The article is concerned with the study of main biological properties of bacteria B. trematum species. In this work the research results of their tinctorial, cultural and biochemical properties are shown, with the view to grounding for setup scheme of isolation and bacteriological test of stated microorganism. According to obtained data bacteria of the given species represents gram-negative, coccoid bacillus that are able to grow both on usual and differentially diagnostic mediums. Bacteria B. trematum grow in temperature range of 17-420С, optimal temperature of cultivation is 37˚С. It should be noticed that the most relevant medium for cultivation is bordetellagar It was established that bacteria grow on the sodium chloride mediums in the range of 3-5%. It was found that studied culture of bacteria B. trematum shows проявляет asugarlytic properties, has positive reaction on catalase, negative reaction on cytochrome oxidase and DNAse. Also during the research it was established that bacteria B. trematum doesn’t utilize urea and citrate, has weak proteolytic activity and doesn’t utilize a number of amino acids. Similar results were obtained when using the set Api 20 E and NEFERMtest 24. Additionally it was established that B. trematum doesn’t reconstruct nitrates to nitrites, doesn’t enzyme β-galactosidase, adrinin hydrolase, triptophane dyaminaze, lysine decarboxylase, ornithine decarboxylase and urea doesn’t utilize citrates, doesn’t produce H2S, indole and acetone, doesn’t oxidate sucrose, melibios, amigdalin . Research results represented in the article provide the basis of isolation scheme and identification of bacteria of given species.

Abstract Background Herbaspirillum camelliae is a gram-negative endophyte isolated from the tea plant. Both strains WT00C and WT00F were found to hydrolyze epigallocatechin-3-gallate (EGCG) and epicatechin-3-gallate (ECG) to release gallic acid (GA) and display tannase activity. However, no tannase gene was annotated in the genome of H. camelliae WT00C. Results The 39 kDa protein, annotated as the prolyl oligopeptidase in the NCBI database, was finally identified as a novel tannase. Its gene was cloned, and the enzyme was expressed in E. coli and purified to homogeneity. Moreover, enzymatic characterizations of this novel tannase named TanHcw were studied. TanHcw was a secretary enzyme with a Sec/SPI signal peptide of 48 amino acids at the N-terminus, and it catalyzed the degradation of tannin, methyl gallate (MG), epigallocatechin-3-gallate (EGCG) and epicatechin-3-gallate (ECG). The optimal temperature and pH of TanHcw activities were 30 °C, pH 6.0 for MG and 40 °C, pH 7.0 for both EGCG and ECG. Na+, K+ Mn2+ and Triton-X100, Tween80 increased the enzyme activity of TanHcw, whereas Zn2+, Mg2+, Hg2+, EMSO, EDTA and β-mercaptoethanol inhibited enzyme activity. Km, kcat and kcat /Km of TanHcw were 0.30 mM, 37.84 s−1, 130.67 mM−1 s−1 for EGCG, 0.33 mM, 34.59 s−1, 105.01 mM−1 s−1 for ECG and 0.82 mM, 14.64 s−1, 18.17 mM−1 s−1 for MG, respectively. Conclusion A novel tannase TanHcw from H. camelliae has been identified and characterized. The biological properties of TanHcw suggest that it plays a crucial role in the specific colonization of H. camelliae in tea plants. Discovery of the tannase TanHcw in this study gives us a reasonable explanation for the host specificity of H. camelliae. In addition, studying the characteristics of this enzyme offers the possibility of further defining its potential in industrial application.

The soil fertility aspect is characterized by the good biological properties of the soil. One of the important elements of the soil biological properties is the bacterial population present in it. This research has been conducted from March to April 2016 on the slopes of Mount Merapi Turgo Sleman Yoyakarta. This study aimed to determine the various characteristics of colonies and cells of microbial organisms that are on the sample soil area of rhizosphere in Puspa trees in the slopes of Mount Merapi and to know the microbes that dominated the slopes of Mount Merapi. The method used in this research is observation. The sampling method used is a composite method, which combines 3 children of soil samples taken from one sample point on the same plot of land on a diagonal basis. The result of purification of culture obtained 14 pure bacteria isolate and 5 isolate fungi. From bacteria, 11 gram positive isolates and 3 gram negative isolates. Further research is needed on the identification of bacteria up to the species level.

