SummaryA field experiment was conducted for 3 years to study the effect of Rhizobium inoculation, mulch and N and P fertilizers on soya bean. The application of wheat straw mulch helped greatly in lowering the maximum soil temperature but failed to increase the yield, particularly on the fields where soya bean was raised for the first time. Seed inoculation gave significantly greater yield than control (no inoculation and no mulching). The combination of mulch and seed inoculation, on average, produced 0·76, 0·43 and 0·84 t/ha more grain than inoculation alone in 1976, 1977 and 1978 respectively. A basal dose of 15 kg N/ha increased the yield when seed was inoculated but no mulch applied whereas the dressing of N did not prove beneficial when mulching was combined with inoculation. The grain yield, however, increased up to 180 kg N/ha in the case of mulch alone and control treatments. The response of soya bean to phosphorus application was inconsistent.

A field experiment was conducted during Rabi season of 2012-13 and 2013-14 to study the effect of integrated phosphorus management on growth, yield and quality of lentil. The trial was laid out in split plot design assigning 8 treatments of three phosphorus levels (0, 40 and 80 Kg ha-1), three mode of PSB application (soil application, seed inoculation including control) and sulphur application of 20 Kg ha-1 with control. The crop was fertilized up to 80 Kg ha-1 of phosphorus application enhanced more growth, yield attributes, seed yield and protein yield than the 40 Kg ha-1 and control, however the initial plant population per running meter was remain non-significant during both the years. The PSB applied by seed inoculation and 20 Kg ha-1 sulphur application were showed similar effects. The economic value also calculated to judge best combination and found that the 80 Kg ha-1 phosphorus application, seed inoculation with PSB and 20 Kg ha-1 sulphur application received more net income (ha-1) and benefit cost ratio also.

A field experiment on a greengram-wheat cropping sequence was carried out under limited water supply conditions in 1997-98 and 1998-99 at the farm of the Indian Agricultural Research Institute, New Delhi. The greengram was sown either on flat beds or on broad beds 2 m in width, divided by furrows, with 0, 30 and 60 kg P2O5/ha. After the harvest of greengram pods, wheat was grown in the same plots, either with the greengram stover removed or with the stover incorporated along with 0, 40, 80 and 120 kg N/ha applied to wheat. The grain yield of greengram was higher when sown on broad beds with furrows compared to flat bed sowing, and the application of 30 or 60 kg P2O5/ha resulted in significantly higher grain yields compared to no phosphorus application. The combination of broad bed and furrows with phosphorus fertilization was found to be ideal for achieving higher productivity in greengram. The land configuration treatments had no impact on the productivity of wheat. The application of phosphorus to the preceding crop had a significant residual effect on the grain yield of wheat. The incorporation of greengram stover also significantly increased the grain yield of wheat. The increasing levels of N increased the grain yield of wheat significantly up to 80 kg/ha. The combination of greengram stover incorporation and 80 kg N/ha applied to wheat significantly increased the grain yield. Further, there was a significant interaction effect between the phosphorus applied to the preceding crop and N levels given to wheat on the grain yield of wheat.

