Abstract The study aimed to determine the immune function markers of clinically healthy cows and cows with subclinical and clinical mastitis caused by the coagulase-negative staphylococci. The materials were milk and blood of cows (n = 20; first group) had the clinical mastitis caused by S. aureus and S. haemolyticus, another 20 (second group) had the subclinical mastitis caused by S. aureus and S. haemolyticus, another 20 (third group) were clinically health cows. The milk was examinated by a Californian mastitis test. The immune markers of blood were researched by ELISA using Bovine Elisa Kit (Cloud-CloneCorp. HoustonTX, USA) and relevance methods (The guidelines for the assessment and correction of animal immune status, Voronezh, Russia, 2005). It has been found that the blood of cows of the 2d group had a high level of Interleukin- 2 (15.9±0.4 pg/mL), TNF-α (639.2±19.1 pg/mL), CIC (0.128±0.01 g/L), Serum Bactericidal Activity (78.5±1.4 %) compared with 3d group. It was the reducing of Ig level by 19.6% (20.6±0.3g/L) and Lysozyme activity by 28.4% (2.097±0.02 µg/mL) (P < 0,05-0,001). The comparative analysis of immune function markers in the blood of 1st and 3d groups showed the reduction of TNF-α by 23,3%, Ig level by 19,6 %, Lysozyme activity by 32,8%, T- lymphocytes by 27,2%, and the increasing of CIC by 52,3%, Serum Bactericidal Activity by 6,8%, B- lymphocytes by 10,1% (P < 0,05-0,001), Phagocytic activity of leucocytes by 6,4%. Mastitis events caused by the coagulase-negative staphylococci were associated with reducing of cellular and humoral immunity by twice increasing of Interleukin-2 level.

Objective: To study the prevalence of Coagulase-negative Staphylococci in clinical and subclinical mastitis in dairy cows. Design: Case-control study. Animals: 415 lactating Holstein cows. Procedures: A total of 896 quarter milk samples were collected aseptically from 415 dairy cows for bacteriological examination (mastitic cows, n=50; healthy cows, n=174). Identification of different bacterial isolates followed by antibiotic sensitivity test for Coagulase-negative Staphylococci (CNS) was carried out, using disc-diffusion method against 13 different antibiotics. Results: 224 out of 415 cows (54%) were proven to be mastitic (12% clinical mastitis, 42 % subclinical mastitis). Coagulase-negative Staphylococci were the prevalent isolate (32%). CNS was recorded in 25.7 % of clinical cases and 34% of subclinical mastitis. The antibiogram of CNS isolates against 13 different antibiotics revealed high level of resistance to ampicillin (85%) and oxacillin (85%). However, they were less resistant to vancomycin (5%). The other antibiotics showed variable levels of resistance ranging from 40-75%. Conclusion and clinical relevance: Coagulase-negative Staphylococci were the most common bacteria isolated from mastitic cases. Multi-drug resistance was observed among CNS with high resistance to ampicillin and penicillin, so the use of such B-lactam antibiotics should be restricted specially if there were suspected cases of Coagulase-negative Staphylococci.

Goat milk ranks fourth in terms of global milk production and lately it has become increasingly popular among consumers. Unfortunately, mastitis is one of the most common diseases that affects dairy goats, with serious economic consequences and food safety matters. The prevalence of clinical mastitis is lower than 5% and the main aetiological agent is S. aureus. On the other hand, the prevalence of subclinical mastitis is between 5-30%, with coagulase negative staphylococci (CNS) representing the most often isolated microorganisms.The aim of this paper is to highlight the main aspects regarding the aetiology of goat mastitis, as well as the importance of the milk somatic cell count (MSCC) in the diagnosis process. Although the inflammation of the mammary gland in goats is not as frequently diagnosed compared to cow mastitis, there are several aspects worth discussing in order to fully understand the pathogenesis of intramammary infections.

Mastitis is a major health problem of the udder in dairy sheep breeds. For diagnosis of subclinical mastitis, somatic cell count (SCC) is commonly used. The presence of pathogens in the udder causes the increase of leukocytes and thus SCC in milk. Therefore, the aim of this study was to evaluate the presence of pathogens in the milk of ewes and the possible relationship with SCC. The changes of leukocytes subpopulation in milk samples with high SCC were evaluated as well. The experiment was carried out on a dairy farm with the Lacaune breed. This study was conducted on 45 ewes (98 milk samples) without signs of clinical mastitis. Based on somatic cell count, samples were divided to five SCC groups: SCC1 &lt; 200 000 cells/ml (45 milk samples); 200 000 ≤ SCC2 &lt; 400 000 cells/ml (10 milk samples); 400 000 ≤ SCC3 &lt; 600 000 cells/ml (six milk samples); 600 000 ≤ SCC4 &lt; 1 000 000 cells/ml (six milk samples); SCC5 ≥ 1 000 000 cells/ml (31 milk samples). No pathogens were observed in the majority of milk samples (60.20%). Coagulase-negative staphylococci (CNS) were the most commonly isolated pathogens from the milk of ewes (86.11%). Staphylococcus epidermidis had the highest incidence from CNS (35.48%). In the SCC5 group, up to 79.31% of bacteriological samples were positive. The percentage of leukocytes significantly increased (P &lt; 0.001) in the samples with higher SCC (≥ 200 × 10<sup>3</sup> cells/ml) in comparison to the group SCC1. Also, the percentage of polymorphonuclear cells (PMNs) was significantly higher with increasing SCC (P &lt; 0.001). In conclusion, the presented results showed that the high SCC was caused by the presence of the pathogen in milk. Thus SCC &lt; 200 000 cells/ml and leukocyte subpopulation, especially PMNs, could be considered as important tools in udder health programs applied in dairy ewes.

