In vitro antimicrobial and antibiofilm activities of the Lippia alba essential oil and its major components (citral and carvone) against Staphylococcus aureus were investigated. Essential oils (LA1EO, LA2EO, and LA3EO) were extracted from the aerial parts of three L. alba specimens by hydrodistillation and analyzed by gas chromatography coupled to a mass spectrometer. Minimum Inhibitory Concentrations (MIC) and Minimum Bacterial Concentration (MBC) were determined by the microdilution method. For the antibiofilm assays, the biomass formation in the biofilm was evaluated by the microtiter-plate technique with the crystal violet (CV) assay and the viability of the bacterial cells was analyzed. All oils and their major components presented antibacterial activity, and the lowest MIC and MBC values were 0.5 mg mL−1 when LA1EO and citral were used. Potential inhibition (100%) of S. aureus biofilm formation at the concentration of 0.5 mg mL−1 of all EOs was observed. However, the elimination of biofilm cells was confirmed at concentrations of 1 mg mL−1, 2 mg mL−1, 2 mg mL−1, and 0.5 mg mL−1 for LA1EO, LA2EO, LA3EO, and citral, respectively. The results obtained in the present research point to the promising antibacterial and antibiofilm potential of L. alba EOs against S. aureus, a species of recognized clinical interest.

ABSTRACT The purpose of the present study was to determine the bioactive compounds in rosemary essential oil (REO) and tea tree essential oil (TEO) and to investigate their antibacterial and antibiofilm activities against Staphylococcus aureus and Escherichia coli in vitro. The MIC and MBC assays were performed to assess the antibacterial activity of these two EOs against S. aureus and E. coli with the broth microdilution method. A crystal violet assay was used to ascertain the effects of EOs on the biofilm formation of the test strains, and a tetrazolium bromide (MTT) assay was used to measure the level of inactivation of mature biofilms by EOs. Gas chromatography–mass spectrometry revealed 15 compounds in REO and 27 compounds in TEO, representing 97.78 and 98.13% of the total EO, respectively. Eucalyptol and α-pinene were found in high concentrations in REO, and the two major compounds in TEO were 4-terpineol and terpinolene. The MICs of REO for the two S. aureus and E. coli test strains were both 0.5 mg/mL, and the MICs of TEO for the two strains were both 0.25 mg/mL. Therefore, these EOs can significantly inhibit the formation of biofilms and induced morphological biofilm changes, as verified by scanning electron microscopy. Both EOs had destructive effects on the mature biofilm of the two test strains. TEO was more inhibitory than REO for biofilm formation by the two test strains. HIGHLIGHTS

A significant antibacterial effect of Galangal (Alpinia galangal) against foodborne bacteria Staphylococcus aureus, and Listeria monocytogenes, was reported with higher quantity of active biomolecule 1’-acetoxychavicolacetate. However, the effect on biofilm formation is yet unknown. Therefore, crude extract of Galangal rhizome was investigated for in-vitro antibiofilm activity against Staphylococcus aureus. Galangal extract was tested for antimicrobial activity against six strains of S. aureus including methicillin resisitant S. aureus. The antibiofilm activity was tested against S. aureus SA113 using microtiter-plate assay and scanning electron microscopic imaging. Galangal extract showed antimicrobial activity with an inhibition zone ranging from 36 - 46 mm against for all tested strains of S. aureus. The minimum inhibitory concentration and the minimum bactericidal concentration of the extract were 1.25 mg/ml and 5 mg/ml, respectively. More than 50% reduction of biofilm adhered to the surface was observed in 2.5 mg/ml representing minimum biofilm resistant concentration MBRC50 of the extract. The antibiofilm assay showed significant (80.68%) reduction at 20 mg/ml of extract. The 1´acetochavicol acetate (82.88%) was found to be the major chemical compound of galangal extract. Galangal rhizome extract possesses antibiofilm activity against S. aureus. Findings of the study could be useful for application of galangal crude extract as a disinfectant in food industry to eliminate the cross contaminations of S. aureus.

