Abstract Mycotoxins are harmful secondary fungal metabolites which can contaminate a variety of feedstuffs worldwide. Various negative effects in vivo are associated with these toxins and severity is dependent on the level, type, and duration of exposure and age, species, and health status of animals. Wheat middlings or “midds” are a by-product of flour milling and are used in a wide range of livestock rations including frequent inclusion in pelleted feeds. The objectives of the current study were to determine the occurrence of mycotoxins in wheat midds samples and to evaluate the potential risk posed to livestock. Since November 2015, 97 samples submitted from 21 states and provinces have been screened via LC-MS/MS technique for the presence of six major mycotoxin groups: aflatoxins, type A trichothecenes, type B trichothecenes (B-Trich), fumonisins, zearalenone, and ochratoxin A (OTA) at Romer Labs (Union, MO, USA) or Activation Laboratories (Ancaster, Ontario, Canada). A subset of samples were also screened for the presence of six ergot alkaloids. Parameters of the main mycotoxins detected are presented in Table 1. On average, 1.7 mycotoxin groups were detected per sample with 91.8% of samples testing positive for at least one mycotoxin. The B-Trich deoxynivalenol was detected in 89.7% of samples (mean 1871.5 ± 157.5 ppb). OTA was also frequently detected (30.9% positive), but at low risk levels (4.2 ± 0.5 ppb). Out of 33 samples screened for ergot alkaloids, 72.7% were positive (493.3 ppb ± 68.3 ppb). Wheat midds are a commonly used feed ingredient for various livestock species. The types and concentrations of mycotoxins detected may pose challenges to livestock health, performance, and reproduction. Special consideration for young animals which are especially susceptible to mycotoxin challenges, such as calves consuming starter feeds, is warranted.

Inositol phosphates (InsPs), especially myo-inositol hexakisphosphate (InsP6), are important binders of phosphorus and minerals in plant seeds. However, they have long been considered as anti-nutritional components of plant foods due to their possible negative effects on the absorption of minerals and proteins in mammals. On the other hand, recent findings have found InsPs to be ubiquitous in eukaryote cells and actively participating in multiple cell functions. In vivo and in vitro studies have also documented the preventive potential of these compounds against the development of a wide range of diseases. In light of these findings, interest in the relationship between these compounds and human health has been renewed. It is suggested that the interactions of InsPs with other nutrients in the gut are complex, that the absorption of dietary InsPs might be implied but is not certain, and that the disease fighting capabilities of InsPs hold both promises and limitations. At the same time, the analysis of these compounds in foods and biological samples still faces many challenges, calling for more advanced modification and developments in the future.

Abstract Mycotoxins are secondary fungal metabolites that are detrimental to animal health and productivity. This study investigated occurrence and contamination levels of mycotoxins in the 2019 US corn harvest, including corn grain and corn byproduct feed ingredients (distillers dried grains, gluten feed, etc.). Corn samples from the 2019 harvest and corn DDGS and other byproducts from October 2019 through April 2020 were screened via the LC-MS/MS technique for the presence of six major mycotoxin groups: aflatoxins, type A trichothecenes, type B trichothecenes (B-Trich), fumonisins (FUM), zearalenone (ZEN) and ochratoxin A. Parameters of the main toxins found were compared to the two prior harvest years using the Kruskal-Wallis Test (Prism7, GraphPad, La Jolla, CA) and are presented in Table 1. Mean toxin count per sample in corn grain was similar in 2019 as 2018, with just over two toxins per sample. Mean B-Trich level (parts per billion, ppb) was elevated in 2019 from 2017. Fumonisin contamination increased in 2019 compared to 2017, and prevalence was similar to 2018. Zearalenone contamination and prevalence were similar in 2019 compared to 2018 levels. Co-contamination in corn byproduct feeds was steady, averaging nearly three toxins per sample. Mean B-Trich contamination was higher than the 2017 crop, and FUM contamination was higher in 2019 than 2017 and 2018. Zearalenone levels in 2019 byproducts are the highest seen in the past three years. Preliminary results of the 2019 corn-based feed ingredients survey suggest mycotoxin occurrence and contamination levels are approaching those observed in the challenging 2018 crop. Furthermore, there are continued concerns for co-contamination and greater ZEN contamination that may impose negative effects on growth, health and reproductive performance in ruminants.

