ABSTRACT A purified recombinant protein from Eimeria acervulina (3-1E) was used to vaccinate chickens in ovo against coccidiosis both alone and in combination with expression plasmids encoding the interleukin 1 (IL-1), IL-2, IL-6, IL-8, IL-15, IL-16, IL-17, IL-18, or gamma interferon (IFN-γ) gene. When used alone, vaccination with 100 or 500 μg of 3-1E resulted in significantly decreased oocyst shedding compared with that in nonvaccinated chickens. Simultaneous vaccination of the 3-1E protein with the IL-1, -15, -16, or -17 gene induced higher serum antibody responses than 3-1E alone. To evaluate protective intestinal immunity, vaccinated birds were challenged with live E. acervulina oocysts 14 days posthatch, and fecal-oocyst shedding and body weight gain were determined as parameters of coccidiosis. Chickens vaccinated with 3-1E protein showed significantly lower oocyst shedding and normal body weight gain than nonvaccinated and infected controls. Simultaneous immunization with 3-1E and the IL-2, -15, -17, or -18 or IFN-γ gene further reduced oocyst shedding compared with that achieved with 3-1E alone. These results provide the first evidence that in ovo vaccination with the recombinant 3-1E Eimeria protein induces protective intestinal immunity against coccidiosis, and this effect was enhanced by coadministration of genes encoding immunity-related cytokines.

Eimeria praecox produced less effect on body-weight gain and food consumption than E. acervulina. No deaths occurred and no weight loss followed infection with doses of oocysts up to 10. However, there was a depression of body-weight gain in birds infected with both species and changes in the permeability of the intestine were noted from as early as 3½ h after infection. This suggests that increased intestinal permeability is not the major factor in the greater pathogenicity of E. acervulina compared with E. praecox. Eimeria acervulina produced considerably greater effects on the host as evidenced by mortality, body weight gain and food consumption.I wish to thank Mr B. J. Millard and Mr M. Abstract Toll-like receptor (TLR) agonists are emerging as promising vaccine adjuvants and immunomodulators in poultry against many diseases. Infectious bursa disease (IBD) still remains as a major threat in poultry industry. Improving the vaccine mediated immune response would help in better protection against IBD virus infection. Adjuvant potential of TLR3 agonist, Poly I:C with different IBD vaccines has been analyzed in chicken in the present study. Intermediate, intermediate plus IBD vaccine, bursaplex vaccine and their respective poly I:C combinations were used for immunization of chicken. IBD specific antibody titre, bursa to body weight ratio, body weight gain and bursal lesion scores were evaluated at weekly interval in different immunization groups. Fold changes in cytokines IL-1β and IFN-γ mRNA expression levels were also analyzed in different groups. Intermediate IBD plus vaccine induced significantly (P≤0.05) higher IBD specific antibody response at 35 days of age than other groups with comparatively lower body weight gain and moderate bursal lesion score. Poly I:C co-administration with intermediate IBD vaccine and bursaplex vaccine improved the IBD specific antibody titres, better body weight gain and moderately less bursal lesion score. However, Poly I:C combination with intermediate plus IBD vaccine did not improve the specific immune response. IL-1β levels were up-regulated in intermediate plus and bursaplex group, whereas IFN-γ m RNA expression levels were upregulated in intermediate IBD with Poly IC group. In conclusion, poly I:C co-administration with intermediate IBD and bursaplex vaccine was beneficial and improved the specific immune response with least immunosuppression and bursal damage.

The hypothesis of the present work was that the effects of in ovo injection may differ in different chicken strains. The influence of in ovo royal jelly (RJ) injection on hatching, growth and blood parameters in two chicken strains (Dokki-4 and El-Salam as example for different strains) was evaluated. A total of 1080 eggs were used. On the seventh day of incubation, the eggs were randomly allocated into six experimental groups in a 2 × 3 arrangement that included the two chicken strains and three concentrations of RJ (0, 0.25 and 0.5 mL RJ/egg). Injection with 0.5 mL RJ/egg improved hatchability compared to the other treatments. The El-Salam strain exhibited significantly higher body weight and body weight gain than the Dokki-4 strain. Injection with 0.5 mL RJ/egg significantly (p < 0.05) improved chicken body weight and daily weight gain compared to the control treatment. RJ injection decreased blood lipid profile parameters and the numbers of monocytes and eosinophils and increased total protein, globulin, haemoglobin (Hb) and lymphocyte levels compared to the control treatment. The Dokki-4 strain showed significantly higher antibody titres against avian influenza virus (AIV) (p < 0.05) and sheep red blood cells (SRBCs) (p < 0.0001) than the El-Salam strain and RJ injection enhanced antibody titres against AIV, Newcastle disease virus (NDV) and SRBCs. Therefore, the Dokki-4 strain was superior to the El-Salam strain for the tested parameters and injection with 0.5 mL RJ/egg produced the best hatching parameters, growth performance and health-related traits. RJ in ovo injection was much more effective in the Dokki-4 strain than in the El-Salam strain, which supported the hypothesis of the study that varying the chicken strain could alter the response to the in ovo injection with RJ.

