:1:2); cryomill-wash/Econospin (5:1:2); rotation/phenol-chloroform (9:10:1); Sequential/mirVana (1:1:3). All methods extracted the same metabolites, yet rotation/phenol-chloroform did not extract lipids. Cryomill/mirVana and sequential/mirVana recovered the highest amounts of RNA, at 70 and 68% of that recovered with mirVana kit alone. sequential/mirVana, involving RNA extraction from the interphase of our established sequential solvent addition and shaking metabolomics-lipidomics extraction method, was the most efficient approach overall. Sequential/mirVana was applied to study a) the biological effect caused by acute serum starvation in BXD-1425 cells and b) primary ependymoma tumour tissue. We found (a) 64 differentially abundant metabolites and 28 differentially expressed metabolic genes, discovering four gene-metabolite interactions, and (b) all metabolites and 62% lipids were above the limit of detection, and RNA yield was sufficient for transcriptomics, in just 10 mg of tissue.

Pandan (Pandanus amaryllifolius Roxb.) leaves are widely used in Malaysia as a source of natural flavoring.  The major compound contributing to the characteristic flavour of Pandan is 2-acetyl-1-pyrroline (2AP). As the consumer requirement for use of natural flavours, extraction of components from natural sources has been sought. In this study, solvent extraction of 2AP from Pandan leaves was performed. The effect of solvent used during extraction process (i.e. methanol, ethanol, propanol) towards the yield of 2AP was investigated. The presence of 2AP was determined using GCMS. The results obtained showed that ethanol was the best solvent to extract 2AP from Pandan leaves compared to methanol as higher 2AP peak arises from ethanol chromatogram.  However there is no 2AP detected when propanol was used as solvent.  It is believed that polarity of the solvent plays an important role in the extraction of 2AP.

Development natural pigment retained a variety of nutrition composition is extremely research value.Water Wax Trees Fruit was as raw materials,citric acid aqueous solution was as extracting agent.Amaranthine pigment extraction method were studied on fully grasping the stability of the natural amaranthine pigment and no damage to the water wax fruit biological activity.The effect of extraction yield were studied on Water Wax Trees Fruit natural amaranthine pigment.The results show that the better extraction conditions are extraction temperature at 70 °C, extraction time for 3h,citric acid concentration1.0 % , the material/liquid ration(g/ml) 1:5.The extraction yield is 2.95%.Ultraviolet absorption maximum wavelength is 280 nm.

Gelatin is an intermediate ingredient which is oftenly used in many field such as food, pharmacy, and cosmetics. It is usually extracted from pig and cow. Halal issue of gelatin sources and the outbreaks of mad cow diseases encouraged people to find an alternative sources of gelatin. One of the alternative sources of gelatin was duck bone. The aim of this research was to describe physicochemical properties of duck bone gelatin which is extracted by using acid extraction method as an alternative sources of halal gelatin. The extraction of duck bone gelatin used 5% concentration of HCl (hydrochloric acid). The extraction process consisted of four steps, they were degreassing, defating, demineralization, and acid extraction. The result showed that gelatin which was extracted from duck bone had these several characteristic: yield of 6.24%, pH 4.0, water content of 13.43%, ash content of 13.42%, protein content of 65.43%, and whiteness degree of 30.35%. Generally, gelatin which was extracted from duck bone had similar characteristic with commercial gelatin and SNI standard. Further researcher had been suggested to reoptimized extraction method in order to reduce ash content.

Biofarmasi 14: 1-9. Curcuma (Curcuma xanthorrhiza Roxb.) is a type of drug plant that has high enough capacity of production in Indonesia. Generally, commerced in the form of fresh curcuma or processed product as simple as simplicia and curcuma powder. Processed products that could be developed is curcuma oleoresin. It is a mixture of essential oils and resins obtained from extraction process of curcuma powder using an organic solvent. Oleoresin has the same flavor and aroma to the extracted material. Due to these characteristics, it is used as a flavor and food coloring, other than as a raw material in pharmaceutical industry. In addition, it also contains active compounds which can support the utilization of drug and food industries. This study aims to determine whether the size of the powder, powdered curcuma immersion time, and interactions between them that can be influenced the content of curcuminoids, total phenol and antioxidant activity of curcuma oleoresin. Selection of solar dryers in the drying process is based on previously studied that compare the natural drying technique with a solar dryer, and the best results of those studies are shown in the solar dryer. This research using completely randomized design with two factors: the size variation of curcuma powder (60, 80 and 100 mesh) and immersion time variation (extraction) of curcuma powder (12, 24 and 36 hours). The results showed that the powder size of curcuma and immersion time has an effect on curcuminoid content, total phenol and antioxidant activity of curcuma oleoresin. However, there are no interaction between both factors. That is, the size and the immersion time of curcuma powder do not affect each other on the content of curcuminoid, total phenol and activity of antioxidant.

