Twenty-four chronic alcoholics admitted to hospital for detoxification after a drinking spree were examined by upper gastrointestinal endoscopy. Biopsy specimens were taken from corpus/fundus, antrum and duodenum for tissue histology (eosin stain). From the duodenum villus index and ultrastructure (scanning electron microscopy, SEM) were also performed. As a control group 12 subjectively healthy non-alcoholics referred to upper gastrointestinal endoscopy mainly for dyspepsia were chosen.Gastrointestinal symptoms were common in alcoholics (88%). Endoscopic and histological gastroduodenitis were not more common in the alcohol group. There was no correlation between gastrointestinal symptoms and endoscopic or histological gastroduodenitis in both groups. In the duodenum, 50% of the alcoholics and 82% in the control group had alterations by scanning electron microscopy. Ten of the 11 alcoholics with an abnormal ultrastructure had diarrhoea. In the control group dyspepsia (ulcus suspect) was correlated to a pathological SEM.

This in vitro study evaluated the effects of dentifrices containing nano-hydroxyapatite (n-HAp) on dentinal tubule occlusion and on mineral deposition. Dentin specimens of ten human teeth were submersed for 30 s in 40% citric acid and then randomly divided into four groups (three study groups and one control group). In the study groups, the dentin samples were exposed to three different n-HAp toothpastes: Karex (Dr. Kurt Wolff GmbH & Co. KG, Bielefeld, Germany), Biorepair Plus Sensitive (Coswell SpA, Bologna, Italy), and Dr. Wolff’s Biorepair (Dr. Kurt Wolff GmbH & Co. KG, Bielefeld, Germany); in the control group no toothpaste was applied. All of the samples were evaluated using scanning electron microscopy (SEM) and energy-dispersive X-ray (EDX) analysis. In the control group all of the samples showed a frank and wide opening of the dentinal tubules, whereas in the study groups different degrees of tubule closure by mineral depositions were observed. Toothpastes containing n-HAp determined a significant occlusion of dentinal tubules and a significant increase of mineral deposition on the dentin surface. All three tested toothpastes showed similar results regarding the degree of dentinal tubule closure. Varying degrees of differences in calcium, phosphate, carbon, and oxygen ion concentrations among the three tested toothpastes were obtained.

The purpose of this work was to verifyin vitroadherence ofE. corrodensandS. oralisto the surface of tongue piercings made of surgical steel, titanium, Bioplast, and Teflon. For this, 160 piercings were used for the count of Colony Forming Units (CFU) and 32 piercings for analysis under scanning electron microscopy. Of these, 96 (24 of each type) were individually incubated in 5 mL of BHI broth and 50 μL of inoculum at 37°C/24 h. The other 96 piercings formed the control group and were individually incubated in 5 mL of BHI broth at 37°C/24 h. Plates were incubated at 37°C/48 h for counting of CFU/mL and data were submitted to statistical analysis (pvalue<0.05). ForE. corrodens, difference among types of material was observed (p<0.001) and titanium and surgical steel showed lower bacterial adherence. The adherence ofS. oralisdiffered among piercings, showing lower colonization (p<0.007) in titanium and surgical steel piercings. The four types of piercings were susceptible to colonization byE. corrodensandS. oralis, and bacterial adhesion was more significant in those made of Bioplast and Teflon. The piercings presented bacterial colonies on their surface, being higher in plastic piercings probably due to their uneven and rough surface.

Objective. To study the effect of curcumol on liver sinusoidal endothelial cells (LSECs) and to analyze the mechanism of antihepatic fibrosis. Methods. The effects of drug intervention on cell proliferation rates were detected by MTT assay. The expression of NF-κB was detected by RT-PCR and WB. The NF-κB expression and entry into the nucleus were detected by immunofluorescence; scanning electron microscopy was used to observe the changes of LSECs fenestrae. Results. MTT results showed that the interference of cell proliferation in each group was small. RT-PCR showed that the expression of NF-κB in the curcumol intervention group was significantly lower than that in the positive control group (P<0.05). The WB detection found that, in the curcumol intervention group, the expression of pNF-κB in the NF-κB signaling pathway was significantly lower than that in the positive control group (P<0.05). Scanning electron microscopy showed that the LSEC fenestrae were significantly improved compared with the positive control group. Conclusion. Curcumol may be one of the mechanisms of antihepatic fibrosis by inhibiting the activity of the NF-κB signaling pathway and increasing the fenestrae of LSECs.

The current study investigates the performance of concrete incorporating ground granulated blast-furnace (GGBF) slag in the presence of colloidal nano-silica. A control group of concrete mixtures is compared with a group of mixtures with 50% slag replacement, with each group examined at two different ratios of colloidal nano-silica (3% and 6% of the total cementitious material). Subsequently, the relative performance of the two groups is compared with ordinary Portland cement concrete in relation to strength and durability properties. Evaluation included experimental examination of compressive and tensile strength, rapid chloride penetration, and porosimetry using mercury intrusion tests. Furthermore, the microstructure of the cementitious matrix was evaluated using scanning electron microscopy imaging. Results of the tested concrete mixtures indicated that nano-silica particles can improve the properties of concrete containing GGBF slag. Improvement in high early strength as well as reduction in permeability are observed. Furthermore, nano-silica caused a refinement of the pore structure and an improvement to the interfacial transition zone (ITZ) as seen through mercury intrusion porosimetry (MIP) results and scanning electron microscopy imaging, respectively.

