The Eugenia genus is rich in bioactive substances with biological potential. Mollicutes are bacteria without cell walls, which are responsible for various human and animal diseases. The aim of this study was to evaluate the chemical composition and antimollicute activity of Eugenia mattosii. Leaves and stems were extracted with methanol, partitioned into fractions of different polarities, and submitted to column chromatography in order to isolate major compounds. Identification and quantification of isolated substances in extracts and fractions were performed by UPLC-PDA/ESI+ - QTOF. Methanolic extracts, ethyl acetate and chloroform fractions and isolated substances were screened for antimycoplasmic activity against five Mycoplasma strains. The methanolic extracts of leaves and stems showed moderate antimycoplasmic activity (MICs = 250-500 μg.mL-1). The fractions exhibited better effect, with MICs = 125-1000 μg.mL-1, especially the ethyl acetate fraction of the leaves, which presented MICs of 125-250 μg.mL-1 for all strains tested. Phytochemical analyses evidenced the presence of some phenolic compounds, including pinostrobin, cryptostrobin and catechin, the first showing promising antimycoplasmic activity. Quantification of the compounds demonstrated higher concentrations of pinostrobin and cryptostrobin in the chloroform fraction. In conclusion, E. mattosii presented antimycoplasmic activity related, at least in part, to the presence of pinostrobin.

Abstract. In the current studies, crude and subsequent fractions of Aconitum violaceum aerial parts were screened for their toxicity, antimicrobial effects as well as antioxidant potential. Phytochemically, the plant is enriched in alkaloids alongside anthraquinones (present in ethyl acetate fraction (AV3) and saponins (detected in chloroform fraction (AV2). In Brine shrimp lethality assay, AV3 was the most potent (59 %) in killing Artemia naupili larvae at a dose of 1000 µg/mL. AV3 exhibited strongest antimicrobial effect against the bacteria E. coli (80 %) and S. flexneri (76 %) as well as against the tested fungi, A. niger (86 %) respectively. However, chloroform fraction (AV2) was the most effective (almost 90 %) antifungal against A. niger and C. albicans. Overall, strong antioxidant activity was observed for AV3 with IC50 values of 120.04 ± 0.4 µg/mL (65.4 ± 0.01 standard) in DPPH and 125.1 ± 0.3 µg/mL (2.0 ± 0.03 standard) in ABTS free radical assays. AV3 showed promising inhibition against acetylcholinesterase (AChE) 70 ± 0.8 % and butyrylcholinesterase (BChE) 57 ± 0.2 % at dose of 100 µg/mL thus confirming a potent invitro cholinesterase inhibitory effect. The overall results indicated strong biological potential of ethyl acetate fraction obtained form A. violaceum and a possible new therapeutic source could be formulated from its pure isolates.   Resumen.

Antioxidants are compounds that can inhibit oxidation reactions by binding to free radicals and highly reactive molecules. One form of reactive oxygen compounds is free radicals, these compounds are formed in the body and are triggered by various factors. Noni fruits have been used by the community as a medicine for high blood pressure, beriberi, urinating, inflammation of bile, inflammation of the intestine, dysentery, constipation, spleen pain, liver pain, diabetes and lumbago. Nevertheless, research on the identification of the active compounds of noni fruit which act as antioxidants yet. This study aims to extract and fractionation of Noni fruit, determine the antioxidant activity of Noni fruit extract, and identify the types of bioactive compounds that act as antioxidants. Noni fruit was extracted used maceration technique with 96% ethanol and then fractionated using n-hexane, chloroform and ethyl acetate solvents. Antioxidant activity from the results of noni fruit fractionation was measured by the DPPH (1.1 diphenyl-2-picrylhydrachyl) method and identification of bioactive compounds was carried out by GCMS. The results showed the greatest antioxidant activity was found in chloroform fraction with a percentage of inhibition of 78.19%. For the first time this study revealed the types of bioactive compounds from noni fruit ethyl acetate fraction which act as antioxidants are n-hexadecanoic acid, squalene, pyridin-3-carboxamide, oxime, n- (2-trifluoro methyl phenyl), and beta-sitosterol