Pastures are key feed sources for dairy production and can be contaminated with several secondary metabolites from fungi and plants with toxic or endocrine-disrupting activities, which possess a risk for the health, reproduction and performance of cattle. This exploratory study aimed to determine the co-occurrences and concentrations of a wide range of mycotoxins, phytoestrogens and other secondary metabolites in grazing pastures. Representative samples of pastures were collected from 18 Austrian dairy farms (one sample per farm) between April to October 2019. After sample preparation (drying and milling) the pastures were subjected to multi-metabolite analysis using LC-MS/MS. In total, 68 metabolites were detected, including regulated zearalenone and deoxynivalenol (range: 2.16–138 and 107–505 μg/kg on a dry matter (DM) basis, respectively), modified (3-deoxynivalenol-glucoside, HT-2-glucoside) and emerging Fusarium mycotoxins (e.g., enniatins), ergot alkaloids and Alternaria metabolites along with phytoestrogens and other metabolites. Aflatoxins, fumonisins, T-2 toxin, HT-2 toxin and ochratoxins were not detected. Of the geo-climatic factors and botanical diversity investigated, the environment temperature (average of 2 pre-sampling months and the sampling month) was the most influential factor. The number of fungal metabolites linearly increased with increasing temperatures and temperatures exceeding 15 °C triggered an exponential increment in the concentrations of Fusarium and Alternaria metabolites and ergot alkaloids. In conclusion, even though the levels of regulated mycotoxins detected were below the EU guidance levels, the long-term exposure along with co-occurrence with modified and emerging mycotoxins might be an underestimated risk for grazing and forage-fed livestock. The one-year preliminary data points out a dominant effect of environmental temperature in the diversity and contamination level of fungal metabolites in pastures.

Cold atmospheric plasma use in clinical studies is mainly limited to the treatment of chronic wounds, but its application in a wide range of medical fields is now the goal of many analyses. It is therefore likely that its application spectrum will be expanded in the future. Cold atmospheric plasma has been shown to reduce microbial load without any known significant negative effects on healthy tissues, and this should enhance its possible application to any microbial infection site. It has also been shown to have anti-tumour effects. In addition, it acts proliferatively on stem cells and other cultivated cells, and the highly increased nitric oxide levels have a very important effect on this proliferation. Cold atmospheric plasma use may also have a beneficial effect on immunotherapy in cancer patients. Finally, it is possible that the use of plasma devices will not remain limited to surface structures, because current endeavours to develop sufficiently miniature microplasma devices could very likely lead to its application in subcutaneous and internal structures. This study summarises the available literature on cold plasma action mechanisms and analyses of its current in vivo and in vitro use, primarily in the fields of regenerative and dental medicine and oncology.

Vitamin K4 was labelled with 99mTc with an efficiency higher than 97%. The compound was stable up to 24 h at room temperature, and its biodistribution in NMRI mice indicated its in vivo stability. Blood radioactivity levels were high over a wide range. 10% of the injected activity remained in blood after 24 h. Excretion was mostly via kidneys. Only the liver and kidneys concentrated appreciable amounts of radioactivity. Testis/soft tissue ratios were 1.4 and 1.57 at 6 and 24 h, respectively. Testis/blood ratios were lower than 1. In vitro studies with mouse blood indicated that 33.9 ±9.6% of the radioactivity was associated with RBCs; it was washed out almost completely with saline. Protein binding was 28.7 ±6.3% as determined by TCA precipitation. Blood clearance of 99mTc-l<4 in normal subjects showed a slow decrease of radioactivity, reaching a plateau after 16 h at 20% of the injected activity. In scintigraphic images in men the testes could be well visualized. The right/left testis ratio was 1.08 ±0.13. Testis/soft tissue and testis/blood activity ratios were highest at 3 h. These ratios were higher than those obtained with pertechnetate at 20 min post injection.99mTc-l<4 appears to be a promising radiopharmaceutical for the scintigraphic visualization of testes.

Voynich-Syanozhentsky, V.A. Golubkova, G.V. Lyapin and others. The department solved a wide range of tasks, the main areas of research were the following: – theoretical and applied hydrodynamics and hydraulics, with reference to the open channel flows that affect the state and level of safety of the hydraulic structures; – integrated use and protection of water bodies – water sources and water sources of water resources used in land reclamation; – development of measures and technical solutions for the protection of objects from the negative effects of water; – theoretical substantiation of works to improve the safety level of the GTS (declaration); – development and implementation of digitalization methods for solving design, construction, operation and control of landreclamation facilities. Currently, promising areas of research is the development of a decision-making algorithm in the designation of measures to rationalize the provision of resources to water amelioration. The algorithm is developed on the basis of a detailed study, systematization and processing of data both on safety and on the efficiency of systems and structures, ensuring the delivery of irrigation water of the required quality and in sufficient quantity from a water source to the field.

