The effect of adding cottonseed hulls to casein- and cottonseed-kernel-based diets on the apparent and true ileal digestibility of N and amino acids, and the proportion of this effect accounted for by condensed tannin (CT), were determined using the growing rat. Sixtyrats were allocated randomly to ten semi- purified diets, containing either casein (four diets) or purified unheated solvent-extracted cottonseed kernel (six diets) as the sole protein source, with Cr2O3 added as an indigestible marker. Two of the casein diets contained no hulls whilst the remaining two diets contained 70 g cottonseed hulls/kg. Two of the cottonseed-kernel-based diets contained no hulls, with two containing 23 g hulls/kg and the remaining two containing 46 g hulls/kg. For each pair of diets, PEG was either included or excluded. The effect of CT was quantified by comparing control rats (-PEG; CTacting) with PEG-supplemented rats (+PEG; CT inactivated) at each level of dietary hulls. The rats were given their respective experimental diets for 14 d. Each rat was given the food ad libitumfor 10 min hourly from 08·00 to 18·00 hours. On day 14, samples of digesta were collected at death from the terminal 150 mm of ileum at 7 h from the first meal. Apparent and true ileal digestibilities were calculated for DM, N and the individual amino acids. The principal finding was that the inclusion of hulls depressed the apparent and true ileal digestibilities of N and amino acids, but with the response differing between diets. With the casein-based diet the mean apparent and true ileal amino acid digestibilities were significantly depressed from 0·89 and 0·96 to 0·85 and 0·92 respectively, by the inclusion of 70 g hulls/kg in the diet, and addition of PEG then restored these to 0·89 and 0·95. All of the depression could be explained by the CT content of the hulls. However, with the cottonseed-kernel-based diet the responses fell into three categories. The apparent and true ileal digestibilities of the essential amino acids cystine and methionine were not affected by hull addition, ileal digestibilities of leucine, isoleucine, lysine, threonine and valine were markedly depressed by hull addition with approximately 50% of the depression being explained by CT, whilst the ileal digestibilities of histidine, arginine and phenylalanine were depressed by hull addition but little or none of this effect could be explained by CT. Thus the effect of hulls on protein digestion clearly differed with source of protein. With the cottonseed-kernel-based diet it seems that components of the hulls other than CT also depressed the apparent and true ileal digestibilities of N and amino acids. The identity of these components is unknown.

The effect of condensed tannins (CT) from heated and unheated cottonseed on the apparent ileal digestibility of amino acids for the growing rat and pig was determined. In Expt 1, twenty-four rats were allocated to four semi-purified diets which contained cottonseed kernel and hulls as the only protein source. Two of the diets contained unheated solvent-extracted cottonseed kernel and hulls, while the remaining two diets contained similar material but which had been heat-treated by autoclaving at 110° for 120 min. In Expt 2, twelve rats and twelve pigs were fed on four semi-purified diets containing commercial cottonseed meal (CSM) as the only protein source. Cr2O3, was added to all diets as an indigestible marker. For each pair of diets in both experiments, PEG was either included or excluded. The effect of CT was assessed by comparing control animals (-PEG; CT acting) with PEG supplemented animals (+ PEG; CT inactivated). Ileal contents from the terminal 150 and 450 mm of ileum were collected at slaughter, 7 h from the start of feeding, for the rats and pigs respectively. Apparent ileal amino acid digestibility for rats fed on the diet containing cottonseed kernel and hulls was significantly depressed by the heat treatment, particularly for lysine and threonine. On average, apparent ileal amino acid digestibility in the diets without PEG was decreased from 0·80 to 0·70 by heat treatment. Dietary cottonseed CT depressed apparent ileal protein digestibility in the pig and in the rat. The addition of PEG to the diets significantly increased the apparent ileal digestibility of N and some amino acids for the pigs and the rats. The mean increase in apparent ileal digestibility due to PEG addition for the fourteen amino acids was 2 percentage units in both species fed on the commercial CSM diets, and 2 or 4 percentage units in rats fed on the unheated or the heated cottonseed kernel and hull diets respectively. The effect of PEG was similar in the heated and unheated cottonseed kernel and hulls for most amino acids, but apparent ileal digestibilities of threonine, tyrosine and lysine were increased more by PEG in heated than in unheated CSM. Apparent ileal N digestibility was lower in the pig than in the rat. For several of the amino acids there were significant animal species differences in apparent ileal digestibility. Studies into the effects of cottonseed CT should be carried out in the target animal species. The commercial CSM had a low apparent ileal amino acid digestibility overall, particularly for the essential amino acids lysine and threonine. It was concluded that effects of heating did not eliminate the reversible reactivity of cottonseed CT on amino acid digestion in rats and pigs but rather appeared to increase it for threoniw, tyrosine and lysine in Expt 1, causing large reductions in apparent ileal digestibility of these amino acids.

