Cymbopogon caesius grass is an aromatic tall grass species which grows in waste field and along the edges of crop fields in Karnataka. From this grass fungal endophytes were isolated on PDA medium and identified as Curvularia lunata. The fungus was cultured on Potato Dextrose broth for production of fungal metabolites. Ethyl acetate extract of the C. lunata was obtained by liquid-liquid partition of broth of endophyte and evaporation. Dried crude extract was tested for antimicrobial activity by agar well diffusion method. The extract showed antimicrobial nature against all the test pathogens. Crude extract of C. lunata is highly active against Staphylococcus aureus with the inhibition percentage of 92 and Candida albicans with 81 percent inhibition.

An endophytic fungus isolated from the plantCinnamomum mollissimumwas investigated for the bioactivity of its metabolites. The fungus, similar to aPhomasp., was cultured in potato dextrose broth for two weeks, followed by extraction with ethyl acetate. The crude extract obtained was fractionated by high-performance liquid chromatography. Both crude extract and fractions were assayed for cytotoxicity against P388 murine leukemic cells and inhibition of bacterial and fungal pathogens. The bioactive extract fraction was purified further and characterized by nuclear magnetic resonance, mass spectral and X-ray crystallography analysis. A polyketide compound, 5-hydroxyramulosin, was identified as the constituent of the bioactive fungal extract fraction. This compound inhibited the fungal pathogenAspergillus niger(IC501.56 μg/mL) and was cytotoxic against murine leukemia cells (IC502.10 μg/mL). 5-Hydroxyramulosin was the major compound produced by the endophytic fungus. This research suggests that fungal endophytes are a good source of bioactive metabolites which have potential applications in medicine.

Background: Quinazolines and quinazolinones derivatives are well known for their important range of therapeutic activities. Objective: The study aims to carry out the synthesis of some derivatives of substituted fluoroquinazolinones based on structure-based design and evaluation of their antibacterial, antifungal, and anti-biofilm activities. Methods: Compounds were chemically synthesized by conventional methods. Structures were established on the basis of spectral and elemental analyses. The antimicrobial potential was tested against various microorganisms using the agar disc-diffusion method. MIC and MBC as well as anti-biofilm activity for the highly active compounds were assessed. Moreover, the computational studies were performed using Auto dock free software package (version 4.0) to explain the predicted mode of binding. Results: All derivatives (5-8), (10a-g), and (A-H) were biologically tested and showed significant antimicrobial activity comparable to the reference compounds. Compounds 10b, 10c, and 10d had a good MIC and MBC against Gram-positive bacteria, whereas 10b and 10d showed significant MIC and MBC against Gram-negative bacteria. However, compounds E and F exhibited good MIC and MBC against fungi. Compound 10c and 8 exhibited significant anti-biofilm activity towards S. aureus and M. luteus. Molecular docking study revealed a strong binding of these derivatives with their receptor-site and detected their predicted mode of binding. Conclusion: The synthesized derivatives showed promising antibacterial, antifungal, and antibiofilm activities. Modeling study explained their binding mode and showed strong binding affinity with their receptor-site. The highly active compounds 5 and 10c could be subjected to future optimization and investigation to be effective antimicrobial agents.

Abstract U. platyphylla is a weedy grass species commonly found in disturbed, open and sandy sites such as crop fields, ditches and roadsides. It is considered a troublesome weed because of its tolerance to some herbicides principally in maize plantations (Chamblee et al., 1982; Gallaher et al.,1999). U. platyphylla is highly adaptable and it is able to germinate and grow throughout a wide range of soil and environmental conditions (Burke et al., 2003). Additionally, its seeds may remain on the crop residue until pre-emergence herbicides are no longer effective in controlling the germinating seeds, at which time the seeds fall to the soil surface and germinate (Alford et al., 2005).

