A study, using four types of water samples ranging from relatively pure stream water to water containing industrial and domestic effluents, was carried out to investigate the effect of storage temperature on four bacterial parameters: total coliform, fecal coliform, fecal streptococcus, and heterotrophic bacteria. In the study, each water sample was cooled immediately after collection to approximately 1.5 °C by storage in crushed ice. At 2-, 24-, 30-, and 48-h intervals, the sample was mixed, and a subsample was removed and tested. Three separate analyses for each parameter were made on each subsample. The data available for statistical analyses contained, in some cases, the values obtained for the three subsamples and, in others, the means of the three values. The data were analysed as replicated data and as part of the entire set. The analysed data indicated (i) that with the exception of heterotrophic populations, more than 75% of the samples were microbiologically stable for at least 24 h, (ii) that at least 50% of samples tested for heterotrophic densities were stable for a minimum period of 24 h, (iii) that the original water temperature and bacterial load do not appear to be consistent factors in the preservation of samples for microbiological analysis, and (iv) that nutrient levels, also, do not seem to be consistent factors in the preservation of water samples for microbiological analyses.

The microbiological study of water samples obtained from Udo Anwankwo River was investigated. Water samples were collected from three different sampling points along the course of the river and analyzed using standard procedures. The total bacterial counts, total coliform counts and total fungal counts of the water samples ranged from 2.6×105 to 4.8×105cfu/ml, 1.2×104 to 1.8×105cfu/ml and 0.24×103 to 1.9×103cfu/ml respectively. A total of nine bacteria species belonging to the following genera, Bacillus, Salmonella, Escherichia, Pseudomonas, Micrococcus, Staphylococcus, Vibrio, Enterobacter and Streptococcus were isolated and identified from the samples, while five fungal isolates including members of the genera Aspergillus niger, Mucor, Penicillum, Rhizopus and Fusarium were isolated. The study through microbial analysis has revealed that the river water sample was not free from pathogens and thereby not suitable for potable use. There is need to put adequate measures towards the control of pollution and proper treatment of the water before usage as it contains pathogenic organisms.

Chattogram is the second most populated city in Bangladesh. This port city faces a serious threat mainly due to the lack of safe drinking water. This study was conducted for determining drinking water quality of groundwater sources in Chattogram city. The study was performed in the BCSIR laboratory, Chattogram. It was carried out for a period of six months from 1st July, 2018 to 31th December, 2018. Total six water samples were collected from three different locations (Baluchora, C&B colony and Khulshi area). Each sampling location consists of two separate sampling points. Physicochemical parameters of the collected samples like Temperature, pH, Electrical Conductivity (EC), Total dissolved solid (TDS), Hardness, Turbidity and concentration of Cl, As, Mn, Fe, Pb, Cr and Cd were examined. Microbial parameters like Total Coliform (TC) were also measured. All the analyzed parameters compared with BSTI and WHO drinking water quality standards to understand the overall ground water quality status of the study area. The results reveal that water samples in almost all locations were contaminated with microbial contamination and that the range of physico-chemical parameters was not adequate for consumption. Preliminary treatments like boiling, filtering etc are required before using groundwater for drinking and the necessary measures must be taken for a safe alternative source of drinking water.

The physicochemical, microbiological and heavy metal studies of water samples obtained from Mbiaso River and Enang Stream were investigated. Water samples were collected from two locations with two sampling points from each location along the course of the water body based on their use by the community and analysed using standard procedures. The results obtained were compared with WHO standards for drinking and recreational water. Aside titratable acidity, titratable alkalinity and BOD, all other physicochemical parameters were within the permissible standards. All the heavy metals concentrations were above the permissible limits except for Lead (Pb) that ranged between 0.01mg/l to 0.05 mg/l for all the sampling points. All the water samples were found to harbor coliforms organisms in numbers greater than the required WHO standards for water except for Enang river that lacked the presence of Vibrio spp. The total viable counts for all the water samples were generally high exceeding the standard limit of 1.0 x 102 cfu/ml for water. The total heterotrophic bacteria count, total coliform count, faecal coliform count and total fungal count ranged between 1.9x104 - 52x104 cfu/ml, 2.3x104 -38 x104 cfu/ml, 1.7x104 - 30x104 cfu/ml and 2.5x104 - 5.3x104 cfu/ml respectively. The total salmonella-shigella counts and total staphylococcus counts were in the range of 0.2 x102-0.4 x102 cfu/ml and 0.3 x102 -0.6 x104 cfu/ml respectively. From the study, it is suggested that some regulations measures be put in place to control pollution of the water while the water be properly treated before use for domestic, potable or recreational purposes.