Production technology of metal casting industry in Indonesia needs to be improved, especially in the manufacturing of spare parts and box engine made of gray cast iron which has various wall thick such as dove tale construction. Microstructure of gray cast iron is influenced by cooling rate during casting, chemical composition and melting treatment process (inoculation). The part which has the thinnest thickness has the fastest cooling therefore, the grain boundary is smaller compared to other section. As a result this part has highest hardness and difficult to be machined. This research is conducted to solve this problem by modifying melting and solidification treatment process. The research starting from micro structure analysis, composition and mechanical properties tests on the product, and then conducting modification treatment through Taguchi method approach. Experimental results obtained show that the best level settings to control factors which affect to the uniformity of the microstructure and mechanical properties in gray cast iron is the addition of seed inoculation super ® 75, as much as 0.25% with the method of inoculation material entering into the Transfer Ladle.Teknologi produksi pada industri pengecoran di Indonesia masih membutuhkan perbaikan terutama dalam pembuatan komponen mesin perkakas dan peralatan pabrik yang terbuat dari besi tuang kelabu yang mempunyai variasi ketebalan yang besar seperti konstruksi ekor burung (dove tale). Pada pengecoran, struktur mikro dari besi tuang kelabu sangat dipengaruhi oleh kecepatan pendinginan, komposisi kimia dan proses perlakuan pada logam cair (inokulasi). Bagian yang mempunyai ukuran paling tipis mempunyai kecepatan pendinigan paling tinggi karena itu ukuran butirnya jauh lebih kecil dari bagian lain, akibatnya bagian ini mempunyai kekerasan lebih tinggi dan sulit dilakukan pengerjaan mesin. Penelitian ini bertujuan untuk memperbaiki hal ini yang terjadi pada dove taledengan cara memodifikasi proses perlakuan pada cairan besi dan proses pendinginan. Penelitian dimulai dari analisa struktur mikro, pengujian komposisi kimia, pengujian sifat mekanis pada produk kemudian dilakukan modifikasi menggunakan pendekatan metode statistik Taguchi. Hasil penelitian menunjukkan bahwa pengaturan terbaik yang dapat diperoleh untuk mendapatkan keseragaman struktur mikro dan sifat mekanis pada pengecoran besi tuang kelabu adalah penambahan seed inoculation super ® 75, sebesar 0.25% dengan metode pemasukan inokulasi kedalam Ladle pengangkut logam cair.Keywords: carbon, micro structure, hardness, inoculation

A total of 14 Rhizobium strains were isolated from lentil accessions grown at the ICARDA experimental research station at Marchouch in Morocco and used for molecular characterization and symbiotic efficiency assessment. Individual phylogenetic analysis using the 16S rRNA gene, house-keeping genes rpoB, recA, and gyrB, and symbiotic genes nodD and nodA along with Multilocus Sequence Analysis (MLSA) of the concatenated genes (16S rRNA-rpoB-recA-gyrB) was carried out for the identification and clustering of the isolates. The symbiotic efficiency of the strains was assessed on three Moroccan lentil cultivars (Bakria, Chakkouf, and Zaria) based on the number of nodules, plant height, plant dry weight, and total nitrogen content in leaves. The results showed that the individual phylogenetic analysis clustered all the strains into Rhizobium laguerreae and Rhizobium leguminosarum with sequence similarity ranging from 94 to 100%, except one strain which clustered with Mesorhizobium huakuii with sequence similarity of 100%. The MLSA of the concatenated genes and the related percentages of similarity clustered these strains into two groups of Rhizobium species, with one strain as a new genospecies when applying the threshold of 96%. For symbiotic efficiency, the Bakria variety showed the best association with 10 strains compared to its non-inoculated control (p-value ≤ 0.05), followed by Chakkouf and Zaria. The present study concluded that the genetic diversity and the symbiotic efficiency of Rhizobium strains appeared to be mainly under the control of the lentil genotypes.

AbstractThe Bradyrhizobium vignae strain ORS3257 is an elite strain recommended for cowpea inoculation in Senegal. This strain was recently shown to establish symbioses on some Aeschynomene species using a cocktail of Type III effectors (T3Es) secreted by the T3SS machinery. In this study, using a collection of mutants in different T3Es genes, we sought to identify the effectors that modulate the symbiotic properties of ORS3257 in three Vigna species (V. unguiculata, V. radiata and V. mungo). While the T3SS had a positive impact on the symbiotic efficiency of the strain in V. unguiculata and V. mungo, it blocked symbiosis with V. radiata. The combination of effectors promoting nodulation in V. unguiculata and V. mungo differed, in both cases, NopT and NopAB were involved, suggesting they are key determinants for nodulation, and to a lesser extent, NopM1 and NopP1, which are additionally required for optimal symbiosis with V. mungo. In contrast, only one effector, NopP2, was identified as the cause of the incompatibility between ORS3257 and V. radiata. The identification of key effectors which promote symbiotic efficiency or render the interaction incompatible is important for the development of inoculation strategies to improve the growth of Vigna species cultivated in Africa and Asia.

