Water borne diseases are the major cause of morbidity and mortality in developing countries like Bangladesh, where disease outbreaks occur due to unsafe drinking water, inadequate sanitation and poor hygienic practices. The objective of this study was to evaluate the quality of water used for drinking as well as household purposes in Dhaka city, Bangladesh. Eighteen water samples were collected from different rivers and canals. Among those, two samples were treated with chlorine. Physico-chemical parameters (pH, salinity, turbidity, conductivity and dissolved oxygen) were monitored during the time of sample collection using portable meters. For microbiological analysis, water samples were collected in sterile screw capped bottle. All Experiments were carried out within 24 hours after collecting the samples. All the water sources were found to be contaminated with total coliform, faecal coliform, Escherichia coli, Klebsiella spp., Salmonella spp., Shigella spp., Vibrio cholerae, Aeromonas spp. and Fungi except treated water samples. Several biochemical tests were performed for confirmation of suspected organisms. According to these results, huge amount of total coliform and faecal coliform were observed in every sample and these amount were very much similar for other organisms. Treated water samples were found to be microbiologically accepted than untreated water samples. Pathogenic microorganisms have been so far reported to contaminate a wide range of foods triggering food borne infections or intoxications including the enteric complications, abdominal pain, fever, bloodstream infection etc. The current study was attempted to observe the microbiological quality of some popular foods collected from different places of Dhaka city, Bangladesh. Conventional cultural, microscopic and biochemical tests were followed for the detection and enumeration of bacterial isolates associated with these food samples. The investigation encompassed detection of total viable bacterial count (TVBC) and presumptive identification of other pathogenic bacteria from these samples. Higher counts of TVBC, coliform, Staphylococcus spp. and fungal load were recorded as 1.46×107 CFU/g (yogurt 1), 4.5×106 CFU/g (yogurt 1), 5.6×105 CFU/g (raw meat) and 2.9×103 CFU/g (sea fish), respectively. Fecal coliform was detected only in one out of ten samples. On the contrary Salmonella spp., Vibrio spp., Shigella spp. and Pseudomonas spp. were completely absent. The antibiogram study showed that all the isolates were sensitive against Kanamycin and Azithromycin. Better sensitivity was also observed against Gentamycin and Ciprofloxacin. Varying degree of antibiotic resistance was also detected against Cefixime, Amikacin and Neomycin. Our study emphasizes the need for continuous monitoring of the various categories of food samples for the safety of public health.

Present study attempted to isolate and quantify the microorganisms from the pear samples collected within Dhaka metropolis. Out of the five samples studied, total viable bacterial count and the total coliform count were near about 105 cfu/g and 104 cfu/g, respectively. Among the specific pathogens, prevalence of Salmonella spp. and Vibrio spp. were found only in two samples. Whereas Klebsiella spp, Pseudomonas spp. and Staphylococcus aureus were detected in all the samples studied. Fungal growth was also observed in each sample within the range of 8.8×104 – 9.5×104 cfu/g. The study of antibiogram revealed that, most of the pathogens were resistant to more than one antibiotic. Interestingly the pear samples showed no anti-bacterial activity against any of the laboratory isolates such as, Escherichia coli, Pseudomonas spp., Salmonella spp. and Bacillus spp.Stamford Journal of Microbiology, Vol.5(1) 2015: 1-4

A total of 300 different milk and yogurt samples were collected from Dhaka city and analyzed for total viable bacterial count (TVBC), total coliform count (TCC) and presence of several pathogens such as Escherichia coli, Salmonella spp., Shigella spp., Vibrio spp. and Listeria monocytogenes. Raw milk samples collected from Moghbazar area showed highest TVBC (4.2 × 106 cfu/ml) and samples from Mohammadpur area showed lowest count (3.5 × 103 cfu/ml). All the pasteurized milk samples showed TVBC of 1.9 × 102 to 2.8 × 103 cfu/ml, and TCC only in one sample Yogurt samples (open) collected from vendor showed TVBC of 9.1 × 103 to 8.2 × 107 cfu/ml, and TCC only in sample collected from Dhanmondi, Dhaka. The packed yogurt samples showed TVBC of 2.2 × 102 to 6.1 × 103 cfu/ml, and TCC only in one sample. All the milk and yogurt samples were found contaminated with E. coli and Shigella-like species. The microbiological study of water samples obtained from Udo Anwankwo River was investigated. Water samples were collected from three different sampling points along the course of the river and analyzed using standard procedures. The total bacterial counts, total coliform counts and total fungal counts of the water samples ranged from 2.6×105 to 4.8×105cfu/ml, 1.2×104 to 1.8×105cfu/ml and 0.24×103 to 1.9×103cfu/ml respectively. A total of nine bacteria species belonging to the following genera, Bacillus, Salmonella, Escherichia, Pseudomonas, Micrococcus, Staphylococcus, Vibrio, Enterobacter and Streptococcus were isolated and identified from the samples, while five fungal isolates including members of the genera Aspergillus niger, Mucor, Penicillum, Rhizopus and Fusarium were isolated. The study through microbial analysis has revealed that the river water sample was not free from pathogens and thereby not suitable for potable use. There is need to put adequate measures towards the control of pollution and proper treatment of the water before usage as it contains pathogenic organisms.