Abstract. Using adapted methods for balanced experiments with poultry, the true digestibility coefficients of the essential amino acids of two high protein sunflower meals – 40+® and 47® have been established. The true digestibility coefficients by using two methods – with intact and randomized (caecectomized) birds have been compared. Six birds were tube fed and six – feed deprived for both of the fodders. The amino acids were established with amino analyzer AAA-881 after preliminary HCl hydrolysis. The mean digestibility of sunflower meal 40+® is higher in intact (88.11 versus 85.73 of sunflower meal 47®) compared to randomized birds (87.20 versus 85.46 of sunflower meal 47®). The differences by lots are statistically significant only in intact birds. The authors recommend for practical needs to use the average true digestibility coefficients between both meals, established with randomized birds (ileal digestibility).

Abstract An experiment was conducted to investigate the effect of rice bran (RB) and Ronozyme® WX on apparent and true ileal amino acid (AA) digestibility coefficients in broiler chicks using a regression approach. Two hundred and forty 21-day-old Abor Acre plus chicks were randomly allotted to six diets with varying levels of RB (100, 200 and 300 g/kg) without (0g/kg) or with Ronozyme® WX (25g/kg) supplementation in a 2x3 factorial arrangement using a randomized complete block design. Each treatment had 5 replicates with 8 birds each. The birds were fed for 5 days (d 22–26). On d 26, digesta was sampled from distal ileum. Data were analyzed using general linear model at α0.05. Lysine had the highest concentration among the essential amino acids (AAs) while the lowest concentration was observed for threonine. Rice bran at 10% for the essential amino acids with enzyme was significantly higher but compared with concentrations of RB at 10% and 20% without enzyme. There were no significant differences among the true ileal amino acid digestibility coefficients except lysine (10% RB) without enzyme (0.91) which was significantly higher than 20% RB with enzyme (0.84) but similar to other treatments. Rice bran with enzyme had a significant (P < 0.05) interactive effect on the apparent ileal digestibility coefficients of arginine, histidine, isoleucine methionine and phenylalanine. When RB was supplemented with enzyme, a significant (P < 0.05) interactive effect on true ileal digestibility coefficients of arginine and methionine was observed. Rice bran-enzyme interaction had no significant effect on the non-essential AAs. The slopes of the regression lines ranged from 0.77 (arginine) to 0.98 (phenylalanine). In conclusion, enzyme supplementation did not significantly influence the digestibility coefficients of AA in rice bran. However, the inclusion of 10% and 20% RB supplemented with Ronozyme® can help to improve the methionine content in rice bran.

1. The effect of supplementing barley diets with urea (U), extracted decorticated groundnut meal (GNM) or Peruvian fish meal (PFM) on plasma free amino acid concentrations in sheep have been examined and the first limiting amino acid has been indicated by measuring the changes in the concentration of the plasma essential amino acids (PEAA) during a rumen infusion of a volatile fatty acid (VFA) mixture.2. Three wethers fitted with rumen and re-entrant duodenal cannulas were given isonitrogenous, isoenergetic diets containing (g/kg dry matter (DM)) U 20, GNM 106 or PFM 78, the crude protein (nitrogen × 6.25) contents being 139, 145 and 148 respectively. The sheep were fed hourly, the mean daily dm intake being 0.634 kg.3. Plasma concentrations of valine, threonine, lysine, isoleucine and leucine were linearly related to their concentrations in duodenal digesta.4. A VFA mixture was infused into the rumen for 6 h to supply (mmol/min) acetate 1.47, propionate 0.22 and n-butyrate 0.27. Blood samples were taken 6 h before, during and 12 h after the end of the infusion.5. The concentration of all PEAA decreased relative to the pre-infusion and post-infusion controls but there were no significant differences between diets.6. The mean decreases in concentration averaged over all three diets showed that the decrease in concentration of methionine (41.5%) was far greater than for any other essential amino acid suggesting that under these conditions methionine was the first limiting amino acid.

