Significance. Black Walnut (Juglans nigra L.) is a species of trees of the Walnut family (Juglandaceae). This plant of the genus Walnut (Juglans L.) has not been sufficiently studied unlike other species e.g. Juglans regia L. This medicinal plant raw material is quite perspective, its preparations have antimicrobial, general tonic effect. We can use its leaves, unripe fruit, and the bark. However they are not widely used in medicine. In order to introduce the plants of the Walnut genus to the State Pharmacopoeia of the Russian Federation (RF State Pharmacopoeia), it is necessary to conduct of pharmacognostic studies, to develop product specification file to confirm the identification and quality of medicinal plant raw materials. The aim of this study is to develop a method of quantitative determination of flavonoids in the bark of the black walnut (Juglans nigra L.). Materials and methods. Material of the study was black walnut bark, stocked during the sap flow period (April) in 2018. The bark was skived up to 15 cm long and 23 cm wide. The bark was air-dried with the protection from direct sun light. The end of the drying was checked by the brittleness of the bark. Results. The methods of the quantitative determination of flavonoids in walnut bark has been developed. We used the differential spectrophotometry taking into consideration state standard sample of rutin at the analytical wavelength of 416 nm. The error of a single determination with a confidence level of 95% is 1.20%. Conclusion. We used the developed technique and analyzed a number of samples of black walnut bark. The content of total flavonoids in the plant raw material is 5.13 0.02% (as calculated on rutin). The flavonoid content should be at least 4.0%.

Introduction. Herbal drugs of black walnut (Juglans nigra L.) are not registered on the territory of the Russian Federation at the present time. However, the State Register of Medicinal Products includes an extract of unripe fruits of a related species walnut (Juglans regia L.) «Yuglanex» (made in Russia) and the complex herbal drug «Tonsilgon» (made in Germany). A number of biologically active food supplements from black walnut raw materials have been registered by Russian Federal Service for Surveillance on Consumer Rights Protection and Human Wellbeing. They are an additional source of phenolic compounds as tannins and flavonoids. The purpose of the article is to study and systematize up-to-date information on the chemical composition of the medicinal plant raw material of black walnut and the pharmacological properties of its main biologically active compounds.Text. All parts of the black walnut are used in the ethnomedicine of the indigenous population of North America according to indications similar to the walnut in Asia and the Manchurian nut in the Far East: snake bites, fever and disorders of the gastrointestinal tract.The chemical composition of the medicinal plant raw material of black walnut is dencently walnut by its qualitative composition. Fruits, bark and leaves of black walnut contain a rich polyphenol complex (naphthoquinones, in particular, juglon and its derivatives, tannins, flavonoids, phenolic acids), vitamins, essential oil, organic acids. However, the quantitative analysis revealed a higher content of biologically active substances in the raw black walnut, especially in relation to polyphenolic compounds.The scientific literature describes the results of experiments on animals, confirming antioxidant, antimicrobial, antifungal, antiviral, antiparasitic, hypoglycemic, antispasmodic, and anti-tumor effect on certain cell lines.Conclusion. As a result of studying the literature and systematizing the current information on the chemical composition of the medicinal plant raw material of the black nut and the pharmacological properties of its main biologically active compounds, it has been established that the main properties are related to the presence of the phenol complex. However, a deeper study of the chemical composition is required. Both the total extracts of the black walnut from plant raw materials and individual compounds show predominantly antimicrobial, antifungal, antioxidant, antiviral, hypotensive, immunomodulatory, antitumor and antispasmodic activities in preclinical trials.

Introduction. Modern pharmacognostic research is aimed at searching for plant biologically active individual compounds (hereinafter referred to as RBAIS) isolated from plant extracts.Aim. Validation of HPLC-UV quantitative determination of sapogenin diosgenin in plant extracts from fenugreek seeds.Materials and methods. The object of study was raw materials-fenugreek seeds produced as medicinal plant raw materials by LLC «Sage» (Irkutsk). Validation of the method was carried out according to the parameters: specificity, linearity, correctness, precision in accordance with the requirements of SP XIV. One series of medicinal plant raw materials was used for the analysis, such as serial number – 010117, release date – 15 february 2017.Results and discussion. Validation characteristics were determined and their compliance with the necessary acceptance criteria was experimentally confirmed.Conclusion. It is established that the developed method of identification and quantitative determination of diosgenin in fenugreek seed extracts by HPLC-UV is correct, precise, specific and linear in the analytical field.

