The antioxidant has an activity to neutralize free radical compound that the body needs to avoid damage cells and tissues. Phenolic is one of the compounds that have an antioxidant activity. The influences of ultrasonic-assisted extraction (UAE) and microwave-assisted extraction (MAE) conditions on phenolic compounds of Parkia speciosa Hassk. leaves were investigated. The effects of temperature (40C, 50C, 60C and 70C), time (10, 30 and 50 minutes) and material-solvent ratio (1:10, 1:13, 1:15 ) were evaluated based on the yield, total phenolic content (TPC) and antioxidant activity. The result showed that the highest yield (15.82%) was obtained at 1:15 (w/w) of material-solvent ratio, 50C of temperature and 50 minutes of extraction time for MAE. The highest yield of UAE is 15.53% that sample was obtained at 1:13 (w/w) of material-solvent ratio, 60C of optimal temperature and 30 minutes extraction time. The highest IC50 of UAE method extract was 52.55 ppm, while the extract obtained using MAE method was 50.44 ppm. UAE is more stable at higher temperatures. Time and solvent which was used more efficient than MAE. Extract of petai leaves (Parkia speciosa Hassk.) were very potential to be used as a source of natural antioxidants because they have IC50 values from 41.39 to 66.00 ppm. Its antioxidants capacity is ranged from strong to very strong capacity.

Abstrak. Kenikir (Cosmos caudatus) adalah tanaman yang sering dimanfaatkan daunnya untuk bahan pangan maupun juga sebagai obat-obatan karena memiliki kandungan senyawa fenolik yang cukup tinggi. Ekstraksi senyawa fenol menggunakan metode konvensional masih memerlukan waktu yang cukup lama dan jumlah pelarut yang banyak. Sehingga perlu dicari alternatif metode ekstraksi yang memiliki waktu ekstraksi dan kebutuhan pelarut yang relatif rendah seperti menggunakan Microwave Assisted Extraction (MAE). Tujuan dari penelitian ini adalah melakukan ekstraksi senyawa fenolik dari daun kenikir dengan metode MAE terhadap total fenol dan aktivitas antioksidan. Penelitian ini menggunakan 2 faktor perlakuan yaitu rasio pelarut terhadap kenikir segar yaitu 1:4, 1:6, dan 1:8 (b/v) dan waktu ekstraksi 2, 3 dan 4 menit. Penelitian ini menjelaskan bahwa nilai total fenol tertinggi (17,162 ± 0,11 mg GAE/g berat segar) diperoleh pada perlakuan rasio pelarut 1:8 (b/v), sedangkan aktivitas antioksidan tertinggi (4,203 ± 0,26 mg/ml) pada perlakuan rasio pelarut 1:4 (b/v). Waktu perlakuan 3 menit menunjukkan hasil yang optimal untuk nilai total fenol dan aktivitas antioksidan. Hasil tersebut juga menunjukkan nilai yang lebih baik jika dibandingkan dengan metode konvensional baik dengan maserasi biasa dan maserasi dengan waterbath. Extraction of Phenolic Compounds from Cosmos caudatus using Microwave Assisted Extraction (MAE) Abstract. Cosmos caudatus, a plant commonly used its leaves for food and also as a medicine since it contains high phenolic compounds. The extraction of phenolic compounds using conventional methods still require more extraction time and need much more of solvent. Therefore, it was necessary to find an alternative extraction method that has a rapid extraction time and a few solvent need such as using Microwave Assisted Extraction (MAE). The aim of this study is to extract of phenolic compounds from Cosmos caudatus with MAE method of total phenol and antioxidant activity. This study using 2 treatment factors, the sampel-solvent ratio of 1:4, 1:6, and 1:8 (w/v) and the extraction time of 2, 3 and 4 minutes. This study explained that the highest total phenolic compounds (17.162 ± 0.11 mg GAE/g fw) and antioxidant activity (4.203 ± 0.26 mg/ml) were obtained at 1:8 and 1:4 (w/v) of sampel-solvent ratio, respectively. The extraction time of 3 minutes was the optimal condition for total phenolic compounds and antioxidant activity. These results also showed the highest parameter value when compared with both conventional methods: maceration and maceration with waterbath.