Aim. To validate the suitability of commercial API 20E test-system (bioMerieux) for the identifi cation and characterization of facultative gram-negative phytopathogenic bacterial isolates. Methods. Conventional mi- crobiological methods, API 20E test-system (bioMerieux) according to the manufacturer’s instructions. Re- sults. The identifi cation results for Erwinia amylovora, Pectobacterium carotovorum and Pantoea agglome- rans isolates were derived from the conventional and API 20E test systems, which, were in line with the literature data for these species. The API 20E test-system showed high suitability for P. agglomerans isolates identifi cation. Although not all the species of facultatively anaerobic phytopathogenic bacteria may be identi- fi ed using API 20E test-system, its application will surely allow obtaining reliable data about their physiologi- cal and biochemical properties, valuable for identifi cation of bacteria, in the course of 24 h. Conclusions. The results of tests, obtained for investigated species while using API 20E test-system, and those of conventional microbiological methods coincided. The application of API 20E test-system (bioMerieux) ensures fast obtain- ing of important data, which may be used to identify phytopathogenic bacteria of Erwinia, Pectobacterium, Pantoea genera.

Activated sludges were acclimated to p-nitrophenol (PNP) in two operational modes, a batch and a continuous. The operational mode of the PNP acclimation of activated sludges strongly affected the physiological characteristics of predominant microorganisms responsible for PNP degradation. Predominant PNP degraders in the sludge in batch mode (Sludge B) had lower PNP affinity and were relatively insensitive to PNP concentration. Those of the sludge in continuous mode (Sludge C), on the other hand, had very high PNP affinity and were sensitive to PNP. MPN enumeration of PNP degraders in sludge B and C using media with different PNP concentrations (0.05, 0.2,0.5 and 2.0 mM) supported the above results. Medium with 0.2 mM of PNP did not recover PNP degraders in sludge C well, while it recovered PNP degraders in sludge B as well as the medium with 0.05 mM did. When switching from one operational mode to the other, the predominant population in sludge B shifted to the sensitive group, but that of sludge C did not shift at the given loading of PNP, showing relative resistance to inhibitive concentration.

The increasing chemicalization of production and life leads to the pollution of water bodies by chemicals, the effect of which on the micro - and macro - organisms is poorly understood. This section of the study in sanitary bacteriology is becoming ever more topical and is an important task of modern hygienic science. One of complicacies of the study of the problem is related with the fact that the presence of only experimental data fails to be sufficient, as the impact of any given chemical substance on different bacteria in the experiment does not mean that under natural conditions, similar results will be obtained. One reason for this may be the inhibitory effect of the given chemical on biological properties of bacteria, while in field conditions in the water several chemicals interacting with each other can exist. In this regard, the aim of the work was to assess the indicator value of sanitary and microbiological indices of epidemic hazard of water use in conditions of chemical pollution of surface water bodies.

The focus of the research presented in the paper is the method for evaluating and assessing tourism events and its applicability which is illustrated with the visitors’ assessment of the Festival of pop music in Vrnjačka Banja (the Festival). The primary data collected for the purpose of the Festival evaluation were gathered by survey method on a sample of 200 respondents, who were classified into two groups, locals and other visitors, and asked to assess the given criteria. The paper aims at using evaluation for improving the Festival development with the ultimate goal of improving the quality of the very event, which would influence an increase in the number of visitors to the Festival, i.e. tourist destination. The research results confirm the employed method to be useful for the evaluation of tourism events.

Gram-negative bacteria, especially Enterobacterales, have emerged as major players in antimicrobial resistance worldwide. Resistance may affect all major classes of anti-gram-negative agents, becoming multidrug resistant or even pan-drug resistant. Currently, β-lactamase-mediated resistance does not spare even the most powerful β-lactams (carbapenems), whose activity is challenged by carbapenemases. The dissemination of carbapenemases-encoding genes among Enterobacterales is a matter of concern, given the importance of carbapenems to treat nosocomial infections. Based on their amino acid sequences, carbapenemases are grouped into three major classes. Classes A and D use an active-site serine to catalyze hydrolysis, while class B (MBLs) require one or two zinc ions for their activity. The most important and clinically relevant carbapenemases are KPC, IMP/VIM/NDM, and OXA-48. However, several carbapenemases belonging to the different classes are less frequently detected. They correspond to class A (SME-, Nmc-A/IMI-, SFC-, GES-, BIC-like…), to class B (GIM, TMB, LMB…), class C (CMY-10 and ACT-28), and to class D (OXA-372). This review will address the genetic diversity, biochemical properties, and detection methods of minor acquired carbapenemases in Enterobacterales.