The aims of this study were to evaluate the contamination of lettuce (Lactuca sativa), produced in Londrina, Paraná (PR), with total coliform, coliform at 45 °C, E. coli, and Salmonella spp.; and to determine the E. coli contamination of irrigation water used at the farms studied. Four farms were evaluated, of which three produced lettuce using a conventional system and one using an organic system. An evaluation of the production practices of the farms was also carried out. A total of 111 samples were analyzed, 71 lettuce samples from the conventional system and 40 samples from the organic system. A total of eight irrigation water samples were collected for analysis. Coliform at 45 °C counts above the limit tolerated by Brazilian legislation were observed in 2.8% (2/71) of conventionally grown lettuce samples, and Salmonella spp. was isolated in 1.4% (1/71) of those samples. In the organic lettuce samples, 12.5% (5/40) had coliform at 45 °C counts above the limit tolerated and Salmonella spp. was not detected. Irrigation water samples from three farms were unsatisfactory, with counts higher than 102MPN of E. coli per 100mL. The results of this study demonstrate that most conventionally grown lettuce samples show good sanitary conditions in production, and that lettuce contamination is not related to contamination found in irrigation water samples. The results also showed that the organic production practices required by Brazilian certification agencies should be applied to ensure that contamination of produced lettuce remains controlled.

Chattogram is the second most populated city in Bangladesh. This port city faces a serious threat mainly due to the lack of safe drinking water. This study was conducted for determining drinking water quality of groundwater sources in Chattogram city. The study was performed in the BCSIR laboratory, Chattogram. It was carried out for a period of six months from 1st July, 2018 to 31th December, 2018. Total six water samples were collected from three different locations (Baluchora, C&B colony and Khulshi area). Each sampling location consists of two separate sampling points. Physicochemical parameters of the collected samples like Temperature, pH, Electrical Conductivity (EC), Total dissolved solid (TDS), Hardness, Turbidity and concentration of Cl, As, Mn, Fe, Pb, Cr and Cd were examined. Microbial parameters like Total Coliform (TC) were also measured. All the analyzed parameters compared with BSTI and WHO drinking water quality standards to understand the overall ground water quality status of the study area. The results reveal that water samples in almost all locations were contaminated with microbial contamination and that the range of physico-chemical parameters was not adequate for consumption. Preliminary treatments like boiling, filtering etc are required before using groundwater for drinking and the necessary measures must be taken for a safe alternative source of drinking water.

