AbstrakKrisan (Chrysanthemum morifolium Ramat) mengandung senyawa kuersetin dengan efek farmakologi yang sangat luas. Penambahan polyethilene glycol (PEG) dapat dimanfaatkan untuk meningkatkan produksi metabolit sekunder secara in vitro. Penelitian ini bertujuan untuk mengetahui pengaruh PEG terhadap pertumbuhan kalus krisan dan kadar kuersetin 3-O-rhamnosida pada pencahayaan yang berbeda. Penelitian menggunakan Rancangan Acak Lengkap (RAL). Eksplan kalus berumur 45 hari setelah tanam (HST) disubkultur pada media MS + 4 ppm 2,4-D dengan penambahan PEG dalam lima taraf konsentrasi yaitu 0, 10, 20, 30, dan 40 ppm. Kultur diinkubasi pada kondisi gelap dan terang. Parameter yang diamati adalah warna, tekstur, ukuran, berat basah, dan berat kering kalus serta kadar kuersetin 3-O-rhamnosida. Data kuantitatif dianalisis menggunakan Analisis Varians dan Uji Jarak Berganda Duncan (α 5%), sedangkan data kualitatif dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa kalus pada kondisi terang berwarna cokelat dan cokelat kehijauan, sedangkan pada kondisi gelap berwarna putih kecokelatan. Perlakuan 10 ppm PEG menghasilkan berat basah dan berat kering kalus tertinggi baik pada kondisi terang maupun gelap berturut-turut 1,97 g dan 2,92 g; 0,94 g dan 1,09 g. Kadar kuersetin 3-O-rhamnosida tertinggi pada kondisi gelap dan terang terdapat pada perlakuan 10 ppm PEG berturut-turut  1,72 µg/g berat kering (BK) dan 2,59 µg/g BK.Abstract Chrysanthemum morifolium Ramat. contains quercetin with very broad pharmacological effects. The addition of PEG can be used to increase the production of secondary metabolites using in vitro method. This study aimed to determine the effect of PEG on the growth of Chrysanthemum callus and quercetin 3-O-rhamnoside content in different lighting conditions. The experimental design used was a Completely Randomized Design. After 45 days, callus were subcultured on MS medium + 4 ppm 2,4-D which PEG was added in five concentration levels (0, 10, 20, 30, and 40 ppm). Culture was incubated in dark and light conditions. Parameters observed were color, texture, size, wet weight and dry weight of callus, also quercetin 3-O-rhamnoside levels. Quantitative data was analyzed using Analysis of Variance and Duncan's Multiple Distance (α 5%). Qualitative data was analyzed descriptively. The results showed that in light condition, the callus has brown and greenish brown color, whereas in dark, it has brownish white color. The 10 ppm PEG treatment produced the highest fresh weight and dry weight in both light and dark conditions of 1.97 g and 2.92 g, 0.94 g and 1.09 g, respectively.The highest quercetin 3-O-rhamnoside content  in dark and light conditions were founded in 10 ppm PEG treatment of 1.72 µg/g dry weight (DW) and 2.59 µg/g DW.

The purpose of this study was to evaluate the response of administration of a combination of 2,4-D and BAP on the growth of soybean callus subculture and determine the combination of 2,4-D and BAP most good for the growth of soybean callus subculture. The study design used completely randomized design (CRD) with a pattern factorials. The factor one e.i 2,4-D consists of 4 levels, namely: 0, 5, 10, and 15 ppm. A factor of 2 e.i BAP consists of 4 levels, namely: 0, 2, 4, dan 6 ppm. Every combination treatment repeated three times. Parameters measured include the percentage is growing callus, type of callus, dry weight and wet weight of soybean callus. The results showed that administration of the hormone 2,4-D and BAP can spur the growth of soybean callus subculture, the combination of BAP 2 ppm and 10 ppm of 2,4-D is the best combination for a percentage of callus and growing callus types, whereas the wet weight and the weight dried callus is not driven by a combination of the hormone 2,4-D and BAP

The objectives of the research were to study the effect of 2,4-dichlorophenoxiacetic acid (2,4-D) and kinetin on callus growth and saponin production from Talinum paniculatum Gaertn. callus. The outline of the research was T. paniculatum as a medicinal plant potentially to be developed with in vitro culture method. The addition of 2,4-D and kinetin in culture’s medium would induce protein synthesis, further more it would influence cell proliferation and cell metabolism with regulation enzyme action, so that both of induced callus growth and secondary metabolism production from the cell that be cultured. The research used factorial completely randomized design with two factors (2,4-D concentration: 0 mgl-1, 0.5 mgl-1, 1 mgl-1, 1.5 mgl-1 and kinetin concentration: 0 mgl-1, 0.5 mgl-1, 1 mgl-1, 1.5 mgl-1) with three replicates. Data that be collected were qualitative data (callus morphology included textur and colour of callus) and quantitative data (callus growth rate, callus wet weight, callus dry weight and saponin content). Data were analyzed by using anova and followed by DMRT 5% confidence level and corelation regresion. The result of the research indicated that the treatment with addition plant regulation (2,4-D and kinetin) on MS medium have significant effect on callus growth and saponin production. Addition 2,4-D 1.5 mgl-1 and kinetin 1.5 mgl-1 as anoptimum combination concentration to induce callus growth and saponin production.

