Objective. To study the effect of Heweianshen decoction (HAD) on orexin-A and cholecystokinin-8 (CCK-8) expression in rat models of insomnia caused by injecting parachlorophenylalanine (PCPA) intraperitoneally.Methods. Fifty male Wistar rats were randomly divided into five groups (10 rats in each group): blank group, model group, and low-, medium-, and high-dose HAD-treated groups. A rat model of insomnia was established by injecting intraperitoneally with PCPA (300 mg/kg body weight). Rats were given normal saline (10 mL/kg) or 5.25, 10.5, and 21 g/kg HAD by intragastric administration once a day for 6 days. After that, the rats were sacrificed to collect the hypothalamus for tests, using radioimmunoassay to detect the expression of orexin-A and CCK-8.Results. Heweianshen decoction reduced the expression of orexin-A and increased the expression of CCK-8 in the hypothalamus of rat model of insomnia.Conclusion. The therapeutic effect of HAD on insomnia is partially attributed to the decreased expression of orexin-A and increased expression of CCK-8.

Abstract BackgroundPlantaginis semen has been widely used as folk medicine and health care food against hyperuricemia (HUA) and gout, but little was known about its pharmacological mechanism. MethodsThe model was established by potassium oxonate intragastric administration. 42 Sprague-Dawley (SD) male rats were randomly divided into the control group, model group, benzbromarone group (10 mg/kg) and three Plantaginis semen groups (n = 7). The Plantaginis semen groups were treated orally with Plantaginis semen at 0.9375, 1.875 and 3.75 g/kg for 28 days. The levels of serum uric acid (UA), creatinine (Cr), triacylglycerol (TG) and tumor necrosis factor-α (TNF-α) were detected using enzyme-linked immunosorbent assay kits. Ultra performance liquid chromatography quadrupole time of flight mass spectrometry (UPLC-Q-TOF/MS) was used as the basis for serum lipidomics analysis, and orthogonal partial least squares discriminant analysis (OPLS-DA) was carried out for the pattern recognition and characteristic metabolites identification. The relative levels of critical regulatory factors of urate anion transporter 1(URAT1) and phosphatidylinositol 3-kinase/ protein kinases B (PI3K/Akt) were determined by quantitative real-time polymerase chain reaction (RT-qPCR). ResultsCompared with the model group, the levels of serum UA, Cr, and TG were significantly (p<0.01) decreased in benzbromarone and three Plantaginis semen groups and the level of serum TNF-α was significantly (p<0.05) decreased in benzbromarone and low dose of Plantaginis semen group. With lipidomics analysis, significant lipid metabolic perturbations were observed in HUA rats, 13 metabolites were identified as potential biomarkers and glycerophospholipid metabolism pathway was mostly affected. These perturbations can be partially restored via treatment of benzbromarone and Plantaginis semen. Additionally, the URAT1 and PI3K/Akt mRNA expression levels were significantly decreased (p<0.05) after treatment with benzbromarone and high dose of Plantaginis semen. ConclusionsPlantaginis semen had significant anti-HUA, anti-inflammatory and renal protection effects and could attenuate potassium oxonate-induced HUA through regulation of lipid metabolism disorder. Objective To observe the effect of Xiaochaihu Decoction on cytokines and MAO in model rats with experimental autoimmune prostatitis (EAP) to provide an experimental basis for the use of Chinese herbal prescriptions in the treatment of chronic prostatitis. Methods Thirty male Wistar rats were randomly divided into blank Group, model group and treatment group (Xiaochaihu Decoction). Except the blank group, rats in model and treatment groups were injected subcutaneously in multiple points on days 0 and 30 with prostatic protein extractive solution (60 mg/mL), and intraperitoneally injected with diph-theria-pertussis and tetanus vaccine (DPT vaccine) to establish the EAP model. Model rats were administrated Xiaochaihu Decoction and were sacrificed after 4 weeks. Pathological changes in the prostate gland were observed with HE staining and changes in serum interleukin-6 (IL-6), interleukin-8 (IL-8), and Tumor necrosis factor-α (TNF-α) levels were detected with enzyme-linked immunosorbent assay (ELISA). Quantitative PCR to observe the expression of IL-17, CCL2 and MAOA/B. Results After treatment with Xiaochaihu Decoction, the serum IL-6, IL-8, and TNF-α levels all significantly lower in the treatment group as compared with the model group (P < 0.05). The intervention of Xiaochaihu Decoction can change the state of abnormally elevated IL-17, CCL2 and MAOA expression levels. Conclusions The therapeutic efficacy remarkable from the theory of heating into the sperm chamber to experimental autoimmune prostatitis rats.