Commercial drinking water may serve as potential threat to public health if these items are contaminated with a number of pathogenic microorganisms due to faulty manufacturing process. Present study attempted to isolate and quantify the microorganisms from various jar and bottle water samples collected from various areas of Dhaka city. Antibiotic susceptibility pattern of suspected bacterial isolates were also determined in this study. Out of the eighteen samples studied, ten were jar water samples and eight were bottled water samples. The range of total viable bacterial count (TVBC) in these samples ranged from 102 to 105 cfu/ml. Specific pathogens such as, Salmonella spp., Shigella spp., Vibrio spp. and fecal coliforms could not be found in these samples. However coliforms could be detected in 10 samples. The antibiogram study showed that all Escherichia coli and Klebsiella spp. isolates found from these samples were sensitive against gentamicin (10 μg) and azithromycin (30 μg). Variable antibiotic resistance among these bacterial isolates was detected against cefotaxime (30 μg), streptomycin (10 μg) and erythromycin (15 μg). A study, using four types of water samples ranging from relatively pure stream water to water containing industrial and domestic effluents, was carried out to investigate the effect of storage temperature on four bacterial parameters: total coliform, fecal coliform, fecal streptococcus, and heterotrophic bacteria. In the study, each water sample was cooled immediately after collection to approximately 1.5 °C by storage in crushed ice. At 2-, 24-, 30-, and 48-h intervals, the sample was mixed, and a subsample was removed and tested. Three separate analyses for each parameter were made on each subsample. The data available for statistical analyses contained, in some cases, the values obtained for the three subsamples and, in others, the means of the three values. The data were analysed as replicated data and as part of the entire set. The analysed data indicated (i) that with the exception of heterotrophic populations, more than 75% of the samples were microbiologically stable for at least 24 h, (ii) that at least 50% of samples tested for heterotrophic densities were stable for a minimum period of 24 h, (iii) that the original water temperature and bacterial load do not appear to be consistent factors in the preservation of samples for microbiological analysis, and (iv) that nutrient levels, also, do not seem to be consistent factors in the preservation of water samples for microbiological analyses.

The current study was aimed to detect the presence of contaminating microorganisms in some commonly available spices collected from different areas of Dhaka city, Bangladesh and also to assess their antibacterial activity. Out of eight samples under this study, total viable bacterial count (TVBC) was observed within the range of 106 to 107cfu/g. Among the specific bacterial isolates Staphylococcus spp., Pseudomonas spp. and Escherichia coli were found in almost all the samples within the margin of 102 to 106 cfu/g. Alarmingly Salmonella spp. and Vibrio spp. were presumptively identified in one and two samples respectively. Besides every sample showed the presence of fungus with load up to 103 cfu/g. Antibacterial activity of these samples was tested against seven laboratory isolates. Syzygium aromaticum showed highest activity against as many as five bacterial isolates while Cinnamomum verum was active against four isolates. On the other contrary, both Zingiber officinale and Laurus nobilis demonstrated their antibacterial activity only against Staphylococcus spp. and Klebsiella spp. respectively. Penelitian ini bertujuan untuk mengetahui kualitas Air di Desa Rarowatu Kabupaten Bombana. Pemeriksaan coliform pada air sumur bor dan air swadaya masyarakat menggunakan Metode Most Probable Number (MPN). Pada perlakuan ini terdiri dari presumptive test yang menggunakan media Lactose Broth (LB) dengan metode 3 seri tabung, Tahap berikutnya confirmative test dengan menggunakan media Brillian Green Lactose Broth (BGLB). Pada identifikasi  Escherichia coli  menggunakan media Eosin Methylene Blue Agar, pewarnaan Gram dan media uji biokimia. Hasil penelitian menunjukkan sampel yang terdiri dari 7 sampel air sumur bor dan I sampel air swadaya masyarakat berdasarkan uji MPN mencapai 2400/100 ml. Hasil identifikasi menggunakan media EMBA menunjukkan koloni yang berwarna hijau metalik pada sampel air swadaya  serta  dilakukan pewarnaan Gram menunjukkan bentuk basil pendek dan gram negative. Hasil uji biokimia menunjukkan hasil uji indol positif, methyl red positif, voges proskauer negatif dan uji sitrat negatif. Uji IMViC dapat disimpulkan bahwa bakteri pada kode sampel D memiliki sifat yang sama terhadap bakteri E. coli. Kata kunci :  Coliform, Identifikasi, Escherichia coli, BombanaABSTRACTThis study aims to determine the quality of water in Rarowatu Village, Bombana Regency. Coliform examination in borehole water and community self-water using the Most Probable Number (MPN) Method. This treatment consists of a presumptive test that uses Lactose Broth (LB) media with the tube 3 series method. The next stage is the confirmative test using Brilliant Green Lactose Broth (BGLB) media. The identification of Escherichia coli using Eosin Methylene Blue Agar media, Gram staining, and biochemical test media. The results of the study showed that samples consisting of 7 samples of bore well water and I community self-water samples based on the MPN test reached 2400/100 ml. The identification results using EMBA media showed that metallic green colonies in self-help water samples and Gram staining performed showed short and gram-negative bacilli forms.
