Abstract A variety of different substances can promote adverse effects on livestock and companion animals, varying from a decrease in overall growth, to frank toxicological response and death. Many substances that are the cause of adverse effects are either components in feed, part of the forage landscape for grazing animals, or even a part of the nutritive feed composition for pets. It has been well-known that vitamin D intake increases bone mineralization, but methionine consumption may also positively impact bone density. Excess consumption of other nutrients may lead to adverse effects, as current research found that excess tryptophan consumption in weaned pigs adversely affects intestinal morphology and tight junction proteins in other species. However, some studies have found that different animals can have opposing effects from consuming the same substances. For example, a study in mice has found that consumption of chitosan impairs intestinal barrier integrity, while twice the intake by weaned piglets improved the average daily gain. This presentation will discuss examples of nutrients and other substances that may adversely impact the growth and reproductive capacity of companion animals and livestock, underscoring the need for the completion of safety studies of potentially new ingredients to evaluate their long-term use in commercial feed formulations.

The effect of glutamine (Gln) on growth performance, intestinal morphology and intestinal barrier function were evaluated in broilers. A total of 320 birds were divided into a control group (CON) and three experimental groups (Gln 1, Gln 2 and Gln 3). Broilers of group CON received basal diet; broilers in group Gln 1, Gln 2 and Gln 3 were supplemented with 0.5%, 1.0% and 1.5 % Gln, respectively, for 42 days. The results indicated that Gln has no influence on the average daily gain (ADG) among the treatments in the periods of 1 to 21 d, 22 to 42 d and 1 to 42 d (P > 0.05). However, Gln improved average daily feed intake (ADFI) and feed intake: average daily gain (F: G), increased the villus height, villus height to crypt depth ratio (V/C) and the activities of sucrose, the ZO1, claudin-1 and occluding mRNA expression levels (P 0.05). Moreover, Gln decreased the crypt depth of jejunum and ileum in broilers at days 21 and 42 (P 0.05). In conclusion, Gln had a positive effect on growth performance and gut parameters by modifying the function of the intestinal mucosa barrier.

Abstract Background Necrotic enteritis (NE), which is caused by Clostridium perfringens (C. perfringens), is an economically important disease in broiler. Among normal flora in the broiler intestinal, Clostridium butyricum (C. butyricum) has been identified as a probiotic agent that reduces the susceptibility of broilers to C. perfringens. It also promotes the repair of broiler intestinal damage from harmful intestinal pathogens. However, the effects of C. butyricum infection on broiler intestinal integrity during NE are largely unknown. In this study, we investigated the effects of C. butyricum on the growth performance, intestinal morphology and barrier function, and the functions of immune-related cytokines under NE in broilers. Results Infected group (PC) showed significant decrease in the average daily gain (ADG, p < 0.01) and increase in feed conversion ratio (FCR, p < 0.01) compared with C. butyricum (NECB1 and NECB2) through dietary supplement. Supplementation of C. butyricum (NECB1 and NECB2) restored the intestinal villus height, increased the crypt depth, and reduced the intestinal damage under NE. Furthermore, broilers infected with NE (PC) had higher serum IgA and endotoxin content, and after addition with C. butyricum (NECB1 and NECB2) returned to normal level. In addition, compared with PC, supplementation of C. butyricum (NECB1 and NECB2) restored intestinal barrier function-related genes (such as CLDN-1, CLDN-3, OCLN, MUC2, ZO-1, and CLDN5), cytokines (such as TNF-α, IL-10, IL-6, and TGFB1) and C. perfringens plc gene expression. Moreover, C. butyricum (NECB1 and NECB2) could restore the decrease in Gt (p < 0.01) and Isc caused by NE (PC). In addition, NECB2 reduced the upregulation of FD4 flux caused by NE infection (p < 0.01). Conclusion Dietary supplementation of C. butyricum into broilers with NE improved chicken growth performance, intestinal morphology, intestinal barrier function, and immune function. Notably, no statistical difference was observed with the addition of C. butyricum at different time points.