ABSTRACTLasers are instrumental in advanced bioimaging and Raman spectroscopy. However, they are also well known for their destructive effects on living organisms, leading to concerns about the adverse effects of laser technologies. To implement Raman spectroscopy for cell analysis and manipulation, such as Raman-activated cell sorting, it is crucial to identify nondestructive conditions for living cells. Here, we evaluated quantitatively the effect of 532-nm laser irradiation on bacterial cell fate and growth at the single-cell level. Using a purpose-built microfluidic platform, we were able to quantify the growth characteristics, i.e., specific growth rates and lag times of individual cells, as well as the survival rate of a population in conjunction with Raman spectroscopy. Representative Gram-negative and Gram-positive species show similar trends in response to a laser irradiation dose. Laser irradiation could compromise the physiological function of cells, and the degree of destruction is both dose and strain dependent, ranging from reduced cell growth to a complete loss of cell metabolic activity and finally to physical disintegration. Gram-positive bacterial cells are more susceptible than Gram-negative bacterial strains to irradiation-induced damage. By directly correlating Raman acquisition with single-cell growth characteristics, we provide evidence of nondestructive characteristics of Raman spectroscopy on individual bacterial cells. However, while strong Raman signals can be obtained without causing cell death, the variety of responses from different strains and from individual cells justifies careful evaluation of Raman acquisition conditions if cell viability is critical.IMPORTANCEIn Raman spectroscopy, the use of powerful monochromatic light in laser-based systems facilitates the detection of inherently weak signals. This allows environmentally and clinically relevant microorganisms to be measured at the single-cell level. The significance of being able to perform Raman measurement is that, unlike label-based fluorescence techniques, it provides a “fingerprint” that is specific to the identity and state of any (unlabeled) sample. Thus, it has emerged as a powerful method for studying living cells under physiological and environmental conditions. However, the laser's high power also has the potential to kill bacteria, which leads to concerns. The research presented here is a quantitative evaluation that provides a generic platform and methodology to evaluate the effects of laser irradiation on individual bacterial cells. Furthermore, it illustrates this by determining the conditions required to nondestructively measure the spectra of representative bacteria from several different groups.

The research work was conducted to isolate and identify the microflora from apparently healthy caged parrots. A total of 45 samples (oral swabs, cloacal swabs and feces) were collected from five types of caged parrots (Gray cockatiels, Rose ringed parakeet, Alexandriane parakeet, Red breast parakeet and Blossom headed parakeet) of Dhaka Zoo during the period from April to August 2009. The samples were cultured on different bacteriological media and the bacteria were identified by their cultural and biochemical properties. All the isolates were allowed for antibiogram study. The bacteria isolated in this study from different types of caged parrots were E. coli (64.44%), Salmonella spp. (46.67%), Staphylococcus spp. (46.67%), Pasteurella spp. (33.33%), Proteus spp. (6.67%) and some unidentified Gram-positive and Gram-negative bacteria. Of these isolates, E. coli was the most frequent isolate. The frequency of Gram-negative bacteria was higher in this study. The percentage of bacterial isolates recovered from each type of parrots was almost similar. Irrespective of types of parrots, the higher percentage of different bacteria was isolated from cloacal swab (77.78%) followed by feces (75.56%). The 68.89% isolates were recovered from oral swab. All the suspected isolates of Salmonella spp. were confirmed by slide agglutination test using Salmonella polyvalent ‘O’ antiserum. Among the 21 Salmonella spp. isolated in this study, 4 (19.05%) isolates were identified as S. Pullorum when tested with specific antisera against S. Pullorum. The results of antibiotic sensitivity tests revealed that ampicillin and amoxicillin were completely resistant to E. coli and Pasteurella spp.; ampicillin to Proteus spp.; and furazolidone to Salmonella spp. and Pasteurella spp. However, the antibiotics of fluoroquinolone group such as ciprofloxacin, norfloxacin and enrofloxacin showed moderate to high sensitivity against almost all the bacterial isolates.