Staphylococcus aureus is a human pathogen that causes serious infections with high mortality rates. The antimicrobial activities of eight phenolic compounds (caffeic acid, chlorogenic acid, ferulic acid, morin, quercetin, isoquercitrin, rutin and hesperidin) were evaluated against planktonic and biofilm forming Staphylococcus aureus cells. Their ability to prevent biofilm formation via interference with bacterial cell adhesion and to reduce biomass of 24 h old biofilms has been addressed in this study. The antibacterial activities (MIC/MBC) were demonstrated using the microdilution method and the investigated compounds showed good activity against S. aureus isolates, with MICs in the range 0.05-0.4 mg/mL. Also, they exhibited promising antibiofilm potential in dose depended manner. Among all tested compounds, morin and quercetin showed the best antibiofilm activity at MIC values. The observed antimicrobial potential of the studied natural products can serve as a starting point towards development of novel plant-based therapeutics for the treatment of common infections such as the ones caused by S. aureus.

AbstractCombination of various compounds and essential oils for pharmaceutical formulations withdraw attention. In this present study, it was aimed to evaluate the in vitro potential synergistic antibacterial effect of Lavandula latifolia (spike lavender) essential oil with camphor by using the checkerboard method against the human pathogens; Staphylococcus aureus and Listeria monocytogenes. Pharmacopoeia quality L. latifolia essential oil and racemic camphor were analyzed and verified by GC-FID and GC/MS, simultaneously. In vitro antibacterial activity of essential oil and camphor (MIC range: 0.16–20 mg/mL) and standard antimicrobial clarithromycin (MIC range: 0.125–16 μg/mL) were carried out by broth microdilution against S. aureus and L. monocytogenes standard strains, respectively. Resulting antibacterial effects were evaluated for their fractional inhibitory concentrations (FICs) as antagonistic, additive and synergistic effects. The analytical results showed that the major component of essential oil was linalool (45.2%) and 1,8-cineole (25.6%). Antibacterial effects of essential oil were determined as MIC 1.25–5 mg/mL. As a result of the experiments, L. latifolia essential oil–camphor combinations were identified as “synergistic (FIC ≤ 0.5), and additive (0.5 < FIC ≤ 1)” in the respective combinations, suggesting further evaluation for formulations for potential antimicrobial applications in food and pharmaceuticals.

Ultra-high performance liquid chromatography/mass spectrometry showed soyasaponin I and the isoflavones daidzein, genistein, and glycitein to be the main components of the methanolic extract of the Korean soybean fermented product doenjang, which is known to be a rich source of naturally occurring bioactive substances, at average contents of 515.40, 236.30, 131.23, and 29.00 ng/mg, respectively. The antimicrobial activity of the methanolic extract of doenjang against nine Staphylococcusaureus strains was determined in vitro by the broth microdilution method to investigate its potential to serve as an alternative antibacterial compound. The results suggest that the extract is an effective antistaphylococcal agent at concentrations of 2048–4096 µg/mL. Moreover, the tested extract also showed the ability to inhibit the growth of both methicillin-sensitive and methicillin-resistant animal and clinical S. aureus isolates. The growth kinetics of the chosen strains of S. aureus at the minimum inhibitory concentration of the methanolic extract of doenjang support the idea that the tested extract acts as an antibacterial compound. To the best of our knowledge, this is the first report on the antistaphylococcal action of the methanolic extract of doenjang thus, additional studies including in vivo testing are necessary to confirm this hypothesis.