The flowers and fruits of various species of the hawthorn genus (Crataegus L.) are widely used in domestic and foreign medical practice as cardiotonic preparations. In our opinion, Crataegus submolls Sarg. is a perspective species for collection of the raw materials. Crataegus submolls Sarg. is successfully cultivated in the Russian Federation and is distinguished by its rapid growth and high yield compared with wild-growing species. Moreover, the fresh fruits of Crataegus submolls Sarg. are used in nutrition. The flowers of Crataegus submollis Sarg. contain flavonoids, among of which, like in the case of Hawthorn blood-red the hyperoside is present. It was determined that the maximum of the absorption curve of the solution of Crataegus submollis Sarg. flowers with a solution of aluminum chloride is 412 nm. The method of quantitative determination by the differential spectrophotometry of the total flavonoids calculated on hyperoside in the Crataegus submollis Sarg. flowers has been developed. It was established that the appropriate extragent for this raw material is 70% ethanol. The level of total flavonoid in Crataegus submollis Sarg. flowers has been studied. It was established that separate parts of Crataegus submollis Sarg. flowers differ markedly in the content of total flavonoids and their specific weight in raw materials. It was established that the content of total flavonoids calculated on hyperoside is varied from 2.05±0.08% tо 3.00±0.12%. It was determined that the content of total flavonoids calculated on hyperoside in different parts of hawthorn flowers ranges from 0.98% (receptacle and calyx) to 4.53% (petals). In our opinion, Crataegus submollis Sarg. flowers are the promising type of medicinal plant raw materials.

Currently, the actual task of modern pharmacy is to study the chemical composition and pharmacological properties of plant objects. Within the framework of this concept, it seems interesting to study Quercus robur L. buds. One of the promising groups of biologically active compounds of Quercus robur L. buds are flavonoids. This group of substances has a wide range of a pharmacological activity, which is significant in the creation of new medicines based on medicinal plant raw materials.The aim of the article was to work out methods for quantitative determination of total flavonoids in Quercus robur l. buds.Materials and methods. The research materials were aqueous-alcoholic extracts from Quercus robur L. buds with 70% ethyl alcohol which were analyzed by differential UV spectrophotometry on spectrophotometer “SF 2000” (Russia).Results. The methods for quantitative determination of total flavonoids in Quercus robur L. buds by differential UV spectrophotometry, has been developed using a standard sample of cynaroside at the analytical wavelength of 400 nm. The optimum parameters for the extraction of total flavonoids from Quercus robur L. buds have been determined. They are: the optimum extractant is 70% ethyl alcohol; the “raw material-extractant” ratio is 1:50; the extraction time is 120 min, the degree of atomization is 2 mm.The content of total flavonoids for Quercus robur L. buds has been determined; it varies from 0.27%±0.01 to 0.44%±0.02. These results make possible to recommend the content of total flavonoids for this type of raw materials not less than 0.25% as a lower limit.Conclusion. The data obtained in the course of the experiment, makes it possible to conclude that a further study of Quercus robur L. buds is promising, and it also contributes to the implementation of medicinal plant raw materials “Quercus robur L. buds” in the State Pharmacopoeia (Russia).

