ABSTRACTLowering the pH in bacterium-based succinate fermentation is considered a feasible approach to reduce total production costs. Newly isolatedEnterobacter aerogenesstrain AJ110637, a rapid carbon source assimilator under weakly acidic (pH 5.0) conditions, was selected as a platform for succinate production. Our previous work showed that the ΔadhE/PCK strain, developed from AJ110637 with inactivated ethanol dehydrogenase and introducedActinobacillus succinogenesphosphoenolpyruvate carboxykinase (PCK), generated succinate as a major product of anaerobic mixed-acid fermentation from glucose under weakly acidic conditions (pH <6.2). To further improve the production of succinate by the ΔadhE/PCK strain, metabolically engineered strains were designed based on the elimination of pathways that produced undesirable products and the introduction of two carboxylation pathways from phosphoenolpyruvate and pyruvate to oxaloacetate. The highest production of succinate was observed with strain ES04/PCK+PYC, which had inactivated ethanol, lactate, acetate, and 2,3-butanediol pathways and coexpressed PCK andCorynebacterium glutamicumpyruvate carboxylase (PYC). This strain produced succinate from glucose with over 70% yield (gram per gram) without any measurable formation of ethanol, lactate, or 2,3-butanediol under weakly acidic conditions. The impact of lowering the pH from 7.0 to 5.5 on succinate production in this strain was evaluated under pH-controlled batch culture conditions and showed that the lower pH decreased the succinate titer but increased its yield. These findings can be applied to identify additional engineering targets to increase succinate production.

Purpose The purpose of this paper is to illustrate the development and testing of an innovative mobile application using design-based research. Design/methodology/approach This paper reports on the process of transformation of existing printed course material into digitized content through design-based research where design, research and practice were concurrently applied through several iterations of the mobile application. For this transformation, one session each from BSc in Nursing, Bachelor of Pharmacy and Bachelor of Medical Laboratory Sciences was selected. In the first phase of the design-based research, the main research question was formulated. In the second phase, a mobile learning application (OUSL MLearn) was designed and developed to address the research question. In the third phase, this application was evaluated by five groups of stakeholders: content experts to validate the content; educational technologists to check the alignment of technical and pedagogical features; novice users to check the overall effectiveness of the application; developer to develop the application, to check the ease of usage; and researchers to identify the impact of this innovation. These stakeholders were closely involved throughout the whole process which lasted over a period of four months. At the end of this development phase, the results were reflected upon and used for further enrichment. Findings It was observed that the developed mobile application was accessible, appealing and pedagogically constructive for users. However, optimization, development time, technical and organizational issues, workload of academics and production costs were identified as major challenges. Research limitations/implications This study was based on the findings of a small sample of potential users. Practical implications The findings have implications for designing culturally adaptive interactive mobile applications. Originality/value This study will benefit practitioners to design culturally sensitive mobile learning courses and researchers to conduct design-based research.

This paper deals with the impact of the most important factors of the total production costs in bioethanol production. The most influential factors are: total investment costs, price of raw materials (price of biomass, enzymes, yeast), and energy costs. Taking into account these factors, a procedure for estimation total production costs was establish. In order to gain insight into the relationship of production and selling price of bioethanol, price of bioethanol for some countries of the European Union and the United States are given.

