Abstract: The objective of this work was to compare wild poinsettia (Euphorbia heterophylla) biotypes as to their morphological characteristics, as well as to determine their genetic similarity in the state of Rio Grande do Sul, Brazil, and to confront the light compensation point of susceptible biotypes and of biotypes with low-resistance level to glyphosate. Besides the morphology analysis, the study included the genetic characterization, by AFLP, of susceptible (GR50= 58.65 g ha-1 a.e.) biotypes and of biotypes with low resistance (GR50= 310.36 g ha-1 a.e.) to glyphosate. The competitive ability of the biotypes was inferred according to their light compensation point. The observed morphological characteristics do not allow to differentiate biotypes as to their geographical location, or their resistance level to glyphosate. The genetic analysis identified low genetic diversity among the 15 tested biotypes, with a large amount of subgroups. The biotypes with low resistance formed an isolated group. The susceptible biotypes showed the lowest values of light compensation point and, possibly, they have a greater competitive ability under low-luminosity conditions. The evaluated characteristics do not enable to differentiate genotypes with different levels of glyphosate resistance.

Psidium guajava L. is a perennial fruit tree in subtropical and tropical areas. In Bangladesh, P. guajava has been used as edible fruits and people use it to treat acute diarrhea, cough and intestinal spasmodic diseases. In the present study, morphological and molecular characterizations were used to display different levels of variability. Molecular marker random amplified polymorphic DNA (RAPD) was used for the molecular identification of 33 P. guajava germplasm from three selected south-western location of Bangladesh. Among them, eleven commercially cultivated germplasm and the rest twenty two were collected from local farmers. The 10-mer and 12-mer oligonucleotide primers were used in RAPD to amplify. Four primers, A02, A03, S07 and S08, were able to direct the amplification and yield a total of 252 band patterns of which 33.19% were polymorphic. The highest percent of polymorphic loci (37.5%) was observed from primer A03 and the lowest (28.57%) was from primer S08. Results were analyzed by molecular algorithm UPGMA and Neighbor-Joining. Thirty-three genotypes on the dendrogram were identified and divided into two major groups and subgroups on the basis of morphological characteristics and also on the uncultivated and commercial cultivars. The range of genetic distance was observed 0.5253 (Jelly and Thai) to 0.6631 (V30 and V 22). Based on the cluster analysis, the P. guajava samples have morphological difference were grouped independently. Due to the oversupply and scarcity cycles of the Agave tequilana Weber blue variety, the effect of agave age (harvested in 4, 5, and 6 years) as raw material for the tequila 100% agave silver class was studied for each stage in a full-scale (industrial) process. Harvested plants showed differences in their morphological characteristics that affected the amount of juice; this had an impact in the fermentation stage since must composition was modified in the nitrogen content and juice/exudate ratio. This was noticed due to an increase in the production of higher alcohols attributed to the odd-chain fatty of the exudate, which affects n-propanol production. The characterization of the final product showed the feasibility to use agaves (less than 7 years) to produce the Tequila 100% agave silver class and to comply with the quality criteria. Furthermore, the final product was analyzed with the gas chromatography-isotope ratio mass-spectrometry technique to determine its authenticity. The δ13CVPDB isotopic parameter (−13.40‰ in average) values show the type of plant used as a raw material for ethanol production, while the δ18OVSMOW (20.52‰ in average) isotopic parameter can be helpful in corroborating and ensuring the traceability of the product and the geographical location of the beverage production.

AbstractThis study deals with the determination of new natural fibers extracted from the Corypha taliera fruit (CTF) and its characteristics were reported for the potential alternative of harmful synthetic fiber. The physical, chemical, mechanical, thermal, and morphological characteristics were investigated for CTF fibers. X-ray diffraction and chemical composition characterization ensured a higher amount of cellulose (55.1 wt%) content and crystallinity (62.5%) in the CTF fiber. The FTIR analysis ensured the different functional groups of cellulose, hemicellulose, and lignin present in the fiber. The Scherrer’s equation was used to determine crystallite size 1.45 nm. The mean diameter, specific density, and linear density of the CTF fiber were found (average) 131 μm, 0.86 g/cc, and 43 Tex, respectively. The maximum tensile strength was obtained 53.55 MPa for GL 20 mm and Young’s modulus 572.21 MPa for GL 30 mm. The required energy at break was recorded during the tensile strength experiment from the tensile strength tester and the average values for GL 20 mm and GL 30 mm are 0.05381 J and 0.08968 J, respectively. The thermal analysis ensured the thermal sustainability of CTF fiber up to 230 °C. Entirely the aforementioned outcomes ensured that the new CTF fiber is the expected reinforcement to the fiber-reinforced composite materials.

