Restrictions on the use of antibiotics in the poultry industry stimulate the development of alternative nutritional solutions to maintain or improve poultry health. This requires more insight in the modulatory effects of feed additives on the immune system and microbiota composition. Compounds known to influence the innate immune system and microbiota composition were selected and screened in vitro, in ovo, and in vivo. Among all compounds, 57 enhanced NK cell activation, 56 increased phagocytosis, and 22 increased NO production of the macrophage cell line HD11 in vitro. Based on these results, availability and regulatory status, six compounds were selected for further analysis. None of these compounds showed negative effects on growth, hatchability, and feed conversion in in ovo and in vivo studies. Based on the most interesting numerical results and highest future potential feasibility, two compounds were analyzed further. Administration of glucose oligosaccharide and long-chain glucomannan in vivo both enhanced activation of intraepithelial NK cells and led to increased relative abundance of lactic acid bacteria (LAB) amongst ileum and ceca microbiota after seven days of supplementation. Positive correlations between NK cell subsets and activation, and relative abundance of LAB suggest the involvement of microbiota in the modulation of the function of intraepithelial NK cells. This study identifies glucose oligosaccharide and long-chain glucomannan supplementation as effective nutritional strategies to modulate the intestinal microbiota composition and strengthen the intraepithelial innate immune system.

Abstract Lactation, feeding conditions, microbial interventions and piglet growth in the first few weeks of life have important impact on the intestinal microbiota establishment and immune system development of piglets. Indeed, colostrum and milk contain various bioactive components such as immune factors, antimicrobial peptides and oligosaccharides that contribute to maintain intestinal homeostasis and regulate interactions between microbiota and host immune system. Recent results revealed that low birth weight piglet (LBWP) with poor weight gain during the first two weeks of life develop different intestinal microbiota and immune response profiles compared to high BWP (HBWP) littermates. Consequently, piglets within litters may have different resilience to infections after weaning and benefit from feed additives in a specific manner. A study has been performed to evaluate the potential of bovine colostrum extract (BC) as replacement to plasma proteins for improving gut health and resilience to Salmonella infection in piglets. Results revealed that in weaned piglets fed BC, intestinal microbiota was differently modulated and bacterial dysbiosis induced by Salmonella was restored faster. Moreover, expression of genes involved in innate immunity such as β-defensin-2 and glutathione peroxidase-2 was respectively down- and up-regulated in BC fed piglets. A combination of dietary supplementation with BC, cupper and vitamins A and D has also been tested in LBWP and HBWP, and there is clear evidence that BC in combination with other feed additives promote growth and gut health in both LBWP and HBWP. The porcine intestinal epithelial cell line IPEC-J2 was used to better understand the functional properties of BC. Results indicated that BC improves wound healing, enhances barrier function and modulates the expression of several genes involved in innate immune response. Finally, as microbial intervention, the potential of fecal transplantation to modulate intestinal microbiota and immune system development of piglets is under investigation and will be discussed.

As a stressor widely existing in daily life, noise can cause great alterations to the immune system and result in many physical and mental disorders, including noise-induced deafness, sleep disorders, cardiovascular diseases, endocrine diseases and other problems. The immune system plays a major role in maintaining homeostasis by recognizing and removing harmful substances in the body. Many studies have shown that noise may play vital roles in the occurrence and development of some immune diseases. In humans, both innate immunity and specific immunity can be influenced by noise, and different exposure durations and intensities of noise may exert various effects on the immune system. Short-term or low-intensity noise can enhance immune function, while long-term or high-intensity noise suppresses it. Noise can lead to the occurrence of noise-induced hearing loss (NIHL) through the production of autoantibodies such as anti-Hsp70 and anti-Hsp60 and exert adverse effects related to other immune-related diseases such as some autoimmune diseases and non-Hodgkin lymphoma. The neuroendocrine system, mainly including the hypothalamic-pituitary-adrenal (HPA) axis and the sympathetic-adrenal-medullary (SAM) system, is involved in the mechanisms of immune-related diseases induced by noise and gut microbiota dysfunction. In addition, noise exposure during pregnancy may be harmful to the immune system of the fetus. On the other hand, some studies have shown that music can improve immune function and alleviate the adverse effects caused by noise.

An allergy is an overreaction of the immune system to a substance called an antigen, e.g., pollen from grasses, dust mites, etc. The drugs used for allergy in allopathy have undesirable side effects. The use of medicinal plants becomes popular due to the adverse effects of allopathic drugs. Nutraceuticals are food playing a significant role in maintaining normal physiological function. Mast cells are immunologically important cells found in almost all parts of our body, and contain histamines, leukotrienes within their granular sacs, along with those of basophils, are responsible for the symptoms of allergy. According to sources mast cell stabilizers can be classified into three categories, e.g., synthetic, semi synthetic and natural. Mast cell stabilising agents from natural resources can be obtained from different group of compounds, e.g., flavonoids, coumarins, phenols, terpenoids, alkaloids. In this book chapter, the active constituents present in them and their mode of action are highlighted using techniques of computational biology, e.g., molecular docking, etc.