Introduction. Walnut (Juglans regia L.) is a medicinal plant containing a large number of biologically active substances. A number of authors propose to use walnut leaves as medicinal raw materials, in which juglone, flavonoids, tannins, ascorbic acid and other substances are identified. Currently, a standardization technique for this type of raw material has been developed and proposed. However, the medicinal plant walnut can be a source of other types of raw materials, such as milky-waxed walnuts, walnut bark, and walnut partitions. These types of medicinal raw materials contain a large number of biologically active substances and have a great potential for pharmacological activity. Therefore, the study with the subsequent standardization of medicinal plant material of walnut fruits in the stage of milk-wax ripeness and walnut bark is relevant.Aim. The aim of the study is to study the external signs, microscopy of raw walnut bark and walnut fruit in the stage of milky-wax ripeness and chemical composition of alcohol extract from these types of raw materials.Materials and methods. Chromato-mass spectrometry and TLC were used in the work.Results and discussion. When describing external signs and microscopy, diagnostic signs of both whole and crushed raw materials, and powder of walnut bark, and fruits of walnut in the stage of milky-wax ripeness were revealed. Alcohol extract obtained from medicinal plant raw materials walnut bark is a clear brown liquid. Alcohol extract obtained from medicinal plant raw materials of walnut fruit at the stage of milky-wax ripeness is a transparent, green-brown liquid with a fragrant odor.Conclusion. Gallic acid was identified by TLC in walnut bark. Chromato-mass spectrometry in alcohol extraction from the fruit of walnut in the stage of milky-wax maturity was able to identify 17 compounds belonging to different classes of biologically active substances. Sugars, flavonoids, coumarin phenolic compounds, organic acids were identified. In alcoholic extraction from the bark of a walnut, marker compounds have been determined, the combination of which makes it possible to quickly identify this type of raw material using chromatography-mass spectrometry.

The purpose of the research is to work out a routine method for the quantitative determination of the total amount of hydroxycinnamic acids in chicory herb growing in Ukraine. The presence of chlorogenic and caffeic acids was experimentally proven by comparing the TLC profiles of solutions of the chicory extract and standard substances of hydroxycinnamic acids. According to the chromatogram, the chemical composition of the ethanol extracts was qualitatively constant and did not depend on the series of plants collected in different places (7 regions of Ukraine) and in different years (2017 and 2018). The water-alcohol solutions of 8 chicory herb samples had the absorption maximum at the wavelength range from 326 to 330 nm, which was typical for phenolic acids. Chlorogenic acid was chosen as a marker substance for the quantitative determination by direct spectrophotometry. The value of the specific absorbance (556.21(λ=327 nm)) of chlorogenic acid in the concentration of 10.08 µg/ml in ethanol 50% was determined. The total amount of hydroxycinnamic acids was found to be 1.87% for the sample of 2017, for the samples of 2018 in the range of 4.00–6.61%. The method proposed is acceptable for standardization of the medicinal plant raw material.

The article is devoted to the development of new and perspective for the State Pharmacopoeia of the Russian Federation (SP RF) techniques for determining of the moisture (loss on drying) of medicinal plant raw materials of the morphological group “Fruit” and medicinal plant preparations by the use of infrared thermogravimetric (IRTGM) method. The techniques for determination of moisture (loss on drying) IRTGM-method as exemplified by the fruit of the following medicinal plants: Silybum marianum (L.) Gaertn., Rosa cinnamomea L. and Anethum graveolens L. were developed. The comparable results for determining of the moisture (loss on drying) of the medicinal plant raw materials of investigated plants were obtained by means of two methods: IRTGM and pharmacopoeial method, which allow us to recommend the developed techniques for inclusion into the relevant monographs of the State Pharmacopoeia of the Russian Federation.

The objects of the study were Mature dried fruits of Chinese birch (Lycium chinense Mill., this. Solanaceae (Solanaceae)). Four samples were examined: No. 1 (Gifts of Pamir, Russia), No. 2 (Gullin Tianhe Pharmaceutical, China), No. 3 (A. J. Alliance, Russia), No. 4 (Globaltorg, Russia). The purpose of this work is to determine the amount of carotenoids in the fruits of Chinese by spectrophotometry. The quantitative determination of the amount of carotenoids in the raw material of Chinese Dereza was carried out. Thin-layer chromatography was used to confirm the presence of carotenoids in the fruits of Dereza. The expediency of using the spectrophotometric method for the detection and quantitative determination of carotenoids in extracts from the fruits of Chinese birch is substantiated. The electron spectra of hexane, acetone and acetone-hexane solutions in the wavelength range 400–500 nm have a maximum optical density at 450±2 nm, characteristic of β-carotene. The optimal conditions of extraction of carotenoids from the raw materials of this plant (extractant – hexane; ratio "raw material–extractant" – 1 : 5; extraction time – 90 minutes; the degree of grinding of raw materials – 0.5 mm). It was determined that the error of a single determination of the content of carotenoids in the fruits of Chinese Dereza with a confidence probability of 95% is ±3,49%. It was revealed that the content of carotenoids in the raw material of Chinese Dereza varies in the range of 33–39 mg%.
