Antibacterial Effect of a Short Peptide, VV18, from Calcineurin-A of Macrobrachium rosenbergii: Antibiofilm Agent Against Escherichia coli and a Bacterial Membrane Disruptor in Pseudomonas aeruginosa

Tagetes lucida Cav. (Asteraceae=Compositae) is used for treating stomach infections. The study focused on evaluating the composition and antimicrobial effect of an extract of T. lucida Cav. The plant extracted with ethanol at 10% w/v, and the extract composition analyzed by Rp-HPLC-MS. The antibacterial effect was evaluated against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Salmonella choleraesuis using disk diffusion, microdilution and bioautography methods. The sytox and comet assays were used to evaluate the mechanism of action. In this way, nine phenolic compounds were detected in the extract of T. lucida. The extract exhibited activity only on S. aureus (MIC of 4.000 mg/ml). The bioautography revealed that the phenolic compounds could act synergistically. The sytox and comet tests showed an antibacterial action of the extract on the bacterial membrane and DNA of this bacterial strain.

An exploratory study based on laboratory experiment was carried out to determine the antibacterial effect of Dimethyl sulfoxide (DMSO) extract of Aloe vera leaf gel (DAE) against standard strains of Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Klebsiella pneumoniae in the Department of Pharmacology & Therapeutics in collaboration with the Department of Microbiology, Mymensingh Medical College, Mymensingh, Bangladesh. DMSO extract was used in five different concentrations (100, 200, 300, 400 and 500 μg/ml). Dose dependent inhibitory effect was seen against the test organisms using disc diffusion method. Zone of inhibition (ZOI) were 8 mm, 13 mm, 15 mm, 16 mm and 21 mm against S. aureus; 0 mm, 8 mm, 13 mm, 15 mm and 18 mm against P. aeruginosa; 8 mm, 11 mm, 13 mm, 16 mm and 20 mm against E. coli; 0 mm, 9 mm, 12 mm, 14 mm and 18 mm against K. pneumoniae at 100, 200, 300, 400 and 500 μg/ml respectively. The minimum inhibitory concentration (MIC) was assessed by broth dilution technique. The MICs of DAE for S. aureus, P. aeruginosa, E. coli and K. pneumoniae were 300 μg/ml, 400 μg/ml, 400 μg/ml and 450 μg/ml respectively. From the study it was observed that DMSO extract of Aloe vera leaf gel possesses antibacterial effect against the test pathogens. The findings highlight the need for further extensive study to detect and isolate the biologically active ingredients present in the Aloe vera leaves which are responsible for antibacterial effect. AbstractA number of nine benzylidene-4-nitroanilines were synthesized by condensation method. The formation of the substituted (E)-N-benzylidene-4-nitrobenzenamines has been confirmed from their physical and Ultra-Violet, Infra-Red, NMR spectral data. The evaluation of antimicrobial screening of substituted (E)-N-benzylidene-4-nitrobenzenamines was conducted by using standard Bauer-Kirby method. Three gram-positive microbes namely Bacillus subtilis, Micrococcus luteus and Staphylococcus aureus, and two gram-negative microbes, Escherichia coli and Pseudomonas aeruginosa, were used for the antibacterial evaluation. The antifungal activities against Aspergillus niger and Penicilium scup fungal species were also performed. A good antibacterial effect has been possessed by some of the substituted (E)-N-benzylidene-4-nitrobenzenamines on the microorganisms utilized in the present study

Due to the lack of raining levels, accretion population growth and increasing pollution in the water resources, the world nowadays is facing a real challenge of providing enough drinking water supplies. Many common species of bacteria could be founded in the water resources like Escherichia coli and Pseudomonas aeruginosa. This paper is an attempt to find an effective agent to eliminate these bacteria. silver nanoparticles (AgNPs) have been tested against these bacteria. Sliver nanoparticles were synthesized by wet chemistry method and were dispersed in an aqueous suspension and prepared in different concentrations. Also, some common antibiotics have been tested against the selected bacteria including; gentamycin (GN), cephalexin (KF), erythromycin (E), ampicillin (AM) and amoxicillin (AMX). The results confirmed that silver nanoparticles had a good antibacterial effect against the selected bacteria in comparison with the tested antibiotics.