Abstract In Platycodi Radix (root of Platycodon grandiflorum), there are a number of platycosides that consist of a pentacyclic triterpenoid aglycone and two sugar moieties. Due to the pharmacological activities of platycosides, it is critical to assess their contents in PR, and develop an effective method to profile various platycosides is required. In this study, an analytical method based on ultra performance liquid chromatography coupled with quadrupole time-of-flight/mass spectrometry (UPLC-QTOF/MS) with an in-house library was developed and applied to profile various platycosides from four different Platycodi Radix cultivars. As a result, platycosides, including six isomeric pairs, were successfully analyzed in the PRs. In the principal component analysis, several platycosides were represented as main variables to differentiate the four Platycodi Radix cultivars. Their different levels of platycosides were also represented by relative quantification. Finally, this study indicated the proposed method based on the UPLC-QTOF/MS can be an effective tool for identifying the detail characterization of various platycosides in the Platycodi Radix.

Treatments of atherosclerosis depend on the severity of the disease at the diagnosis time. Non-invasive diagnosis techniques, capable of detecting stenosis at early stages, are essential to reduce associated costs and mortality rates. We used computational fluid dynamics and acoustics analysis to extensively investigate the sound sources arising from high-turbulent fluctuating flow through stenosis. The frequency spectral analysis and proper orthogonal decomposition unveiled the frequency contents of the fluctuations for different severities and decomposed the flow into several frequency bandwidths. Results showed that high-intensity turbulent pressure fluctuations appeared inside the stenosis for severities above 70%, concentrated at plaque surface, and immediately in the post-stenotic region. Analysis of these fluctuations with the progression of the stenosis indicated that (a) there was a distinct break frequency for each severity level, ranging from 40 to 230 Hz, (b) acoustic spatial-frequency maps demonstrated the variation of the frequency content with respect to the distance from the stenosis, and (c) high-energy, high-frequency fluctuations existed inside the stenosis only for severe cases. This information can be essential for predicting the severity level of progressive stenosis, comprehending the nature of the sound sources, and determining the location of the stenosis with respect to the point of measurements.

Pestalotiopsis-like species have been reported affecting strawberry worldwide. Recently, severe and unprecedented outbreaks have been reported in Florida commercial fields where leaf, fruit, petiole, crown, and root symptoms were observed, and yield was severely affected. The taxonomic status of the fungus is confusing because it has gone through multiple reclassifications over the years. Morphological characteristics, phylogenetic analyses, and pathogenicity tests were evaluated for strawberry isolates recovered from diseased plants in Florida. Phylogenetic analyses derived from the combined internal transcribed spacer, β-tub, and tef1 regions demonstrated that although there was low genetic diversity among the strawberry isolates, there was a clear separation of the isolates in two groups. The first group included isolates recovered over a period of several years, which was identified as Neopestalotiopsis rosae. Most isolates recovered during the recent outbreaks were genetically different and may belong to a new species. On potato dextrose agar, both groups produced white, circular, and cottony colonies. From the bottom, colonies were white to pale yellow for Neopestalotiopsis sp. and pale luteous to orange for N. rosae. Spores for both groups were five-celled with three median versicolored cells. Mycelial growth and spore production were higher for the new Neopestalotiopsis sp. isolates. Isolates from both groups were pathogenic to strawberry roots and crowns. However, the new Neopestalotiopsis sp. proved more aggressive in fruit and leaf inoculation tests, confirming observations from the recent outbreaks in commercial strawberry fields in Florida.