Water is very crucial for the survival and maintenance of most living organisms. The quality of water available for plant, animal or human use cannot therefore be compromised. This study therefore examined the physicochemical and microbiological characteristics of water samples in Okomu National Park, Edo State, Nigeria. Water samples were collected from four waterholes (three rivers and one stream) for two seasons (dry and wet) using grab sampling technique. These water samples were analysed for physicochemical [temperature, pH, total dissolved solid, electrical conductivity, total suspended solids, dissolved oxygen (DO), nitrate, chloride, phosphate, sulphate, biological oxygen demand (BOD) and chemical oxygen demand (COD)] and microbiological (total coliform and fungal counts) analyses using standard methods. Data collected were subjected to descriptive statistics and T-test at α0.05 and compared with WHO permissible limits. The result showed that the mean COD (both seasons) and BOD (dry season) were above the comparable WHO permissible limit while there was no significant difference in the values of all the physicochemical parameters across the seasons of sampling. Salmonella / Shigella and Staphylococcus aureus were observed to be absent while the total coliform and fungi counts were observed to be higher than the WHO permissible limit for drinking water. The detection of Escherichia coli in the water samples (except Arakhuan stream) is an indication of faecal contamination either of animal or human origin. There was significant difference in total heterotrophic bacteria (t=-4.936) and total coliform counts (t=-2.417) in the waters sampled. Constant monitoring of the waterholes and intensified sensitization of the surrounding communities on ecosystem conservation is needed to protect the park’s ecosystems.

Crude oil exploration has been beneficial to our economy but detrimental to our environment with the artisanal refineries further compounding the challenge. This research was aimed at investigating the microbiology and heavy metal pollution of three crude oil polluted rivers in Rivers State, Nigeria and effects on living organisms inhabiting that environment. This study was carried out in three locations in South-South Nigeria (Eagle Island, Iwofe and Chokocho rivers). A total of 64 water samples (upstream and downstream points) were collected using appropriate containers and sterile polyethene bags for 6 crab samples for a period of three months. Water and crab samples were analyzed for heavy metals using Atomic Absorption Spectrophotometric method while microbiological analysis involved isolation and enumeration of microbial populations of the water and crab samples as well as characterization and identification of the isolates using standard methods Results showed Total Heterotrophic bacteria (THB) ranged from 6.0 × 106 cfu/ml to 9.0 × 108cfu/ml for the downstream locations and 1.7 × 106cfu/ml to 3.5 × 107cfu/ml for the upstream locations. Total Heterotrophic Fungi (THF) ranged from 2.0 × 104cfu/ml to  1.1 × 105cfu/ml  for downstream locations and 0.1 × 104cfu/ml to  4.0 × 104cfu/ml  for upstream locations, Hydrocarbon Utilizing Bacteria (HUB) ranged from 0.8 × 103cfu/ml to  4.0 × 103cfu/ml in downstream locations and 2.0 × 103cfu/ml to  7.4 × 103cfu/ml in upstream locations, Hydrocarbon Utilizing Fungi (HUF) ranged from 1.0 × 103cfu/ml to  6.0 × 103cfu/ml for downstream locations and 5.0 × 102cfu/ml to  8.0 × 103cfu/ml for upstream locations.   The bacteria identified biochemically included Serratia sp., Enterobacter sp. and Salmonella sp. for the crab samples and Bacillus sp. was dominant in the water samples (Bacillus subtilis, Bacillus cereus, Bacillus carboniphilus). The heavy metals (Fe, Ni, Zn, Cd, Cu, Mn, Cr and Pb) were below the DPR permissible limit but are likely to increase since the activities leading to pollution are still ongoing. The crab as a filter feeder, had higher concentration of heavy metals and microbial population and the location with the highest crude oil pollution (14.5mg/l) had the lowest THB (6.0 × 106cfu/ml) as physicochemical parameters like the amount of Dissolved Oxygen had been altered.

The use of unsafe water supplies and microbial contaminated water may pose serious health challenge to users. The aim of the study was to determine the level of bacterial contamination of the various water sources and the suitability for human use and consumption in University of Calabar Community, Nigeria. A total of 30 water samples were obtained from the University storage tanks and analysed for the presence of bacteria. The level of feacal coliform count, total coliform count and heterotrophic bacterial count was analysed using membrane filtration method and standard culture method on a differential and selective media. The samples were cultured on MacConkey and Nutrient agar. The isolates obtained from the above media were subcultured into slants of nutrient agar. Isolates were subjected to Gram staining and biochemical tests. The feacal coliform, Escherichia coli was isolated from all the water samples. Total coliform counts ranged from 1cfu/ml - 92cfu/ml while total heterotrophic bacterial count ranged from 1cfu/ml to 161cfu/ml. The bacteria species isolated were Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, Pseudomonas aeruginosa, Streptococcus species, Enterobacter aerogenes, Coagulase negative Staphylococcus, Proteus species, Lactobacillus species and Listeria species. The results obtained from this study have shown high level of bacterial load which makes the water unsuitable for human consumption. Further treatment may be needed as the bacteria isolates from the water samples point to feacal contamination which may be due to inadequate treatment of water, contamination while in the storage tanks or passage through contaminated pipes supplying the community. The consumers may be at high risks of enteric bacterial infections.