This study was carried out to determine the prevalence of coagulase-negative staphylococci in clinical and subclinical mastitis in commercial and small-scale farms in Zimbabwe. Thirty five quarter milk samples from clinical mastitis cases and 371 quarter milk samples from cows with subclinical mastitis were cultured for bacterial pathogens. The most frequent pathogens isolated in clinical mastitis were the enteric bacteria (31.4 %), followed by coagulase negative staphylococci (22.9 %) and then Staphylococcus aureus (17.1 %), whereas in subclinical mastitis S. aureus (34.2 %) and coagulase-negative staphylococci were (33.2 %) the most common. Bacillus species were only isolated in milk samples from subclinical mastitis. Coagulase-negative staphylococci were observed in mixed infections with other bacteria in only 2.2 % of the 406 milk samples from clinical and subclinical mastitis where they were isolated together with Bacillus species in 6 of the 9 mixed infection cases. About 95 % of the milk samples from which 131 coagulase-negative staphylococci were isolated had correspondingly high somatic cell counts. The coagulase-negative staphylococci isolated most frequently were S. chromogenes (7.9 %), S. epidermidis (7.4 %) and S. hominis (5.9 %). They were all associated with high somatic cell counts. All the coagulase-negative staphylococci isolates were susceptible to cloxacillin and erythromycin, and more than 90 %of the isolates were susceptible to neomycin, penicillin and streptomycin. The highest resistance was to tetracycline (17.6 %), followed by lincomycin (13.7 %). About 8 % of the isolates were resistant to both penicillin and streptomycin.

Six hundred and thirty clinically-normal milk samples from dairy goat flocks comprising a mixed population of German Alpine, Toggenburg, Saanen and Galla crosses were exam-ined over a 3-month period to determine the prevalence of bacterial organisms. Bacteria were isolated in 28.7 % of the milk samples (181/630) either singly (92.8 %) or in combination (7.2 %). The most prevalent bacterial organisms were Staphylococcus spp. (60.3 %), followed by Micrococcus spp. (17.7 %), Acinetobacter spp. (5 %), Actinomyces spp. (5%) and Streptococcus spp. (1.1 %). The Staphylococcus spp. were mainly coagulase negative (64.3 %). Coagulase-- negative staphylococci and coagulase-positive staphylococci accounted for 37.5 % and 22.7 % respectively of the total bacteria isolated. The isolation of bacteria, some of which are important in clinical and subclinical mastitis, in apparently normal caprine milk, indicates that particular attention should be given to the management of these dairy goat flocks in order to avoid the development of cases of clinical mastitis.

The responses of five lactating East Friesian milk ewes to experimental mammary infection with Staphylococcus epidermidis and of five control ewes were examined over a period of 10 weeks. Infection caused an influx of neutrophils into milk, the numbers of which started to rise 4 h post infection and peaked 24 h after infection. The initial response was accompanied by mild fever and mild leucopaenia in blood (8 h after infection). No other signs of systemic infection were observed. Milk appeared normal at all times, although the milk yield of infected ewes tended to decline. Staphylococci were absent in milk from four ewes at 2 d and at 3 d after infection, but re-emerged intermittently in four of five ewes at subsequent samplings. Cytokines in milk were measured by ELISA. IL-8 was elevated in infected glands at 2 h and peaked at 8 h. In the four ewes intermittently shedding bacteria, IL-8 remained elevated until the final sampling at 10 weeks. IL-1β was transiently elevated at 1 d and 2 d and showed a pronounced peak in one sheep. Milk samples from this ewe were bacteriologically negative, somatic cell count (SCC) was within the normal range and the concentrations of IL-1β, as well as IL-8, were similiar to the control group (n=5) from 1 week after infection until the final sampling. Histological examination revealed leucocytic infiltrates in the four glands remaining infected at the end of the experiment, and a high level of CD5+ lymphocytes in three ewes. The results suggest that the relationship between the initial neutrophil influx and the proinflammatory cytokines may be responsible for determining the course of infection. Subclinical mastitis due to coagulase-negative staphylococci leads to minor changes in milk yield and milk constituents.