The present study aimed at evaluating the effectiveness of different natural deep eutectic solvents (NADES) on the extraction of phenolic compounds from Lavandula pedunculata subsp. lusitanica (Chaytor) Franco, on the antioxidant activity, and acetylcholinesterase (AChE), butyrylcholinesterase (BChE) and tyrosinase (Tyr) inhibitory capacities. Ten different NADES were used in this research and compared with conventional solvents. Ultrasound-assisted extraction (UAE) for 60 min proved to be the best extraction condition, and proline:lactic acid (1:1) and choline chloride:urea (1:2) extracts showed the highest total phenolic contents (56.00 ± 0.77 mgGAE/gdw) and antioxidant activity [64.35 ± 1.74 mgTE/gdw and 72.13 ± 0.97 mgTE/gdw in 2.2-diphenyl-1-picrylhydrazyl (DPPH) and 2.2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) methods, respectively]. These extracts also exhibited enzymes inhibitory capacity particularly against Tyr and AChE. Even so, organic acid-based NADES showed to be the best extractants producing extracts with considerable ability to inhibit enzymes. Twenty-four phenolic compounds were identified by HPLC-HRMS, being rosmarinic acid, ferulic acid and salvianolic acid B the major compounds. The results confirmed that the combination of UAE and NADES provide an excellent alternative to organic solvents for sustainable and green extraction, and have huge potential for use in industrial applications involving the extraction of bioactive compounds from plants.

Objective: This work aimed to determine the optimal extraction conditions of Mahkota dewa using different extraction methods: microwave-assistedextraction with ethanol (MAE-ethanol), ultrasonic-assisted extraction using a choline chloride natural deep eutectic solvent (UAE-NADES), andSoxhlet and reflux extractions.Methods: Experimental design was performed through Box-Behnken. The efficiency of the methods was evaluated based on the total phenolic content(TPC) and antioxidant activity. The presence of relevant compounds was qualitatively assessed by high-performance liquid chromatography (HPLC).Results: For the MAE-ethanol method, optimum extraction was achieved with 65% ethanol, in 1 min under 30% microwave power, and a 1:12 sample–solvent ratio, which yielded a TPC and antioxidant activity of 62.79±0.74 mg gallic acid equivalents (GAE)/g powder and 30.48±0.32%, respectively. InUAE-NADES, the optimal conditions were choline chloride: lactic acid ratio of 1:4 with a 50% water addition, 20 min extraction (TPC and antioxidantactivity of 65.25 mg GAE/g powder and 26.45±0.02%). The Soxhlet and reflux methods gave a TPC of 49.68±0.59 and 50.98±0.54 mg GAE/g powder,and antioxidant activity of 22.87±0.13% and 25.97±0.34%, respectively. Qualitative HPLC analysis showed the presence of quercetin, mahkoside A,4,6-dihydroxy-4-methoxybenzophenone-2-O-β-D-glucopyranoside, and mangiferin.Conclusion: MAE-ethanol was more efficient than the conventional Soxhlet and reflux methods and performed similarly to UAE-NADES in terms of TPC.

In a completely green approach to the exploitation of kiwi juice pomace (KP), a microwaved-assisted extraction (MAE) process was performed to extract antioxidant compounds present in KP, evaluating the influence of four independent process variables (temperature (T), extraction time (E), solvent composition (C), and solid-to-solvent ratio (R)) on the response of total phenolic content (TPC). The optimal conditions for the green extraction of total polyphenols from KP were obtained using a three-level fractional factorial design under response surface methodology (RSM) coupled with desirability optimization, and a feed-forward multilayered perceptron artificial neural network (ANN) with a back-propagation algorithm. Data were analyzed by ANOVA and fitted to a second-order polynomial equation using the regression method. Results showed that T was the most influential factor, followed by R and C, whereas the extraction time (E) was not shown to have a significant linear effect on the extraction yield of total polyphenols (TPs). The optimal conditions based on both individual and combinations of all responses were found out (T: 75 °C; E: 15 min; C: 50% ethanol:water; R: 1:15), and under these conditions the obtained extract showed both a high bioactive compound content and a high antioxidant potential, pointing out how this by-product could become an inexpensive source of compounds with high added value. A very good agreement was observed between experimental and calculated extraction yields, thus supporting the use of these models to quantitatively describe the recovery of natural antioxidants from KP. Finally, the ANN model exhibited more accurate prediction and better generalization capabilities than the RSM model (R2: 0.90 and 0.99, for RSM and ANN, respectively).

ABSTRACT Yacon (Smallanthus sonchifolius) is a tuberous root from the Andean region in the South America rich em water, fructooligosaccharides and phenolic compounds, some of which are natural antioxidants and may help prevent the deleterious action of free radicals in the body. The yacon has attracted much attention due to their potential health benefits to humans. In this study the levels of total phenolics, tannins, phenolic acids, and total antioxidant activity were measured in the peel and pulp of yacon tubers both in the fresh and flour forms. The flours of yacon presented higher concentrations of total phenolics and tannins, especially peel flour. The yacon pulp flour stood out as the main source of phenolic acids, mainly caffeic and chlorogenic acid. The total antioxidant activity assessed by DPPH (2,2-Diphenyl-1-picrylhydrazyl) and ABTS (2,2′-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) assays was higher in the yacon peel flour. The total antioxidant activity was correlated with the total phenolic content and tannins by the DPPH and ABTS assays. These results suggest that yacon can be used as an alternative food source of phenolic compounds that help prevent degenerative processes caused by oxidative stress, especially in the flours form.

