T2 and T4 bacteriophage have been exposed to various treatments which are known to release the encapsulated DNA. The unseparated reaction products have been examined by autoradiography. The results indicate the presence of one large subunit of DNA (molecular weight 45 x 106) for each former phage particle. Some smaller subunits of molecular weight 12 x 106 have been observed. The large subunit is sensitive to very small amounts of DNAase, and is resistant to mixed proteases and cannot be dispersed by banding in cesium chloride density gradients. The sensitivity to fragmentation by P32 decay and the increase in this sensitivity following heat treatment are best explained by assuming that the large subunit is a duplex of polynucleotide strands over most of its length. The presence of hypothetical non-DNA interconnections is considered.

GP 32 (molecular weight 35000) is a T4 bacteriophage protein that destabilizes the DNA helix. The fragment GP32*I (77% of the total weight), which destabilizes helices better than does the parent molecule, crystallizes as platelets thin enough for electron diffraction and electron imaging. In this paper we discuss the structure of this protein as revealed in images reconstructed from stained and unstained crystals.Crystals were prepared as previously described. Crystals for electron microscopy were pelleted from the buffer suspension, washed in distilled water, and resuspended in 1% glucose. Two lambda droplets were placed on grids over freshly evaporated carbon, allowed to sit for five minutes, and then were drained. Stained crystals were prepared the same way, except that prior to draining the droplet, two lambda of aqueous 1% uranyl acetate solution were applied for 20 seconds. Micrographs were produced using less than 2 e/Å2 for unstained crystals or less than 8 e/Å2 for stained crystals.

Boron trimethyl reacts with each of the organometalloid-hydrazine derivatives Me2NN(R)GeMe3 (R = H, Me, SiMe3) except Me2NN(SiMe3)GeMe3, and with Me2NNHSiMe3, forming 1:1 adducts. With Me2NN(SiMe3)GeMe3 and BMe3, no adduct was formed even at low temperatures. On the basis of p.m.r. spectra, the metalloid-substituted nitrogen atom rather than the terminal nitrogen, is indicated as the basic center at which adduct formation occurs. With BF3, the Si—N and Ge—N bonds of the hydrazine derivatives were cleaved, again indicating attack at the central nitrogen atom. In a series of experiments involving CY2 (Y = O, S) and Me2NN(R)MMe3 (R = H, Me, SiMe3; M = Si, Ge), reaction occurred readily for R = H, but less readily for R = Me and for the compound Me2NN(GeMe3)2, while no reaction was observed for the compounds Me2NN(SiMe3)2 and Me2NN(SiMe3)GeMe3, even when heat or basic catalysts were employed. Reaction products, carbamate and thiocarbamate derivatives, Me2NN(R)C(Y)YMMe3, were characterized on the basis molecular weight, i.r., p.m.r., and mass spectroscopy, and m.p. data. Ease of formation is discussed in terms of electronic and steric effects.

Light scattering measurements performed on DNA after irradiation in the presence of psoralen clearly show that inter strand cross linkings are present in the macromolecule. In fact after heat denaturation and successive cooling irradiated macromolecule shows a molecular weight practically unchanged while a DNA sample after the same treatment shows a molecular weight half of the intact native DNA. Also the general conformation of irradiated DNA undergoes practically to no modifications after the same heat treatment while native DNA shows itself to have been strongly modified. Moreover, on the basis of flow dichroism determinations, DNA cross-linked by psoralen after heat denaturation showed to be able to restore its ordered double helix structure, during the successive cooling.

Salmonella newport phages 16–19 and 7–11 have very long heads and are members of two rare and so far little-known phage groups. Both produce various morphological aberrations. Preparations of phage 7–11 contain numerous polyheads and about 0.4% short heads belonging to nine size classes. In addition, one giant phage particle was observed. The head of phage 7–11 seems to be an icosahedron which became elongated by adding successive rows of subunits. Phages 16–19 and 7–11 have buoyant densities in CsCl of 1.43 and 1.48 g/mL and particle weights of 103 and 204 × 106 respectively. Both viruses contain double-stranded DNA, internal proteins, and sugars. Phage 16–19 contains 46.5% DNA of 35 × 106 molecular weight, and glucose. Phage 7–11 contains 47.5% DNA of 108 × 106 molecular weight, and mannose. Base compositions of phage and S. newport DNAs were determined from buoyant densities, melting point, and acid hydrolysis. Phage 16–19 contains 5.4% 5-methylcytosine.