ABSTRACT Clostridioides difficile is a pathogenic bacterium that infects the human colon to cause diarrheal disease. Growth of the bacterium is known to be dependent on certain bile acids, oxygen levels, and nutrient availability in the intestine, but how the environmental pH can influence C. difficile is mostly unknown. Previous studies indicated that C. difficile modulates the intestinal pH, and prospective cohort studies have found a strong association between a more alkaline fecal pH and C. difficile infection. Based on these data, we hypothesized that C. difficile physiology can be affected by various pH conditions. In this study, we investigated the impact of a range of pH conditions on C. difficile to assess potential effects on growth, sporulation, motility, and toxin production in the strains 630Δerm and R20291. We observed pH-dependent differences in sporulation rate, spore morphology, and viability. Sporulation frequency was lowest under acidic conditions, and differences in cell morphology were apparent at low pH. In alkaline environments, C. difficile sporulation was greater for strain 630Δerm, whereas R20291 produced relatively high levels of spores in a broad range of pH conditions. Rapid changes in pH during exponential growth impacted sporulation similarly among the strains. Furthermore, we observed an increase in C. difficile motility with increases in pH, and strain-dependent differences in toxin production under acidic conditions. The data demonstrate that pH is an important parameter that affects C. difficile physiology and may reveal relevant insights into the growth and dissemination of this pathogen. IMPORTANCE Clostridioides difficile is an anaerobic bacterium that causes gastrointestinal disease. C. difficile forms dormant spores which can survive harsh environmental conditions, allowing their spread to new hosts. In this study, we determine how intestinally relevant pH conditions impact C. difficile physiology in the two divergent strains, 630Δerm and R20291. Our data demonstrate that low pH conditions reduce C. difficile growth, sporulation, and motility. However, toxin production and spore morphology were differentially impacted in the two strains at low pH. In addition, we observed that alkaline environments reduce C. difficile growth, but increase cell motility. When pH was adjusted rapidly during growth, we observed similar impacts on both strains. This study provides new insights into the phenotypic diversity of C. difficile grown under diverse pH conditions present in the intestinal tract, and demonstrates similarities and differences in the pH responses of different C. difficile isolates.

Purpose – The purpose of this paper is to examine the use of sustainability as strategic cornerstone for a marketing channel in a mature market, particularly in the hog industry in the Canadian Province of Quebec. Due to the growing attention to sustainability, and the international trend toward agricultural trades and stakeholder involvement in food, there is a need for a system-based approach in the field of food systems sustainability. Design/methodology/approach – In total, 12 semi-structured individual interviews were conducted among marketing managers working for different organizations within the Quebec hog marketing channel. The organizations chosen vary depending on the size, level of integration and involvement in the industry, to create a sample that adequately represents the industry. For the purpose of this research, three primary producers, one veterinarian, three licenced abattoirs, one food processor, one distributor, one independent retailer and two types of food service facility, one from a franchise system and the other from a high-end restaurant were questioned regarding the current state of the industry, as well as the impact of sustainable development on their strategic plans. Findings – The Canadian hog and pork industry is currently facing strong competition from several fronts: the USA and South America, a population with increasingly sophisticated demands, a strong Canadian dollar and a significant increase in input costs, particularly in respect to oil and corn. To be able to meet this competition, and in order to prosper in an uncertain marketplace, marketing channel believes that it must reduce its production costs, increase product awareness in the domestic market and promote cooperation among industry members. Research limitations/implications – This research is mainly concerned with the Quebec hog industry. External validity has not been achieved with the current research. In addition, a lack of distributor availability to answer the interview questions limits researchers’ capacity to extrapolate results to all retailers. Meetings with other food distributors would be required to verify the accuracy of results. Although this study is exploratory in nature, several appealing research avenues emerge. First, the research focusses primarily on members of the distribution network, but does not consider the consumers’ point of view. It would be fruitful to study the impact of sustainable development on the perceived product quality by measuring consumers’ intent to purchase. Practical implications – The principal point of interest that makes this case worthy of study and of potential application to future business modeling is how sustainability and sustainable development is perceived throughout various marketing channels. The Quebec hog industry faces strong competition from several countries that are able to offer a similar product at a lower cost. The mature market consists of a limited number of producers interested in offering a product on the market at the lowest possible price. Social implications – When it comes to sustainable development and the hog industry, the economic and environmental aspects seem to be well understood by members of the industry. On the other hand, the social aspect of sustainability is not mentioned frequently and is often not a part of the leaders’ main concerns. Nevertheless, members of the industry agree that sustainable development affects the Quebec hog value chain, and will remain a topic of interest in the coming years. Originality/value – To the knowledge, no study has been conducted to evaluate the use of sustainability as strategic cornerstone for a marketing channel in a mature market, particularly in the hog industry. Many studies have been conducted in a context of emerging markers. However, very few studies addressed the issue in an established economic environment.

