In this paper an attempt has been made to highlight the physicochemical study of methnolic extract of leaves of Tilkor carried out by soxhlet extraction process, phytochemical analysis of the extract, separations, isolation of bioactive components through Thin Layer Chromatography (TLC) as well as column chromatography respectively and characterisation of isolated compound by the means of several spectral analysis such as 1H NMR, 13C NMR, IR, U.V. Mass spectroscopy. The methanolic extract of leaves of the plant (in tropical conditions of Mithilanchal, Bihar, India) reveal the presence of phytochemicals like alkaloids, flavanoids, tannins, saponins, cardiac glycosides, steroids, terpenoids etc. The secondary metaboilities showed antimicrobial activity. The two isolated compounds were characterised by spectroscopic techniques which revealed the structure of compound A as - stigmosterol and compound B as tritriaconatane and is also found to have antimicrobial activity.

The present study was conducted to identify the phenols from methanol extracts (MHI) of medicinally and economically useful leaves of Holoptelea integrifolia (Roxb.) plant using High Performance Thin Layer Chromatography (HPLC) technique. Preliminary phytochemical screening was done and HPTLC studies were carried out on CAMAG HPTLC system equipped with Linomat V applicator (Switzerland). Densitometric scanning was performed with Camag TLC scanner IV in the reflectance absorbance mode at 540 nm and operated by Win CATS software (1.4.6 Camag) with the help of tungsten lamp. Preliminary phytochemical screening of methanolic extract of Holoptelea integrifolia showed the presence of steroids, alkaloids, flavonoids, proteins, phenols and carbohydrates. HPT LC finger printing of phenols of methanolic extract of leaf revealed seven polyvalent phytoconstituents (7 peaks) and corresponding ascending order of Rf values in the range of 0.15 to 0.75. From the results of preliminary phytochemical analysis and above Rf values, we have concluded the presence of phenols in methanol extracts.

Comparative study of plant extracts crude and aqueous, methanolic and ethanolic with antibiotics, provide evidence that calotropis procera extracts has the similar antibacterial activity as these antibiotics against test pathogens i.e. Salmonella typhi and E.coli. The analysis of antimicrobial activity of aqueous, methanolic and ethanolic extract of leaves and flower of Calotropis procera was carried out in disc method and also determined MIC value at 600nm through optical density using spectrophotometer. The zone of inhibition produced by extracts was examined and compares it with zone produced by antibiotics. The effect exhibited by ethanolic extract of leaves and flower was significantly greater than the aqueous and methanolic extract of leaves and flower. Crude extracts i.e. latex, leaves, fruit and flower crude extracts. Among them, flower crude extracts shows similar zone of inhibition to test anitibiotics.while in MIC value, we made different concentration of extracts and antibiotics.i.e for crude we made 25%, 50%.75% and 100% concentration of crude juice and for the aqueous, methanolic and ethanolic we made 0.5mg/mL,1mg/mL,2mg/mL,3mg/mL,4mg/mL, 5mg/mL and 6mg/mL in Dimethyl salfoxide, and same antibiotics concentration. Aqueous leaves extracts show MIC at 0.5mg/ml against E.coli while against Salmonella it shows MIC at 1mg/ml... We also determine phytochemical analysis for presences of different compounds in crude extracts. The obtained results provide a support for the use of Calotropis procera, in traditional medicine and suggest its further advance investigation.

Objective: This study was focused on to compare the antimicrobial activity of methanolic leaf extracts of tulsi and neem.Methods: We have chosen tulsi (Ocimum sanctum) and neem (Azadirachta indica) to compare their antimicrobial activity toward Gram-positive (Staphylococcus aureus) and Gram-negative (Escherichia coli) bacteria. Phytochemical extracts have been made by the use of methanol as solvent and dried leaf powder by the cold maceration extraction process. Phytochemical analysis for some secondary metabolites has been done using standard protocols. Nutrient agar plates were inoculated with the above-mentioned microorganisms by spreading bacterial inoculum on the surface of the media. Wells (6 mm in diameter) were punched in the agar. The phytochemical extracts of neem and tulsi were allowed to diffuse into the medium, and after incubation of 24 h at 37°C, the zones of inhibition were observed.Results: Statistical analysis showed that tulsi was more effective toward S. aureus while neem was more effective toward E. coli. Minimum inhibitory concentration (MIC) of tulsi for both the microorganisms was 0.4 g/ml, and the MIC of neem for both the microorganisms was 0.2 g/ml. When both the tulsi and neem extract were mixed with each other for every concentration at equal volume, they have shown better effects in comparison to individual neem or tulsi extract and also the MIC got reduced to 0.2 g/ml for both the bacteria.Conclusions: Leaf extracts of both the A. indica and O. sanctum have shown antimicrobial activity against E. coli and S. aureus. A. indica has higher antimicrobial activity against S. aureus, whereas the O. sanctum was found to be more effective against E. coli (indicated by the zone of inhibition). When both the extracts were mixed with each other, they have shown a better effect toward both bacteria.