Research on the response of roots of palm sugar palm seeds (Arenga pinnata) in nurseries at doses and intervals of Nasa liquid organic fertilizer. Aims to determine the dosage, interval and combination of dosages and fertilization intervals with Nasa liquid organic fertilizer which can provide the best dry weight of the roots in the nursery. The research design used was a Completely Randomized Design (CRD) with 3x3 factorial experiments and each treatment was repeated 8 (eight) times, consisting of: the first factor was the treatment of POC Nasa dose in a concentration of 3 cc POC Nasa per liter of water (D) consists of 3 levels, namely: d1 = 300 ml POC Nasa, d2 = 400 ml POC Nasa, d3 = 500 ml POC Nasa, while the second factor is the treatment of POC Nasa Interval (I) consisting of 3 levels, namely: i1 = 2 once a week, i2 = once every 3 weeks, i3 = once every 4 weeks. The results of the POC Nasa dose study had a significant effect on leaf wet weight, leaf dry weight, root wet weight and root dry weight. The best dose at this level are: d2 (400 ml of Nasa liquid organic fertilizer), the interval of liquid organic fertilizer Nasa has a significant effect on leaf wet weight, leaf dry weight, root wet weight and root dry weight. The best dose at this level is: i1 (once every 2 weeks). The treatment combination has no significant effect on all parameters.

Dendrobium anosmum is one of natural orchids in Indonesia. Optimization of medium composition for orchid propagation through in vitro culture is necessary to enhance propagule multiplication capabilities and quality. This study was aimed to study the influence of concentration of coconut water in culture medium on in vitro growth and development of D. anosmum orchid species and to determine the optimal coconut water concentration in culture media.  The experiment were arranged in a Completely Randomized Design with four treatments and eight replications. The treatments consisted of the addition of coconut water with concentrations: 0 ml•l -1 (control), 50 ml•l-1, 100 ml•l-1 and 150 ml•l-1. The results showed that addition of coconut water in culture medium gave different effect on shoot growth and multiplication of D. anosmum orchids.  Coconut water concentration of 100 ml•l-1 was the best concentration for growth and multiplication of D. anosmum orchids, based on both shoots and roots growth, plantlet height and wet weight.

Meizotropis pellita is a rare, endangered and threatened plant species found in Patwadanger forest. In nature this species grows from rootstock in precise and sensitive habitats. However, phenolic compounds provoke browning reaction which is responsible for lethal browning in plant tissue culture and hindering in vitro germination. The aim of this study was to minimize the effect of phenolic compound during seed germination. We evaluate different antibrowning treatments in Meizotropis pellita seeds. The seeds were pretreated in Polyvinylpyrrolidone and then treated seeds were cultured on MS media and sterilized seeds were directly inoculated on PVP containing MS media. These culture media incubate under dark and light conditions at different durations respectively. Our results showed that addition of antibrowning agent, PVP inhibit onset of browning. Dark and light condition also played a crucial role on onset of browning. Delayed and low intensity of browning were observed in case of seeds that were inoculated on MS media containing PVP and were kept in dark. The present research offers a positive in vitro seed germination protocol for Meizotropis pellita conservation.

This study aims to know the effect of Benzyl Amino Purine (BAP) and Coconut Water on shoot bud growth and Broccoli Sulforaphane content (Brassica oleracea L. var italic Plenck). The study was conducted in the laboratory of Biotechnology Department of Aquaculture, Faculty of Agriculture of Sam Ratulangi University, Manado, that conducted from August-December 2017. This study used a Complete Randomized Design (RAL), consisting of 8 treatments and each repeated as many 4 times, so we get 32 unit experiment. The variables observed were number of buds, number of leaves, plant height, wet weight, root number and Sulforaphane content analysis. The result of research shows that analysis of variance showed that in the use of Benzyl Amino Purine (BAP) concentration 3 ppm tends to increase the number of leaves aged 4 Weeks After Culture (MSK) and increase the number of shoots age 2 and 6 Weeks After Culture (MSK). Benzyl Amino Purine (BAP) 3 ppm can increase the wet weight of age 6W eeks After Culture ((MSK). Coconut water 20% tends to increase the number of leaves at age 6 Weeks After Culture (MSK) and increase the number of shoots aged 6 Weeks After Culture (MSK), while for combination of 3 ppm Benzyl Amino Purine (BAP) and coconut water 20% tends to increase the number of leaves aged 2 Weeks After Culture (MSK) and the number of shoots aged 2 Weeks After Culture (MSK). Combination of coconut water and Benzyl Amino Purine (BAP) is not detected by the content of Sulforaphane.

