Time course of adaptation to hypoxia in newborn rats

In newborn rats after a few minutes of hypoxia, ventilation is similar to the normoxic value. Nevertheless, after a few days in hypoxia, newborn rats have a sustained hyperventilation. In this study we examined the time course of the newborn rat's adaptation to hypoxia. Measurements of body size, hematocrit, lung and heart mass, and breathing pattern have been performed on newborn rats exposed to hypoxia (10% O2) for different time intervals from 4 to 60 h (hypoxic, H), and on same-age rats growing in air (controls, C). Ventilation measured by flow plethysmography was increased in H rats above the C value from about 8 h; this was due to a higher breathing rate and, from 24 h, also to a larger tidal volume. During the early hours of hypoxia, oxygen consumption measured manometrically was about 50% of C, while after 3 days in hypoxia it was almost like the C value. These observations indicate that the lack of sustained hyperventilation, characteristic of the newborn's acute exposure to hypoxia, is an immediate but transient phenomenon that is resolved after a few hours, and suggest a tight link between metabolic and ventilatory hypoxic responses. Body weight of H rats was less than in C, owing to an immediate decrease below the prehypoxic value. Dry heart and lung weight changed in proportion with the rest of the body during the first 36–48 h of hypoxia, then they increased disproportionately more. Hence, these temporal changes suggest that the large heart and lung weight – body weight ratios of the chronic hypoxic animals result from their smaller body mass and the stimulated growth of cardiac and pulmonary tissues.

We asked 1) whether newborn rats respond to chronic hypercapnia with a persistent increase in ventilation and 2) whether changes in lung mass were accompanying the respiratory adaptation to chronic hypercapnia, as previously observed during neonatal chronic hypoxia. Five litters of rats were kept in 7% CO2 (with 21% O2) from day 1 to 7 after birth (CO2exp) and compared with six litters of control rats growing in normocapnia-normoxia (C). Body weight was similar between the two groups. Ventilation, measured by flow plethysmography, increased in CO2exp from day 2 and remained steadily elevated, and at day 7 it almost doubled (174%) the C value because of the large increase in tidal volume and mean inspiratory flow (192 and 189%, respectively) with no changes in respiratory frequency. Two days after return to normocapnia, ventilation was still 33% higher than in C; at this time, acute exposure to hypercapnia increased ventilation relatively less in the CO2exp than in C because of a lower increase in tidal volume. Neither the lung weight-to-body weight nor the heart weight-to-body weight ratios increased in CO2exp. We conclude that 1) chronic hypercapnia in newborn rats induces a steady increase in ventilation, which persists at least 2 days after return to normocapnia with a reduction in the acute response to CO2, and 2) hyperventilation per se is not the cause of the increased lung mass observed during chronic neonatal hypoxia.

We characterized the time course of the left ventricular (LV) geometric and functional changes after aortic banding, validated them by necropsy, and investigated the sensitivity of echocardiographic findings on LV hypertrophy. C57BL/6 mice were subjected to transverse aortic constriction (TAC) or sham operation; echocardiographic assessments were performed before or at 2, 4, 6, and 11 wk after surgery; and some of the mice were euthanized at the corresponding time points. There was a progressive increase in diastolic posterior wall thickness and LV systolic dimension; the percentage of LV fractional shortening (LV%FS) decreased progressively at 4 wk, whereas these parameters remained stable in sham-operated mice. Echo LV mass and LV%FS correlated well with actual whole heart mass and ratio of lung weight to body weight, respectively ( r = 0.765 and −0.749, respectively; P < 0.0001). These results suggest that the development of myocardial hypertrophy and systolic dysfunction is a time-dependent process. Echocardiographic assessment of myocardial hypertrophy and functional changes correlate well with the actual heart mass and lung mass. Echocardiography is sensitive enough to assess myocardial hypertrophy and heart functional changes induced by pressure overload in mice.