Objective: To evaluate the antimicrobial activity of methanolic extract from the peel of the fruit of Citrus Limon (Family-Rutaceae) in conjugation with phytochemical analysis.Methods: The methanolic extract from the peel of the fruit of Citrus Limon (Family-Rutaceae) was separated from fruits, shade dried, powdered and extracted using methanol, analysed for phytochemical constituents using standard methods. The antimicrobial activity of the plant extract was examined against 2 bacterial strains among one is Gram-positive (Staphylococcus aureus) and other is Gram-negative (Escherichia coli) and 1 fungal strains (Candida albicans) using agar well diffusion method.Results: The present investigation shows the phytochemical analysis, antimicrobial activity of the methanolic extract of the fruits peel Citrus limon. Various phytochemical analyses revealed the presence of alkaloids, saponin, flavonoids, carbohydrates, glycosides and citric acids and tannins. The antimicrobial activity of the methanolic extract of the plant showed significant result against all the of the test organisms.Conclusion: The present study concluded that methanolic extract of the peel of Citrus Limon contains the high presence of phytochemicals. The methanolic extract of the plant was found to possess promising antimicrobial activity when compared with the standards.

Objective: This investigation involves the extraction, isolation, and characterization of flavonoid from a Euphorbiaceae family plant Chrozophoraplicata followed by evaluation of its antioxidant principles.Methods: The dried leaves were subjected to sequential soxhlation with polar and nonpolar solvents. Methanolic extract reveals the presence of largeamount of flavonoids. Methanolic extract was subjected to isolation using column chromatographic analysis with solvents such as petroleum ether,chloroform, hexane, ethyl acetate, methanol, and water. Further, the isolated compound was subjected to thin layer chromatography technique andspectral analysis such as infrared, 1HNMR, 13CNMR, mass spectroscopy, and high performance thin layer chromatography (HPTLC) finger printingtechniques. The compound was evaluated for in vitro antioxidant studies using 2,2-diphenyl-1-picrylhydrazyl (DPPH), NO assay, reducing power assay,H2O2 scavenging assay, superoxide anion scavenging assay and β-Carotene linoleate system and in vivo antioxidative studies using carbon tetrachloride(CCl4), and acetaminophen intoxicated rats.Results: The compound was isolated in methanol:water in the ratio of 80:20 using column chromatographic technique. On the basis of phytochemical,chromatographic, and spectral analysis, the isolated compound was identified as kaempferol and finally with HPTLC finger printing technique it wasfound that the Rf value of the isolated compound was found to be 0.58 which is nearly similar to the Rf value of standard kaempferol (0.55). Hence,the isolated compound was confirmed as kaempferol and is structurally elucidated as 3,5,7-trihydroxy-2-(4-hydroxyphenyl)chromen-4-one. Thiscompound was isolated for the first time from the C. plicata leaves. The in vitro antioxidant assay of isolated flavonoid has shown a dose-dependentincrease in free radical scavenging activity using DPPH, no assay, reducing power assay, H2O2 scavenging assay, superoxide anion scavenging assay, andβ-carotene linoleate system. Further, the methanolic extract of C. plicata (MECP) leaves was subjected to single dose acute toxicity study for 14 days infemale rats on the basis of OECD guidelines 423 and the therapeutically selected doses were 200 mg/kg and 400 mg/kg. In vivo antioxidant studies inCCl4 and acetaminophen intoxicated rats indicated that the MECP leaves have significantly decreased lipid peroxidation in a dose-dependent mannerand increased the levels of catalase, superoxide dismutase, and glutathione.Conclusions: By the above results, it was concluded that the isolated compound from C. plicata leaves was confirmed as kaempferol and it possessessignificant antioxidative potentials.Keywords: Chrozophora plicata leaves, Flavonoids, Extraction, Isolation, Characterization, Methanolic extract, Antioxidant activity, Carbontetrachloride, Acetaminophen.