Abstract The aim of this study was to evaluate the occurrence of mastitis and its impact on the reproductive parameters in a herd of 180 dairy cows. Based on the herd records 127 cows of Slovak spotted cattle and their crosses with red Holstein were selected for study between 1—2 months after calving. The examination of the health status of the mammary glands consisted of: the clinical examination of the udder, the California mastitis test (CMT) supplemented by the collection of mixed milk samples, and the laboratory examination of bacterial pathogens causing the mastitis. In addition to the mam­mary investigation, reproduction indicators such as the length of the insemination interval, the service period, the intercalving period and the insemination index were also analyzed. The results of this study indicated: a high incidence of mastitis (41.6 %), especially latent (21.2 %), subclinical (15.7 %) and clinical (4.7 %) forms were most common in the herd. The most frequently isolated bacteria from the infected milk samples were: coagulase negative staphylococci (54.1 %), S. aureus (16.9 %), Streptococcus spp. (15.0 %), A. viridans (7.5 %) and Ent. faecalis (6.4 %). According to the available literature, the optimum values of the intercalving period were 365—400 days, the insemination interval 55—80 days, the insemination index 1.2—2 and the service period 60—110 days. In comparison, our results showed increased, unsatisfactory reproductive values in the group of dairy cows with clinical mastitis. While in healthy cows as well as in groups of cows with latent and subclinical mastitis, all of the reproductive indicators were within the optimal levels.

Bacterial mastitis is a major health problem in dairy sheep worldwide. It is associated with reduced milk yield and occasionally involuntary culling of affected ewes, as well as insufficient growth and mortality of lambs. In general, the incidence of clinical mastitis in ewes during lactation is lower than 5%. However, the prevalence of subclinical mastitis is variable ranging from 10-30% or more. In clinical cases of bacterial intramammary infections, Staphylococcus aureus is dominant. In the case of subclinical mastitis the prevailing isolates are Coagulase-Negative Staphylococci(CNS). Moreover, predisposing factors related to the environment, genetics, udder morphology and nutrition, are likely to contribute to the occurrence of mastitis. Hence, control of the latter factors and bacteriological contamination during milking are the main preventive measures. The notion is that detection of the ovine genome regions involved in mastitis resistance will also facilitate the most effective control of mastitis in flock level. Appropriate genetic selection, together with the implementation of preventive measures, could reduce the negative consequences of bacterial mastitis.

AbstractThe aim of the present study was to determine the proportions of individual coagulase-negative Staphylococcus species in clinical and subclinical mastitis. The material consisted of 100 CNS isolates obtained from 223 milk samples collected from cows with clinical and subclinical mastitis. Coagulase- negative staphylococci constituted 44.8% of all isolated microorganisms. CNS were isolated from the mammary gland secretions of 86 cows from farms in the Lublin region (Poland). Clinical mastitis was found in 20 whereas subclinical mastitis in 66 study cows (23.3% and 76.7%, respectively). The symptoms of clinical mastitis were mild. The clinical forms of mastitis concerned mainly the first or second lactation.Subclinical mastitis was most commonly observed during the second lactation. Four CNS species (S. xylosus, S. chromogenes, S. haemolyticus and S. sciuri ) were isolated from clinical and subclinical mastitis. S. xylosus was the commonest CNS species isolated from cows with clinical mastitis whereas S. chromogenes was the most prevalent one in subclinical mastitis cases. The three CNS species (S. warneri, S. hominis and S. saprophyticus) caused only subclinical mastitis.

Thirty-two farms (n=535cows) located in the state of Guanajuato, Mexico, were sampled. Pathogens from bovine subclinical mastitis (SCM) and clinical mastitis (CLM) were identified by 16S rDNA and the sensitivity to both antibiotics and bacteriocins ofBacillus thuringiensiswas tested. Forty-six milk samples were selected for their positive California Mastitis Test (CMT) (≥3) and any abnormality in the udder or milk. The frequency of SCM and CLM was 39.1% and 9.3%, respectively. Averages for test day milk yield (MY), lactation number (LN), herd size (HS), and number of days in milk (DM) were 20.6 kg, 2.8 lactations, 16.7 animals, and 164.1 days, respectively. MY was dependent on dairy herd (DH), LN, HS, and DMP<0.01, and correlations between udder quarters from the CMT were around 0.49P<0.01. Coagulase-negative staphylococci were mainly identified, as well asStaphylococcus aureus,Streptococcus uberis,Brevibacterium stationis,B. conglomeratum, andStaphylococcus agnetis. Bacterial isolates were resistant to penicillin, clindamycin, ampicillin, and cefotaxime. Bacteriocins synthesized byBacillus thuringiensisinhibited the growth of multiantibiotic resistance bacteria such asS. agnetis,S. equorum,Streptococcus uberis,Brevibacterium stationis, andBrachybacterium conglomeratum, but they were not active againstS. sciuri, a microorganism that showed an 84% resistance to antibiotics tested in this study.