By the chemical sequential extraction method, various chemical speciation of phosphorus were extracted from shallow sediments of Nansi Lake. Aiming at the existing problems of lower concentration of phosphorus, the method combined the original sequential extraction and Mo-Sb Anti spectrophotometric method was improved to reduce the interference caused by stratification and chromogenic reaction. In addition, due to the presence of excess sodium dithionite, the extracting solution need to be diluted during occlude phosphorus (Oc-P) extraction process.

Objective: The determination and ratio of ferulic acid and gastrodin in Xiongma Decoction extract by different extraction process were compared, to provide scientific research basis on creating new drugs, controlling the quality combination of modern Compound Chinese, determining the main effect and compatibility. Methods: Chuanxiong and Tianma were extracted by different extraction method such as the boiling method and ethanol extraction by different concentrations to make ​​Xiongma Decoction. The content of ferulic acid and gastrodin were determination by HPLC to make Xiongma Decoction and their ratio was calculated. Chromatographic conditions of ferulic acid: mobile phase: methanol-0.01 % acetic acid solution (30:70, V/V), the detection wavelength: 310nm; chromatographic conditions of Gastrodin: mobile phase: acetonitrile-0.05 % phosphoric acid (3:97, V/V), detection wavelength: 220nm. Results: The determination of ferulic acid was 0.976mg • ml-1, gastrodin was 1.586mg • ml-1, the ratio of ferulic acid and gastrodin was1:1.6 in boiling method; while the determination of ferulic acid was 6.08mg • ml-1, gastrodin was 3.57mg • ml-1, the ratio was 1.7:1 in ethanol extraction. Conclusion: There are different determination and ratio, when there is different extraction process in the same prescription. This may bring about changes the main effect in prescription. This suggests that the ratio of the main active ingredient should be included in the quality inspection, when preparation process of the traditional recipe was changed.

Evaluated with the yield of anise star oil, steam extraction method assisted by microwave is studied for the extraction of anise star oil from Illicium verum Hook. f. in contrast with the traditional steam distillation (SD) method in this paper. The new optimal steam extraction technology assisted by microwave of anise star oil is obtained as: extracting time 80 minutes, solid-liquid ratio(g/mL) 1:7, size of material 80~100 mesh and extracting temperature 80°C. Average yield of anise star oil is up to 11.6% (g/g) under the above technology. Size of material is the most important influencing factor in the extraction process. The optimal traditional steam distillation technology is: extracting time 120 minutes, solid-liquid ratio(g/mL) 1:6, size of material 80~100 mesh and extracting temperature 100°C , the average yield of anise star oil is only 9.81% (g/g). The steam extraction technology assisted by microwave has advantages in both the technological conditions and the yield of anise star oil than the traditional steam distillation technology.

Pectin are polymer of D-galacturonic acid connected by β-1,4 glucosidic. Pectin are widely used in pharmaceutical, food and beverage industries. This research was expected to took advantage of banana peels (Musa paradisiaca) by producing pectin. The research used an extraction method by using acid solvent in additional to ethanol to form the sediment of pectin and the last step is drying in order to get dry pectin. The fixed variables which were used in this research were 10 grams banana peels, 10% water content of sample, and chloride acid solvent. Extraction process was carried out by pH 1; 1,5; 2; extraction time of 70, 80, 90, 100 minute, and temperature of 80 oC, 90 oC. The results showed that the highest yield of pectin extraction was obtained at temperature of 80 oC, pH 1,5 during 80 minute with 11,88% water content, 0,98% ash content, and 3,72% the methoxyl content.

Abstract An enhanced and different method for the active coagulant agent extraction from Moringa Oleifera seeds powder (MOSP) was established and compared to the conventional extraction method in distillate water. In the improved method, MOSP were extracted using sodium chloride as solvent at different concentrations to extract more coagulant agent from Moringa Oleifera and enhance coagulation activity. In this study, MOSP were initially processed and oil content was removed to minimize coagulant concentration usage (MOSP-EO). Moringa Oleifera seeds powder was characterized by both X-ray and FTIR analysis. Ultrasound treatment as well was considered as an additional treatment for MOSP-EO to investigate its effect on coagulant agent extraction process improvement. Coagulation/flocculation experiments were conducted to assess coagulant extraction performance realized through various conditions. The effect of coagulant dosage, solvent concentration and ultrasound exposition duration were investigated for a real effluent of municipal wastewater treatment. Among the three studied NaCl concentrations, 1.0 M was found to be the best solvent concentration for high turbidity removal of more than 97% using 140 mg/L of MOSP-EO compared to extraction in distillate water 88% using 170 mg/L of the same coagulant. NaCl 1.0 M demonstrated the best performance in biochemical oxygen demand (BOD5) removal as well, where more than 98% of municipal wastewater initial BOD5 was eliminated. Mixing MOSP-EO assisted with ultrasound waves at different treatment periods did decrease the active coagulant agent extraction and thus showed its inconvenient for Moringa Oleifera coagulation activity usage.
