Abstract Background Drugs currently used for controlling onchocerciasis and lymphatic filariasis (LF) are mainly microfilaricidal, with minimal or no effect on the adult worms. For efficient management of these diseases, it is necessary to search for new drugs with macrofilaricidal activities that can be used singly or in combination with existing ones. Daniellia oliveri and Psorospermum febrifugum are two plants commonly used in the local management of these infections in Bambui, a township in the North West Region of Cameroon, but there is currently no documented scientific evidence to support their claimed anthelmintic efficacy and safety. The aim of this study was to provide evidence in support of the search for means to eliminate these diseases by screening extracts and chromatographic fractions isolated from these plants for efficacy against the parasitic roundworms Onchocerca ochengi and Brugia pahangi. Methods The viability of O. ochengi adult worms was assessed using the MTT/formazan assay. Fully confluent monkey kidney epithelial cells (LLC-MK2) served as the feeder layer for the O. ochengi microfilariae (mfs) assays. Viability of the mfs was assessed by microscopic examination for mean motility scoring (relative to the negative control) every 24 h post addition of an extract. The Worminator system was used to test the effects of the extracts on adult B. pahangi motility, and mean motility units were determined for each worm. Cytotoxicity of the active extracts on N27 cells was assessed using the MTS assay. Results Extracts from D. oliveri and P. febrifugum were effective against the adult roundworms O. ochengi and B. pahangi. Interestingly, extracts showing macrofilaricidal activities against O. ochengi also showed activity against O. ochengi mfs. The hexane stem bark extract of D. oliveri (DOBHEX) was more selective for adult O. ochengi than for mfs, with a half maximal and 100% inhibitory concentration (IC50 and IC100, respectively) against adult O. ochengi of 13.9 and 31.3 μg/ml, respectively. The in vitro cytotoxicity of all active extracts on N27 cells showed selective toxicity for parasites (selectivity index > 1). Bioassay-guided fractionation of the extracts yielded fractions with activity against adult B. pahangi, thus confirming the presence of bioactive principles in the plant extracts. Conclusions Our study supports the use of D. oliveri and P. febrifugum in the traditional treatment of onchocerciasis and LF. The further purification of active extracts from these plants could yield lead compounds for filarial drug discovery and development. Graphic abstract

Telah dilakukan penelitian secara kualitatif terhadap pengaruh antibakteri dari ekstrak kulit batang matoa (Pometia pinnata. Spp.) terhadap bakteri Gram positif Staphylococcus aureus . Sebelum dilakukan uji antibakteri, sampel yang telah diekstrak secara maserasi diuji fitokimia terlebih dahulu untuk menentukan kandungan metabolit sekunder yang telah diketahui berperan sebagai agen antibakteri. Setelah dilakukan uji fitokimia, ekstrak diuji aktivitas antibakterinya terhadap bakteri Staphylococcus aureus dengan menggunakan teknik difusi agar dengan cara sumuran. Ekstrak dilarutkan pada aquades steril dengan perbandingan 2 g ekstrak pada 2 mL air. Untuk kontrol positif, digunakan ciprofloxacin dan aquades steril sebagai kontrol negatif. Hasil penelitian ini menunjukkan bahwa ekstrak kulit batang matoa memiliki aktivitas antibakteri setelah diinkubasi selama 24 jam pada 37 OC. Dari 3 kali pengulangan dengan masing – masing 3 sumuran, didapat zona hambat masing – masing 16.84 mm, 12.5 mm dan 14.5 mm dengan kontrol positif 29.67 mm serta kontrol negatif 0 mm. Hasil yang diperoleh didukung oleh keberadaan metabolit sekunder hasil uji fitokimia yaitu tanin, flavonoid, terpenoid dan saponin.A qualitative study has done  of the antibacterial effect of matoa (Pometia pinnata) stem bark extract against Gram-positive bacteria Staphylococcus aureus . Before the antibacterial test , samples were extracted by maceration and then  phytochemical  tested to measuring the content of secondary metabolites  that have been known to act as an antibacterial agent . After being tested of phytochemical , extracts were tested the antibacterial effect  against Staphylococcus aureus using agar diffusion technique. Extract was dissolved in sterile distilled water with a ratio of 2 g of extract in 2 mL of water . For the positive control , use of ciprofloxacin and sterile distilled water as a negative control . Results of this study indicate that matoa bark extract has antibacterial effect after incubation for 24 h at 37OC . Of 3 times with each repetition - each 3 wells, the inhibition zone obtained - each 16.84 mm , 12.5 mm and 14.5 mm with 29.67 mm of positive control and a negative control by 0 mm . The results are supported by the presence of secondary metabolites by phytochemical test such as tannins , flavonoids , terpenoids and saponins.