Abstract We report for the first time the concentrations of free amino acids in human intestinal biopsies obtained by routinely performed endoscopy. We studied 15 medical patients with no changes of the mucosa and six HIV-infected persons with duodenitis. The mean (and SD) sum of all amino acids, taurine excepted, was 61.9 (5.4) mmol/kg dry weight in duodenal biopsies of HIV-negative subjects (n = 11) and 82.9 (0.6) mmol/kg in colonic specimens: 50% (44%) of the total (minus taurine) consisted of aspartate and glutamate and 14% (12%), of the essential amino acids. The relative amino acid pattern in duodenum and colon differed completely from that for muscle: aspartate was fourfold higher; glutamate, phenylalanine, glycine, valine, leucine, and isoleucine were about twofold higher. In contrast, glutamine amounted only to 4% (duodenum) to 14% (colon) of muscle glutamine. In duodenal biopsies of the HIV-infected persons, we found significantly (P less than 0.01, except glutamine: P less than 0.025) increased concentrations of glutamate (24.1 vs 17 mmol/kg dry weight), ornithine (1.4 vs 0.4), valine (2.2 vs 1.7), and glutamine.

1. Sheep were used to evaluate the nutritional consequences of a low condensed-tannin concentration (22 g/kg dry matter (DM)) in lotus (Lotus corniculatus L.) (control group) compared with lotus given to sheep receiving intraruminal polyethylene glycol (PEG) infusion (PEG group). PEG selectively binds to tannins and prevents tannins from binding proteins.2. DM intakes (1430 (SE 28) g/d) and digestibility of energy (663 (SE 4.5) kJ/MJ intake) were similar for both groups but the apparent digestion of nitrogen was lower in the control sheep (0.70) than in the PEG sheep (0.78; P < 0.001).3. The proportion of N apparently digested before the abomasum (i.e. in the rumen) was lower (P < 0.05) in control sheep (0.12) than in PEG sheep (0.21; P < 0.05). Rumen ammonia concentrations were lower (P < 0.001) in control sheep than in PEG sheep. The proportion of neutral-detergent fibre (NDF) digested in the rumen was similar for both groups (0.48 (SE 0.012)) but less energy was digested in the rumen of the control (0.42) than of the PEG sheep (0.47; P < 0.05).4. The flux of essential amino acids (EAA) through the abomasum of control sheep was 50% greater than that in PEG sheep; flux of non-essential amino acids (NEAA) was 14% higher in control than in PEG sheep. Apparent digestibility of EAA in the small intestine was similar for both treatments (0.67), but NEAA were less well digested in the control (0.55) than in the PEG sheep (0.69).5. The presence of tannins in the control group increased net apparent absorption of threonine (57%), valine (89%), isoleucine (94%), leucine (30%), tyrosine (41%), phenylalanine (93%), histidine (90%) and lysine (59%), and reduced NEAA absorption by 10%, compared with PEG sheep.

Abstract The objectives were to determine standardized ileal digestibility (SID) of amino acids (AA) in hatchery byproducts fed to nursery pigs and to test the additivity of ileal digestible AA concentrations in hatchery byproducts. Ten barrows (initial body weight = 11.3 ± 1.3 kg) surgically fitted with T-cannulas at the distal ileum were individually housed in pens. Four diets were prepared to contain 30% of infertile eggs, unhatched eggs, culled chicks, or a mixture (20% dried infertile eggs + 20% dried unhatched eggs + 60% dried culled chicks) as the sole source of nitrogen. A nitrogen-free diet was additionally prepared to measure basal endogenous losses of AA. All experimental diet contained 0.5% chromic oxide as an indigestible index. Ten pigs were assigned to a replicated 5 × 4 Latin square design with 5 diets and 4 periods. Each period consisted of a 5-d adaptation period and a 2-d collection period. The hypothesis was that the determined digestible AA concentrations of the mixture are not different from the calculated values based on digestible AA in the individual ingredients. A t-test was used to determine the additivity of digestible AA concentrations in individual ingredients. Infertile eggs had greater (P &lt; 0.05) SID of all indispensable AA except for lysine and methionine compared with culled chicks. The SID of all indispensable AA except for tryptophan was not different between culled chicks and the mixture. The calculated apparent and standardized ileal digestible AA in the mixture were not different from the determined values, but calculated ileal digestible tryptophan in the mixture was less (P &lt; 0.05) than the measured value. In conclusion, ileal digestibility of most indispensable AA in infertile eggs is greater than that in culled chicks and ileal digestible AA values are additive in the mixture except for tryptophan.