Objectives: This study was aimed to evaluate different water samples in terms of Total bacterial load and Total Coliform Load in comparison with different parameters such as pH, type of samples, chlorination status, turbidity, temperature and collection areas. Methods: Altogether 250 water samples were collected; 110(44%) were ground water samples, 60(24%) were public tap water samples and 80(32%) were bottled water samples. Total Coliform load was evaluated using Membrane Filtration Technique and Total Bacterial Load was evaluated using Aerobic Plate Count Technique. pH was measured using pH meter, turbidity was measured using nephelometer, temperature was measured using temperature probe and bottled water were considered chlorinated. Results: Among these 250 samples, the highest mean bacterial load was seen in public tap water samples (137×103 cfu/ml) and least mean bacterial load was seen on bottled water sample (28×103). Similarly, highest mean coliform load was seen on ground water samples (81 cfu/ml) and least mean coliform load was seen on bottled water samples (6 cfu/ml). Conclusion: This study has concluded that ground water samples contain large number of coliforms which suggest it might be fecally contaminated. Also the result had shown even bottled water contained coliforms. Therefore, it is recommended to treat ground water before using. Proper care must be taken during manufacture of bottled water.

Water borne diseases are the major cause of morbidity and mortality in developing countries like Bangladesh, where disease outbreaks occur due to unsafe drinking water, inadequate sanitation and poor hygienic practices. The objective of this study was to evaluate the quality of water used for drinking as well as household purposes in Dhaka city, Bangladesh. Eighteen water samples were collected from different rivers and canals. Among those, two samples were treated with chlorine. Physico-chemical parameters (pH, salinity, turbidity, conductivity and dissolved oxygen) were monitored during the time of sample collection using portable meters. For microbiological analysis, water samples were collected in sterile screw capped bottle. All Experiments were carried out within 24 hours after collecting the samples. All the water sources were found to be contaminated with total coliform, faecal coliform, Escherichia coli, Klebsiella spp., Salmonella spp., Shigella spp., Vibrio cholerae, Aeromonas spp. and Fungi except treated water samples. Several biochemical tests were performed for confirmation of suspected organisms. According to these results, huge amount of total coliform and faecal coliform were observed in every sample and these amount were very much similar for other organisms. Treated water samples were found to be microbiologically accepted than untreated water samples. Forty samples of freshwater clam (Batissa violacea), popularly known as Kai, collected from the Suva market were analysed for total aerobic plate count and total coliforms. The heterotrophic bacteria isolated by plate count were also characterised up to genera. The results indicated higher than acceptable level of plate count and coliform bacteria. While total plate count of aerobic heterotrophic bacteria ranged from 5.5 x 105 colony forming units (cfu) per gram to more than 107 cfu/ gram, total coliform load varied between 1.1 x 104 to 1.1 x 105 per 100 gm of Kai flesh. The characterisation of the heterotrophic bacteria revealed the predominance of the genera Micrococcus (34%) and Bacillus (24%). Other genera encountered included Acinetobacter, Vibrio, Aeromonas, Alcaligenes, Pseudomonas, Streptococcus and members of the family Enterobacteriaceae. The results revealed the need for depuration of the Kai in clean running water to reduce the bacterial load to acceptable levels. Temperature control soon after harvesting by proper icing and thorough cooking of the Kai before consumption is also recommended.

In this study, the sanitary quality of tap water sources within the University of Port Harcourt was investigated in order to determine its suitability for drinking and other domestic applications. Eight composite samples of tap water were collected from Eight different locations within the three campuses of the University of Port Harcourt using 300 ml-capacity sterile containers. These locations included NDDC Hostel, Sports Hostel, Medical Hostel, NUH Hostel, Dan Etete Hostel, Delta A Hostel, Delta B Hostel and Choba Campus. After collection, water samples were taken to the laboratory for enumeration and identification of Total heterotrophic bacteria (THB), Total coliform (TC) and Faecal coliform (FC) using the membrane filtration method. Result showed that the average THB, TC and FC counts recorded in the tap water samples across the eight locations ranged from 12.4 CFU to 36.7 CFU, 4.3 CFU to 10.1 CFU and 2.8 CFU to 5.2 CFU per 100ml respectively. Bacterial isolates were identified as probably belonging to genera such as Bacillus spp., Klebsiella spp., Enterobacter spp., Staphylococcus spp., Proteus spp., Citrobacter spp., Serratia spp. and Escherichia spp., respectively. Samples from Delta B hostel and Choba campus recorded the lowest and highest THB, TC and FC counts respectively. According to WHO standard, the result of THB, TC and FC counts indicated that the tap water in all eight locations were above the limit acceptable for drinking but within acceptable limit for other domestic use such as bathing and washing.