ABSTRACTStaphylococcus aureusis a prolific human pathogen capable of causing severe invasive disease with a myriad of presentations. The ability ofS. aureusto cause infection is strongly linked with its capacity to overcome the effects of innate immunity, whether by directly killing immune cells or expressing factors that diminish the impact of immune effectors. One such scenario is the induction of lactic acid fermentation byS. aureusin response to host nitric oxide (NO·). This fermentative activity allowsS. aureusto balance redox during NO·-induced respiration inhibition. However, little is known about the metabolic substrates and pathways that support this activity. Here, we identify glycolytic hexose catabolism as being essential forS. aureusgrowth in the presence of high levels of NO·. We determine that glycolysis supportsS. aureusNO· resistance by allowing for ATP and precursor metabolite production in a redox-balanced and respiration-independent manner. We further demonstrate that glycolysis is required for NO· resistance during phagocytosis and that increased levels of extracellular glucose limit the effectiveness of phagocytic killing by enhancing NO· resistance. Finally, we demonstrate thatS. aureusglycolysis is essential for virulence in both sepsis and skin/soft tissue models of infection in a time frame consistent with the induction of innate immunity and host NO· production.IMPORTANCEStaphylococcus aureusis a leading human bacterial pathogen capable of causing a wide variety of diseases that, as a result of antibiotic resistance, are very difficult to treat. The frequency ofS. aureustissue invasion suggests that this bacterium has evolved to resist innate immunity and grow using the nutrients present in otherwise sterile host tissue. We have identified glycolysis as an essential component ofS. aureusvirulence and attribute its importance to promoting nitric oxide resistance and growth under low oxygen conditions. Our data suggest that diabetics, a patient population characterized by excess serum glucose, may be more susceptible toS. aureusas a result of increased glucose availability. Furthermore, the essential nature ofS. aureusglycolysis indicates that a newly developed glycolysis inhibitor may be a highly effective treatment forS. aureusinfections.

PurposeThe purpose of this paper is to ascertain the relative impact of different Tony Award nominations and wins on the financial success of a Broadway theater production, as defined by the length of the production’s run.Design/methodology/approachCox hazard regression was used to identify the impact of Tony Award nominations and wins (time-varying covariates), while controlling for several time-invariant covariates: type of production (play or musical, revival or original Broadway production), production costs (operationalized via the cast size), the month and year of opening, and initial marketing success (defined as the percentage of first full week’s tickets sold).FindingsThe award with the strongest relationship with production longevity was the Tony Award for Best Musical (nomination OR=0.566,p=0.110; win OR=0.323,p=0.020). Several other awards had a relationship with production longevity, but most were not statistically significant.Research limitations/implicationsThe limitations include the low statistical power for many time-varying covariates and the cumulative impact of multiple awards was not investigated. Future researchers interested in the Broadway industry should not combine Tony Awards because of the varying impact on economic outcomes for a production.Originality/valueThis study is the first to investigate all 22 Tony Award nominations and wins and their individual impact on an economic outcome. This paper includes the study’s raw data and SPSS syntax to comply with open science practices. The author encourages readers to replicate the analysis.