AbstractThe phytochemical and antimicrobial activity of the petroleum ether and crude methanol extracts, chloroform and ethyl acetate fractions of the leaves of Lannea kerstingii were investigated. Phytochemical screening revealed the presence of steroids and triterpenes in the petroleum ether extract, steroid, triterpene, flavonoids and tannins in both crude methanol extract and chloroform fraction while the ethyl acetate fraction contained only flavonoids and tannins. The extracts exhibited antimicrobial activities with zones of inhibition ranging from 17.00 to 21.03, 20.10 to 25.24, 25.32 to 34.02 and 22.28 to 27.20 mm for petroleum ether extract, methanol extract, chloroform and ethyl acetate fractions respectively. The minimum inhibitory concentration was between 5 and 10mg/ml, 5mg/ml for the petroleum ether and methanol extract respectively, and between 2.5 and 5 mg/ml, 5mg/ml for the acetate fractions. The minimum bactericidal concentration for all the extracts was 40mg/ml respectively except for chloroform fraction which ranged from 20 to 40mg/ml. The minimum fungicidal concentration for all the extracts was found to be 40mg/ml respectively. This result indicates the broad spectrum antimicrobial potential of L. Kerstingii and justifies the use of this plant in traditional medicine.

The antibacterial activity of methanol extract from the root bark of Akanda (Calotropis gigantea L.) and its petroleum ether, chloroform and ethyl acetate fractions were investigated. Both of methanol extract and its chloroform fraction showed activity against Sarcina lutea, Bacillus megaterium and Pseudomonas aeruginosa. Petroleum ether fraction showed activity against Bacillus subtilis and Shigella sonnei whereas ethyl acetate fraction showed activity against Pseudomonas aeruginosa and Escherichia coli at 20ìg/disc, 30ìg/disc and 40ìg/disc doses. Among the tested materials, methanol extract and its chloroform fraction showed comparatively better results. Minimum inhibitory concentration (MIC) for methanol extract and each fraction were also determined by serial dilution technique. Abstract Background Diabetes mellitus is a metabolic disorder of glucose metabolism and management of blood glucose level is the hallmark in the treatment of this disease. The present study investigated chemical composition, in vitro antioxidant and antidiabetic activity of different fractions of 80% methanol Piper guineense leaves extract. Materials and methods The crude methanolic extract of P. guineense was obtained following 80% methanol cold extraction and was successively partitioned with dichloromethane (DCM), ethyl acetate (EtOAc), n-butanol (nBuOH) and aqueous solvents to give four fractions. The chemical composition of the fractions from P. guineense was determined using gas chromatography coupled with mass spectrometry (GC-MS) and their potentials as antioxidant and anti-diabetes were evaluated. Results The percentage yields were 3.16, 2.22, 0.68 and 0.66% (w/w) in n-butanol, DCM, aqueous and ethyl acetate fractions of P. guineense methanolic extract, respectively. The GC-MS analyses identified a total of 71 and 34 phytochemicals in n-butanol and ethyl acetate fractions respectively. Tributyl acetylcitrate (10.95%) and phytol (9.11%) were the major components in the n-butanol fraction while ethyl piperonyl cyanoacetate (27.35%) and phytol (15.17%) were the major constituents in the ethyl acetate fraction. Ethyl acetate fraction had the highest ferric reducing antioxidant power with a value of 53.96 ± 0.40 mgAAE/g while n-butanol fraction possessed highest total antioxidant power (9.98 ± 0.15) followed by aqueous fraction (9.72 ± 0.02). The ethyl acetate and n-butanol fractions with IC50 value of 0.24 ± 0.07 and 0.83 ± 0.15 μg/mL respectively elicited significant inhibitory activities against α-glucosidase while only n-butanol fraction (IC50 = 0.33 ± 0.09 μg/ml) exhibited appreciable inhibition against α-amylase activity. However, none of the four fractions showed significant inhibitory activity towards dipeptidyl-peptidase-IV. Conclusion n-butanol and ethyl acetate fractions of 80% methanol P. guineense leaves extract can be a potential source of bioactive compounds of pharmacological importance in the management of diabetes.