In newborn rats, lipoprotein lipase (LPL) activity was higher in the liver than in several other tissues, such as heart, diaphragm or lungs, and accounted for about 3% of total LPL activity in the body. There was no significant correlation between LPL activity in liver and in plasma. Thus transport of the enzyme from extrahepatic tissues was probably not the major source of LPL in liver. To study LPL biosynthesis directly, newborn rats were injected intraperitoneally with [35S]methionine, and LPL was isolated by immunoprecipitation and separation by SDS/polyacrylamide-gel electrophoresis. Radioactivity in LPL increased with a similar time course in all tissues studied, including the liver. Substantial synthesis of LPL was also demonstrated in isolated perfused livers from newborn rats, whereas synthesis was low in livers from adult rats. There was strong LPL immunofluorescence in livers from newborn rats, mainly within sinusoids and along the walls of larger vessels. This labelling disappeared after perfusion with heparin, which indicates that much of the enzyme is in contact with blood and can take part in lipoprotein metabolism.

Modifications in load-related sensory input during unloaded walking can lead to recalibration of the body schema and result in aftereffects. The main objective of this study was to identify the adaptive changes in gait and body-weight perception produced by unloaded walking. Gait performance during treadmill walking was assessed in 12 young participants before and after 30 min of unloaded walking (38% body weight) by measuring lower limb kinematics, temporal gait measures, and electromyography (EMG). A customized weight-perception scale was used to assess perception of body weight. Participants perceived their body weight to be significantly heavier than normal after unloading while walking. Angular displacement about ankle and knee was significantly reduced immediately after unloaded walking, while temporal gait parameters remained unchanged. The EMG activity in some muscles was significantly reduced after unloading. These findings indicate that walking at reduced body weight results in alterations in segmental kinematics, neuromuscular activity, and perception of body weight, which are the aftereffects of motor adaptation to altered load-related afferent information produced by unloading. Understanding the adaptive responses of gait to unloading and the time course of the aftereffects will be useful for practitioners who use body-weight unloading for rehabilitation.

Abstract. The effects of the restriction of the advised daily feed intake (ADFI) during 1 or 2 weeks on broiler body weight (BW) and carcass composition were studied. A total of 150 1-day-old male broilers (Ross-308) were randomly allotted to a control and four experimental treatments with three replicates per treatment. Broilers were subjected to a feeding restriction equal to 75 and 50 % of ADFI, either for a 7- or a 14-day period, between the 7th and 14th and 7th and 21th day of age, respectively. The BW and carcass composition were determined on day 42. From the findings, neither of the two levels of restriction nor the two time intervals had significant effects (P > 0.05) on the BW or breast and leg weight and proportion, liver and most of the body components. However, the feeding treatments show a tendency (P = 0.078) to influence the BW, with birds fed 75 % of ADFI for 14 day being heavier than the control group. For eviscerated carcass weight, an effect (P = 0.002) was observed, with birds fed 75 % of ADFI for 14 day having heavier carcasses (P < 0.05) than those in control group. The abdominal fat, both with regard to weight and proportion, was slightly influenced by dietary treatments (P > 0.05). From our results, it can be concluded that the applied feeding restrictions have a neutral or slightly beneficial effect on broiler growth traits and carcass composition.

Previously, we have found that the addition of L-ascorbic acid to chitosan enhanced the reduction in body weight gain in guinea pigs fed a high-fat diet. We hypothesized that the addition of L-ascorbic acid to chitosan would accelerate the reduction of body weight in humans, similar to the animal model. Overweight subjects administered chitosan with or without L-ascorbic acid for 8 weeks, were assigned to three groups: Control group (N = 26, placebo, vehicle only), Chito group (N = 27, 3 g/day chitosan), and Chito-vita group (N = 27, 3 g/day chitosan plus 2 g/day L-ascorbic acid). The body weights and body mass index (BMI) of the Chito and Chito-vita groups decreased significantly (p < 0.05) compared to the Control group. The BMI of the Chito-vita group decreased significantly compared to the Chito group (Chito: -1.0 kg/m2 vs. Chito-vita: -1.6 kg/m2, p < 0.05). The results showed that the chitosan enhanced reduction of body weight and BMI was accentuated by the addition of L-ascorbic acid. The fat mass, percentage body fat, body circumference, and skinfold thickness in the Chito and Chito-vita groups decreased more than the Control group; however, these parameters were not significantly different between the three groups. Chitosan combined with L-ascorbic acid may be useful for controlling body weight.