Hyperuricemia (HUA) is a metabolic disorder of purine metabolism which leads to the increase of serum uric acid. Tongfeng Lian granule (TFLA) is a clinical empirical traditional herb prescription in China, is biobased natural product material. To study its regulatory mechanism on uric acid transporter in rats with HUA, the rat model of hyperuricemic was established by oral administration of potassium oxazinate (1.5 g/kg) and adenine (0.05 g/kg). The related indexes were detected to evaluate the uric Acid Transporter after treat with TFLA. In the results, compared with model group, the high dose TFLA can reduce the levels of Related serum biochemical indexes (*P < 0.05, **P < 0.01). TFLA could reduce the levels of URAT1, GLUT9 and increase the expression of OAT3 in kidney. In conclusion, TFLA can effectively inhibit the level of serum uric acid with hyperuricemia rats, and the possible mechanism related to TFLA inhibiting the reabsorption of uric acid by URAT1 and GLUT9, promoting the secretion of OAT3 and uric acid into urine.

Objective: To study the effects of the ethnic medicine Polygala fallax Hemsl with Guangxi characteristics on the sex hormones and ?-EP in research objective perimenopausal rat models. Methods: 40 female SPF rats were randomly divided into 4 groups, including the normal, model, high-dose and low-dose groups. Rats of three groups except for the normal one were treated with perimenopausal modelling through the method of subcutaneous injection of compound 4-VCD for 15 consecutive days. Rats of the normal and model group were normally fed without any treatment. Rats of the high-dose and low-dose groups were administered by high- and low-dose intragastric administration of the extract of Polygala fallax Hemsl. According to the menstrual cycle of the vaginal smear of the rat, each menstrual cycle is a course of treatment and 6 consecutive courses of treatment would be given. The indexes of serum sex hormones (E2, FSH, LH) and ?-EP of rats in each group were observed after treatment. Results: After the treatment of 6 cycles, for the levels of ?-EP and E2, the model group was lowest (P<0.05), the normal group was highest (P<0.05); and the high-dose group was higher than the low-dose group; For the levels of FSH and LH, the normal group was lowest (P<0.05), the model group was highest (P<0.05), and the high-dose group was lower than the low-dose group. Conclusion: Guangxi characteristic national medicine Polygala fallax Hemsl can effectively improve the levels of serum sex hormones and ?-EP in perimenopausal rat models and relieve the related symptoms with a certain dose-effect relationship.

Background: Moxibustion treatment has been found to ameliorate clinical symptoms including abdominal pain, diarrhoea and constipation in patients with irritable bowel syndrome (IBS). Herein we investigated the mechanisms underlying the use of moxibustion in a rat model of IBS. Methods: In our study, an IBS model was established in rats by colorectal distension (CRD) stimulus and mustard oil enema. The rats were randomly divided into a normal group, model group, mild moxibustion group, electroacupuncture group, probiotic group and dicetel group. Abdominal withdrawal reflex (AWR) scores were determined within 90 min of the last treatment. The expression of GDNF/GFRα3 protein and mRNA in the colon and spinal cord were detected by immunohistochemistry and quantitative real-time-PCR, respectively. Results: The IBS model rats had significantly higher AWR scores than the normal group ( P<0.01). After mild moxibustion treatment, the AWR score was significantly reduced (20 mm Hg, P<0.05; 40 mm Hg, 60 mm Hg and 80 mm Hg, P<0.01). The model group showed significantly more colonic glial cell line-derived neurotrophic factor (GDNF/GFRα3 (GDNF family receptor α3) protein and mRNA expression in the colon and spinal cord than the normal group ( P<0.01). Compared with the model group, the expression of GDNF/GFRα3 protein and mRNA in the colon and spinal cord of the rats were significantly decreased in the mild moxibustion group (colon: GDNF and GFRα3 protein, P<0.01; GDNF and GFRα3 mRNA, P<0.01; spinal cord: GDNF and GFRα3 protein, P<0.01; GDNF mRNA, P<0.05, GFRα3 mRNA, P<0.01). Conclusions: Our data suggest that moxibustion therapy may mitigate CRD-induced increases in the expression of GDNF and its receptor GFRα3 in the colon and spinal cord in a rat model of IBS.