Abstract- The essential oil composition from the aerial parts of Ormenis africana (Asteraceae), an endemic species from Morocco, has been investigated by GC/MS. A total of 31 compounds were identified, representing 77%. After fractionation by column chromatography, the main compound was isolated and its structure elucidated by NMR spectroscopy. The essential oil was dominated by oxygenated compounds with spathulenol (45.8%) followed by camphor (7.1%), -cadinol (5.9%) and -bisabolol (5.9%) as the main compounds. This oil can be classified as spathulenol-type according to its spathulenol content. In vitro the antibacterial activity of the whole essential oil against three Gram positive (Bacillus cereus, Enterococcus faecalis, Streptococcus C) bacteria and three Gram negative (Proteus vulgaris, Escherichia coli, Pseudomonas aeroginosa) bacteria, showed significant results. The essential oil obtained from the leaves of Lippia alba (Mill.) N.E. Brown (Verbenaceae) has shown great pharmacological potential as an analgesic, antispasmodic, and antimicrobial agent. The aim of this study was to evaluate the modulatory effect of Lippia alba essential oil (LaEO I) on the activity of clinically used antimicrobial agents on Salmonella enterica serovar Typhi (Salmonella typhi) and Shigella dysenteriae biofilms. The Minimum Inhibitory Concentration of LaEO I (MICLaEO I) was determined by the microdilution method, and the effect of LaEO I on the activity of clinically used antimicrobials was assessed by the Checkboard method. The values obtained from MICLaEO I and ciprofloxacin were used to evaluate the effect of time of exposure on cell viability. LaEO I main components were geranial (34.2%), neral (25.9%), and myrcene (12.5%). The MICLaEO I was 1 mg/mL for both strains. LaEO I positively modulated the action of ciprofloxacin, cefepime, and ceftriaxone. After the first hour of treatment with MICLaEO I, the cell viability of the strains showed a 5 log10 CFU/mL reduction, and the LaEO I-CIP association was able to inhibit growth during the first 6 h of the test. Regarding the anti-biofilm activity, MICLaEO I was able to reduce the biofilm mass of Salmonella typhi by 61.2% and of Shigella dysenteriae by 38.9%. MICLaEO I was not able to eradicate the preformed biofilm; however, there was a reduction in the biofilm microbial viability. LaEO I has the potential to be used as an antimicrobial agent and interferes with biofilm formation; also, it is able to reduce cell viability in preformed biofilm and synergistically modulate the activity of ciprofloxacin.

Dental plaque prevention can be achieved by inhibition of mouth cavity microbes to built biofilm. Kaempferia galanga rhizome has been known as a potential antibacterial agent. This research aimed to reveal the potency of Kaempferia galanga extract and essential oil as anti plaque active agents, based on their in vitro inhibitory activity against the planktonic growth and biofilm of Streptococcus mutans ATCC 21752. Kaempferia galanga extract was obtained by defatting dried-pulverized samples in petroleum ether prior to immersion in 70% ethanol. The fresh rhizome was steam-hydro distilled for 6 h to yield the essential oil. Antibacterial and anti biofilm assays were measured by micro dilution technique on polystyrene 96-wells micro titer plates at 37°C. The percentage of inhibition was calculated by comparing the absorbance of samples to the vehicle (control) measured by micro plate reader at 595 nm. Biofilms formed were first stained by 1% crystal violet. The above assays were performed in triplicates. This study revealed that both K. galanga rhizome essential oil and ethanolic extract showed antibacterial and antibiofilm activity towards S. mutans. The ethanol extract showed MIC90 value at 0.091% w/v and MBC at 2.724% w/v for antibacterial activity; IC50 at 0.048 % w/v for anti biofilm formation activity; and EC50 at 0.052%w/v for biofilm degradation activity. Until the highest concentration tested (0.6%w/v), the MIC90 and MBC values of the essential oil were not revealed, but higher biofilm inhibitory activity i.e. IC50 at 0.025 % w/v; and EC50 at 0.034 %w/v were observed. Key words: biofilm inhibitor, antibacterial, Kaempferia galanga

The aim of present study was, to investigate the essential oil of Craniotome furcata for its antimicrobial and antioxidant activities. The essential oil was obtained by steam distillation of the flowering aerial parts of C. furcata. The oil was tested for its antimicrobial activity in vitro against four Gram-positive, four Gram-negative bacteria and three fungi, by using disk diffusion and tube dilution methods. The antioxidant potential of the essential oil was investigated by two different models DPPH free radical and ABTS free radical cation scavenging. The essential oil was effective against all the tested microorganisms, with minimum inhibitory concentrations (MICs) ranged from 4.11±1.44 to 0.30±0.10 mg/ml. The oil showed antioxidant activity in DPPH free radical and ABTS free radical cation scavenging model. The results of this study revealed that the essential oil of C. furcata possesses antimicrobial and antioxidant activities.