While vitamins naturally occurring in plant and animal based feed ingredients can supply a reasonable proportion of the birds daily needs, their contribution is rarely considered during formulation. This situation arises due to variability, especially in cereals and vegetable proteins, for example, the vitamin E content of corn can vary from 10 to 40 IU/kg. Because of this uncertainty in the natural supply of vitamins within a feed, the bird’s vitamin supply is therefore met by the addition of synthetic vitamins, usually in the form of a so called premix, that contains all vitamins and perhaps some other micronutrients and feed additives. This premixes use according to producer suggestions and in some cases their doses can be higher than real requirements. Skinner et al (1992) removed vitamin premixes from 21 day of age and did not report any adverse effects. Gwyther et al (1992) showed sever performance reduction by vitamin premix withdrawal from 21 to 49 day of age. This experiment was conducted to determine the effects of Vitamin supplement levels on broiler chickens from 28 to 49 days of age.

This study was designed to determine the effect of feeding deoxynivalenol (DON) contaminated wheat to growing pigs on growth performance, digestibility and retention of nitrogen (N), phosphorus (P) and calcium (Ca). Secondly, we wished to evaluate the potential of four commercial anti-mycotoxin additives in preventing the adverse effects of DON. Sixty piglets (6.0 kg body weight) were assigned to six different corn-soybean meal-wheat diets (control diet: <0.5 mg kg−1 DON; contaminated diet (DON-contaminated): 4 mg kg−1; and four contaminated diets supplemented with a different anti-mycotoxin additive: DON+GLUC (glucomannan), DON+YBP (yeast, live bacteria, enzymes, plant extracts), DON+ALU (aluminosilicate) and DON+PC (preservation components). Piglets were housed individually in pens for 7 d and then transferred to metabolic crates for urine and feces collection for 7 d. At the end of these 2 wk, mannitol and lactulose doses were given by oral administration. Urine was collected for 24 h for an evaluation of barrier integrity of intestinal mucosa. The piglets were then euthanatized and intestinal samples collected for morphology studies. Feeding DON-contaminated wheat reduced average daily feed intake, average daily gain and G:F ratio compared with the control diet (P<0.05). Only DON+PC diet restored the growth performance of piglets fed DON-contaminated diet. Daily retention of N and P was not affected by DON contamination or anti-mycotoxin additives, but retention of Ca was higher in piglets fed the DON-contaminated diets than the control diet. DON-contaminated diet reduced digestibility of dry matter, gross energy and fat, villi height in jejunum, and recovery of mannitol and lactulose compared with the control diet (P<0.05). Finally, DON concentrations in serum from piglets fed the DON-contaminated diets were higher than in the control diet (P<0.05). Piglets fed DON-contaminated wheat had reduced growth possibly caused by impaired jejunal morphology and decreased digestibility of energy and fat.

After the initial description of extrarenal synthesis of 1,25-dihydroxyvitamin D (1,25-(OH)2D) three decades ago, extensive progress has been made in unraveling the immunomodulatory roles of vitamin D in the pathogenesis of granulomatous disorders, including sarcoidosis. It has been shown that 1,25-(OH)2D has dual effects on the immune system, including upregulating innate immunity as well as downregulating the autoimmune response. The latter mechanism plays an important role in the pathogenesis and treatment of sarcoidosis. Vitamin D supplementation in patients with sarcoidosis has been hampered owing to concerns about the development of hypercalcemia and hypercalciuria given that extrarenal 1-α hydroxylase is substrate dependent. Recently, a few studies have cast doubt over the mechanisms underlying the development of hypercalcemia in this population. These studies demonstrated an inverse relationship between the level of vitamin D and severity of sarcoidosis. Consequently, clinical interest has been piqued in the use of vitamin D to attenuate the autoimmune response in this disorder. However, the development of hypercalcemia and the attendant detrimental effects are real possibilities. Although the average serum calcium concentration did not change following vitamin D supplementation, in two recent studies, hypercalciuria occurred in one out of 13 and two out of 16 patients. This review is a concise summary of the literature, outlining past work and newer developments in the use of vitamin D in sarcoidosis. We feel that larger-scale placebo-controlled randomized studies are needed in this population. Since the current first-line treatment of sarcoidosis is glucocorticoids, which confer many systemic adverse effects, and steroid-sparing immunosuppressant treatment options carry additional risks of adverse effects, adjunct management with vitamin D in combination with potent anti-osteoporotic medications could minimize the risk of glucocorticoid-induced osteoporosis and modulate the immune system to attenuate disease activity in sarcoidosis.