We investigated the effects of rapamycin (RAPA) and chloroquine (CQ) in supporting growth performance and the intestinal mucosal barrier in response to deoxynivalenol (DON) in piglets. A total of 32 healthy weaned piglets (bodyweight 7.10±0.58 kg) were divided into four groups and treated daily with RAPA (1 mg/kg BW), CQ (10 mg/kg BW), or a control volume of normal saline (two groups) until the end of the experiment. After feeding a basal diet for seven days, three groups were then switched to mildewed feed containing 1 mg kg/DON for a further seven days. In contrast to the control group, DON-treated piglets showed decreased average daily gain (ADG) and daily feed intake (ADFI), as well as negatively affected intestinal morphology as indicated by villus height, crypt depth, and tight junction protein expression. A group treated with RAPA and DON showed increased intestinal autophagy, aggravated inflammatory responses, and damage to the intestinal mucosa and permeability, leading to reduced growth performance. Meanwhile, a group treated with CQ and DON showed indices comparable to the non-DON control group, with alleviated inflammatory cytokines and healthy intestinal morphology and structure. They also showed better growth performance compared to DON treatment alone. These findings have important implications for mediating autophagy against DON in vivo, as well as the potential for CQ in improving growth performance and maintaining intestinal barrier integrity in weanling piglets.

Abstract To optimize the use of dietary P by pigs, 5 feeding strategies were studied in a 3-phase feeding trial on 240 pigs (initial bodyweight (BW) of 31 kg): 1) C-C-C providing 100% of digestible phosphorus (Pdig, 4.3 g/kg STTD) and calcium (Ca, 9.7 g/kg) requirement to maximize bone mineralization, 2) L-L-L 60% of the Pdig and Ca requirements of C-C-C, 3) Phyt-Phyt-Phyt (phosphate-free, with phytase, 750, 686, 390 FTU/kg), providing 60% of Pdig and Ca requirements in phase 1, then 100%, 4) and 5) C in phases 1 and 3, and 60% of the need for Pdig in phase 2, associated with 65% of the requirements for Ca (N) or 80% (H), namely C-N-C and C-H-C. The BW and bone mineral content (BMC) were measured at the beginning and end of each phase. The BMC gain (gBMC), average daily gain (ADG) and average daily feed intake (ADFI) were calculated by phase. In phase 1, ADG was lower in the Phyt group than the C group (1.05 vs 1.10 kg/d, P &lt; 0.01) and the BMC of group C and gBMC were higher than those of the Phyt and B groups (P &lt; 0.05). In phase 2, C-C and Phyt-Phyt groups had similar BMC due to higher gBMC in the Phyt-Phyt (27.1 vs 18.4 g/d, P &lt; 0.01). At the end of phases 2 and 3, C-C-C, C-N-C and C-H-C groups had similar BMC. The Phyt and B groups showed an increased phosphorus-use efficiency during phases 1 and 2 (+20% vs C). Phosphorus retention was also higher in the C-N-C and C-H-C groups, during the depletion in phase 2 (+24% vs C, P&lt; 0.05). These results showed the potential of a depletion-repletion strategy including free phosphate diet to reduce phosphorus intake and excretion without affecting final growth performance and bone mineralization because of increased minerals utilization efficacies.