A common problem related to the production of potatoes in Indonesia is the use of poor quality seed sources and declining seeds. Plant tissue culture is one of the best methods used for the provision of quality seeds. This study aims to establish the best combination of BAP concentration and the concentration of paclobutrazol in spurring the growth of cuttings and initiation of potato tubers in vitro.This research was conducted in November 2017 until June 2018 at Agronomy Laboratory of Biotechnology Division and Plant Culture Network of Agriculture Faculty of Bengkulu University. The  research  design  used  was  Completely  Randomized  Design  (CRD)  which  consist  of  18 combination of BAP treatment and Paclobutrazol was repeated 6 times. The concentration of BAP used was 0 mg / l, 5 mg / l, 10 mg / l and the concentration of Paclobutrazol 0 mg / l; 2.5 mg / l; 5 mg / l; 7.5 mg / l; 10 mg / l; 12.5 mg / l. The data of the research were analyzed by F test of 5% level and if there were different between treatments followed by Duncan's Multiple Range Test (DMRT) test of5% level. The results showed that the combination of BAP and  Paclobutrazol concentration had significant effect on all observed variables. Treatment of 5 mg / l BAP and 7.5 mg / l Paclobutrazol is the best medium in inducing potato micro tube as indicated by the fastest tuber formation time, highest percentage of productive crops, largest number of tubers per bottle, number of tubers per productive plant, tuber diameter the largest and highest wet weight of tuber per bottle. Keywords : in vitro, BAP, paclobutrazol, Potato

Availability of quality seeds in production of krisan (Chrysanthemum morifolium Ramat cv Dewi ratih) cultivation is still rare, therefore research on seed multiplication through tissue culture is needed. The media used in tissue culture is relatively expensive for home industry. This study aims to determine the respond of leaf and stem explants using foliar fertilizers (Growmore, Gandasil D and Mutiara) as an alternative media for callus inductions. This study used a Completely Randomized Design (CRD) consisted of 4 treatments: P0: ½ MS + 0,25 mg/l BAP, P1 (Growmore + 0,25 mg/l BAP), P2 (Gandasil D + 0,25 mg/l BAP), P3 (Mutiara + 0,25 mg/l BAP). The variables observed in this study included callus appearance time, callus color and callus texture. The result of this study indicated that the use of BAP (6-Benzyl Amino Purine) affected the time of callus formation and callus morphology. Callus was formed on leaf explants 13 days after planting while on stem explants 7 days after planting and compact texture. Growmore + 0,25 mg/l BAP treatment yields the best callus on leaf explant, while Gandasil D + 0,25 mg/l BAP treatment yields the best callus on stem explant.

ABSTRACT Some factors may influence the in vitro culture of native orchids, especially the culture medium, sealing system and light condition. This study aimed to evaluate the addition of banana pulp and peel in the formulation of culture media, sealing system and irradiance, in the in vitro growth of Cattleya nobilior Rchb. f. A completely randomized design was used in a factorial scheme with five culture media [Murashige & Skoog supplemented or not with banana pulp or peel (100 or 200g L-1)], two light conditions [3,000 K LED lamps (43 µmol m-2 s-1 or 86 µmol m-2 s-1)] and two sealing systems (natural and conventional ventilation), with six replicates of one vial each. The plant height, diameter of the largest pseudobulb, length of the largest root and of the largest leaf, fresh weight and number of leaves, roots and shoots were evaluated. The use of both the banana pulp and peel as additives in the culture medium formulation promoted the C. nobilior growth. The natural ventilation system increased the plant height, pseudobulb diameter and leaf length, while the conventional system promoted tillering.

The objectives of this study were to determine the effects of yellow light (Y), green light (G), and two blue lights (B) at different wavelengths in conjunction with red light (R) on the growth and morphogenesis of potato plantlets in vitro. Randomized nodal explants were cut into 1.0–1.5 cm pieces and were grown under five different light conditions: fluorescent white light (FL); the combined spectra of R, Y, and B at 445 nm (R630B445Y); the combined spectra of R, G, and B at 445 nm (R630B445G); the combined spectra of R, Y, and B at 465 nm (R630B465Y); and the combined spectra of R, G, and B at 465 nm (R630B465G). Morphogenesis and physiological parameters were investigated. The results showed that R630B445Y and R630B465Y increased the fresh weight (FW), dry weight (DW), stem diameter, blade number, leaf area, specific leaf weight (SLW), and the health index of potato plantlets in vitro; root activity increased significantly; and soluble sugar, soluble protein, and starch also increased. The addition of Y to the combined spectra of R and B contributed to the growth, development, and morphogenesis more than the combined spectra of R and B with G, and B at 445 nm was more effective at promoting plant growth than was B at 465 nm.