Alumina refineries are among the leading sources of atmospheric air pollution with a wide range of pollutants hazardous to human respiratory organs. It is relevant to study and evaluate the occurrence of the risks for development of respiratory diseases in children living in the area affected by the emission components of an alumina refinery. We assessed air quality of the area under observation and comparison according to monitoring observations, risk of non-carcinogenic effects from the respiratory organs. The content of chemicals in the blood and urine adequate to risk factors was quantified. The structure of individual groups of respiratory diseases was analyzed. The causal relationships of violations of laboratory parameters with an increased content of chemicals in biological media were evaluated. It was found that an aerogenic exposure of chemical pollutants is formed on the territory with the production of metallurgical alumina. It determines the risk for development of respiratory diseases, exceeding an acceptable level up to 49.9 times. In the exposed children, the content of manganese, chromium, nickel, copper, xylenes, formaldehyde and aluminum, fluoride ion in the urine was increased to 4.2 times in relation to the indices in the comparison group. A high level of additional respiratory morbidity(1.8 times) was revealed. Chronic lymphoproliferative diseases of the nasopharynx and inflammatory diseases of the upper respiratory tract (up to 6.6 times more often), inflammatory diseases with a predominance of the mechanism of allergic inflammation ( up to 2.1 times more often)are more often detected in the framework of the respiratory diseases. Negative effects on the part of the respiratory system in the form of activation of antioxidant processes, the development of an inflammatory reaction, local, general and specific sensitization of the respiratory tract were established. It confirms the occurrence of the risks for the development of respiratory diseases in children in the exposure area of the chemical factors of alumina refinery-associated economic activity.

Background: Genus Berberis (family Berberidaceae), which contains about 650 species and 17 genera worldwide, has been used in folklore and various traditional medicine systems. Berberis Linn. is the most established group among genera with around 450-500 species across the world. This comprehensive review will not only help researchers for further evaluation but also provide substantial information for future exploitation of species to develop novel herbal formulations. Objective: The present review is focussed to summarize and collect the updated review of information of Genus Berberis species reported to date regarding their ethnomedicinal information, chemical constituents, traditional/folklore use, and reported pharmacological activities on more than 40 species of Berberis. Conclusion: A comprehensive survey of the literature reveals that various species of the genus possess various phytoconstituents mainly alkaloids, flavonoid based compounds isolated from different parts of a plant with a wide range of pharmacological activities. So far, many pharmacological activities like anti-cancer, anti-hyperlipidemic, hepatoprotective, immunomodulatory, anti-inflammatory both in vitro & in vivo and clinical study of different extracts/isolated compounds of different species of Berberis have been reported, proving their importance as a medicinal plant and claiming their traditional use.

Cancer is the second leading cause of morbidity and mortality worldwide. Although chemotherapy and radiotherapy enhance the survival rate of cancerous patients but they have several acute toxic effects. Therefore, there is a need to search for new anticancer agents having better efficacy and lesser side effects. In this regard, herbal treatment is found to be a safe method for treating and preventing cancer. Here, an attempt has been made to screen some less explored medicinal plants like Ammania baccifera, Asclepias curassavica, Azadarichta indica, Butea monosperma, Croton tiglium, Hedera nepalensis, Jatropha curcas, Momordica charantia, Moringa oleifera, Psidium guajava, etc. having potent anticancer activity with minimum cytotoxic value (IC50 >3μM) and lesser or negligible toxicity. They are rich in active phytochemicals with a wide range of drug targets. In this study, these medicinal plants were evaluated for dose-dependent cytotoxicological studies via in vitro MTT assay and in vivo tumor models along with some more plants which are reported to have IC50 value in the range of 0.019-0.528 mg/ml. The findings indicate that these plants inhibit tumor growth by their antiproliferative, pro-apoptotic, anti-metastatic and anti-angiogenic molecular targets. They are widely used because of their easy availability, affordable price and having no or sometimes minimal side effects. This review provides a baseline for the discovery of anticancer drugs from medicinal plants having minimum cytotoxic value with minimal side effects and establishment of their analogues for the welfare of mankind.

: Allostery is an efficient and particular regulatory mechanism to regulate protein functions. Different from conserved orthosteric sites, allosteric sites have distinctive functional mechanism to form the complex regulatory network. In drug discovery, kinase inhibitors targeting the allosteric pockets have received extensive attention for the advantages of high selectivity and low toxicity. The approval of trametinib as the first allosteric inhibitor validated that allosteric inhibitors could be used as effective therapeutic drugs for treatment of diseases. To date, a wide range of allosteric inhibitors have been identified. In this perspective, we outline different binding modes and potential advantages of allosteric inhibitors. In the meantime, the research processes of typical and novel allosteric inhibitors are described briefly in terms of structureactivity relationships, ligand-protein interactions and in vitro and in vivo activity. Additionally, challenges as well as opportunities are presented.