Background. In Ethiopia, stem bark of Periploca linearifolia is used for the treatment of malaria by the local community and demonstrated antimalarial activity in vitro. Despite its in vitro antimalarial activity, no scientific study has been carried out to verify its activity in vivo. Therefore, the aim of the study was to evaluate the antimalarial activity of Periploca linearifolia stem bark extract in mice. Methods. The dried stem bark of Periploca linearifolia was extracted with 80% methanol and evaluated for its antimalarial activity on both early and established Plasmodium berghei infected mice. The extract was prepared at graded doses of 200, 400, and 600 mg/kg. Chloroquine and distilled water were administered to the positive and negative control groups, respectively. Results. The crude extract, at all tested doses, suppressed parasitemia significantly (p<0.05) for 200 and 400 mg/kg and (p<0.001) for 600 mg/kg. The suppression values at these doses were 56.98, 43.33, and 38.17 percent, respectively. Periploca linearifolia extract also demonstrated schizonticidal activity in the established malaria infection. Conclusion. The plant Periploca linearifolia has a promising antimalarial activity in mice, supporting its in vitro finding. Thus, it could be considered as a potential source to develop new antimalarial agent.

Garcinia husor is one of the folk medicines in Maluku-Indonesia. This species has been used for the treatmet of Malaria disease. The phytochemical contents and antiplasmodial activity not reported yet. In this study we evaluated the quantitative phytochemicals, in vitro and in vivo antiplasmodial activity of stem bark ethyl acetate extract. In vitro assay was done using P. falciparum 3D7 strain sensitive of chloroquine. For in vivo analysis, four groups of M. musculus were infected by P. berghei and their parasitemia levels were for 7 days of treatment with ethyl acetate extract; hematological and biochemical parameter were analyzed at the end of experiment. The result showed ethyl acetate extract with the TPC (169.47 mg GAE/100 g ±0.61) and TPC (167.37 mg QE/100 g ±1.05) was active against P. falciparum 3D7 strain (IC50 value of 0.31±0.43 μg/ml). The animal treated with extract showed suppression of parasitemia to 87.57±1.41% compared with the P. berghei infected-mice (negative control), ED50 value of 22.30 mg/kg BW. The dose of extract in 200 mg/kg BW was reduce parasitemia of infected mice with P. berghei more potential. The ethyl acetate of the stem bark G. husor with has antiplasmodial properties and future investigation are necessary to elucidate its mechanism of action.

This study was designed to determine the effect of Carica papaya L. stem bark extracts on cholesterol concentration in rats induced with glibenclamide. A completely randomized design was used for the experiment which consisted of 6 treatment groups, each group consisted of four rats, as follows:1) KN (negative control, non-diabetic rats); KP, diabetic rats given glibenclamide 10 mg/kg body weight; EP 1, diabetic rats given 0 mg/kg body weight/day extract; EP2, diabetic rats given 100 mg/kg body weight/day extract; and EP3, diabetic rats given 200 mg/kg body weight/day extract, EP4, diabetic rats given 300 mg/kg body weight/day extract for 28 day. The results showed that C. papaya L. stem bark extract decreased (P<0.05) cholesterol levels in diabetic rats. It was concluded that C. papaya L. stem bark extract had potential as anti-hypercholesterolemic in diabetic rats.

Foeniculum vulgareMill commonly called fennel has been used in traditional medicine for a wide range of ailments related to digestive, endocrine, reproductive, and respiratory systems. Additionally, it is also used as a galactagogue agent for lactating mothers. The review aims to gather the fragmented information available in the literature regarding morphology, ethnomedicinal applications, phytochemistry, pharmacology, and toxicology ofFoeniculum vulgare. It also compiles available scientific evidence for the ethnobotanical claims and to identify gaps required to be filled by future research. Findings based on their traditional uses and scientific evaluation indicates thatFoeniculum vulgareremains to be the most widely used herbal plant. It has been used for more than forty types of disorders. Phytochemical studies have shown the presence of numerous valuable compounds, such as volatile compounds, flavonoids, phenolic compounds, fatty acids, and amino acids. Compiled data indicate their efficacy in severalin vitroandin vivopharmacological properties such as antimicrobial, antiviral, anti-inflammatory, antimutagenic, antinociceptive, antipyretic, antispasmodic, antithrombotic, apoptotic, cardiovascular, chemomodulatory, antitumor, hepatoprotective, hypoglycemic, hypolipidemic, and memory enhancing property.Foeniculum vulgarehas emerged as a good source of traditional medicine and it provides a noteworthy basis in pharmaceutical biology for the development/formulation of new drugs and future clinical uses.