The objective of this study is to investigate if sinomenine hydrochloride (SIN-HCl) could be effective against adriamycin-induced renal fibrosis by regulating autophagy in a rat model. Forty male Sprague-Dawley (SD) rats were randomly divided into control group, model group, telmisartan group, and SIN-HCl group; rat model was induced by adriamycin; all rats were given intragastric administration for 6 weeks. Urine was collected from rats in metabolic cages to determine 24 h protein level. This was done after intragastric administration for the first two weeks and then once for every two weeks. Renal pathological changes were examined by the staining of HE, Masson, and PASM. Expressions and distributions of fibronectin (FN), laminin (LN), light chain 3 (LC3), and Beclin-1 were observed by immunohistochemistry. SIN-HCl ameliorates proteinuria, meanwhile attenuating the renal pathological changes in adriamycin-induced rats and also attenuating renal fibrosis and excessive autophagy by reducing the expression of FN, LN, LC3, and Beclin-1. SIN-HCl attenuates renal fibrosis by inhibiting excessive autophagy induced by adriamycin and upregulates the basal autophagy.

Objective: To evaluate the hyperglycemic effects of Kursi Ziyabet (KZ) tablets on the rat models of diabetes mellitus. Methods: In total, 58 male SD rats were assigned randomly to six groups. All except the normal group were transformed into experimental diabetes mellitus rat models by injecting streptozocin. The hyperglycemic effect and the mechanism of Ziyabet were evaluated by body weight, food and water intake, fasting blood sugar, and related parameters by measuring the oxidative stress-related factors and lipid metabolism indicator level by the corresponding kits using the rat experimental models. Results: Compared with the model group, body weight markedly increased after 3–6 weeks of intragastric administration of Ziyabet tablets ([Formula: see text]), while the water intake significantly decreased in the same period of time ([Formula: see text]). Food intake and fasting blood sugar level also decreased with the high dosage of Ziyabet tablets ([Formula: see text]). There is no significant difference in pancreas’ MDA content of the Ziyabet groups when compared to the model group ([Formula: see text]), while significant increase in SOD level was observed in high-dosage KZ group ([Formula: see text]). The blood serum insulin and free fatty acid level also decreased in the high-dosage KZ group compared with the model group ([Formula: see text]). Conclusion: We conclude that Ziyabet tablets demonstrated protective effects on the diabetic rat models.

Objectives: In this study, the effects of triiodothyronine (T3) on neurospheres isolated from SVZ of AD induced rats were examined. Methods: Eighteen male Wistar rats were classified into two groups: Sham (Sh) and STZ (Streptozotocin injected, 1.5 mg/kg in each lateral ventricle on days 1 and 3 after recovery). On day 21, the SVZ was extracted and neurospheres were cultured. T3 (50 nM) was added to the culture medium (STZ+T3 group) and then, the morphology and seladin-1 gene expression of neurospheres were evaluated. Results: The diameter and the number of neurospheres along with the gene expression of seladin-1 were significantly decreased in the STZ group compared to Sh group (P˂0.05) while the administration of T3 significantly (P˂0.05) increased all these parameters in the STZ group. Conclusion: STZ decreases the proliferation of stem cells extracted from SVZ and administration of T3 to the culture media improves the morphology and up-regulates the gene expression of seladin-1 of neurospheres.