The Norwegian Scientific Committee for Food Safety (Vitenskapskomiteen for mattrygghet, VKM) has, at the request of the Norwegian Food Safety Authority (Mattilsynet; NFSA), assessed the risk of "other substances" in food supplements and energy drinks sold in Norway. VKM has assessed the risk of doses given by NFSA. These risk assessments will provide NFSA with the scientific basis while regulating "other substances" in food supplements. "Other substances" are described in the food supplement directive 2002/46/EC as substances other than vitamins or minerals that have a nutritional and/ or physiological e ffect. It is added mainly to food supplements, but also to energy drinks and other foods. In this series of risk assessments of "other substances" the VKM has not evaluated any claimed beneficial effects from these substances, only possible adverse effects. The present report is a risk assessment of specified doses of L-threonine in food supplements, and it is based on previous risk assessments and articles retrieved from literature searches. According to information from NFSA, L-threonine is an ingredient in food supplements sold in Norway. NSFA has requested a risk assessment of 1000, 1200, 1500, 2000 and 2400 mg/day of L-threonine from food supplements.  L-threonine is an essential amino acid not known to cause any adverse health effects. Previous reports do not indicate a tolerable upper intake level, apart from an approval of a dose of 1150 mg/day by the Scientific Committee of the Spanish Agency for Food Safety and Nutrition (AESAN). Long-term studies in humans were not found. The only available human studies were: a small uncontrolled one-year pilot study with doses ranging from 0.5 to 2.5 g/day, one eight-week randomised controlled trial (RCT) using a dose of 7.5 g/day, and two 2-week RCTs using doses of 6 and 4.5 g/day. No adverse effects (diary method of registration of adverse effects) were reported in the eight-week clinical trial, and the only adverse effects observed in the two-week trials were one case of indigestion and one case of diarrhoea. A four-week rodent toxicity study indicated a no observed adverse effect level (NOAEL) of 854.3 mg/kg bw per day (only dose tested, no adverse effects observed).  The value used for comparison with the estimated exposure in the risk characterisation is the NOAEL defined in an 8-week randomised placebo controlled study in humans, 7500 mg/day. For a 70-kg individual, this corresponds to 107 mg/kg bw per day. Two human two-week studies and a small one-year pilot study support the notion that this dose will be well tolerated. The overall mean threonine intake according to NHANES III (3 g/day) is slightly larger than the doses requested for evaluation in the present risk assessment. No studies in children (10 to <14 years) and adolescents (14 to <18 years) were identified. Based on the included literature there was no evidence indicating that age affects the tolerance for relevant doses of threonine. Therefore, in this risk characterisation a tolerance as for adults, based on body weight, was assumed for these age groups. VKM concludes that: In adults (≥18 years), the specified doses 1000, 1200, 1500, 2000 and 2400 mg/day L-threonine in food supplements are unlikely to cause adverse health effects. In adolescents (14 to <18 years), the specified doses 1000, 1200, 1500, 2000 and 2400 mg/day L-threonine in food supplements are unlikely to cause adverse health effects. In children (10 to <14 years), the specified doses 1000, 1200, 1500, 2000 and 2400 mg/day L-threonine in food supplements are unlikely to cause adverse health effects. Children younger than 10 years were not within the scope of the present risk assessment.

Abstract BACKGROUND: The effects of inorganic trace minerals (ITM) replaced by low-dose glycine-complexed trace minerals (GCM) on growth, serum parameters, immunity, intestinal morphology, and mineral excretion in piglets were investigated. One hundred and twenty-eight weaned piglets (14.18 ± 0.33kg body weight (BW)) were randomly assigned to 4 treatments with 4 replicates, 8 piglets per replicate. Treatments consist of: (T1) basal diet + 100% inorganic trace mineral (ITM) as the control group (20 ppm Cu, 150 ppm Fe, 150 ppm Zn, and 30 ppm Mn from sulfates); (T2) basal diet + 50% ITM (Cu, Fe, Zn, and Mn from sulfates, 50% of control) + 50% organic trace minerals (OTM, Cu, Fe, Zn, and Mn from glycine complexed trace minerals (GCM), 50% of control); (T3) basis diet + 50% OTM from GCM; (T4) basal diet + 70% OTM from GCM. The feeding period lasted 28 d and was divided into 2 stages (0 to 14 d and 15 to 28 d). After feeding trial , 6 pigs per treatment were randomly selected to slaughter for sampling. RESULTS: Average daily gain, feed intake, and G:F were not affected by dietary treatments during the overall period. During the second, and the overall feeding phases, the digestibility of Zn and Fe in T3 and T4 was higher than that of T1 ( P < 0.05). The concentration of serum ferritin in T2 was significantly higher than T3 and T4. Serum immunoglobulin A concentration in the ileal mucosa of T2 was higher than that of T1 ( P < 0.05), and the higher duodenum villus height was observed in T4 compared with the rest treatments ( P < 0.05). The lowest trace mineral excretion was overserved in T3 ( P < 0.01); in addition, the urinary concentrations of Zn and Fe in T2 were lower than that in T1 ( P < 0.05). CONCLUSION: These results indicate that GCM have higher bioavailability than ITM, and that supplementation of low-dose GCM to replace full dose ITM could reduce mineral excretion without affecting performance, blood profiles, immune responses, and intestinal morphology in piglets.