AbstractThe grape pomace is a by-product from the industrial processing of grape juice, which can be used as a source of bioactive compounds. The aim of this study was to separate the phenolic compounds from grape pomace using an acidic aqueous solution with 2 % citric acid as a solvent, using both ultrasound-assisted extraction, with powers of 250, 350 and 450 W and times of 5, 10 and 15 min, and microwave-assisted extraction using powers of 600, 800 and 1,000 W and times of 5, 7 and 10 min. The results showed that for both methods of extraction, the contents of total phenolic compounds and antioxidant activity by ABTS and DPPH increased with time, and microwave at 1,000 W for 10 min corresponded to the best extraction condition. However, the contents of phenolic compounds and antioxidant activity were lower than exhaustive extraction using acidified methanol solution.

Two optimized methods for ultrasound-assisted extraction were evaluated for the extraction of two types of acai bioactive compounds: Total anthocyanins (TAs) and total phenolic compounds (TPCs). For the extraction optimization, a Box Behnken factorial design of different variables in the following intervals was used: Methanol-water (25%–75%) for solvent composition, temperatures between 10 and 70 °C, amplitude in the range between 30% and 70% of the maximum amplitude −200 W), extraction solvent pH (2–7), the ratio for sample-solvent (0.5 g:10 mL–0.5 g:20 mL), and cycle between 0.2 and 0.7 s. The extraction kinetics were studied using different periods between 5 and 30 min. TA and TPC were analyzed by UHPLC and the Folin–Ciocalteu method, respectively. Optimized conditions for TA were: 51% MeOH in water, 31 °C temperature, pH 6.38, cycle 0.7 s, 65% amplitude, and 0.5 g:10 mL of sample-solvent ratio. Optimized conditions for the TPC were: 49% MeOH in water, 41 °C temperature, pH 6.98, cycle 0.2 s, 30% amplitude, and 0.5 g:10 mL of sample-solvent ratio. Both methods presented a relative standard deviation below 5% in the precision study. The suitability of the methods was tested in real samples. It was confirmed that these methods are feasible for the extraction of the studied bioactive compounds from different açai matrices.

In the present work, the ultrasonic-assisted extraction (UAE) of total flavonoids (TF) from Pteris cretica L. was optimized by response surface methodology (RSM) on the basis of a single-factor experiment. The optimized UAE parameters were as follows: Ethanol concentration 56.74%, extraction time 45.94 min, extraction temperature 74.27 °C, and liquid/solid ratio 33.69 mL/g. Under the optimized conditions, the total flavonoids yield (TFY) was 4.71 ± 0.04%, which was higher than that obtained by heat reflux extraction (HRE). The extracts were further analyzed by HPLC, and five major flavonoids, including rutin, quercitrin, luteolin, apigenin, and luteolin-7-O-glucoside, were identified and quantified. Furthermore, the results of the antioxidant test showed that the TF extract obtained under optimized UAE conditions exhibited good 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical (ABTS+•), nitric oxide radical (NO•) scavenging activities, and ferrous ion (Fe2+) chelating capacity, with IC50 values of 74.49, 82.92, 89.12, and 713.41 µg/mL, respectively. Results indicated that the UAE technique developed in this work was an efficient, rapid, and simple approach for the extraction of flavonoids with antioxidant activity from P. cretica.

Abstract The aim of the present study was to reveal an effective extraction procedure for maximization of the yield of steviol glycosides and total phenolic compounds as well as antioxidant activity in stevia extracts. Ultrasound assisted extraction was compared with conventional solvent extraction. The examined solvents were water (100°C/24 h) and 70% ethanol (at 70°C for 30 min). Qualitative and quantitative analyses of steviol glycosides in the extracts obtained were performed using high performance liquid chromatography. Total phenolic compounds, flavonoids, and radical scavenging capacity by 2, 2-azino-di-3-ethylbenzothialozine- sulphonic acid) assay were also determined. The highest content of steviol glycosides, total phenolic compounds, and flavonoids in stevia extracts were obtained when ultrasound assisted extraction was used. The antioxidant activity of the extracts was correlated with the total amount of phenolic compounds. The results indicated that the examined sonication parameters represented as the probe diameter (7 and 22 mm) and treatment time (2, 4, 6, 8, and 10 min) significantly contributed to the yield of steviol glycosides, total phenolic compounds, and flavonoids. The optimum conditions for the maximum yield of steviol glycosides, total phenolic compounds, and flavonoids were as follows: extraction time 10 min, probe diameter 22 mm, and temperature 81.2°C.