AbstractIn order to study the allelic variation of Glu-1 gene (High molecular weight glutenin), 100 Iranian wheat cultivars including imported and domestic genotypes were analyzed using ALP-PCR technique. Four specific primer pairs were used based on the genetic loci of Glu-A1, Glu-B1, and Glu-D1 to perform the chain polymerase reactions. PCR reaction products were resolved on 2% agarose gel. Since allele “a” had the largest relative frequency (0.707), two alleles (a 344bp; b 362bp) were identified by P1-P2 primer for Glu-A1 locus. Three alleles (a 800bp; b 500bp; c 300bp) were detected for Glu-B1 locus by P5-P6 primer, and allele “b” was assumed as the highest relative frequency (0.618). Two primer pairs were applied for Glu-D1 locus. Ultimately, four alleles were identified, where allele “c” had the highest relative frequency (0.525). The observed genetic variation value for Glu-D1 locus (H=0.648) exhibits the maximal polymorphism. Using cluster analysis, the relationship between the observed polymorphism and geographical variation was investigated. The results indicated that there exists a remarkable variation in Glu- A1 locus between the Iranian wheat cultivars.

Transformations of high-molecular-weight compounds of oil sand natural bitumen under the heat treatment were studied in this work. For that purpose the natural bitumen isolated from oil sand taken from the Beke field (Kazakhstan) was used as a substrate. Thermal processing of natural bitumen leads to a general change in the chemical composition of components and to an increase in the output of certain fractions. The contents of oil, tar and asphaltenes were determined and the elemental composition of tar-asphaltene compounds was evaluated. Molecular structures of the tar and asphaltene components of natural bitumen before and after cracking have been defined from the data of elemental analysis, NMR spectroscopy and molecular weight. The high molecular compounds were presented as giant molecules containing small aromatic islands some of which were linked by aliphatic chains, that was proved by infrared spectroscopy.

Abstract The application of phenol-formaldehyde resins as crosslinking agents is increasing in importance due to the good high temperature properties of the corresponding vulcanizate and the use in thermoplastic vulcanizates. With respect to the chemistry of phenol-formaldehyde cure (reaction mechanism and structure of crosslink) there are still problems that have to be resolved. The reaction products of the phenol-formaldehyde resin curing of EPDM, contain 2-ethylidene norbornene (ENB) as the third monomer, have been studied. Since such an investigation is rather difficult to perform for the polymer system, a low molecular weight model for EPDM was used: 2-ethylidene norbornane (ENBH). Reaction of ENBH and a resole results in scission of the dimethylene ether bridges, i.e. in degradation of the resole into mono-, bis- and terisooctylphenol units. These are consequently converted into products, consisting of two ENBH molecules linked by mono-, bis- and terisooctylphenol units. The solid resole seems to be a technological solution for storing phenol in combination with formaldehyde. These results support the use of 2-hydroxymethylphenol (HMP) as a low molecular weight model for the resole. At low temperatures and/or short reaction times HMP oligomers (= resoles) and HMP oligomers linked to one ENBH molecule are formed, which are converted into ENBH/HMP (1:1) condensation products. The reaction products of ENBH with both the resole and HMP are shown to contain methylene linked structures, as demonstrated by the formation of monisooctylphenol crosslinks and the presence of residual unsaturation and hydroxyl groups, besides chroman linked structures. This is the first experimental evidence that during phenol-formaldehyde resin cure of rubber, formation of methylene bridges occurs.

1. Several substances of high molecular weight were examined for their suitability as suspension media in the formation of density gradients for the zonal centrifugation of avian erythrocytes. None proved satisfactory. 2. The behaviour of pigeon erythrocytes in rate-sedimentation experiments in a type A zonal rotor with density gradients of sucrose was examined. The mature cells sediment more rapidly than the younger cells and have a lower RNA/DNA ratio. Maturation is accompanied by a greater loss of RNA from the nucleus than from the cytoplasm. 3. The base composition of the nuclear RNA and of the two species of cytoplasmic ribosomal RNA is reported. 4. The RNA of erythrocytes may be labelled in vivo by injection of inorganic [32P]phosphate. The cells most active in the synthesis of RNA sediment less rapidly than the bulk of the cells. 5. Reticulocyte nuclei sediment more slowly than those from erythrocytes. Reticulocyte nuclei have a mean volume of 35μ3 and are isopycnic with sucrose of density 1·2871 (measured at 20°). Maturation of the nuclei causes them to shrink to a volume of 25μ3 and the density to increase to 1·2944.
