Objective: The aim of this study was to evaluate the potential effect of the methanolic extract of plantGlycyrrhiza glabraroots on bone mineral density and femoral bone strength of ovariectomized rats. Methods: Thirty 10-month-old Wistar rats were randomly separated into three groups of ten, Control, Ovariectomy and Ovariectomy-plus-Glycyrrhiza in their drinking water. Total and proximal tibial bone mineral density was measured in all groups before ovariectomy (baseline) and after 3 and 6 months post ovariectomy. Three-point-bending of the femurs and uterine weight and histology were examined at the end of the study. Results: No significant difference was noted in bone density percentage change of total tibia from baseline to 3 months between Control and Ovariectomy-plus-Glycyrrhiza groups (+5.31% ± 4.75 and +3.30% ± 6.31 respectively,P = non significant), and of proximal tibia accordingly (+5.58% ± 6.92 and +2.61% ± 13.62,P = non significant) demonstrating a strong osteoprotective effect. There was notable difference in percentage change of total tibia from baseline to 6 months between groups Ovariectomy and Ovariectomy-plus-Glycyrrhiza (−13.03% ± 5.11 and −0.84% ± 7.63 respectively,P < 0.005), and of proximal tibia accordingly (−27.9% ± 3.69 and −0.81% ± 14.85 respectively,P < 0.001), confirming the protective effect ofGlycyrrhiza glabraextract in preserving bone density of the Ovariectomy-plus-Glycyrrhiza group. Three-point-bending did not reveal any statistically significant difference between Ovariectomy and Ovariectomy-plus-Glycyrrhiza groups. Uterine weights of the Ovariectomy-plus-Glycyrrhiza group ranged between the other two groups with no statistically significant difference to each. Conclusions:Glycyrrhiza glabraroot extract notably protected tibial bone mineral density loss in Ovariectomy-plus-Glycyrrhiza rats in comparison with ovariectomized rats, but did not improve biomechanical strength.

Osteoporosis is a common problem in menopause woman. The main cause is lack of estrogen hormone. Commonly, prevention therapy is by consuming high calcium milk, but it is not effective. Bali orange’s peel (Citrus maxima Merr.) is a waste material but known contains phytoestrogen according to previous study. Considering of this result, fortification of high calcium milk and Bali orange’s peel is expected to be an effective solution for osteoporosis in menopause woman. This research began with extraction of Bali orange’s peel (BPE) using ethanol 70% by maceration method. Ovariectomized Sprague Dawley female rats as the model of post menopausal woman were treated by BPE for 28 days. The doses of BPE was given to rats is 500 and 1000 mg/KgBW combined with high calcium milk. Bone density was determined using digital microradiography, the profile showed the increase of bone density in group that treated with combination of BPE 1000 mg/Kg BW and high calcium milk compare to control and given only milk groups. Docking molecular showed that BPE’s active compound which are hesperidin and naringin have interaction with estrogen receptor α and β. Docking score of naringin with ER α and β are -19,97; -18,99 respectively. Meanwhile the docking score of hesperidin with ER α and β are -19,98; +49,92 respectively. Overall, the result of this research showed that fortification of BPE with high calcium milk has good prospect to develop as effective therapy of osteoporosis. Keywords : Citrus maxima Merr., phytoestrogen, osteoporosis, high calcium milk, estrogen receptor

Osteoporosis has become a major public health concern, due to its rising incidence every year. Osteoporosis mostly occurs in women at menopause, especially in Asia. Secang (Caesalpinia sappan L.) heartwood contains brazilin and brazilein which are known to suppress expression of NF-ĸB, an osteoclastogenesis protein playing an important role in osteoporosis.  Thus, the aim of this research is to investigate the inhibition of osteoporosis by ethanolic extract of secang (Caesalpinia sappan L.) (EES) in ovariectomized (OVX) rats. Thirty female Sprague Dawley rats were divided into five groups treated for 20 days:  group I were OVX and treated with 1000 mg/kgBW EES; group II were OVX and treated with 500 mg/kgBW EES; group III were OVX and treated with CMC-Na; group IV were shame OVX baseline; and group V were non-OVX-baseline. Bone density was examined by using X-Ray rontgen and was strengthen by qualitative data of Giemsa staining. Moreover, total number of osteoclasts in bone slide was observed by TRAP expression using double staining immunohistochemistry with hematoxylin eosin (HE) – TRAP. Treatment of 500 mg/kgBW EES in ovariectomized rats showed the highest bone density of 0.60 gr/cm3 and the lowest number of osteoclasts was 4.66 osteoclasts per field of view. These results showed that EES is potent to be developed as an antiosteoporosis agent. Further study on its dose adjustment and molecular mechanism needs to be conducted. Keywords: Caesalpinia sappan L., osteoporosis, RANK, bone density