Isolates of Sphaceloma manihoticola, the asexual stage of Elsinoe brasiliensis, were collected from several regions of south-central Brazil. The isolates were obtained from samples of leaves, stems, and petioles of cassava (Manihot esculenta) and the weedy Euphorbia heterophylla (“amendoim bravo”) by directly plating infected tissue onto acidified potato dextrose agar. For pathogenicity studies, 19 isolates were inoculated onto each of two cassava cultivars, MBRA 703 as a susceptible cultivar and MBRA 12 as a resistant cultivar to S. manihoticola. MBRA 703, with the greatest pathogenicity to 58% (11) of the isolates, showed an intermediate pathogenic reaction to 16% (3) of the isolates, and was less pathogenic to 26% (5) of the isolates. MBRA 12, with a less pathogenic reaction to 63% (12) of the isolates, showed an intermediate pathogenic reaction to 16% (3) of the isolates, and was highly pathogenic to 21% (4) of the isolates. The isolates were verified as belonging to the genus Sphaceloma based on their morphological characteristics, including conidia and hyphae of monoconidial isolate. Conidia of isolates were small, thin-walled, ellipsoid to (rarely) globose, commonly with one or two gut-tules. Conidiophores were phialides, hyaline to slightly pigmented 0-to-1 septate; conidiophores from the weedy specie were phialides, hyaline to brown 0-to-2 septate producing hyaline conidia. The isolates also were verified as belonging to the genus Sphaceloma by using a poly-merase chain reaction (PCR) assay, which detected a 645-bp band in all isolates except two (1 and 6) for which the PCR product had 600 bp. Digestion of the amplified product with the enzymes MspI and CfoI allowed differences to be detected in restriction patterns among isolates. A homogeneous banding pattern was obtained for 17 of the isolates but a different restriction pattern was obtained for isolates 1 and 6 of E. heterophylla. This suggests the possibility of another species within this group of isolates. The results indicate the presence of pathogenic variation among isolates of the fungus and an isolate-host interaction, because statistically significant differences were observed between the two cassava cultivars in response to inoculation with the isolates of S. manihoticola.

Apple fruit in Chile are primarily produced for export to Asia, Europe, and the United States, which typically requires 15 to 40 days of maritime transportation. Therefore, Chilean apple production must fulfill the sanitization requirements imposed by the receiving countries. Under these circumstances, it was important to clarify the etiology of bull's eye rot that can severely affect ‘Cripps Pink’ apple and other late-harvest cultivars in Chile. Based on morphological characteristics and the partial sequence analysis of the internal transcribed spacer sequences and β-tubulin genes, Neofabraea alba was identified as the causal agent of the bull's eye rot of Chilean apple. These results were further corroborated using species-specific primers. The incidence of bull's eye rot varied considerably; for instance, in 2009, 0.0 to 58.7% in 38 Cripps Pink orchards surveyed in the relatively arid and humid apple-growing areas of Chile, respectively. There was no evidence for the presence of N. malicorticis or N. perennans, which are commonly identified as causal agents of bull's eye rot in other apple-producing countries. Altogether, these data suggest that N. alba might represent the predominant and possibly the only cause of bull's-eye rot of Chilean apple.

Abstract Drawing from a Sellarsian realist-naturalist epistemology, we trace different levels of cognitive hierarchy procedures through which a representational system learns to update its own states and improve its ‘map-making’ capabilities from pre-conscious operations which modulate base-localization functions, to patterns of epistemic revision and integration at the conceptual and theoretical levels, producing a nomological double of its world. We show how ontological theorization becomes diachronically coordinated with and constrained by empirical science, and how the formal-quantitative kernel of scientific theories corresponds to qualitative-conceptual determinations at the structural level. Following Johanna Seibt’s characterization of ontology as a theory of categorial inference, we trace the preservation of inferential semantic structure across ontological theories in relation to model languages and provide provisional indications to coordinate Seibt’s account with a convergent realist assessment of systematic modeling, defining the epistemological conditions for articulating the preservation of formal structure in theories toward a limit-point of enquiry.