Objectives: We assess the association between the presence of biofilms and cilial damage in patients with chronic rhinosinusitis (CRS), describe the microorganisms associated with samples that exhibited cilial loss and biofilms, and demonstrate the absence of ciliary injury and biofilms in similarly prepared “normal” controls. Methods: We examined samples of ethmoid mucosa obtained from 24 patients who underwent functional endoscopic sinus surgery for CRS. Samples from a control group (20 healthy subjects) were also examined. The specimens were divided into 2 fragments; the first was processed for bacterial cultures, and the second was subjected to scanning electron microscopy. Statistical analysis was performed. Results: All CRS samples had positive bacterial cultures. The scanning electron microscopy analysis showed bacterial biofilms in 10 of the 24 specimens. A marked destruction of the epithelium was observed in samples positive for biofilms (p < 0.001), and the presence of Haemophilus influenzae was associated with ciliary abnormalities (partial damage in 55.6% and absence of cilia in 50%; p = 0.041). Conclusions: The high percentage of biofilms in our specimens confirms the association between biofilms and CRS. Our data support the hypothesis that biofilm formation represents the latter phase of an inflammatory process that leads to complete epithelial destruction.

This study aimed at evaluating the use of scanning electron microscopy in the study of the post-vaccinal respiratory reaction of the tracheal epithelium of quails (Coturnix coturnix japonica) immunized against Newcastle disease. A number of 36 quails were distributed into four groups: T1 <FONT FACE=Symbol>¾</FONT> control birds (non-vaccinated); T2 <FONT FACE=Symbol>¾</FONT> birds vaccinated with Ulster 2C strain; t3 <FONT FACE=Symbol>¾</FONT> birds vaccinated with B1 strain; t4 <FONT FACE=Symbol>¾</FONT> birds vaccinated with LaSota strain. Regardless the experimental group, birds did not show detectable clinical signs of post-vaccinal respiratory reaction. However, the analysis of tracheal fragments by scanning electron microscopy showed that birds vaccinated with B1 and LaSota strains developed epithelial sloughing of the trachea, whereas those vaccinated Ulster 2C strain did not develop this change, demonstrating intact tracheal epithelium, similar to the control group.

The bond strength by three point bending strength of two metal substrates (commercially pure titanium or grade II, and Ti-6Al-4V alloy or grade V) combined to three distinct low-fusing ceramic systems (LFC) and the nature of porcelain-metal fracture by scanning electron microscopy (SEM) were evaluated. The results were compared to a combination of palladium-silver (Pd-Ag) alloy and conventional porcelain (Duceram VMK68). Sixty metal strips measuring 25x3x0.5mm were made - 30 of titanium grade II and 30 of titanium grade V, with application of the following types of porcelain: Vita Titankeramik, Triceram or Duceratin (10 specimens for each porcelain). The porcelains were bonded to the strips with dimensions limited to 8x3x1mm. The control group consisted of ten specimens Pd-Ag alloy/Duceram VMK68 porcelain. Statistical analyses were made by one-way analysis of variance (ANOVA) and Tukey test at 5% significance level. Results showed that the bond strength in control group (48.0MPa ± 4.0) was significantly higher than the Ti grade II (26.7MPa ± 4.1) and Ti grade V (25.2MPa ± 2.2) combinations. When Duceratin porcelain was applied in both substrates, Ti grade II and Ti grade V, the results were significantly lower than in Ti grade II/Vitatitankeramik. SEM analysis indicated a predominance of adhesive fractures for the groups Ti grade II and Ti grade V, and cohesive fracture for control group Pd-Ag/Duceram. Control group showed the best bond strength compared to the groups that employed LFC. Among LFC, the worst results were obtained when Duceratin porcelain was used in both substrates. SEM confirmed the results of three point bending strength.

AbstractOur aim was to test the hypothesis that the use of mouthwashes, consumption of soft drinks, as well as the type of light curing unit (LCU), would change the surface roughness (Ra) and morphology of a nanofilled composite resin (Z350® 3M ESPE). Samples (80) were divided into eight groups: Halogen LCU, group 1, saliva (control); group 2, Pepsi Twist®; group 3, Listerine®; group 4, Colgate Plax®; LED LCU, group 5, saliva; group 6, Pepsi Twist®; group 7, Listerine®; group 8, Colgate Plax®. Ra values were measured at baseline, and after 7 and 14 days. One specimen of each group was prepared for scanning electron microscopy analysis after 14 days. The data were subjected to multifactor analysis of variance at a 95% confidence followed by Tukey's honestly significant difference post-hoc test. All the treatments resulted in morphological changes in composite resin surface, and the most significant change was in Pepsi Twist® groups. The samples of G6 had the greatest increase in Ra. The immersion of nanofilled resin in mouthwashes with alcohol and soft drink increases the surface roughness. Polymerization by halogen LCU (reduced light intensity) associated with alcohol contained mouthwash resulted in significant roughness on the composite.

Dall-Miles cables are widely used for trochanteric re-attachment in hip arthroplasty, but their effects on the cement mantle have not been reported. We have carried out an ex vivo study to investigate the influence of cables on the mantle. Charnley femoral components were implanted in eight proximal human femora using Palacos cement. Wires (control group, n=4) or 2mm stainless steel Dall-Miles cables (n=4) were passed through holes drilled in the proximal femur before the cement was introduced. The wires or cables were tightened using the appropriate instruments after the cement had hardened. The specimens were sectioned using a diamond saw, and examined by scanning electron microscopy and light microscopy before and after staining with penetrant dye. Deformation of the strands at the tension edge of each cable, with debonding from the cement in two specimens, was observed. There was no damage to cement at the compression edge. Tensioning of Dall-Miles cables passing through Palacos cement does not appear to damage the mantle.