Plant phenolic extracts are widely recognized as an important source of natural antioxidant substances and potential compounds for cosmetic formulations. This study aimed to evaluate the chemical profile, photoprotective and antioxidant activities of stem bark extract of Spondias purpurea L. (ciriguela) and its application in photoprotective formulations. Thirty phenolic constituents were annotated by ultra-performance liquid chromatography coupled with an electrospray ionization quadrupole time-of-flight mass spectrometry in mode negative (UPLC‑QTOF-MS2). The stem bark extract antioxidant and chelation potential, expressed in half maximal inhibitory concentration (IC50), showed 6.25 and 352.22 μg mL−1, respectively. The phenolic extract was used as an active ingredient in six sunscreen formulations, with concentrations ranging from 0.2 to 10%. The ultraviolet (UV) protection properties of the formulations were evaluated by sun protection factor (SPF) values obtained in 0.2 mg mL−1 (0.495 to 2.27) and 2.0 mg mL−1 (2.29 to 15.87). The SPF value for the extract (14.37 and 26.16) was high, but there was a reduction in the base formulation. However, these results suggested that the bioactive extracted of stem bark of Spondias purpurea L. has interesting potential to reduce the damage caused by UV radiation and may be utilized as an active ingredient in a sunscreen formulation.

Abstract Background Bacteria belonging to the Salmonella genus are major concern for health, as they are widely reported in many cases of food poisoning. The use of antibiotics remains a main stream control strategy for avian salmonellosis as well as typhoid and paratyphoid fevers in humans. Due to the growing awareness about drug resistance and toxicities, the use of antibiotics is being discouraged in many countries whilst advocating potent benign alternatives such as phyto-based medicine. The objective of this work was to isolate, characterise the bioactive compounds of Canarium schweinfurthii; and evaluate their anti-salmonellal activity. Methods The hydro-ethanolic extract of Canarium schweinfurthii was fractionated and tested for their anti-salmonellal activity. The most active fractions (i.e. chloroform and ethyl acetate partition fractions) were then explored for their phytochemical constituents. Fractionation on normal phase silica gel column chromatography and size exclusion chromatography on Sephadex LH-20 led to the isolation of four compounds (maniladiol, scopoletin, ethyl gallate and gallic acid) reported for the first time in Canarium schweinfurthii. Results Result indicated that scopoletin and gallic acid had greater activity than the crude extracts and partition fractions. Among the isolated compounds, scopoletin showed the highest inhibitory activity with a MIC of 16 μg/ml against Salmonella Typhimurium and Salmonella Enteritidis. Conclusions The overall results of this study indicates that the hydro-ethanolic extract as well as some of isolated compounds have interesting anti-salmonellal activities that could be further explored for the development of potent therapy for salmonellosis. Furthermore, the study adds credence to the folkloric applications of the plant.

Objective: Knema attenuata (Myristicaceae), popularly known as “wild nutmeg,” is an endemic tree species from Western Ghats, which has been used in folk medicine. Conventionally, the stem bark of K. attenuata is used for treating inflammatory conditions without any scientific information available for the same. The present study was undertaken to evaluate the anti-inflammatory activity of the ethanolic stem bark extract (ESBE) of K. attenuata using in vivo and in vitro screening models. Methods: The ethanolic extract of stem bark was prepared by soxhlation, and its cytotoxicity in RAW 264.7 cell line was assessed using MTT assay method. In vivo anti-inflammatory effect of extract was estimated in rats using carrageenan-induced paw edema model and cotton pellet-induced granuloma model. The in vitro anti-inflammatory activity of the extract was evaluated by cyclooxygenase and lipoxygenase inhibition assay, estimation of myeloperoxidase activity, and determination of cellular nitrite levels in lipopolysaccharide-stimulated RAW 264.7 macrophage cells. Results: Toxic symptoms were not observed for the ESBE. The extract demonstrated significant anti-inflammatory activity in both in vivo and in vitro models. The anti-inflammatory action exhibited by the extract was a result of the inhibition of leukocyte migration and nitric oxide pathway and partially by inhibition of mediators such as prostaglandins and leukotrienes. Conclusion: Findings from the study provide the evidence for the popular use of stem bark extract of K. attenuata as a potential anti-inflammatory agent.
