Abstract Austropuccinia psidii is an invasive pathogenic rust that infects the Myrtaceae family. This rust is a threat to Myrtaceae plantations around the world due to its widespread distribution. In this study, we observed the presence of A. psidii in three species of Myrtaceae, i.e. Melaleuca cajuputi, Syzygium myrtifolium, and Syzygium polyanthum planted in Yogyakarta and Sukabumi. The symptoms of infection were yellow-reddish spot in young leaves, presence of urediniospores in infected spot, foliage, and branch dieback. To confirm the presence of A. psidii on those trees, a molecular detection was performed using specific primer for A. psidii (Ppsi1/Ppsi6) on DNA samples extracted from diseased leaves. The presence of A. psidii was proved by the presence of DNA amplicon sized around 500bp in all samples collected from three different hosts. In this study, S. myrtifolium was firstly reported to be infected by this rust in Indonesia. Further study about the presence and the economic impact of this pathogen in Indonesia should be conducted. Indonesia has many species numbers of Myrtaceae and some species are important for medicines, herbs, foods, and as industrial plants. A strategy to control this pathogen should be established to avoid large economic losses in Myrtaceae plantations in Indonesia.

Megalocytivirus is a genus of piscine viruses that belongs to the Iridoviridae family, and this family comprises red seabream iridovirus (RSIV), turbot reddish body iridovirus (TRBIV), and infectious spleen and kidney necrosis virus. RSIV causes high mortality and economic losses in the rock bream (Oplegnathus fasciatus) Korean aquaculture industry. The World Organization for Animal Health’s Office International des Epizooties has provided a manual for RSIV detection. However, detection against TRBIV has not been confirmed. In this study, a multiplex PCR method was established to detect two genotypes of Megalocytivirus, RSIV and TRBIV. New primer pairs were optimized for the PCR reaction. A mixture of one universal and two specific primer pairs could amplify three distinct products targeting three different genes. The sensitivity of the primer pairs was evaluated and results showed a detection limit of 2.0  105 copies for each target gene. Moreover, the primer pairs did not amplify any other viruses. The evaluation of multiplex PCR using 21 RSIV Korean isolates has shown that it can distinguish two genotypes of Megalocytivirus from 21 RSIV Korean isolates. Finally, we describe a new multiplex PCR method to detect different genotypes of Megalocytivirus simultaneously, which makes the diagnosis of viral diseases occurring in the Korean aquaculture industry more convenient.

This paper deals with state of the art risk and resilience calculations for industrial plants. Resilience is a top priority issue on the agenda of societies due to climate change and the all-time demand for human life safety and financial robustness. Industrial plants are highly complex systems containing a considerable number of equipment such as steel storage tanks, pipe rack-piping systems, and other installations. Loss Of Containment (LOC) scenarios triggered by past earthquakes due to failure on critical components were followed by severe repercussions on the community, long recovery times and great economic losses. Hence, facility planners and emergency managers should be aware of possible seismic damages and should have already established recovery plans to maximize the resilience and minimize the losses. Seismic risk assessment is the first step of resilience calculations, as it establishes possible damage scenarios. In order to have an accurate risk analysis, the plant equipment vulnerability must be assessed; this is made feasible either from fragility databases in the literature that refer to customized equipment or through numerical calculations. Two different approaches to fragility assessment will be discussed in this paper: (i) code-based Fragility Curves (FCs); and (ii) fragility curves based on numerical models. A carbon black process plant is used as a case study in order to display the influence of various fragility curve realizations taking their effects on risk and resilience calculations into account. Additionally, a new way of representing the total resilience of industrial installations is proposed. More precisely, all possible scenarios will be endowed with their weighted recovery curves (according to their probability of occurrence) and summed together. The result is a concise graph that can help stakeholders to identify critical plant equipment and make decisions on seismic mitigation strategies for plant safety and efficiency. Finally, possible mitigation strategies, like structural health monitoring and metamaterial-based seismic shields are addressed, in order to show how future developments may enhance plant resilience. ABSTRACT: The aim of this study was to investigate the importance of an acute disease that was characterized by sudden death associated with exercise that affected cattle in the region of the Araguaia River, especially in the municipality of Torixoréu, southeastern Mato Grosso, Brazil. Between August and September 2013, eighty farms in the municipality were visited and the pastures of these farms were inspected. Epidemiological questionnaires were completed from 65 farms. A plant identified as Niedenzuella stannea (N. stannea) of the Malpighiaceae family was attributed to the cause of sudden death in cattle, what causes major economic losses. Sudden death reports have occurred since 1970, when livestock was introduced into the region, and sudden death reports still occur. Outbreaks were reported to occur from May to October, but mainly during the dry season between June and August. N. stannea was identified at 41 farms and was always found in near to rivers. The most effective plant control methods included plowing the pastures and fields followed by manual application of Tordon® (Picloram and 2.4D). The administration of 5g/kg of young leaves which were collected from a farm with a sudden death history in cattle also caused sudden death in a sheep. Thus, it is concluded that the sudden death of cattle in the region of the Araguaia River is caused by the consumption of N. stannea.