Mucormycosis is an uncommon but life-threatening invasive fungal infection, mostly occurs in immunocompromised patients. Lacking the appropriate antifungal drugs is one of the reasons that lead to difficulties in the management of mucormycosis. Curcuma longa has been used traditionally and widely to treat various diseases, including fungal infections. In the search for novel antifungal compounds from natural resources, we evaluated the effect of rhizome crude extract of C. longa on Mucor circinelloides – a causal agent of mucormycosis. The results of screening, using broth dilution method and agar-well diffusion method, showed that the C. longa extract exhibited promising antifungal activity against the fungus M. circinelloides. In liquid medium, C. longa extract decreased the ability of spore germination and the speed of hyphae formation of M. circinelloides decreased by up to approximately 70% and 90%, respectively. Besides, in a solid medium, the crude extract presented similar activity with amphotericin B (400 μg\mL) in decreasing the growth of M. circinelloides by nearly 77%. Moreover, the extract of C. longa also likely to induce the yeast-like type of growth of the dimorphic M. circinelloides in the early stage. These results suggest the plant could be a potential source for further study on biochemical components and the mechanism of its antifungal activity.

Asan wetland is a man-made wetland located in Uttarakhand state. In the given study, efforts have been made to study the floristic diversity and species composition of the aquatic macrophyte in Asan wetland and provide a check list. Sixty seven species were recorded under 32 families being maximum in Cyperaceae followed by Poaceae and Asteraceae. Remaining families comprised either one or two species each. Among all the recorded species, monocots (54%) were dominated over dicots (42%) and remaining 4% comprised ferns. Herbaceous macrophytes were found growing abundantly (54%) which are followed in degree of prevalence by the grasses (21%), sedges (18%), shrubs (4%) and climbers (3%). In addition, this study also revealed that pollution resistant species like Bacopa monnieri, Hydrilla verticillata, Vallisneria spiralis and Eichhornia crassipes along with tall grass species which grow in shallow water are spreading considerably that indicates an increasing pollution in the wetland. The present study will provide an important set of informations to support research work like limnological studies, productivity, conservation and management programs of this wetland.

Fungal endophytes of the genus Epichloë live symbiotically in cool season grass species and can produce alkaloids toxic to insects and vertebrates, yet reports of intoxication of grazing animals have been rare in Europe in contrast to overseas. However, due to the beneficial resistance traits observed in Epichloë infected grasses, the inclusion of Epichloë in seed mixtures might become increasingly advantageous. Despite the toxicity of fungal alkaloids, European seed mixtures are rarely tested for Epichloë infection and their infection status is unknown for consumers. In this study, we tested 24 commercially available seed mixtures for their infection rates with Epichloë endophytes and measured the concentrations of the alkaloids ergovaline, lolitrem B, paxilline, and peramine. We detected Epichloë infections in six seed mixtures, and four contained vertebrate and insect toxic alkaloids typical for Epichloë festucae var. lolii infecting Lolium perenne. As Epichloë infected seed mixtures can harm livestock, when infected grasses become dominant in the seeded grasslands, we recommend seed producers to test and communicate Epichloë infection status or avoiding Epichloë infected seed mixtures.

Objective: The objective of this study was to investigate the antimicrobial activity of Phellinus linteus against skin infectious pathogens, Staphylococcus epidermidis ATCC12228 and Propionibacterium acnes DMST 14916.Methods: Fungal fruiting bodies were extracted with 95% ethanol and ethyl acetate, and then, vaporized. The antimicrobial activities were determined by the disc diffusion method against Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228 skin infectious pathogens. A minimum inhibitory concentration (MIC) and a minimum bactericidal concentration (MBC) for those crude extracts were determined. Finally, the chemical profile of crude extract was determined by using thin layer chromatography and GC-MS.Results: The result demonstrated that the ethanolic extraction had more active fractions with an MIC of 0.5 mg/ml against the growth of Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228 and also showed a minimum inhibitory concentration (MBC) at a concentration of 1.0 mg/ml, while ethyl acetate-based solvents failed to develop on TLC according to Retention factor (Rf) values of 0.71-0.76. The GC-MS was applied to investigate the chemical profile of crude extract of Phellinus linteus, revealing a component of hexadecanoic acid and 9, 12-octadecadienoic acid.Conclusion: Phellinus linteus fruiting body extracts have great potential as antimicrobial compounds against Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228. Thus, they can be used in the treatment of infectious diseases caused by bacterial pathogens.

