Does larval food affect cuticular profiles and recognition in eusocial bees? a test on Scaptotrigona gynes (Hymenoptera: Meliponini)

Abstract Social information is widely used in the animal kingdom and can be highly adaptive. In social insects, foragers can use social information to find food, avoid danger or choose a new nest site. Copying others allows individuals to obtain information without having to sample the environment. When foragers communicate information they will often only advertise high quality food sources, thereby filtering out less adaptive information. Stingless bees, a large pantropical group of highly eusocial bees, face intense inter- and intra-specific competition for limited resources, yet display disparate foraging strategies. Within the same environment there are species that communicate the location of food resources to nest-mates and species that do not. Our current understanding of why some species communicate foraging sites while others do not is limited. Studying freely foraging colonies of several co-existing stingless bee species in Brazil, we investigated if recruitment to specific food locations is linked to (1) the sugar content of forage, (2) the duration of foraging trips and (3) the variation in activity of a colony from one day to another and the variation in activity in a species over a day. We found that, contrary to our expectations, species with recruitment communication did not return with higher quality forage than species that do not recruit nestmates. Furthermore, foragers from recruiting species did not have shorter foraging trip durations than those from weakly-recruiting species. Given the intense inter- and intraspecific competition for resources in these environments, it may be that recruiting species favour food resources that can be monopolised by the colony rather than food sources that offer high-quality rewards.

New plant pathogen invasions typified by cryptic disease symptoms or those appearing sporadically in time and patchily in space, might go largely unnoticed and not taken seriously by ecologists. We present evidence that the recent invasion of Pyrenopeziza plantaginis (Dermateaceae) into the Pacific Northwest USA, which causes foliar necrosis in the fall and winter on Plantago lanceolata (plantain), the primary (non-native) foodplant for six of the eight extant Taylor’s checkerspot butterfly populations (Euphydryas editha taylori, endangered species), has altered eco-evolutionary foodplant interactions to a degree that threatens butterfly populations with extinction. Patterns of butterfly, larval food plant, and P. plantaginis disease development suggested the ancestral relationship was a two-foodplant system, with perennial Castilleja spp. supporting oviposition and pre-diapause larvae, and the annual Collinsia parviflora supporting post-diapause larvae. Plantain, in the absence of P. plantaginis disease, provided larval food resources throughout all butterfly life stages and may explain plantain’s initial adoption by Taylor’s checkerspot. However, in the presence of severe P. plantaginis disease, plantain-dependent butterfly populations experience a six-week period in the winter where post-diapause larvae lack essential plantain resources. Only C. parviflora, which is rare and competitively inferior under present habitat conditions, can fulfill the post-diapause larval feeding requirements in the presence of severe P. plantaginis disease. However, a germination timing experiment suggested C. parviflora to be suitably timed for only Washington Taylor’s checkerspot populations. The recent invasion by P. plantaginis appears to have rendered the ancestrally adaptive acquisition of plantain by Taylor’s checkerspot an unreliable, maladaptive foodplant interaction.

Deformed wing virus (DWV) is a honeybee viral pathogen either persisting as an inapparent infection or resulting in wing deformity. The occurrence of deformity is associated with the transmission of DWV through Varroa destructor during pupal stages. Such infections with DWV add to the pathology of V. destructor and play a major role in colony collapse in the course of varroosis. Using a recently developed RT-PCR protocol for the detection of DWV, individual bees and mites originating from hives differing in Varroa infestation levels and the occurrence of crippled bees were analysed. It was found that 100 % of both crippled and asymptomatic bees were positive for DWV. However, a significant difference in the spatial distribution of DWV between asymptomatic and crippled bees could be demonstrated: when analysing head, thorax and abdomen of crippled bees, all body parts were always strongly positive for viral sequences. In contrast, for asymptomatic bees viral sequences could be detected in RNA extracted from the thorax and/or abdomen but never in RNA extracted from the head. DWV replication was demonstrated in almost all DWV-positive body parts of infected bees. Analysing individual mites for the presence of DWV revealed that the percentage of DWV-positive mites differed between mite populations. In addition, it was demonstrated that DWV was able to replicate in some but not all mites. Interestingly, virus replication in mites was correlated with wing deformity. DWV was also detected in the larval food, implicating that in addition to transmission by V. destructor DWV is also transmitted by feeding.