Introduction: Streptococcus sanguis is an initial cause of dental plaque formation which is the initial cause of caries. One of the preventive treatments can be done by using the mouthwash containing antibacterial substances. Along time, natural remedies are proven to be having more antibacterial properties. Ant-plant (Myrmecodia pendens Merr. & Perry) tubers are types of epiphytic plant that grows in many parts of Papua, with many health benefits, and are known to contain flavonoids, tannins, and tocopherols. This study was aimed to determine the antibacterial activity of ethyl acetate fraction from methanolic extracts of ant-plant tubers on the growth of Streptococcus sanguis ATCC 10566. Methods: The ant-plant tubers were extracted with soxhletation method using the methanol solvent. The fractionation was then performed using ethyl acetate to obtain the ethyl acetate fraction. Result: The phytochemical test showed that the ethyl acetate fraction of the ant-plant contained phenolics, tannins, flavonoids, and terpenoids. The bacterial test in this study was using the microdilution method using the ELISA Reader by measuring the Minimum Inhibitory Concentration (MIC) of ethyl acetate fraction of ant-plant tubers towards Streptococcus sanguis with positive control was using the chlorhexidine gluconate as the golden standard medication. The ethyl acetate fraction of ant-plant tuber had an antibacterial effect towards Streptococcus sanguis in the MIC of 31.25 ppm, while chlorhexidine gluconate was 0.49 ppm. Conclusion: The ethyl acetate fraction of ant-plant had a lower antibacterial activity compared to chlorhexidine gluconate on the growth of Streptococcus sanguis ATCC 10566.Keywords: Ant-plant tubers, Myrmecodia pendens Merr. & Perry, antibacterial activity, Streptococcus sanguis ATCC 10566.

Background. The leaves of Hagenia abyssinica have been used in the management of diabetes mellitus in Ethiopian folk medicine. Thus, this study is aimed at investigating the in vitro α-amylase and α-glucosidase inhibitory and antioxidant activities of the crude extract and solvent fractions of H. abyssinica leaves. Methods. The in vitro α-amylase and α-glucosidase inhibitory and antioxidant activities of the plant extract were assessed using 3,5-dinitrosalicylic acid (DNSA), p-nitro-phenyl-a-D glucopyranoside (p-NPG), and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays, respectively. Each value of percent inhibition of α-amylase, α-glucosidase, and DPPH scavenging effect was presented as means ± SEM ( n = 3 ). Results. The α-amylase inhibitory activity of the crude extract and solvent fractions was found to be concentration-dependent. The strongest activity was exhibited by the crude extract at the highest concentration with a percentage inhibition of 74.52% (IC50, 14.52 μg/ml) followed by water fraction 68.24% (IC50, 16.31 μg/ml), ethyl acetate fraction 61.57% (IC50, 18.73 μg/ml), and chloroform fraction 56.87% (IC50, 21.57 μg/ml) of H. abyssinica leaves. In the α-glucosidase inhibition assay, the maximum activity was exhibited by the aqueous fraction 62.54% (IC50, 11.67 μg/ml) followed by ethyl acetate fraction 54.97% (IC50, 15.89 μg/ml), crude extract 46.79% (IC50, >16.5 μg/ml), and chloroform fraction 36.44% (IC50, >16.5 μg/ml). In the antioxidant assay, the crude extract exhibited the highest antioxidant activity 86.36% (IC50, 10.25 μg/ml) followed by water fraction 78.59% (IC50, 13.86 μg/ml), ethyl acetate fraction 71.58% (IC50, 16.34 μg/ml), and chloroform fraction 63.65% (IC50, 18.83 μg/ml). Conclusion. This study has revealed that H. abyssinica leaves possess noticeable in vitro α-amylase and α-glucosidase inhibitory and antioxidant activities.