The genus Prymnesium includes several species that produce toxins with cytotoxic, ichthyotoxic, neurotoxic and haemolytic activity. Bloom episodes of Prymnesium species have been reported from several parts of the world (North America, Europe, Africa, Asia and Australia), especially from temperate and subtropical regions and most of them from brackish waters. Blooms cause great economic losses to aquaculture and fisheries around the world. The ichthyotoxic and allelopathic effects of Prymnesium have been linked to the presence of Haemolysin 1, Prymnesins 1 and 2 and, more recently, fatty acids and fatty acid amides. The toxicology of this genus with regard to different growth conditions such as light, nutrients and other parameters has been well documented. It is unknown, however, whether different species and strains from the Prymnesium genus all produce the same types and level of toxins. In this study, we have determined the haemolytic activity of eight different strains from the genus Prymnesium in both exponential and stationary phases of growth. We have also evaluated the efficiency of the extraction solvent.

Abiotic stresses comprise all nonliving factors, such as soil salinity, drought, extreme temperatures, and metal toxicity, posing a serious threat to agriculture and affecting the plant production around the world. Peanut (Arachis hypogaea L.) is one of the most important crops for vegetable oil, proteins, minerals, and vitamins in the world. Therefore, it is of importance to understand the molecular mechanism of peanut against salt stress. Six transcriptome sequencing libraries including 24-hour salt treatments and control samples were constructed from the young leaves of peanut. A comprehensive analysis between two groups detected 3,425 differentially expressed genes (DEGs) including 2,013 upregulated genes and 1,412 downregulated genes. Of these DEGs, 141 transcription factors (TFs) mainly consisting of MYB, AP2/ERF, WRKY, bHLH, and HSF were identified in response to salinity stress. Further, GO categories of the DEGs highly related to regulation of cell growth, cell periphery, sustained external encapsulating structure, cell wall organization or biogenesis, antioxidant activity, and peroxidase activity were significantly enriched for upregulated DEGs. The function of downregulated DEGs was mainly enriched in regulation of metabolic processes, oxidoreductase activity, and catalytic activity. Fourteen DEGs with response to salt tolerance were validated by real-time PCR. Taken together, the identification of DEGs’ response to salt tolerance of cultivated peanut will provide a solid foundation for improving salt-tolerant peanut genetic manipulation in the future.

Aim of study: The honeybee, Apis mellifera, is one of the most important pollinators in the world. Apicultural activity and pollination services have been affected by the decline in the honeybee population, which may be due to the interaction of multiple risk factors, such as changes in agricultural production, use of pesticides and presence of pathogens. Viruses, in particular, are suspected to be drivers of colony mortality. In this scenario, the aim of this study was to determine the presence of honeybee viruses (IAPV, DWV, SBV, ABPV, BQCV, CBPV) in A. mellifera populations using a RT-mPCR assay.Area of study: Apiaries were situated in Pichincha, Ecuador.Material and methods: Samples were collected from seventeen apiaries that exhibited mortality but without specific clinical signs. Each sample comprised 15 individuals. After RNA extraction, a multiplex PCR analysis was performed for presence of six viruses (IAPV, DWV, SBV, ABPV, BQCV, CBPV).Main results: Four of the viruses (ABPV, DWV, BQCV and SBV) were found in co-infections in these colonies, with ABPV and SBV also being found in simple infections.Research highlights: To our knowledge, this is the first molecular detection of BQCV and SBV in Ecuador. These findings suggest that some of the above viruses could be involved in weakening these colonies.

Apples are the most productive fruits in the world with a lot of medicinal and nutritional value. Significant economic losses occur frequently due to various diseases that occur on a huge scale of apple production. Consequently, the effective and timely discovery of apple leaf infection becomes compulsory. The proposed work uses optimal deep neural network for effectively identifying the diseases of apple trees. This work utilizes a convolution neural network to capture the features of Apple leaves. Extracted features are optimized with the help of the optimization algorithm. The optimized features are utilized in the leaf disease identification process. Here the traditional DNN algorithm is modified by means of weight optimization using adaptive monarch butterfly optimization (AMBO) algorithm. The experimental results show that the proposed disease identification methodology based on the optimized deep neural network accomplishes an overall accuracy of 98.42%.

The involvement of picornaviruses in calf diarrhoea was evaluated by the analysis of 127 faecal samples collected from diarrhoeic calves during 2014–2016. Virus detections were carried out by PCR using generic or specific primer pairs. One-third of the faecal samples (33.86%) were found to be positive for one or more of the studied viruses. Bovine kobuvirus was detected in 22.83%, bovine hungarovirus in 11.02%, while bovine enterovirus 1 in 5.51% of the samples. The sequences of the PCR products indicated the existence of novel variants in all the three virus species. When comparing the partial sequences, the nucleotide sequence identities between our newly detected viruses and those previously deposited to the GenBank ranged between 76 and 99%. Phylogenetic analyses revealed a novel lineage within the species Hunnivirus A. Our findings suggest that these viruses should be regarded as possible aetiological agents of calf diarrhoea. Based on the newly determined sequences, we designed and tested a new generic PCR primer set for the more reliable detection of bovine hungaroviruses. This is the first report on the molecular detection of the presence of bovine hungarovirus, bovine kobuvirus and bovine enterovirus 1 in the faecal samples of diarrhoeic calves in Turkey.