Objective: To estimate the antimicrobial activity of methanolic extract of Adenium obesum (family Apocyanaceae) in association with phytochemical analysis. Methods: The methanolic extract of the flowers of Adenium obesum was prepared and investigated for phytochemical constituents using standard methods. Antimicrobial activity of methanolic extract Adenium obesum has been carried out against one gram-positive bacteria Staphylococcus aureus and one gram-negative bacteria Escherichia coli and the antifungal activity of the plant extract was evaluated on Candida albicans species. The testing was done by the disc diffusion method to visualize the zone of inhibition of methanolic extract was compared with that of standard drug i.e., Ciprofloxacin for antibacterial activity and Fluconazole for antifungal activity. The minimum inhibition concentration was determined by micro dilution method (cup-plate method). Results: The present investigation shows the phytochemical analysis confirmed the presence of alkaloids, saponin glycosides, flavonoids, phenols, proteins and amino acids, cardiac glycosides, steroids, tannin, and phenolic compounds. The anti-microbial activity of the methanolic extract of the plant showed significant results against all the three of the test organisms in compare to the standard drugs. Conclusion: The present study concluded that methanolic extract of the flowers of adenium obesum proved the existence of phytochemical constituents. The methanolic extract of the was found to possess promising antimicrobial activity when compared with the standards and it can be useful for the development of antibiotics.

Present study aims to investigate phytochemical analysis as well as in vitro antimicrobial activity of crude aqueous, methanol, ethanol, chloroform and petroleum ether extracts from leaves of C. roseus. Antimicrobial activity of extract was studied against various bacterial strains (Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, Staphylococcus aureus and Mycobacterium smegmatis). Qualitative preliminary phytochemical analysis revealed that alkaloids, phenols, flavonoids, amino acid and cardiac glycosides are present in the extracts. The result for total phenol and flavonoids content was the highest in methanol and the lowest in petroleum ether crude extract. The study revealed that inhibition significantly depend upon the solvent used for extraction and bacterial strain tested for susceptibility. Aqueous extracts were found less efficient as compared to organic solvent. Methanolic extract were found more effective against tested microbes.

 Objective: The present study is concerned with the detection, isolation, identification, and structural elucidation of the bioactive phytochemical from the bulbs of Eleutherine bulbosa.Methods: Ethyl acetate is the solvent used in Soxhlet extraction. Column chromatography and thin layer chromatography is employed in the isolation procedure. Identification and structural elucidation of the bioactive compound is determined using ultraviolet-visible (UV-vis), Fourier transform-infrared (FTIR) spectroscopy, liquid chromatography–mass spectrometry (LCMS-MS), and 1H nuclear magnetic resonance (1HNMR) spectroscopy.Results: A compound was isolated from the bulbs of E. bulbosa by solvent extraction and chromatography. UV-vis, FTIR spectroscopy, LCMS-MS, and 1HNMR studies showed that the isolated compound was bis (2-6,7-diamino-5,8-dioxo naphthalene-1yl) propanal.Conclusion: An yellowish-brown crystalline needle-shaped compound was isolated from the bulbs of E. bulbosa and characterized chemically. Spectroscopic analysis identified the compound as bis (2-6,7-diamino-5,8-dioxo naphthalene-1yl) propanal. Mass of the compound is m/z 244 (MH+=245).

 Objective: The objective of this study is to identify medicinally important phytochemicals and evaluate the antimicrobial potential of Trachyspermum ammi seeds. Methods: Four different extracts (methanol, acetone, chloroform, and water) were prepared using a soxhlet apparatus, antimicrobial activity was tested using agar well-diffusion technique. Results: The results revealed the presence of flavonoids and saponins in all the extracts prepared. Similarly, alkaloids and phenols presence were obtained in methanol and aqueous extracts. Glycosides and carbohydrates in methanol, chloroform, and aqueous extracts. Further, proteins, terpenoids, and tannins presence were found in methanol, chloroform, and aqueous extracts, respectively. The maximum zone of inhibition was found in the methanolic extract (13.5 mm). Acetone, chloroform, and water extracts showed 9 mm, 10.5 mm, 11 mm respectively, while ciprofloxacin (control) showed 17.5 mm of the zone of inhibition. Conclusion: T. ammi seeds exert biological properties due to the presence of various chemical constituents. Thus, it can be used to obtain novel antibacterial compounds for the treatment of infectious diseases in the future.

Plants used for medicinal practices which were discovered since prehistoric stone ages are termed Medicinal plants, which are also referred to as medicinal herbs, since plants produces bioactive chemical compounds (phytochemicals), this research however, is concerned with the extraction using Soxhlet extraction technique, phytochemical screening using various test methods, which reveals the presence of anthraquinones (free anthraquinones and combined anthraquinones), carbohydrates, cardiac glycosides, glycosides, flavonoids, saponins, steroids/ terpenes, phenolic compounds and tannins, and absence of alkaloids for extracts of senna occidentalis and also, thin layer chromatography profiling which gives probable foundation for further structural elucidation amongst others. This research shows the presence of potent secondary metabolites present in the leaves of senna occidentalis (leaves).