Introduction: Endophyte is considered as a source of natural bioactive secondary metabolites that provides an array of bioactive lead compounds. The present study was aimed to determine the antimicrobial and anti-inflammatory potential of fungal endophytes isolated from Catharanthus roseus. Methods: A total of seven fungal endophytes crude extract were screened against bacterial pathogens. Of these, Curvularia geniculata CATDLF7 crude extract exhibited the most potent inhibitory activity against bacterial pathogen. Hence, CATDLF7 crude extract was subjected to the chromatographic separation. This purification leads to the isolation of six pure compounds (1PS - 6PS). Of these, 3PS was found to be a major constituent and most effective against clinical isolates of methicillin resistant Staphylococcus aureus (MRSA) with minimum inhibitory concentration (MIC) values ranging from 100 to 200 μg/ml. Based on the spectroscopic data, 3PS was characterized as α,β-dehydrocurvularin. This compound also showed synergistic interaction with norfloxacin, and reduced its MIC up to 32-folds with fractional inhibitory concentration index (FICI) of 0.09. Results: To understand the possible antibacterial mechanism of action, α,β-dehydrocurvularin alone (100 μg/ml) exhibited efflux pump inhibitory potential by 0.84 fold decreasing in ethidium bromide (EtBr) fluorescence. In addition, α,β-dehydrocurvularin inhibited inflammatory cytokines TNF-α and IL-6 production which is further validated by molecular docking score -4.921 and -5.641 respectively for understanding orientation and binding affinity. Conclusion: Overall the results highlighted to identifying bioactive compound α,β-dehydrocurvularin which could be used as an antimicrobial and anti-inflammatory agents.

Echinodorus grandiflorus has pharmacological properties due to its secondary metabolism, such as anti-inflammatory, antioxidant, diuretic, analgesic, anti-rheumatic, antihypertensive, and cardioprotective effects. The aim of this study was to determine the phytochemical profile and evaluate the antimicrobial activity of crude extract of E. grandiflorus form its leaves. In the analysis of the phytochemical profile, qualitative tests were performed to identify tannins, alkaloids, flavonoids, anthraquinones, steroids, triterpenes, saponins, polysaccharides, and coumarins. Antimicrobial tests were performed using the disk diffusion method and minimum inhibitory concentration (MIC) in 96-well microplates, using hydroalcoholic crude extract obtained by maceration in the proportions 1:5 and 1:10. The higher content of crude extract was observed by maceration 1:5 (3.26%). In phytochemical tests, the presence of tannins, alkaloids, flavonoids, and saponins was detected. The microbial strains evaluated were Staphylococcus aureus, S. epidermidis, Lactobacillus casei, Bacillus cereus, Escherichia coli, Pseudomonas aeruginosa, and Candida albicans. The antimicrobial activity of crude extract has not apparent against the tested organisms. It is concluded that the crude extract present several phytochemical, however did not show antimicrobial activity, and furthermore studies should be carried out researching isolated chemical compounds and the antimicrobial activity leaves crude extract of Echinodorus grandiflorus plant.

 Hispid hare (Caprolagu shispidus) is an endangered lagomorph which had a wide range of distribution in the past, but currently it is known to occur only from few isolated tall grass pockets of India and Nepal. We explored the population status, habitat utilization and existing threats of the Hispid hare at Shuklaphanta National Park (SNP) by the strip transect and quadrate sampling methods from November 2015 to May 2016 covering both winter and summer seasons. Four study sites of total 1.58ha were selected within SNPand strip transects were randomly laid to determine presence and absence of pellets of Hispid hare and their counts were used to estimate the population density. The Population density of Hispid hare was found to be 0.1820/ha in winter and 0.2268/ha in summer with the statistically significant preference to the tall grassland habitat. Twelve grass species were identified in the habitat of Hispid hare with dominancy of Saccharum spontaneum, Imperata cylindrica, Narenga porphyrocoma, and Saccharum munj. Among the grasses, Imperata cylindrica and Narenga porphyrocoma were the food species of higher preference for Hispid hare.