Abstract The objective of this study was to investigate the effects of xylo-oligosaccharides (XOS) supplementation on growth performance, serum parameters, small intestinal morphology, intestinal mucosal integrity, and immune function in weaned piglets. A total of 240 weaned piglets with an average body weight (BW) of 8.82 ± 0.05 kg (28 d of age) were assigned randomly to 4 dietary treatments in a 28-d trial, including a control diet (CON), 3 diets with XOS supplementation at the concentration of 100, 500 and 1000 mg/kg (XOS100, XOS500, and XOS1000). There were 4 replicates per treatment with 15 pigs per pen. From d 1 to 14, there were no differences (P &gt; 0.05) in average daily gain (ADG), average daily feed intake (ADFI), and gain to feed ratio (G:F) during the different treatments. The different doses of XOS showed a quadratic effect on BW on d 28, ADG and G:F d 1-28 of piglets (P &lt; 0.05). From d 15 to 28, ADG of pigs fed the XOS500 diet was higher (P &lt; 0.05) than pigs fed the CON diet. During the overall period (d 1 to 28), pigs fed the XOS500 diet had a higher BW, ADG and G:F than pigs fed the CON diet (P &lt; 0.05). In addition, compared with the CON group, the XOS500 group had significantly higher serum total antioxidant capacity (T-AOC), total superoxide dismutase (T-SOD) and catalase (CAT) levels and lower malondialdehyde (MDA) levels on d 14 and 28 (P &lt; 0.05). The serum immunoglobulin G (IgG) concentration in the XOS500 group was also significantly higher compared with the CON group on d 14 and 28 (P &lt;0.05). However, serum immunoglobulin A (IgA) and immunoglobulin M (IgM) were not affected by the dietary treatments. Supplementation of XOS500 to the feed significantly increased the villus height (VH) and villus height to crypt depth ratio (VH:CD) in the jejunum and ileum in comparison with the CON and XOS1000 group. Moreover, the XOS500 group significantly elevated the expression levels of Occludin and zonula occludens protein-1 (ZO-1) in the ileum compared to the CON group. The ileal interleukin (IL)-1β, IL-8 and interferon (IFN)-γ mRNA expression levels in the XOS100 and XOS500 group were markedly lower than in the CON group. In contrast, the ileal IL-10 mRNA expression levels were remarkably higher in the XOS500 than CON group. In conclusion, xylo-oligosaccharides have a beneficial effect on growth performance by improving serum antioxidant defense system, serum IgG, small intestinal structure and intestinal barrier function in weaned piglets.