Vicarchika is a type of Kushta Roga (skin disease). It can be correlated with eczema. Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa are some of the bacteria which cause skin diseases. Siddhartaka Snana is a herbal bath that is described in Charaka Samhita as a treatment for Kushta Roga. Siddhartaka Snana is a herbal preparation which consists ten herbs namely; Cyperus rotundus, Catunaregum spinosa, Phyllanthus emblica, Terminalia chebula, Terminalia bellarica, Cassia fistula, Pongamia pinnata, Holarrhena antidysentrica, Alastonia scholaris and Coscinum fenestratum. Antibacterial effect of Siddhartaka Snana against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa that are responsible for skin diseases was studied using disc diffusion method and well diffusion methods. In disc diffusion method, no inhibition zone was observed with any of the tested bacteria. However, an inhibition zone of 9.67mm was observed against Staphylococcus aureus in well diffusion method with Siddhartaka Snana. Minimum Inhibitory Concentration (MIC) of Siddhartaka Snana against Staphylococcus aureus was 0.9225 mg/1ml. Ingredients of Siddhartaka Snana contain tannins, phenols and flavonoids, all of which possess antibacterial properties. Antibacterial effect of Siddhartaka Snana may have been exhibited due to actions of these phytochemicals. It was concluded that Siddhartaka Snana has an antibacterial effect against Staphylococcus aureus.

Background. Autologous platelet-rich gel (APG) is an effective method to improve ulcer healing. However, the mechanisms are not clear. This study aimed to investigate the antibacterial effect of APG in vitro.Methods. Platelet-rich plasma (PRP), platelet-poor plasma (PPP) and APG were prepared from whole blood of sixteen diabetic patients with dermal ulcers. Antibacterial effects againstStaphylococcus aureus,Escherichia coli, andPseudomonas aeruginosawere evaluated by bacteriostasis assay of APG, PRP, and APG-APO (APG combined with apocynin), with phosphate-buffered saline (PBS) and PPP as the control group.Results. (1) Compared to the PBS and PPP, the APG and APG-APO groups showed strong antibacterial activity againstStaphylococcus aureus. There was no significant difference(P>0.05)between APG and APG-APO. (2) Compared to PBS, APG, APG-APO, and PRP showed obvious antibacterial effects againstEscherichia coliandPseudomonas aeruginosa. No significant difference(P>0.05)was revealed among the three groups. Compared to the PPP group, they did not show antibacterial effect againstEscherichia coliandPseudomonas aeruginosa(P>0.05).Conclusions. APG has antibacterial effect againstStaphylococcus aureusmediated by platelet activation in the diabetic patients with dermal ulcer, and does not present obvious antibacterial effect againstEscherichia coliorPseudomonas aeruginosa. Combination of APG and antibiotics may have synergistic antibacterial effect.

Copper is a superior antibacterial metal, which is used for various antibacterial products. It is known that the antimicrobial property is generated by direct contact of bacterial cell to the copper surface. Since porous copper has a benefit to increase the surface area contacted to atmospheric medium, it is expected that antibacterial effect can be enhanced. Two kinds of lotus-type porous copper with different directional open pores were used as samples. In order to elucidate such enhancement, four kinds of culture tests on porous copper plates were carried out using bacteria of Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Salmonella enteritidis. While the counts of viable bacteria of Staphylococcus aureus and Escherichia coli cultured on non-porous copper do not change significantly, the counts of Pseudomonas aeruginosa and Salmonella enteritidis decreases. The counts of the viable bacteria of Staphylococcus aureus and Escherichia coli cultured on porous copper drastically decrease, while the counts of Salmonella enteritidis decreases to some degree. However, the porosity does not affect the decrease of counts of Pseudomonas aeruginosa. Thus, apparent antibacterial property of porous copper is attributed to increase of contact area of the bacteria to porous copper for Staphylococcus aureus, Escherichia coli and Salmonella enteritidis.

Piper crocatum is one of medicinal herbal plants with a large number of benefits. Usually herbal plants have activity as antibacterial agent. Therefore, the objectives of this research were to obtain information on antibacterial activities of the leaf extracts of Piper crocatum againts four types of bacteria, in that Staphylococcus, Bacillus substilis, Escherichia coli, and Pseudomonas aeruginosa and then to analyze the phytochemistry of the leaf extracts of Piper crocatum. The leaves of Piper crocatum were extracted by maceration and reflux using ethanol 30%. The assays of the antibacterial activities and phytochemistry on the extracts were carried out using the method of Maria Bintang. Results showed that the yield of the extraction using ethanol by maceration method was 20.8%. Meanwhile, using the reflux method, the yield was obtained about 26.25%. The phytochemistry analysis showed that the leaf extracts of Piper crocatum contained alkaloid, steroid and tanin. According to this study, it was found that the leaf extract of Piper crocatum can be used to inhibit the growth of B. subtilis and P. aeuruginosa, but can not inhibit the growth of E.coli and S. aureus.