In three experiments broiler chickens were inoculated with sporulated Eimeria acervulina oocysts at 18 d of age. Feed intake, body-weight gain, brush-border enzyme activities, fat digestion, protein digestion and protein retention were measured. Body-weight gain was reduced during the acute phase of the infection and increased during the recovery phase of the infection. Feed intake was decreased on day 4 and day 5 postinfection (PI) and increased from day 7 to day 11 PI. Maltase (EC 3.2.1.20) and sucrase (ECEC 3.2.1.48) activities were decreased on day 5 PI in all intestinal segments. In Expts 2 and 3, however, maltase activity was increased in the ileum. Fat digestion was decreased from day 2 to day 11 PI. N digestion and retention were decreased from day 2 to day 11 PI.

AbstractOver the years, due to genetic selection, improvement of the characteristics of laying and meat poultry with less feed consumption per kilogram of body weight gain has been achieved (FCR). As the productivity of poultry increased, the demand of embryos for nutrients changed. However, the chemical composition of an egg has remained practically unchanged, and therefore, it began providing the embryos with suitable substances with the help of in ovo technology. Improvements in hatching were achieved through the administration of fructose, sucrose and grape seed extract (GSE), while the weight of a one-day-old chick is affected by the injection of amino acids with glucose and magnesium. In addition, amino acids and carbohydrates applied to an egg have contributed to an increase in the activity of digestive enzymes and maturation of the intestine. In connection with early stimulation of the intestinal tract of broiler chicks, they obtained higher weight gain. Supplementation with vitamins positively affects the increase of birds’ immunity and body weight at the end of breeding. On the other hand, the injection of an insulin-like growth factor (IGF-I) influenced the growth and development of muscle tissue during the first weeks of life.

The aim of the study was to determine the development of ramie plants (Boehmeria nivea L. Gaud) and the effect of using ramie leaves on feed on the body weight gain of Wonosobo Sheep (Dombos). Research on the development of ramie plants using survey methods in the area of ramie plant development in Wonosobo Regency. While the research on the use of ramie leaves for fattening was carried out in Butuh Village, Kalikajar District, Wonosobo Regency in 2018. 21 male Dombos were divided into 3 feed treatments with forage proportions of 70%, 50% and 30 ramie leaves respectively. %. The results showed that currently ramie plants were being developed in Wonosobo Regency by CV. Ramindo Berkah Persada Sejahtera in Gandok Village, Kalikajar District, Wonosobo Regency, Central Java. Until now the area of the crop has reached 13 ha. Of this area will produce ramie leaves 195,000 kg / year. If one sheep needs 4 kg of ramie / tail / day leaves, then the potential capacity of sheep is 135 heads / year, if the given one is 50% then the Jurnal Litbang Provinsi Jawa Tengah, Volume 16 202 Nomor 2 – Desember 2018potential capacity is 270 heads / year and if it is reduced again to 25% of ramie leaves then the potential capacity 440 heads / year. The use of ramie leaves as a feed for Wonosobo Sheep fattening can be given as much as 30% in fresh form.

Background and Objective: Yoghurt, especially bio-yoghurt has long been recognized as a product with many health benefits for consumers. Also, honey and olive oil have considerable nutritional and health effects. So, the effect of administration of yoghurt made using ABT culture and fortified with honey (2 and 6%), olive oil (1 and 4%) or honey + olive oil (2+1 and 6+4% respectively) on some biological and hematological properties of rats was investigated.Methods:The body weight gain, serum lipid level, blood glucose level, serum creatinine level, Glutamic Oxaloacetic Transaminase (GOT) activity, Glutamic Pyruvic Transaminase (GPT) activity, leukocytes and lymphocytes counts of rats were evaluated.Results:Blending of bio-yoghurt with rats&#039; diet improved body weight gain. Concentrations of Total plasma Cholesterol (TC), High-Density Lipoprotein cholesterol (HDL), Low-Density Lipoprotein cholesterol (LDL), Very Low-Density Lipoprotein cholesterol (VLDL) and Triglycerides (TG) significantly lowered in plasma of rats fed bio-yoghurt. Levels of TC, LDL, VLDL, and TG also decreased in rat groups feed bio-yoghurt supplemented with honey and olive oil. LDL concentrations were reduced by 10.32, 18.51, 34.17, 22.48, 43.30% in plasma of rats fed classic starter yoghurt, ABT yoghurt, ABT yoghurt contained 6% honey, ABT yoghurt contained 4% olive oil and ABT yoghurt contained 6% honey + 4% olive oil respectively. The blood glucose, serum creatinine, GOT and GPT values of rats decreased while white blood cells and lymphocytes counts increased by feeding bioyoghurt contained honey and olive oil.Conclusion:The findings enhanced the multiple therapeutic effects of bio-yoghurt supplemented with honey and olive oil.
