Abstract Cultivation of tiger grouper (Epinephelus fuscoguttatus) in Floating Net Cages in several locations in Bireuen Regency has developed. The potential disease issue can be caused by bacteria, viruses, fungi, or parasites. The purpose of this study was to determine the effectiveness of mangosteen peel extract capsules (Garcinia mangostana L) against bacterial disease in tiger grouper. This research was carried out in April 2019 at the MIPA Laboratory of Al Muslim University and the laboratory of PT. Centra Proteina Prima, Bireuen Regency. The design used was a completely randomized design with four treatments (0%, 10%, 20%, and 30% mangosteen peel extract capsules) with three replications. The parameters studied in this study including the diameter of the inhibition and the observation of MBC (Minimum Bacterial Concentration). The result showed that the administration of mangosteen peel extract capsules with doses of 10%, 20%, and 30% has no significant effect on reducing disease-causing bacteria in grouper grown on TSA media. Positive results were obtained on positive (+) MBC observations, presumably due to the small dose of mangosteen peel extract capsules given so that it could not kill bacteria grown in dishes with an inhibition range of 0 mm.Introduction

An experiment to evaluate the efficacy of inactivated polyvalent bacterial vaccines to increase specific immunity of tiger grouper (Epinephelus fuscoguttatus) against bacterial infection was conducted in floating net cages located in Sumbawa Island, West Nusa Tenggara. Polyvalent bacterial vaccine was prepared by mixing three species of killed-bacteria (Vibrio harveyi, V. alginolyticus, and Photobacterium leiognathi) with ratio of 1:1:1. The density of bacterial vaccine was adjusted at 1010 CFU/ml. The vaccines were delivered by immersion. The control fish were immersed in sea water without vaccine. Fish with ± 6 cm of total length and ± 7 g of body weigth were used in this experiment. The fishes were reared in concrete tanks in hatchery of Gondol, Bali until they reached ± 9 cm of total length. Later, the fishes were reared for 6 months in floating net cages located in Sumbawa. Re-vaccinations were delivered 60 days and 120 days post initial vaccination. The results showed that titer antibody and survival rate of the vaccinated fish were 1/128 and 77.11%, higher than the control (titer antibody=1/4-1/8, survival=45.56%, respectively). The relative percent survival of the vaccinated fish was 57.95%. It was suggested that the vaccines were immunogenic and had protective ability against bacterial infection in tiger groupers. Keywords: bacterial disease, polyvalent bacterial vaccine, tiger grouper

Tiger Grouper x Cammouflage Grouper (TGCG) (Epinephelus fuscoguttatus x Epinephelus microdon) production is still low nowadays due to the insufficient price of the live feed supply during the larval phase, which is nauplii Artemia sp. One of the live feed types which is able to become a substitute of nauplii Artemia sp. is Moina sp., which has better lysine, histidine, methionine, and leucine contents, besides having the sufficient price for the supply itself. However, Moina sp. is unable to survive in the high salinity, thus it has to be produced as a powder. This condition leads to a further research whether to observe that the Moina sp. powder supplementation is able to become nauplii Artemia sp. substitute through the increased growth and the optimum better lysine, histidine, methionine, and leucine contents in TGCG larvae. The research was conducted using the complete randomized design experimental method. The result showed that Moina sp. powder is compatible as good substitute candidate of nauplii Artemia sp., as it increased the larval length growth, even it had no difference in weight growth, but it was also able to increase the larval lysine, histidine, methionine, and leucine contents.

Background: Lactobacillus takes role in the formation of dental plaque which is the main factor that cause dental caries. The bacteria take role in the metabolism of glucose in the mouth, producing organic acids which lower the pH in the oral cavity. This situation can lead to the formation of dental caries because these bacteria can form biofilm as a defense of microorganisms to antibiotics and the immune response. Mangosteen peel has active ingredients such as flavonoid that can be used to inhibit biofilm. The ability of flavonoid compounds in the phenol group can make the bacterial enzyme becomes inactive, causing the activity of glucosyltransferase enzyme that usually used by bacteria to synthesize sucrose in the medium becomes glucan. As a result, bacterial biofilm formation is inhibited because the amount of glucan as a medium of bacterial attachment is limited, but until now the effectivity dose of flavonoid from mangosteen peel extract (Garcinia mangostana Linn.) is has not known yet Purpose: The aim of this study was to determine the inhibitory efficacy of flavonoid of mangosteen peel extract against the formation of Lactobacillus acidophilus bacterial biofilm. Methods: Bacteria that had been formed into biofilms was studied in two times treatment, one concentration reviewed by flavonoid from mangosteen peel extract and without reviewed by flavonoid from mangosteen peel extract. The treatment group was incubated at 37°C until day 8 since the first day incubated. The treatment group was washed with phosphate buffered saline (PBS) 4 times and dried. The treatment group stained with 0.2 ml 0.1% crystal violet and 15 min incubation. The treatment group was rinsed with distilled water 3 times and then dried. Solvent DMSO 100% was added as much as 0.1 ml in each well. Microtiter plate was shaken for 1 minute and then placed in to the microplate reader and then the OD (Optical Density) can be read. Results: There were significant differences between the inhibitory efficacy of flavonoid of mangosteen peel extract against Lactobacillus acidophilus biofilm bacteria, the control group and group treatments (p <0.05). Conclusion: Flavonoid of mangosteen peel extract with the concentration of 0.78% had inhibitory effect against Lactobacillus acidophilus biofilm bacteria