Purpose The purpose of this paper is to examine whether audit quality influence real earnings management activities using a sample of UK listed firms that have strong incentives to manage earnings upward through meeting past year’s earnings as a benchmark in the post-adoption period of International Financial Reporting Standards (IFRS). Design/methodology/approach The authors use a sample of 4,774 firm-year observations of UK listed firms during the period 2005–2018. Univariate and multivariate analyses have been conducted to test the association after controlling for firm characteristics and institutional variables. Findings The study reports that the presence of Big 4 auditors is significantly and positively related with greater levels of sales and discretionary expenses manipulation. Though the authors do not find any conclusive evidence on production costs manipulation, the aggregated measure of real earnings management shows a significant positive association with the presence of Big 4 auditors. Practical implications The study implies that managers who have incentives to manage earnings upward around the UK firms take advantage of the accounting flexibility in defining policies while reducing information asymmetry among the investors to signal better future performance. The approach to detect earnings manipulation as described in the auditing standards fails to limit the managerial use of real activities due to limited scope and unclear guidance. Thus, due to the significant impact on public policies, the results should, therefore, be of interest to the regulators and standard setters. Originality/value To the best of the authors’ knowledge, this is the first study that examines the association between audit quality and real earnings management for the UK all-purpose operational firms in sampled data that just meet past year’s earnings as a benchmark in the post-IFRS period.

GDP is considered as a critical infrastructure that characterizes the global interaction of consumers and manufacturers of the final product in the national economy. For the analysis and modelling of GDP, the Leontiev model is used to analyze and to model GDP as a critical infrastructure, where costs are interpreted in two ways - as consumer spending and as producer costs. This allows you to determine the degree of their influence on each other through the established production structure. Based on Leontiev's aggregated three-sector model, two scenarios were modelled: a) the impact of final consumer spending on output and the formation of added value and b) the impact of changes in added value on output and final consumer spending. The model includes three sectors – the agricultural sector, industry and service sectors. In the first scenario, the increase in the final consumed product in the agricultural sector by 1000 yen causes an increase in output in the agricultural sector by 1153.5 yen, in industry by 594.4 yen and in the service sector by 358.1 yen. At the same time, total production costs will increase by 1106.0 yen; added value will increase in the agricultural sector by 549.8 yen, in industry – 216.8 yen, in the service sector – 233.4 yen. The volume of production growth can be determined only through the Leontiev model. In the second scenario, an increase in added value in the agricultural sector by 1000 yen brings about an increase in output in the agricultural sector by 1153.5 yen, in industry by 1470.4 yen and in the service sector by 480.7 yen. For both scenarios, the assessment and verification of calculations of GDP growth and output are carried out by the balance method – output and GDP growth is described within the Leontiev model. The three-sectoral model is useful for educational purposes, scientific research; to reach the method in a specific example. The approach can be extended to models of greater dimension and large size.