The midgut microbiota of the yellow fever mosquito Aedes aegypti impacts pathogen susceptibility and transmission by this important vector species. However, factors influencing the composition and size of the microbiome in mosquitoes are poorly understood. We investigated the impact of larval diet abundance during development on the composition and size of the larval and adult microbiota by rearing Aedes aegypti under four larval food regimens, ranging from nutrient deprivation to nutrient excess. We assessed the persistent impacts of larval diet availability on the microbiota of the larval breeding water, larval mosquitoes, and adult mosquitoes under sugar and blood fed conditions using qPCR and high-throughput 16S amplicon sequencing to determine bacterial load and microbiota composition. Bacterial loads in breeding water increased with increasing larval diet. Larvae reared with the lowest diet abundance had significantly fewer bacteria than larvae from two higher diet treatments, but not from the highest diet abundance. Adults from the lowest diet abundance treatment had significantly fewer bacteria in their midguts compared to all higher diet abundance treatments. Larval diet amount also had a significant impact on microbiota composition, primarily within larval breeding water and larvae. Increasing diet correlated with increased relative levels of Enterobacteriaceae and Flavobacteriaceae and decreased relative levels of Sphingomonadaceae. Multiple individual OTUs were significantly impacted by diet including one mapping to the genus Cedecea, which increased with higher diet amounts. This was consistent across all sample types, including sugar fed and blood fed adults. Taken together, these data suggest that availability of diet during development can cause lasting shifts in the size and composition of the microbiota in the disease vector Aedes aegypti.

Eusocial insects live in teeming societies with thousands of their kin. In this crowded environment, workers combat disease by removing or burying their dead or diseased nestmates. For honey bees, we found that hygienic brood-removal behavior is triggered by two odorants – β-ocimene and oleic acid – which are released from brood upon freeze-killing. β-ocimene is a co-opted pheromone that normally signals larval food-begging, whereas oleic acid is a conserved necromone across arthropod taxa. Interestingly, the odorant blend can induce hygienic behavior more consistently than either odorant alone. We suggest that the volatile β-ocimene flags hygienic workers’ attention, while oleic acid is the death cue, triggering removal. Bees with high hygienicity detect and remove brood with these odorants faster than bees with low hygienicity, and both molecules are strong ligands for hygienic behavior-associated odorant binding proteins (OBP16 and OBP18). Odorants that induce low levels of hygienic behavior, however, are weak ligands for these OBPs. We are therefore beginning to paint a picture of the molecular mechanism behind this complex behavior, using odorants associated with freeze-killed brood as a model.

Abstract Honey is valued for its therapeutic qualities which are attributed among others to its antibacterial multifactorial properties. However, all the factors that influence these properties have not been identified. The present study is focused on the antibacterial action of fatty acids originating from royal jelly, the larval food of honeybees. Aliphatic C8-C12 acids characteristic of this bee product had previously been identified in more than fifty different samples of honey originating from seven countries and in eleven samples of Polish herbhoney. Experiments were performed to ascertain the influence of acidity on the antimicrobial activity of the acids. In acidic nutrient media all tested aliphatic hydroxyacids and unsaturated dicarboxylic acids demonstrated antibacterial action against different microbes with minimal inhibitory concentrations between 0.048 and 3.125 mM. Our results confirm that part of the antibacterial activity of honey contributes to these compounds of bee origin.

Black soldier fly larvae are a sustainable protein source and play a vital role in the emerging food-waste recycling industry. One of the challenges of raising larvae in dense aggregations is their rise in temperature during feeding, which, if not mitigated, can become lethal to the larvae. We propose applying air-fluidization to circumvent such overheating. However, the behavior of such a system involves complex air-larva interactions and is poorly understood. In this combined experimental and numerical study, we show that the larval activity changes the behavior of the ensemble when compared to passive particles such as dead larvae. Over a cycle of increasing and then decreasing airflow, the states (pressure and height) of the live larva aggregates are single-value functions of the flow speed. In contrast, dead larva aggregates exhibit hysteresis characteristic of traditional fluidized beds, becoming more porous during the ramp down of airflow. This history-dependence for passive particles is supported by simulations that couple agent-based dynamics and computational fluid dynamics. We show that the hysteresis in height and pressure of the aggregates decreases as the activity of simulated larvae increases. To test if air fluidization can increase larval food intake, we performed feeding trials in a fluidization chamber and visualized the food consumption via x-ray imaging. Although the food mixes more rapidly in faster airflow, the consumption rate decreases. Our findings suggest that providing moderate airflow to larval aggregations may alleviate overheating of larval aggregations and evenly distribute the food without reducing feeding rates.