In specific plant organs, namely the root nodules of alfalfa, fixed nitrogen (ammonia) produced by the symbiotic partner Sinorhizobium meliloti supports the growth of the host plant in nitrogen-depleted environment. Here, we report that a derivative of S. meliloti carrying a mutation in the chromosomal ntrR gene induced nodules with enhanced nitrogen fixation capacity, resulting in an increased dry weight and nitrogen content of alfalfa. The efficient nitrogen fixation is a result of the higher expression level of the nifH gene, encoding one of the subunits of the nitrogenase enzyme, and nifA, the transcriptional regulator of the nif operon. The ntrR gene, controlled negatively by its own product and positively by the symbiotic regulator syrM, is expressed in the same zone of nodules as the nif genes. As a result of the nitrogen-tolerant phenotype of the strain, the beneficial effect of the mutation on efficiency is not abolished in the presence of the exogenous nitrogen source. The ntrR mutant is highly competitive in nodule occupancy compared with the wild-type strain. Sequence analysis of the mutant region revealed a new cluster of genes, termed the “ntrPR operon,” which is highly homologous to a group of vap-related genes of various pathogenic bacteria that are presumably implicated in bacterium-host interactions. On the basis of its favorable properties, the strain is a good candidate for future agricultural utilization.

The present study was aimed at exploring the effect of soil application of different concentrations of orthophosphate (P) (0, 10, 20, 30, and 40 mg kg−1) on rice agronomic and yield parameters, arsenic (As) species accumulation, and polyphenol levels in the grain of rice grown under As spiked soil (10 mg kg−1). The contents of As species (As(V), As (III), MMA and DMA) and polyphenols in rice grain samples were estimated using LC-ICP-MS and LC-MS/MS, respectively. P treatments significantly reduced the toxic effects of As on agronomic parameters such as root weight and length, shoot and spike length, straw, and grain yield. Among the treatments studied, only the treatment of 30 mg kg−1 P helps to decrease the elevated levels of As (V), As (III), and DMA in rice grains due to As application. The study revealed that 30 mg kg−1 was the optimal P application amount to minimize AS accumulation in rice grains and As-linked toxicity on agronomic parameters and chlorophyll biosynthesis. Furthermore, the levels of trans-ferulic acid, chlorogenic acid, caffeic acid, and apigenin-7-glucoside increased in response to accumulation of As in the rice grain. In conclusion, the precise use of phosphorus may help to mitigate arsenic linked phytotoxicity and enhance the food safety aspect of rice grain.

A growing interest in soybean cultivation in Poland has been observed in the recent years, however it faces a lot of difficulties resulting from a poorly understood effectiveness of plant nitrogen fertilization and from the introduction of Bradyrhizobium japonicum to the environment. The aim of the study was to evaluate the consistency of response of two soybean cultivars to three different rates of mineral N fertilization and two seed inoculation treatments with B. japonicum in field conditions over four years regardless of previous B. japonicum presence in the soil. A highly-diversified-over-years rainfall and temperature in the growing season do not allow for a definite statement of the differences resulting from seed inoculation and mineral N fertilization applied separately or jointly in soybean. A high sensitivity of the nodulation process to rainfall deficits was noted, which resulted in a decreased amount of B. japonicum DNA measured in qPCR and dry matter of nodules. ‘Annushka’ demonstrated a higher yield of seeds and protein, higher plants and the 1st pod setting. ‘Aldana’, due to a significant decrease in plant density, produced a higher number of pods, seeds per pod and the 1000 seed weight per plant. Both cultivars responded with an increase in the seed yield after seed inoculation with HiStick, also with an application of 30 and 60 kg N, as well as with Nitragina with 60 kg N.