PurposeThis study investigates the competencies required for quality management professionals to meet the needs of industry 4.0. The authors use a case study strategy at an electronics manufacturer in southern Malaysia, to adapt their role to be relevant in the industry 4.0 environment. In doing so, this study answers the following four questions: (1) How are the changing technological trends expected to impact the future role of quality in industry 4.0? (2) What are the competencies gap between current and future roles of quality professionals? (3) What are the views and practices related to quality roles? (4) How can the gaps identified be closed to meet the quality challenges of industry 4.0?Design/methodology/approachThe research methods consist of a comprehensive review of literature on the technological trends towards industry 4.0 and the impact on the role of quality and competence that may be required in the future, as well as internal document review on the current roles of quality professionals in an electronics manufacturer in southern Malaysia, to identify the competence gap. Empirical data was collected based on surveys conducted on 64 quality professionals with a response rate of 96.88%. Interviews were conducted on three decision-makers from critical areas in the electronics manufacturer for viewpoints from three different perspectives: finance, operations and talent development.FindingsQuality professionals will require technical competencies to interpret large amounts of data from processes to make strategic decisions, the use of new AR tools and be aware of data security risks. Methodological competencies will be required to use data to identify the source of problems, to access reliable sources of learning and the ability to use new tools for solving complex problems efficiently. Social competencies will be required in communications across multi-sites, suppliers and customers in new collaborative virtual platforms, with the ability to retain tacit and explicit knowledge, in a decentralized environment that will require leadership ability to make decisions. Personal competencies required will be the ability to work in a flexible workplace and time and more frequent work-related changes.Research limitations/implicationsThe limitation of the study is based on what the authors currently know of the future, which may not be much for the quality professionals in the electronics manufacturer, who have not been exposed much to the technology yet. The potential for the future landscape to change dramatically with rapid technology changes may also result in a different set of skills for future quality professionals. The quality professionals who were involved in this study were the quality executives, engineers and managers, irrespective of their gender, age, length of service and experience in the field of quality. Therefore, these variables were not taken into consideration for this research.Practical implicationsThis research helped to identify the role of quality in industry 4.0 and key competencies that the quality professionals in the electronics manufacturer will require to adapt to their role in industry 4.0. However, based on the questionnaire and the interview comments of key personnel, it can be concluded that quality professionals lack awareness of their new roles in industry 4.0. This could be due to the fact that the new technology is not implemented by quality professionals but by the innovation team based in Singapore headquarters, as was also advised by the operations head.Social implicationsThe benefit of industry 4.0 technology is clearly shown by Philips's new Dutch factory with robotized technology that was able to produce the same output with one-tenth of the workers of its China factory (Rifkin, 2014, chapter 8). Rojko (2017, p. 80) also shared a similar view that industry 4.0 is expected to reduce production costs by 10–30%, logistics costs by 10–30% and quality management costs by 10–20%. The importance of this research can be seen from the findings of “The Future of Jobs” (2018, p. 22), which suggests that the window of opportunity for organizations to leverage the new technology to re-skill is within the period of 2018–2022, in order to enable employees to reach full potential in the high value-added tasks. The electronics manufacturer may need to keep to this timeline to maintain its competitive advantage.Originality/valueThe purpose of this paper was to determine the competence gap of current quality professionals in the electronics manufacturer with the competencies required in industry 4.0. This led to the third objective, to identify the views of stakeholders based on the propositions derived from the gaps identified, to triangulate the findings, to conclude the competency gaps of the current quality professionals in the electronics manufacturer. Finally, the objective of this paper was to make a recommendation on how to prepare the quality professionals in the electronics manufacturer for their role in industry 4.0. The research identified the technical, methodological, social and personal competencies gap of the quality professionals in the electronics manufacturer by looking at the changes expected in industry 4.0 from four aspects, factory (people and process), business, product and customers.

Purpose This paper aims to present a new model for solving the integrated production planning and scheduling. Usually, the two decision levels are treated sequentially because of their complexity. Scheduling depends on the lot sizes calculated at the tactical level and ignoring scheduling constraints generates unrealistic and inconsistent decisions. Therefore, integrating more detail scheduling constraint in production planning is important for managing efficiently operations. Therefore, an integrated model was developed, and two evolutionary optimization approaches were suggested for solving it, namely, genetic algorithm (GA) and the hybridization of simulated annealing (SA) with GA HSAGA. The proposed algorithms have some parameters that must be adjusted using Taguchi method. Therefore, to evaluate the proposed algorithm, the authors compared the results given by GA and the hybridization. The SA-based local search is embedded into a GA search mechanism to move the GA away from being closed within local optima. The analysis shows that the combination of simulated annealing with GA gives better solutions and minimizes the total production costs. Design/methodology/approach The paper opted for an approached resolution method particularly GA and simulated annealing. The study represents a comparison between the results found using GA and the hybridization of simulated annealing and GA. A total of 45 instances were studied to evaluate job-shop problems of different sizes. Findings The results illustrate that for 36 instances of 45, the hybridization of simulated annealing and GA HSAGA has provided best production costs. The efficiency demonstrated by HSAGA approach is related to the combination between the exploration ability of GA and the capacity to escape local optimum of simulated annealing. Originality/value This study provides a new resolution approach to the integration of planning and scheduling while considering a new operational constrain. The model suggested aims to control the available capacity of the resources and guaranties that the resources to be consumed do not exceed the real availability to avoid the blocking that results from the unavailability of resources. Furthermore, to solve the MILP model, a GA is proposed and then it is combined to simulated annealing.