Background: In ruminants, the weaning period is a critical phase, because these young animals are allocated to various facilities and receive different feeds. Alternatively, supplements and additives can be used. The tannins are polyphenols produced by plants; constitute a group of complex secondary metabolic factors that inhibit the consumption by herbivores or the development of microbial agents. The objective of this study was to determine whether the addition of condensed tannin extract from quebracho trees (Schinopsis lorentzii) in lamb diets after weaning would have beneficial effects on growth and health.Materials, Methods & Results: Forty lambs were assigned randomly to 1 of 4 treatments (10 lambs/treatment): T1 (0%; control), T2, T3, and T4 was 0.3, 0.6 and 0.9 % of quebracho extract in the diet, respectively. The T2 (0.3%) lambs had greater body condition scores (P = 0.005), and although they did not differ significantly, they had numerically greater body weight (P = 0.067) and weight gain (P = 0.072), than the control lambs. The anthelmintic and anticoccidial effect was not possible to verify because no animals in any group were naturally infected by helminths or coccidia. The T4 lambs had greater numbers of erythrocytes (days 10 and 40) [P < 0.05], greater hemoglobin concentrations (days 10 and 40), and greater numbers of total leukocytes (day 10) as a result of greater numbers of neutrophil at the end of the adaptation period (day 10) compared to T1. The T3 and T4 lambs had greater (P < 0.05) serum total protein (days 10 and 40) because of greater concentration of serum globulins (days 10 and 40) compared to T1, suggesting that the addition of tannin stimulated the immune system. The addition of quebracho in the diet affected serum concentrations of glucose, triglycerides, and cholesterol at some times and treatments (P < 0.05). However, these changes did not follow a pattern; this finding only suggested that there was an influence on the metabolism of carbohydrates and lipids. The T2 lambs showed no differences on hemogram or serum clinic biochemistries (P > 0.05); however, they had a greater body condition scores than T1 (P < 0.05).Discussion: According to literature, if condensed tannins are supplied in moderate quantities (≤ 5% of diet) to ruminants, they can prevent bloating, increase the supply of undegraded protein in the rumen and improve the use of essential amino acids. It is also known that tannins in diet may increase urea recycling efficiency by increasing the concentration of urea in saliva and salivary flow into the rumen. Studies observed that cattle and sheep given feed containing condensed tannins had higher production of wool, meat, milk, and ovulation rates in females, desirable effects on animal production. According to the literature, dietary supplementation with tannins from chestnut and quebracho (2 g/kg of feed) to Holstein cattle changed the composition of the ruminal microbiota, particularly the bacteria that degrade fiber and starch, reducing the abundance of Prevotella and Fibrobacter, favoring Ruminococcaceae and other members of the phylum Firmicutes, increasing the pH and decreasing urease activity in ruminal fluid. Therefore, when tannins are supplemented at low concentrations, they can stimulate the intake and utilization of essential amino acids in the small intestine, increasing performance and exerting an anthelmintic effect. Our study found that the addition of 0.3% of quebracho in the diet improved lamb growth and health after weaning. It is therefore an additive that can be used to feed lambs in critical post weaning period.

1. Ten calves (50–58 kg live weight) were given a diet consisting of diluted whole milk, wheat gluten and supplemented with appropriate nutrients including amino acids but deficient in lysine. The lysine requirements of these calves, which were growing at approximately 0.25 kg/d, were estimated from responses to lysine supplementation of this diet. From plasma urea, plasma lysine, nitrogen retention and apparent digestibility of N responses the estimated lysine requirements were 8.5, 7.5, 7.2 and 7.6 g/d respectively.2. From the mean lysine requirement (7.8 g/d) and the ratio, lysine: other essential amino acids in carcasses of similar calves the estimated requirements were (g/d): methionine 2.1, cystine 1.6, threonine 4.9, valine 4.8, isoleucine 3.4, leucine 8.4, tyrosine 3.0, phenylalanine 4.4, histidine 3.0, arginine 8.5, tryptophan 1.0.