Мета роботи — вивчити резистентність до антибіотиків бактеріальних збудників інфекцій сечових шляхів (ІСШ), виділених у пацієнтів урологічного стаціонару в м. Києві. Матеріали і методи. Досліджено 1612 штамів бактерій, виділених із сечі хворих з ІСШ (цистит, уретрит, пієлонефрит), госпіталізованих в урологічне відділення ДУ «Інститут урології НАМН України» у м. Києві протягом 2016 р. Серед пацієнтів переважали жінки — 1201 (74,5 %). Вік хворих становив від 17 до 74 років. Для збору даних використано медичну документацію лікарні. Мікробіологічні дослідження виконано у лабораторії мікробіології ДУ «Інститут урології НАМН України». Аналізували результати культурального дослідження зразків сечі, зібраних за наявності клінічних ознак ІСШ. Дослідження клінічного матеріалу та інтерпретацію отриманих результатів проводили загальноприйнятими методами. Вивчено чутливість уропатогенів до 31 антибіотика дискодифузійним методом відповідно до рекомендацій Інституту клінічних та лабораторних стандартів США (Clinical and Laboratory Standards Institute (CLSI)). Результати та обговорення. Аналіз мікробного спектра сечі виявив домінування серед уропатогенів штамів Escherichia coli (32,0 %), Enterococcus faecalis (19,5 %), Klebsiella pneumoniae (10,9 %), Staphylococcus epidermidis (8,9 %), S. haemolyticus (6,5 %) та Pseudomonas aeruginosa (6,4 %). Частка Enterococcus faecium, Enterobacter aerogenes і Streptococcus viridans становила відповідно 2,5, 2,2 і 1,6 %, Enterobacter cloacae, Klebsiella oxytoca, Acinetobacter baumannii, Proteus vulgaris та Providencia rettgeri — менше 1,0 %. У більшості випадків (69,7 %) мікроорганізми виділено у монокультурі, у решті випадків — у мікробних асоціа- ціях. Високу резистентність до тестованих антибіотиків виявили штами E. aerogenes (45,1 %), E. cloacae (45,7 %), E. faecium (40,9 %), E. faecalis (40,7 %), E. coli (39,9 %), P. aeruginosa (34,0 %), K. pneumoniae (28,6 %). Найбільш активними до уропатогенів були іміпенем (E. coli — 87,6 %, P. aeruginosa — 75,7 %, E. cloacae — 67,3 %, E. aerogenes — 72,6 %, K. pneumoniae — 93,2 %), меропенем (E. coli — 89,1 %, P. aeruginosa — 76,7 %, K. pneumoniae — 82,6 %), лефлоцин (E. coli — 74,5 %, ентерококи — 78,7 %, P. aeruginosa — 76,7 %, E. cloacae — 73,9 %, E. aerogenes — 80,4 %, K. pneumoniae — 83,5 %), амоксицилін/клавуланат (ентерококи — 84,6 %), фурагін (ентерококи — 82,6 %), цефоперазон (K. pneumoniae — 89,2 %, P. aeruginosa — 73,8 %), цефтріаксон (K. pneumoniae — 80,1 %). Висновки. Антибіотикорезистентність збудників ІСШ — важлива терапевтична проблема. Найбільшою активністю до уропатогенів характеризуються іміпенем, меропенем, лефлоцин, амоксицилін/ клавуланат, фурагін, цефоперазон, цефтріаксон, які можна розглядати як препарат вибору для призначення стартової терапії ІСШ. Необхідно здійснювати постійний моніторинг за резистентністю до дії антибіотиків. Політику використання антибіотиків у кожному стаціонарі слід визначати залежно від локальних даних щодо резистентності до протимікробних препаратів.

Ce travail vise l’étude de l’activité antibactérienne de l’huile essentielle (HE) de Pelargoniumx asperum et de la bactériocine, la nisine seul et en combinaison vis-à-vis de six bactéries dont quatre sont multirésistantes d’origine clinique. L’activité antibactérienne in vitro a été évaluée par la méthode de diffusion sur gélose. La concentration minimale inhibitrice (CMI) est aussi déterminée pour HE. Les résultats ont révélé une activité antibactérienne significative exercée par HE visà-vis de Staphylococcus aureus (ATCC 43300), Staphylococcus aureus et Escherichia coli avec des diamètres d’inhibition de 36,00 ; 22,50 et 40,00 mm, respectivement. Cependant, l’HE de Pelargonium asperum a montré une activité antibactérienne supérieure par rapport à la nisine. Les valeurs des CMI rapportées dans cette étude sont comprises entre 1,98–3,96 μl/ml. Les combinaisons réalisées entre HE et la nisine ont montré un effet additif vis-à-vis de Escherichia coli (ATCC 25922) avec (50 % HE Pelargonium asperum + 50 % nisine). Par contre, nous avons enregistré une synergie vis-à-vis de Klebsiella pneumoniae avec (75 % HE Pelargonium asperum + 25 % nisine) et contre Pseudomonas aeruginosa avec les trois combinaisons testées. Les résultats obtenus permettent de dire que l’HE de Pelargonium asperum possède une activité antibactérienne ainsi que sa combinaison avec la nisine pourrait représenter une bonne alternative pour la lutte contre l’antibiorésistance.