Summary Aim: The influence of physiological and pharmacological amounts of iodine on the uptake of radioiodine in the thyroid was examined in a 4-compartment model. This model allows equations to be derived describing the distribution of tracer iodine as a function of time. The aim of the study was to compare the predictions of the model with experimental data. Methods: Five euthyroid persons received stable iodine (200 μg, 10 mg). 1-123-uptake into the thyroid was measured with the Nal (Tl)-detector of a body counter under physiological conditions and after application of each dose of additional iodine. Actual measurements and predicted values were compared, taking into account the individual iodine supply as estimated from the thyroid uptake under physiological conditions and data from the literature. Results: Thyroid iodine uptake decreased from 80% under physiological conditions to 50% in individuals with very low iodine supply (15 μg/d) (n = 2). The uptake calculated from the model was 36%. Iodine uptake into the thyroid did not decrease in individuals with typical iodine supply, i.e. for Cologne 65-85 μg/d (n = 3). After application of 10 mg of stable iodine, uptake into the thyroid decreased in all individuals to about 5%, in accordance with the model calculations. Conclusion: Comparison of theoretical predictions with the measured values demonstrated that the model tested is well suited for describing the time course of iodine distribution and uptake within the body. It can now be used to study aspects of iodine metabolism relevant to the pharmacological administration of iodine which cannot be investigated experimentally in humans for ethical and technical reasons.

Introduction: Prevalence of obesity is 30 % in the Czech Republic and is expected to increase further in the future. This disease complicates surgical procedures but also the postoperative period. The aim of our paper is to present the surgical technique called hand-assisted laparoscopic nephrectomy (HALS), used in surgical management of kidney cancer in morbid obese patients with BMI >40 kg/m2. Methods: The basic cohort of seven patients with BMI >40 undergoing HALS nephrectomy was retrospectively evaluated. Demographic data were analyzed (age, gender, body weight, height, BMI and comorbidities). The perioperative course (surgery time, blood loss, ICU time, hospital stay and early complications), tumor characteristics (histology, TNM classification, tumor size, removed kidney size) and postoperative follow-up were evaluated. Results: The patient age was 38−67 years; the cohort included 2 females and 5 males, the body weight was 117−155 kg and the BMI was 40.3−501 kg/m2. Surgery time was 73−98 minutes, blood loss was 20−450 ml, and hospital stay was 5−7 days; incisional hernia occurred in one patient. Kidney cancer was confirmed in all cases, 48–110 mm in diameter, and the largest removed specimen size was 210×140×130 mm. One patient died just 9 months after the surgery because of metastatic disease; the tumor-free period in the other patients currently varies between 1 and 5 years. Conclusion: HALS nephrectomy seems to be a suitable and safe surgical technique in complicated patients like these morbid obese patients. HALS nephrectomy provides acceptable surgical and oncological results.

ABSTRACT The effect of three doses of prednisolone and ACTH respectively on the weight of the body, the lungs and the hilar lymph nodes was studied on rats killed one month after the intratracheal (i.t.) injection of 50 mg of fine-particulate quartz. The prednisolone was administered via the drinking water, and the ACTH was injected intraperitoneally during the period between the i.t. injection of quartz dust and the killing of the animals. Prednisolone causes the rats to become cachectic and reduces the weight of the hilar lymph nodes. It also retards the transport of quartz dust from the lungs via the lymphatics. All these effects increase with increasing doses of prednisolone. However, its effect on the lung weight is insignificant. ACTH does not affect the body weight, but retards the weight increase of the lungs and the hilar lymph nodes. These effects increase with increasing doses of ACTH, and seem to be connected with an ability of ACTH to promote the clearance of quartz dust from the lungs and hilar lymph nodes. The method used, however, does not make it possible to differentiate quantitatively between any ACTH effects on the bronchogenie and lymphatic lung-clearance mechanisms. Available data, however, indicate that the stimulation of the dust transport from the lungs and hilar lymph nodes is, at least to some extent, related to the lymphatic system.

The article presents the data on the study of the influence of mycotoxins combination (T-2 toxin at the concentration of 0.095 mg/kg and aflatoxin B1 in the concentration of 0.019 mg/kg) on the body of quails and the results of pharmacocorrection of toxicosis with a complex consisting of beet pulp and lecithin. Structural changes in the intestines of quais at fodder mycotoxicosis are described. The use of antitoxic feed additives in poultry led to a weakening of the action of xenobiotics, which was confirmed by an increase in the safety of poultry and increase in body weight of quails, a decrease in the clinical manifestations of intoxication, as well as in positive changes in the structure of the intestine of the poultry during histological examination.