Abstract Background Bisphenol A (BPA), one of the highest-volume chemicals produced worldwide, has been identified as an endocrine disruptor. Many peer-reviewing studies have reported adverse effects of low dose BPA exposure, particularly during perinatal period (gestation and/or lactation). We previously demonstrated that perinatal oral exposure to BPA (via gavage of mothers during gestation and lactation) has long-term consequences on immune response and intestinal barrier functions. Due to its adverse effects on several developmental and physiological processes, BPA was removed from consumer products and replaced by chemical substitutes such as BPS or BPF, that are structurally similar and not well studied compare to BPA. Here, we aimed to compare perinatal oral exposure to these bisphenols (BPs) at two doses (5 and 50 mg/kg body weight (BW)/day (d)) on gut barrier and immune system in female offspring mice at adulthood (Post Natal Day PND70). Methods Pregnant female mice were orally exposed to BPA, BPS or BPF at 5 or 50 μg/kg BW/d from 15th day of gravidity to weaning of pups at PostNatal Day (PND) 21. Gut barrier function and the humoral and cellular immune responses of adult offspring (PND70) were analysed at intestinal and systemic levels. Results In female offspring, perinatal oral BP exposure led to adverse effects on intestinal barrier and immune response that were dependant of the BP nature (A, S or F) and dose of exposure. Stronger impacts were observed with BPS at the dose of 5µg/kg BW/d on inflammatory markers in feces associated with an increase of anti-E. coli IgG, revealing a defect of gut barrier. BPA and BPF exposure induced prominent changes at low dose in offspring mice, in term of gut barrier functions and cellular immune responses, provoking an intestinal and systemic Th1/Th17 inflammation. Conclusion These findings provide, for the first time, a comparative study of long-time consequences of BPA, S and F perinatal exposure by oral route in offspring mice. This work warms that it is mandatory to consider immune markers and dose in risk assessment associated to new BPA’s alternatives. In pigs, high protein diets have been related to post-weaning diarrhoea, which may be due to the production of protein fermentation metabolites that were shown to have harmful effects on the intestinal epithelium in vitro. In this review, we discussed in vivo effects of protein fermentation on the microbial composition and their protein catabolic activity as well as gut and overall health. The reviewed studies applied different dietary protein levels, which was assumed to result in contrasting fermentable protein levels. A general shift to N-utilisation microbial community including potential pathogens was observed, although microbial richness and diversity were not altered in the majority of the studies. Increasing dietary protein levels resulted in higher protein catabolic activity as evidenced by increased concentration of several protein fermentation metabolites like biogenic amines in the digesta of pigs. Moreover, changes in intestinal morphology, permeability and pro-inflammatory cytokine concentrations were observed and diarrhoea incidence was increased. Nevertheless, higher body weight and average daily gain were observed upon increasing dietary protein level. In conclusion, increasing dietary protein resulted in higher proteolytic fermentation, altered microbial community and intestinal physiology. Supplementing diets with fermentable carbohydrates could be a promising strategy to counteract these effects and should be further investigated.

Birth is one of the most important events of animal production agriculture, as newborns are abruptly forced to adapt to environmental and nutritional disruptions that can lead to oxidative damage and delay in growth. Taurine (Tau) is an important regulator of oxidative stress and possesses growth-enhancing properties. In the present study, we investigated the effects of dietary Tau supplementation in gilts during late gestation and lactation on the growth performance of piglets by assessing intestinal morphology and barrier function, and oxidative stress status. Sixteen gilts were randomly allocated to the Con (basal diet) and Tau (basal diet with 1% Tau) groups from 75 d of gestation to weaning. Maternal dietary Tau supplementation significantly increased weaning weight and average daily gain weight in piglets. Piglets in the Tau group had higher villus height and villus height-to-crypt depth ratio (VCR), ZO-1 protein expression, and secretory immunoglobulin A (sIgA) content in the jejunum. Meanwhile, Tau bebeficial affected the milk quality of gilts, as indicated by decreased malondialdehyde (MDA) concentration and increased total superoxide dismutase (T-SOD), total antioxidative capability (T-AOC), glutathione peroxidase (GPx), and catalase (CAT) activity. Furthermore, Tau supplementation increased T-SOD activity in plasma and SOD2 protein expression in the jejunum in the piglets. In conclusion, this study provides evidence that dietary Tau supplementation to gilts improves growth performance in piglets, owing to improved intestinal morphology and barrier function, as well as inhibition of oxidative stress.
